［０００１１
【産業上の利用分野］この発明はインシュリン様成長因
子Ｉ （以下、ＩＧＦ−Ｉと称する）の製剤に関するも
のである。 ［０００２］ 【従来の技術および発明が解決しようとする課題ＩＩＧ
Ｆ−Ｉは血中に存在するインシュリン類似のペプチドで
あり、種々の細胞の増殖促進作用を有しており、その産
生および分泌は成長ホルモンに依存している。 ［０００３］ ＩＧＦ−Ｉは成長ホルモンの骨成長促
進作用を仲介する物質であるソマトメジン群に含まれる
が、特にＩＧＦ−Ｉは成長ホルモン依存性が高く、ソマ
トメジンＣと同一の物質であり、成長ホルモンの作用の
発現に深く関与していることが知られている［０００４
］近年、このＩＧＦ−Ｉは遺伝子工学的手法により比較
的容易に生産することが可能になり、成長促進作用から
下垂体性小人症の治療剤、低身長者の成長促進剤として
、軟骨細胞増殖作用から骨粗鬆症の予防治療剤として、
あるいは骨折の治療剤として、インシュリン様作用から
糖尿病の治療剤として、さらにたんばく同化作用から潰
瘍、外傷、火傷の治療剤などとしての用途が知られてい
る。 ［０００５］Ｌかしながら、ＩＧＦ−Ｉは、高分子量の
ポリペプチドであり、その乾燥製剤は、再溶解時にゲル
が生じて溶解性不良となったり、経時により、その構成
アミノ酸の一部が酸化や脱アミド反応等を受けて別の物
質に変化する等の問題があった。 ［０００６］ 【発明の構成および効果】この様な現状において、本発
明者は、ＩＧＦ−Ｉの溶解性が良好で製剤安定性の優れ
た乾燥製剤を開発すべく種々研究した結果、Ｉ ＧＦ−
Ｉと塩酸、臭化水素酸、硝酸、メタンスルホン酸、硫酸
、リン酸およびシュウ酸より選ばれた強酸とを含む溶液
を乾燥することによって得られる乾燥製剤は溶解性が良
好であり製剤安定性が優れていることを見出し、さらに
鋭意研究の結果本発明の完成に至った。 ［０００７］この発明の乾燥製剤はＩＧＦ−Ｉと強酸と
を含有する溶液を乾燥することにより製造される。 ［０００８］この発明で使用されるＩＧＦ−Ｉとしては
、遺伝子組換え技術（例えば特開昭６１−１３９６号公
報参照）、ペプチド合成法、細胞培養法等により製造さ
れたヒト、ウシ等の哺乳動物由来のＩＧＦ−Ｉが含まれ
、さらに遺伝子組換え技術等により製造された、ＩＧＦ
−Ｉのアミノ酸配列の１部がアミノ酸の挿入、置換、付
加、欠失を起こしたＩＧＦ−Ｉ活性を有するムティンも
含まれる。 ［０００９］ ＩＧＦ−Ｉと強酸を含む溶液中のＩＧ
Ｆ−■と強酸との好ましい量比としては、塩酸、臭化水
素酸および硝酸にあっては、ＩＧＦ−１１ｍｇに対して
約０．５μｍｏｌから約１０μｍｏｌ、好ましくは約１
μｍｏｌから約５μｍｏｌであり、メタンスルホン酸お
よびリン酸にあっては、約１μｍｏｌ程度であり、また
硫酸およびシュウ酸にあっては、約０． ５μｍｏｌ程
度が好ましい。 ［００１０１乾燥法としては、凍結乾燥法、減圧乾燥法
等が用いられ、これらは常法により行なうことができる
。これらＩＧＦ−Ｉの乾燥製剤には、例えば水溶性ゼラ
チン、人血清アルブミン等の蛋白質、デキストラン等の
中性高分子、ソルビトール、マンニトール等の糖類、ト
リプトファン、リジン、グルタミン酸等のアミノ酸、食
塩等の塩類等の賦形剤の他、インシュリン、成長ホルモ
ン等の他の薬剤が適宜添加されることがある。 ［００１１１この発明のＩＧＦ−Ｉの乾燥製剤は溶解性
、製剤安定性はともに良好である。 ［００１２］

硫酸およびシュウ酸にあっては、約０． ５μｍｏｌ程

。これらＩＧＦ−Ｉの乾燥製剤には、例えば水溶性ゼラ

【実施例】以下、実施例により本発明を説明する。
［００１３］下記組成の溶液工ないし９を１０ｍ１容量
のバイアルに充填し均一に混合後凍結乾燥を行った。こ
れをゴム栓に施栓、キャップ巻締を行いそれぞれバイア

注入量 １０μｌ 波長２１４ｎｍ ［００１９］これらの結果により、本発明のＩ ＧＦ−
Ｉの製剤は、溶解性、安定性が共に優れていることが明
らかである。

Claims (2)
- 【請求項１】インシュリン様成長因子 I と塩酸、臭化
水素酸、硝酸、メタンスルホン酸、硫酸、リン酸および
シュウ酸より選ばれた強酸とを含有する溶液を乾燥した
乾燥製剤。 - 【請求項２】強酸が塩酸である請求項１の乾燥製剤。
