Description
広範囲の疾患を標的とするバイオ医薬品に対する増加した需要に伴い、現代の開発努力は、処理関連不純物を制御しながら、モノクローナル抗体(mAb)産物収量を最大化することに重く当てられる。より高い宿主細胞密度、力価、および生産性の予期せぬ副作用の1つは、採取された細胞培養液(HCCF)中の宿主細胞タンパク質(HCP)の増加した存在である。増加したHCPは、下流不純物クリアランス努力をさらに困難にするだけでなく、精製が始まる前であってもmAb安定性に影響を与えることもできる。増加したHCPの1つのかかる結果は、インタクトなmAbのチオレドキシン(Trx)誘導ジスルフィド結合還元および低分子重量種(LMW)の後続の形成である。ジスルフィド還元は、産物安定性、効力、および患者安全性への直接的なリスクであり;還元の酵素メカニズムによりよく確立されているが、HCCF中の増加したレダクターゼ活性および発現につながる細胞メカニズムは不明なままである。さらに、ジスルフィド還元におけるバッチ間変動性は、かかるメカニズムを研究することを困難にする。

宿主細胞中で発現可能な任意のポリペプチドまたはタンパク質は、本開示に従って目的のポリペプチドまたはタンパク質として生産されてもよい。該ポリペプチドは、宿主細胞に内因性である遺伝子から、または遺伝子工学によって宿主細胞中に導入される遺伝子から発現されてもよい。該ポリペプチドは、自然に生じるものであってもよく、またあるいは人間の手により操作または選択された配列を有してもよい。操作されたポリペプチドは、自然に個々に生じる他のポリペプチドセグメントから組み立てられてもよく、また天然でない1つ以上のセグメントを含んでもよい。
医薬品または他の市販の薬剤として現在使用されているかまたは調査下にある多数の抗体を考えると、本開示に従う抗体の生産は興味深い。抗体は、特定の抗原に特異的に結合する能力を有するタンパク質である。宿主細胞中で発現されることができる任意の抗体またはその抗原結合断片は、本開示に従って使用されてもよい。いくつかの実施形態において、発現されるべき抗体は、モノクローナル抗体またはその抗原結合断片である。特定の実施形態において、抗体は、ポリクローナル抗体またはその抗原結合断片である。

本開示の特定の実施形態において、実施者は、増殖中の細胞培養の特定の条件を定期的にモニターすることが有益または必要であることを見出してもよい。細胞培養条件をモニターすることは、実施者が、該細胞培養が組換えポリペプチドまたはタンパク質を準最適レベルで生産しているかどうか、または該培養が準最適生産フェーズに入ろうとしているかどうかを決定することを可能にする。特定の細胞培養条件をモニターするために、分析のために培養の少量のアリコートを除去することが必要であるだろう。当業者は、かかる除去が潜在的に細胞培養柱にコンタミネーションを導入してもよいことを理解し、かかるコンタミネーションのリスクを最小限にするために適切な注意をするであろう。
バッチおよび流加培養によるタンパク質またはポリペプチドの生産のために哺乳類細胞を調製する種々の方法は、当該技術分野において周知である。発現に到達するのに十分な核酸(典型的に、目的のポリペプチドまたはタンパク質をコードする遺伝子および任意の作動可能に連結された遺伝子制御要素を含有するベクター)は、任意の数の周知の技術により宿主細胞株中に導入されてもよい。典型的に、細胞はスクリーニングされて、どの宿主細胞が実際にベクターを取り込み、目的のポリペプチドまたはタンパク質を発現するかが決定される。哺乳類細胞により発現される特定の目的のポリペプチドまたはタンパク質を検出する従来の方法は、免疫組織化学、免疫沈降、フローサイトメトリー、免疫蛍光顕微鏡法、SDS-PAGE、ウェスタンブロット、酵素結合免疫吸着測定法(ELISA)、高性能液体クロマトグラフィー(HPLC)技術、生物活性アッセイおよびアフィニティークロマトグラフィーを含むが、これらに限定されない。当業者は、発現されたポリペプチドまたはタンパク質を検出するための他の適切な技術を知っているであろう。複数の宿主細胞が目的のポリペプチドまたはタンパク質を発現する場合、列挙された技術のいくつかまたは全てが使用されて、どの細胞がそのポリペプチドまたはタンパク質を最高レベルで発現するかが決定されることができる。
上記のように一度生産バイオリアクターが播種されると、細胞培養は、細胞培養の生存、増殖および生存性を助長する条件下で、初期増殖フェーズに維持される。正確な条件は、細胞型に応じて変わり、細胞が由来する生物、および発現されたポリペプチドまたはタンパク質の性質および特徴に応じて変わるだろう。
本開示の教示に従って、初期増殖フェーズの終わりに、培養条件の少なくとも1つはシフトされて、第2のセットの培養条件が適用され、培養における代謝シフトが生じてもよい。阻害性代謝物、最も特に乳酸およびアンモニアの蓄積は、増殖を阻害する。例えば細胞培養の温度、pH、浸透圧または化学的誘導物質レベルにおける変化により達成される代謝シフトは、特異的グルコース消費速度に対する特異的乳酸塩生産速度の比における減少を特徴としてもよい。1つの非限定的な実施形態において、培養条件は、培養の温度をシフトさせることによりシフトされる。しかしながら、当該技術分野において既知であるように、温度をシフトさせることは、適切な代謝シフトが到達されることができる唯一のメカニズムではない。例えば、かかる代謝シフトは、pH、浸透圧、および酪酸ナトリウムレベルを含むがこれらに限定されない他の培養条件をシフトさせることにより到達されることもできる。上記のように、培養シフトのタイミングは、ポリペプチドまたはタンパク質生産要求または細胞自体の必要に基づいて、本開示の実施者により決定されるであろう。
本開示に従って、細胞培養の条件が上記のように一度シフトされると、細胞培養は、細胞培養の生存および生存性を助長し、商業的に適切なレベルでの所望のポリペプチドまたはタンパク質の発現に適切である第2のセットの培養条件下で、後続の生産フェーズの間維持される。
一般に、典型的には、本開示に従って発現される目的のタンパク質またはポリペプチドを単離および/または精製することが望ましいであろう。好ましい実施形態において、発現されたポリペプチドまたはタンパク質は培地中へ分泌され、ゆえに、細胞および他の固形物は、例えば精製処理における第1の工程として、遠心分離または濾過により除去されてもよい。
独自の細胞培養培地および異なるIgG抗体を発現する2つの独自の組換えCHO細胞株を、以下の実施例に記載の実験のために使用した。細胞培養培地は、化学的に定義され、完全に無血清であった。組換えIgG生産CHO細胞を、250mL、1L、および3Lの振とうフラスコ中で懸濁培養中で維持し、36.5°C、150rpm、CO2濃度5%でのインキュベーションのためにCO2シェーカーインキュベーター(Kuhner)を使用した。
モノクローナル抗体(mAb A)についての還元リスクを評価するために、2つの別個の製造キャンペーンの間に生産されたバッチからのHCCFアリコートを、低溶存酸素条件下で一晩保持し、タンパク質Aにより精製し、そしてジスルフィド還元の存在についてキャピラリー電気泳動によりスクリーニングした(結果を表1に要約する)。試験された11ロットのうち、9バッチのタンパク質A溶出液中で還元mAb種を検出し、2バッチ(C1L2およびC2L2)のみが還元を示さなかった。
採取された細胞培養液(HCCF)中のジスルフィド結合還元電位に影響を及ぼしてもよい細胞培養条件をよりよく特徴付けるために、2つの培養を別個の点で採取して、モノクローナル抗体の生産に関連する平常(NC D14)および低生存性(LVC D15)条件を比較した。ジスルフィド結合還元を、キャピラリー電気泳動により複数の貯蔵条件下で評価した。簡潔には、実験室規模、パイロット規模、および/または臨床製造規模培養からのHCCFを室温まで温め、バイオセーフティキャビネット中で滅菌濾過し、ルアーロック滅菌エアフィルターを備えた滅菌単回使用培地ボトルアセンブリに分割した。ジスルフィド結合還元について試験されるべき試料に窒素を30-60分間散布して溶存酸素を除去し、その後ボトルの入口および出口を密閉した。試料を、エアオーバーレイ有りまたは無しで、2-8°Cまたは室温のいずれかで24時間貯蔵し、次いでタンパク質A精製の前に20mMヨードアセトアミド(Sigma)を用いて処理した。ジスルフィド結合還元の程度を、Caliper LabChip GXIIシステム(Perkin Elmer)を使用したキャピラリー電気泳動により測定し、LabChip GXソフトウェアを使用して分析した。細胞生存性条件を表2に示す。
実施例3に記載のプロテオミクスデータに基づいて、チオレドキシン(Trx)およびチオレドキシンレダクターゼ1(TrxR)の発現を、試料±ジスルフィド結合還元にわたってさらに分析した。TrxおよびTrxRは(主に一酸化窒素シグナル伝達の文脈内で)酸化ストレス反応に関与し、Trxシステムは、HCCFにおけるモノクローナル抗体減少に直接関係する。従って、2つのモノクローナル抗体からのベンチ、パイロット、および製造規模試料中のTrxおよびTrxRの発現を、ウェスタンブロット分析により分析した。簡潔には、HCCF材料を、BCAキット(Thermo Fisher)を使用して総タンパク質濃度について分析した。ライセートを、20μg総タンパク質に正規化し、10%またはAnyKD PROTEAN TGX Stain-Freeゲル(Bio-Rad)上で泳動し、ポリビニリデンジフルオリド膜(Bio-Rad)に転写し、5%ウシ血清アルブミン(Sigma)を用いてブロックし、TrxまたはTrxRに対する一次抗体(Cell Signaling Technology)の1:1,000希釈液を用いて、4°Cで一晩プローブした。次いで、ブロットを、西洋ワサビペルオキシダーゼ結合ヤギ抗ウサギまたはヤギ抗マウス二次抗体(Jackson Immunoresearch)の1:20,000希釈液を用いてプローブした。化学発光を、Chemidoc MPシステム(Bio-Rad)上でClarity Western ECL substrate(Bio Rad)を使用して視覚化した。バンド密度を、Image Labソフトウェア(Bio-Rad)を使用して定量し、IgG4軽鎖を各ブロットについてローディング対照として使用した。
NADPHがTrxRおよびTrx活性についての重要な補因子であることを考え、ペントースリン酸経路における最初のNADPH生成酵素であるグルコース-6-リン酸デヒドロゲナーゼ(G6PD)の発現を調べた。2つのモノクローナル抗体培養からの試料を、ウェスタンブロットにより分析し(図6A)、バンド強度を、実施例4に記載の手順(G6PDに対する一次抗体(Cell Signaling Technology)を使用)に従って定量した(図6B)。いくつかの試料は、非ジスルフィド還元試料と比較しておよそ130%の発現を示した一方、G6PD発現およびジスルフィド結合還元の間に明確な相関関係はなかった。
GAPDHは、重要な細胞ストレスセンサーであることが知られており、細胞レダクターゼシステムと相乗的に作用して、酸化的損傷を防止してもよい。核内において、GAPDHは細胞の代謝状態に反応して遺伝子転写を誘導することもできる。さらに重要なことに、GAPDHシステイン残基は、その一時的な不活性化を引き起こす過剰な過酸化物およびスーパーオキシドの存在に対して高い感受性を示す。この不活性化は解糖を停止し、G6Pをペントースリン酸経路中にシャトルして増加したNADPH生産および後続のレダクターゼ機能を可能にする主要なメカニズムである。酸化ストレスが低い場合、GAPDHはTrxにより十分に減少され、その活性状態に戻り、ゆえに解糖を再活性化し、レダクターゼ活性を低下させる。
生産細胞培養の間のGAPDH発現を、その時間的発現パターンを視覚化するために、いくつかの異なる培養条件下で試験した。mAb Bのための実験室規模バイオリアクターを、4つの異なる条件下で培養した:中心点DO2培養(対照)、バリアントN-1中心点DO2条件(Reducing1)、高細胞培養DO2条件(Reducing2)および低細胞培養DO2条件(Reducing3)。該対照およびReducing1バイオリアクター両方からの細胞を、GAPDH発現への変化をモニターするために、0、1、4、7、および9-14日にサンプリングした。全RNAを各バイオリアクターについての細胞ペレットから単離し、分光光度法で定量し、各反応について同じ開始テンプレート濃度に正規化し、そしてΔΔCt法を使用してq-RT-PCRにより分析した(図11A)。Reducing1条件の結果は、対照よりも1.3倍高くから始まる1日から相対的GAPDH mRNA発現において全体的により高い傾向を示し、9日に2.2倍より高い発現のピークまで徐々に増加し、その後10-14日から急激に低下した。これらの結果を検証し、追加の還元バイオリアクター条件を試験するために、対照、Reducing1、Reducing2、およびReducing3バイオリアクターの新しいセットからの細胞ペレットも、培養の7日および9日に分離し、q-RT-PCRによりGAPDH発現について分析した(図11B)。Reducing1条件について、GAPDH発現は、9日に該対照より2.3倍より高く増加した。Reducing2条件について、GAPDH発現は、7日に該対照より1.5倍より高く増加したが、9日に1.2倍のみより高く増加した一方、Reducing3条件は、9日に該対照より1.7倍より高かった。
Claims (16)
- 目的のタンパク質を含む組成物中のジスルフィド結合還元の発生を予測する方法であって、該方法は、グリセルアルデヒド-3-リン酸デヒドロゲナーゼ(GAPDH)である1つの宿主細胞タンパク質の発現レベルまたは活性レベルを測定することを含み、ここでベンチマーク値より上のGAPDH発現レベルまたはベンチマーク値より下の活性レベルが目的の該タンパク質のジスルフィド結合還元の発生に関連し、ここで目的の該タンパク質を含む組成物が、細胞培養から生産される、方法。
- 目的のタンパク質を生産する方法であって、該方法は、
a)目的の該タンパク質を生産することができる宿主細胞を培養すること;
b)グリセルアルデヒド-3-リン酸デヒドロゲナーゼ(GAPDH)である1つの宿主細胞タンパク質の発現レベルまたは活性レベルを測定すること;および
c)GAPDH発現レベルがベンチマーク値より下であるかまたは活性レベルがベンチマーク値より上であるかのいずれかの場合に、目的の該タンパク質を単離すること
を含み、ここで目的の該タンパク質が、抗体またはその抗原結合断片である、方法。 - 発現レベルが、細胞培養液中で、または細胞内で測定される、請求項1-2のいずれか一項に記載の方法。
- 発現レベルが、ウェスタンブロット分析またはqPCR分析により測定される、請求項1-3のいずれか一項に記載の方法。
- グリセルアルデヒド-3-リン酸デヒドロゲナーゼ(GAPDH)活性レベルが、GAPDH活性アッセイにより測定される、請求項1-2のいずれか一項に記載の方法。
- ジスルフィド結合還元の発生が、キャピラリー電気泳動により検出される、請求項1-5のいずれか一項に記載の方法。
- GAPDHの発現レベルは、測定され、かつベンチマーク値がウェスタンブロット分析により決定されるとき約140%のGAPDH相対バンド強度である、請求項1-4のいずれか一項に記載の方法。
- グリセルアルデヒド-3-リン酸デヒドロゲナーゼ(GAPDH)の活性レベルは、測定され、かつ目的のタンパク質中のジスルフィド結合還元の発生は、GAPDH活性アッセイにより決定されるときベンチマーク値と比較して正規化されたGAPDH活性における減少により示される、請求項1-2のいずれか一項に記載の方法。
- 目的のタンパク質が、抗体またはその抗原結合断片である、請求項1に記載の方法。
- 宿主細胞が、哺乳類細胞である、請求項1-9のいずれか一項に記載の方法。
