Description
本発明は、無機-有機複合医科材料の製造方法に関する。 The present invention relates to a method for producing an inorganic-organic composite medical material.
医療において、人体の欠損部位の形態および機能を人工物で補うこと(補綴)が知られている。例えば、歯科治療では、虫歯などにより欠損した歯冠を、歯科材料により修復する。そのような歯科材料として、要求される機械特性を確保すべく、無機材料と有機材料との複合材料が検討されている。 In medicine, it is known to supplement the form and function of a defective part of the human body with an artificial object (prosthesis). For example, in dental treatment, a tooth crown lost due to tooth decay or the like is repaired using dental materials. As such dental materials, composite materials of inorganic and organic materials are being considered in order to ensure the required mechanical properties.
しかるに、歯冠を修復する歯科材料は、咬合において、対合歯のエナメル質(天然ヒトエナメル質)と接触する。このとき、歯科材料の硬さがエナメル質よりも小さいと、歯科材料が摩耗(咬耗)し、歯科材料の硬さがエナメル質よりも大きいと、対合歯のエナメル質が摩耗(咬耗)する。そこで、エナメル質と同程度の硬さを有する複合材料が望まれている。 However, the dental material restoring the crown comes into contact with the enamel of the opposing tooth (natural human enamel) in occlusion. At this time, if the hardness of the dental material is lower than the enamel, the dental material will wear away (occlusal wear), and if the hardness of the dental material is higher than the enamel, the enamel of the opposing tooth will wear away (occlusal wear). )do. Therefore, a composite material having a hardness comparable to that of enamel is desired.
そのような複合材料として、例えば、連通孔を有する無機物と、連通孔内に充填される有機高分子との2相共連続構造を有する、無機-有機複合医科材料が提案されている(例えば、特許文献1参照)。 As such a composite material, an inorganic-organic composite medical material has been proposed, for example, which has a two-phase co-continuous structure of an inorganic material having communicating pores and an organic polymer filled in the communicating pores (for example, (See Patent Document 1).
そのような無機-有機複合医科材料は、無機ナノ粒子および水溶性高分子を分散媒に配合して、無機ナノ粒子を分散媒に分散させて無機粒子分散液を調製した後、無機粒子分散液を乾燥させて、ゲル状の多孔質前駆体を調製し、次いで、多孔質前駆体を焼成して、連通孔を有する無機焼成体を形成し、その後、無機焼成体の連通孔に有機高分子を充填することにより製造される。 Such inorganic-organic composite medical materials are prepared by mixing inorganic nanoparticles and a water-soluble polymer in a dispersion medium, dispersing the inorganic nanoparticles in the dispersion medium to prepare an inorganic particle dispersion, and then preparing an inorganic particle dispersion. is dried to prepare a gel-like porous precursor, then the porous precursor is fired to form an inorganic fired body having communicating pores, and then an organic polymer is injected into the communicating pores of the inorganic fired body. It is manufactured by filling.
しかし、特許文献1に記載の無機-有機複合医科材料の製造方法では、無機-有機複合医科材料の製造効率の向上を図るには限度があり、かつ、無機-有機複合医科材料を所望の形状に形成することが困難である。
However, in the method for producing an inorganic-organic composite medical material described in
そこで、本発明は、無機物を所望の形状に形成できながら、無機-有機複合医科材料の製造効率の向上を図ることができる無機-有機複合医科材料の製造方法を提供する。 Therefore, the present invention provides a method for manufacturing an inorganic-organic composite medical material that can improve the manufacturing efficiency of the inorganic-organic composite medical material while forming an inorganic substance into a desired shape.
The present invention [1] is a method for producing an inorganic-organic composite medical material having a two-phase bicontinuous structure of an inorganic material having communicating pores and an organic polymer filled in the communicating pores, the method comprising: A step of preparing a liquid material containing a polymerizable monomer, a polyoxyalkylene group-containing photopolymerizable monomer, and an alcohol; a step of dispersing inorganic nanoparticles in the liquid material; and a step of dispersing the hydroxyl group-containing photopolymerizable monomer. and the polyoxyalkylene group-containing photopolymerizable monomer are photopolymerized, and the inorganic nanoparticles are dispersed in a polymer matrix of the hydroxyl group-containing photopolymerizable monomer and the polyoxyalkylene group-containing photopolymerizable monomer. a step of preparing a dispersion; a step of removing the alcohol from the particle dispersion; a step of firing the particle dispersion from which the alcohol has been removed to form an inorganic substance having communicating pores; The present invention includes a method for producing an inorganic-organic composite medical material, including a step of filling the communicating pores of an inorganic substance with an organic polymer.
このような方法によれば、水酸基含有光重合性モノマーおよびポリオキシアルキレン基含有光重合性モノマー(以下、モノマー成分とする。)と、アルコール類とを含有する液材に、無機ナノ粒子を分散させる。そして、モノマー成分を光重合して、ポリマーマトリックスに無機ナノ粒子が分散する粒子分散体を調製する。そのため、モノマー成分の光重合により、粒子分散体を所望の形状に形成することができる。 According to such a method, inorganic nanoparticles are dispersed in a liquid material containing a hydroxyl group-containing photopolymerizable monomer, a polyoxyalkylene group-containing photopolymerizable monomer (hereinafter referred to as monomer component), and an alcohol. let Then, the monomer components are photopolymerized to prepare a particle dispersion in which inorganic nanoparticles are dispersed in a polymer matrix. Therefore, the particle dispersion can be formed into a desired shape by photopolymerization of the monomer components.
その後、所望の形状を有する粒子分散体からアルコール類を除去し、その粒子分散体を焼成して、連通孔を有する無機物を形成する。そのため、無機物は、粒子分散体と同じ所望の形状を有する。そして、その無機物の連通孔に有機高分子を充填して、無機-有機複合医科材料を製造する。 Thereafter, the alcohol is removed from the particle dispersion having a desired shape, and the particle dispersion is fired to form an inorganic substance having communicating pores. Therefore, the inorganic material has the same desired shape as the particle dispersion. Then, the communicating pores of the inorganic material are filled with an organic polymer to produce an inorganic-organic composite medical material.
その結果、無機物を所望の形状に形成できながら、無機-有機複合医科材料の製造効率の向上を図ることができる。 As a result, it is possible to form an inorganic substance into a desired shape and to improve the production efficiency of an inorganic-organic composite medical material.
本発明[2]は、前記液材に無機ナノ粒子を分散させる工程において、前記無機ナノ粒子の分散割合は、前記液材および前記無機ナノ粒子の総和に対して、20質量%以上である、上記[1]に記載の無機-有機複合医科材料の製造方法を含んでいる。 The present invention [2] provides that, in the step of dispersing inorganic nanoparticles in the liquid material, the dispersion ratio of the inorganic nanoparticles is 20% by mass or more with respect to the total sum of the liquid material and the inorganic nanoparticles. It includes the method for producing the inorganic-organic composite medical material described in [1] above.
このような方法によれば、無機ナノ粒子の分散割合が上記下限以上であるので、無機物の形成不良を安定して抑制できる。 According to such a method, since the dispersion ratio of inorganic nanoparticles is equal to or higher than the above lower limit, formation defects of inorganic substances can be stably suppressed.
本発明[3]は、前記液材に無機ナノ粒子を分散させる工程において、前記無機ナノ粒子の分散割合は、前記液材および前記無機ナノ粒子の総和に対して、60質量%以下である、上記[1]または[2]に記載の無機-有機複合医科材料の製造方法を含んでいる。 In the present invention [3], in the step of dispersing inorganic nanoparticles in the liquid material, the dispersion ratio of the inorganic nanoparticles is 60% by mass or less with respect to the sum total of the liquid material and the inorganic nanoparticles. It includes the method for producing the inorganic-organic composite medical material described in [1] or [2] above.
このような方法によれば、無機ナノ粒子の分散割合が上記上限以下であるので、粒子分散体を安定して所望の形状に形成できながら、無機物の形成不良をより安定して抑制できる。 According to such a method, since the dispersion ratio of the inorganic nanoparticles is below the above upper limit, the particle dispersion can be stably formed into a desired shape, and defective formation of the inorganic substance can be more stably suppressed.

表1に示す処方となるように、2-ヒドロキシエチルメタクリレート(HEMA、水酸基含有光重合性モノマー)と、トリエチレングリコールジメタクリレート(商品名3G、新中村化学工業社製、ポリオキシアルキレン基含有光重合性モノマー)と、フェノキシエタノール(POE、アルコール類)と、1-プロパノール(PrOH、アルコール類)とを混合して、液材を調製した。
(Examples 1-3, 5-8 and 10)
2-Hydroxyethyl methacrylate (HEMA, a hydroxyl group-containing photopolymerizable monomer) and triethylene glycol dimethacrylate (trade name 3G, manufactured by Shin-Nakamura Chemical Co., Ltd., a polyoxyalkylene group-containing photopolymerizable monomer) were prepared as shown in Table 1. A liquid material was prepared by mixing a polymerizable monomer), phenoxyethanol (POE, an alcohol), and 1-propanol (PrOH, an alcohol).
Next, while irradiating the liquid material with ultrasonic waves, silica particles (trade name OX50, manufactured by Nippon Aerosil Co., Ltd., average primary particle diameter 40 nm, spherical shape) were added in multiple portions so as to have the formulation shown in Table 1. and mixed. Then, the ultrasonic irradiation was continued until there were no lumps of silica particles to disperse the silica particles in the liquid material, and then the ultrasonic irradiation was stopped. After that, phenylbis(2,4,6-trimethylbenzoyl)phosphine oxide (BAPO, photopolymerization initiator) was added to the liquid material in which silica particles were dispersed (inorganic particle dispersion) as shown in Table 1. Add and dissolve.
次いで、BAPOを含む無機粒子分散液を、円柱状のモールド(直径20mm、高さ20mm)に注入した後、光照射装置(商品名α―ライトV,モリタ製作所社製)により、15分間光照射して、HEMAおよび3Gを光重合および硬化させた。
Next, the inorganic particle dispersion containing BAPO was injected into a cylindrical mold (
これにより、HEMAおよび3Gのポリマーマトリックスに、シリカ粒子が分散する粒子分散体を調製した。 In this way, a particle dispersion in which silica particles were dispersed in a polymer matrix of HEMA and 3G was prepared.
なお、粒子分散体の成形性を下記の基準で評価した。その結果を表1に示す。
〇:円柱形モールドに無機粒子分散液を容易に充填できる。
△:円柱形モールドへの無機粒子分散液の充填が困難であるが、手指などによって押し込むことで充填できる。
×:円柱形モールドへの充填が手指などによる押し込みでも困難である。
The moldability of the particle dispersion was evaluated based on the following criteria. The results are shown in Table 1.
○: The inorganic particle dispersion liquid can be easily filled into a cylindrical mold.
Δ: It is difficult to fill the cylindrical mold with the inorganic particle dispersion, but it can be filled by pushing the mold with fingers or the like.
×: It is difficult to fill the cylindrical mold even when pressed with fingers or the like.
次いで、粒子分散体を室温(25℃)にて24時間静置して乾燥した後、オーブンにより80℃まで昇温して24時間乾燥させて、粒子分散体から、POEおよびPrOHを除去した。これにより、粒子分散体の全体が白濁した。 Next, the particle dispersion was allowed to stand at room temperature (25° C.) for 24 hours to dry, and then heated to 80° C. in an oven and dried for 24 hours to remove POE and PrOH from the particle dispersion. As a result, the entire particle dispersion became cloudy.
次いで、乾燥後の粒子分散体を焼成した。具体的には、粒子分散体を焼成炉内に配置して、焼成炉内を100℃/hの昇温速度で室温(25℃)から220℃まで昇温した後、4時間維持した。その後、さらに600℃まで昇温して3時間維持し、次いで、さらに1150℃まで昇温して3時間維持した。 Next, the dried particle dispersion was fired. Specifically, the particle dispersion was placed in a firing furnace, and the temperature inside the firing furnace was raised from room temperature (25°C) to 220°C at a temperature increase rate of 100°C/h, and then maintained for 4 hours. Thereafter, the temperature was further increased to 600°C and maintained for 3 hours, and then further increased to 1150°C and maintained for 3 hours.
これにより、ポリマーマトリックスを燃焼させるとともに、複数のシリカ粒子を互いに接合させて、連通孔を有するシリカガラスの(多孔質シリカ)を得た。その後、焼成炉内を室温(25℃)まで降温して、多孔質シリカを焼成炉から取り出した。 As a result, the polymer matrix was burned, and the plurality of silica particles were bonded to each other to obtain silica glass (porous silica) having communicating pores. Thereafter, the temperature inside the firing furnace was lowered to room temperature (25° C.), and the porous silica was taken out from the firing furnace.
Note that the state of porous silica was evaluated based on the following criteria. The results are shown in Table 1.
Good: No cracks (defective formation) were generated in the porous silica, and the porous silica was able to maintain the same desired shape as the particle dispersion.
Δ: Cracks (defective formation) occur in some parts of the porous silica, but the porous silica maintains the same shape as the particle dispersion.
×: Many cracks (defective formation) occur in the porous silica, and the porous silica cannot maintain the same shape as the particle dispersion.
次いで、多孔質シリカを5質量%のγ―メタクリロキシプロピルトリメトキシシラン(γ-MPTS)のエタノール溶液に、室温(25℃)で3時間浸漬した。その後、多孔質シリカを、γ-MPTSのエタノール溶液から取り出し、80℃にて3時間乾燥させた。これによって、多孔質シリカの表面をシランカップリング処理した。 Next, the porous silica was immersed in an ethanol solution of 5% by mass of γ-methacryloxypropyltrimethoxysilane (γ-MPTS) at room temperature (25° C.) for 3 hours. Thereafter, the porous silica was taken out from the γ-MPTS ethanol solution and dried at 80° C. for 3 hours. As a result, the surface of the porous silica was subjected to silane coupling treatment.
次いで、メチルメタクリレート(MMA、原料単量体)10gを準備し、そのMMAに、過酸化ベンゾイル(BPO、重合開始剤)0.05gを溶解した。 Next, 10 g of methyl methacrylate (MMA, raw material monomer) was prepared, and 0.05 g of benzoyl peroxide (BPO, polymerization initiator) was dissolved in the MMA.
次いで、多孔質シリカを、BPOが溶解されたMMAに、大気圧下において、室温(25℃)で12時間浸漬させた。これによって、MMAが、多孔質シリカの連通孔内に導入された。 Next, the porous silica was immersed in MMA in which BPO was dissolved under atmospheric pressure at room temperature (25° C.) for 12 hours. As a result, MMA was introduced into the communicating pores of the porous silica.
その後、MMAが導入された多孔質シリカを、60℃に加熱して、48時間保持した。これによって、MMAが重合して、ポリメチルメタクリレート(PMMA、有機高分子)が形成した。 Thereafter, the porous silica into which MMA had been introduced was heated to 60° C. and held for 48 hours. As a result, MMA was polymerized to form polymethyl methacrylate (PMMA, an organic polymer).
以上によって、多孔質シリカ(連通孔を有するシリカガラスの焼結体)と、連通孔内に充填されるPMMAとの2相共連続構造を有するナノコンポジットレジンを製造した。 As described above, a nanocomposite resin having a two-phase co-continuous structure of porous silica (a sintered body of silica glass having communicating pores) and PMMA filled in the communicating pores was manufactured.
