Description

b)可溶性CD40LおよびTLR7/8アゴニストで樹状細胞を刺激するステップと
を含む方法に関する。
・TLR7/8アゴニスト、
・可溶性CD40L、
・IL-10に対して特異的な一次結合性タンパク質、
・IL-12に対して特異的な一次結合性タンパク質、
・IL-10に対して特異的な二次結合性タンパク質、および
・IL-12に対して特異的な二次結合性タンパク質
を含むキットに関する。
(i)TLR7/8アゴニストおよびCD40Lを含む組成物、
(ii)IL-10に対して特異的な一次結合性タンパク質およびIL-12に対して特異的な一次結合性タンパク質を含む組成物、ならびに
(iii)IL-10に対して特異的な二次結合性タンパク質およびIL-12に対して特異的な二次結合性タンパク質を含む組成物
を含み得る。
MDGカクテルまたはJonuleitカクテルを用いて成熟化され、CD40L単独で、ならびにR848/IFNγおよびLPSと組み合わせて刺激された樹状細胞
単球を、付着性単離によって単離した後、IL-4およびGMSCFを使用して3日間かけて樹状細胞を発生させた。その後、樹状細胞を、MDGカクテルとIL-4およびGMCSFとを用いて、24時間かけて成熟させた。Jonuleitカクテルの場合は、細胞を、IL-4およびGMSCFを使用して、6日間インキュベートした。次いで、細胞を、Jonuleitカクテル(ならびにIL-4およびGMCSF)を使用して、24時間インキュベートした。全てのアッセイに対して、凍結した樹状細胞を、アッセイの前日に解凍して、アッセイを開始するまでDC培地においてインキュベートして使用した。刺激は、96ウェルプレートにおいて24時間行われる。したがって、ウェルあたり11,000の樹状細胞を播種した。110μlのDC培地(1.5%ヒト血清を伴うVLERPMI1640)中における1.1μlのCD40L、1μg/mlのLPS、およびR848を加えた。
IL-10/IL-12Elispotアッセイを、(C.T.L. Cellular technology Limited、クリーブランド、USA)のヒトIL-10/IL-12Double-Color ELISPOTキットのプロトコルに従って実施した。
10ng/mlのTNFα、10ng/mlのIL-1γ、15ng/mlのIL-6、および1,000ng/mlのプロスタグランジンE2(=PGE2)
10ng/mlのTNF-α、10ng/mlのIL-1β、5,000U/mlのIFNγ、1μg/mlのR848、および250ng/mlプロスタグランジンE2
なお、本発明は、以下の態様をも含むものである。
<1> 樹状細胞の効力を決定する方法であって、
(a)可溶性CD40LおよびTLR7/8アゴニストとのインキュベーションによって樹状細胞を刺激するステップと、
(b)(a)の前記樹状細胞からのマーカータンパク質IL-10およびIL-12の分泌を測定するステップと
を含む方法。
<2> 前記測定が、単一細胞レベルにおいて行われる、上記1に記載の方法。
<3> 前記TLR7/8アゴニストが、4-アミノ-2-エトキシメチル-α,α-ジメチル-1H-イミダゾール[4,5-c]キノリン-1-エタノール(R848)である、上記1または2に記載の方法。
<4> さらに、
(c)IL-12およびIL-10の分泌プロファイルに基づく前記樹状細胞の効力の分類のステップを含む、上記1から3のいずれかに記載の方法。
<5> 1を超える、IL-10分泌に対するIL-12分泌の比率を示す樹状細胞が、T細胞およびナチュラルキラー(NK)細胞を活性化する高い能力を有する樹状細胞として分類される、上記1から4のいずれかに記載の方法。
<6> T細胞およびNK細胞を活性化する高い能力を有する前記樹状細胞が、高いCD80発現レベル、高いCD86発現レベル、低いCD14発現レベル、および低いB7H1発現レベルの表現型を有する、上記5に記載の方法。
<7> T細胞を活性化する高い能力を有する前記樹状細胞が、T細胞を、Th1/Tc1表現型へと極性化する、上記5または6に記載の方法。
<8> 前記Th1/Tc1表現型が、前記T細胞によるIFNγの分泌と、全くないかまたは低いIL-4の発現とによって特徴付けられる、上記7に記載の方法。
<9> NK細胞を活性化する高い能力を有する前記樹状細胞が、NK細胞を活性化して、高レベルのCD69を発現させ、IFNγを分泌させる、上記5から8に記載の方法。
<10> ステップ(b)が、
i)IL-10に対する一次結合性タンパク質およびIL-12に対して特異的な一次結合性タンパク質と共に前記樹状細胞をインキュベートするステップと、
ii)前記一次結合性タンパク質への前記マーカータンパク質の結合を、二次結合性タンパク質によって検出するステップと
を含む、上記1から9のいずれかに記載の方法。
<11> 前記刺激が、前記樹状細胞への可溶性CD40LおよびTLR7/8アゴニストの結合によってのみ生じる、上記1から10のいずれかに記載の方法。
<12> CD40Lが、細胞株によって前記樹状細胞に提示されない、上記1から11のいずれかに記載の方法。
<13> 前記樹状細胞が、異なる細胞株と共培養されない、上記1から12のいずれかに記載の方法。
<14> 放射線照射ステップが適用されない、上記1から13のいずれかに記載の方法。
<15> IL-10およびIL-12に対して特異的な前記一次結合性タンパク質が、担体上に固定化される、上記10から14に記載の方法。
<16> 前記担体が、IL-10およびIL-12に対する前記一次結合性タンパク質で一様にコーティングされる、上記15に記載の方法。
<17> 前記担体が、マルチウェルプレートである、上記15または16に記載の方法。
<18> 前記一次結合性タンパク質が抗体である、上記10から17のいずれかに記載の方法。
<19> 前記二次結合性タンパク質が抗体である、上記10から18のいずれかに記載の方法。
<20> 前記二次結合性タンパク質が蛍光標識される、上記10から19に記載の方法。
<21> 前記樹状細胞が、成熟させた樹状細胞である、上記1から20のいずれかに記載の方法。
<22> 前記樹状細胞の前記成熟化が、成熟化カクテルとのインキュベーションによって行われる、上記21に記載の方法。
<23> 樹状細胞を刺激する方法であって、
a)樹状細胞を提供するステップと、
b)可溶性CD40LおよびTLR7/8アゴニストで前記樹状細胞を刺激するステップとを含む方法。
<24> 樹状細胞を刺激するための可溶性CD40LおよびTLR7/8アゴニストの使用。
<25> TLR7/8アゴニスト、
可溶性CD40L、
IL-10に対して特異的な一次結合性タンパク質、
IL-12に対して特異的な一次結合性タンパク質、
IL-10に対して特異的な二次結合性タンパク質、および
IL-12に対して特異的な二次結合性タンパク質
を含むキット。
<26> (i)TLR7/8アゴニストおよびCD40Lを含む組成物、
(ii)IL-10に対して特異的な一次結合性タンパク質およびIL-12に対して特異的な一次結合性タンパク質を含む組成物、ならびに
(iii)IL-10に対して特異的な二次結合性タンパク質およびIL-12に対して特異的な二次結合性タンパク質を含む組成物
を含む、上記25に記載のキット。
<27> 前記一次結合性タンパク質が抗体であり、および/または前記二次結合性タンパク質が抗体である、上記25または26に記載のキット。
<28> 前記TLR7/8アゴニストがR848である、上記23に記載の方法、上記24に記載の使用、上記25から27に記載のキット。
Claims (16)
- 樹状細胞の効力を決定する方法であって、
(a)可溶性CD40LおよびTLR7/8アゴニストとのインキュベーションによってインビトロで樹状細胞を刺激するステップと、
(b)(a)の前記樹状細胞からのマーカータンパク質IL-10およびIL-12の分泌を測定するステップと、ここで、該測定は、単一細胞レベルにおいて行われる、
(c)IL-12およびIL-10の分泌プロファイルに基づく前記樹状細胞の効力の分類のステップと、ここで、1を超える、IL-10分泌に対するIL-12分泌の比率を示す樹状細胞が、T細胞およびナチュラルキラー(NK)細胞を活性化する高い能力を有する樹状細胞として分類される、
を含み、
T細胞を活性化する高い能力を有する前記樹状細胞が、T細胞を、Th1/Tc1表現型へと極性化する、前記方法。 - 前記TLR7/8アゴニストが、4-アミノ-2-エトキシメチル-α,α-ジメチル-1H-イミダゾール[4,5-c]キノリン-1-エタノール(R848)である、請求項1に記載の方法。
- T細胞およびNK細胞を活性化する高い能力を有する前記樹状細胞が、高いCD80発現レベル、高いCD86発現レベル、低いCD14発現レベル、および低いB7H1発現レベルの表現型を有する、請求項1または2に記載の方法。
- 前記Th1/Tc1表現型が、前記T細胞によるIFNγの分泌と、全くないかまたは低いIL-4の発現とによって特徴付けられる、請求項1から3のいずれか一項に記載の方法。
- NK細胞を活性化する高い能力を有する前記樹状細胞が、NK細胞を活性化して、高レベルのCD69を発現させ、IFNγを分泌させる、請求項1から4のいずれか一項に記載の方法。
- 樹状細胞の効力を決定する方法であって、
(a)可溶性CD40LおよびTLR7/8アゴニストとのインキュベーションによってインビトロで樹状細胞を刺激するステップと、
(b)(a)の前記樹状細胞からのマーカータンパク質IL-10およびIL-12の分泌を測定するステップと、ここで、該測定は、単一細胞レベルにおいて行われる、
(c)IL-12およびIL-10の分泌プロファイルに基づく前記樹状細胞の効力の分類のステップと、ここで、1を超える、IL-10分泌に対するIL-12分泌の比率を示す樹状細胞が、T細胞およびナチュラルキラー(NK)細胞を活性化する高い能力を有する樹状細胞として分類される、
を含み、
NK細胞を活性化する高い能力を有する前記樹状細胞が、NK細胞を活性化して、高レベルのCD69を発現させ、IFNγを分泌させる、前記方法。 - ステップ(b)が、
i)IL-10に対する一次結合性タンパク質およびIL-12に対して特異的な一次結合性タンパク質と共に前記樹状細胞をインキュベートするステップと、
ii)前記一次結合性タンパク質への前記マーカータンパク質の結合を、二次結合性タンパク質によって検出するステップと
を含む、請求項1から6のいずれか一項に記載の方法。 - 前記刺激が、前記樹状細胞への可溶性CD40LおよびTLR7/8アゴニストの結合によってのみ生じる、請求項1から7のいずれか一項に記載の方法。
- CD40Lが、細胞株によって前記樹状細胞に提示されない、請求項1から8のいずれか一項に記載の方法。
- 前記樹状細胞が、異なる細胞株と共培養されない、請求項1から9のいずれか一項に記載の方法。
- 放射線照射ステップが適用されない、請求項1から10のいずれか一項に記載の方法。
- 前記一次結合性タンパク質が抗体である、請求項7に記載の方法。
- 前記二次結合性タンパク質が抗体である、請求項7または12に記載の方法。
- 前記二次結合性タンパク質が蛍光標識される、請求項13に記載の方法。
- 前記樹状細胞が、成熟させた樹状細胞である、請求項1から14のいずれか一項に記載の方法。
- 前記成熟させた樹状細胞が、成熟化カクテルとのインキュベーションによって得られる、請求項15に記載の方法。
