Description
T細胞レセプター(TCR)は、CD4+及びCD8+ T細胞により天然に発現される。TCRは、主要組織適合性複合体(MHC)分子(ヒトでは、MHC分子はヒト白血球抗原又はHLAとしても知られる)との複合体で、抗原提示細胞の表面にディスプレイされる短いペプチド抗原を認識するように設計されている(Davisら(1998), Annu Rev Immunol 16:523-544)。CD8+ T細胞は、細胞傷害性T細胞とも呼ばれ、MHCクラスIに結合するペプチドを特異的に認識し、一般には、感染細胞又はガン細胞の発見及びその破壊の仲介を担っている。
NY-ESO-1を標的するTCRは、以前に報告されている(WO05113595;WO08039818;McCormackら(2013), Cancer Immunol Immunother 62(4):773-785)。
本発明者らは、下記の鎖を含んでなる天然型TCRを同定した:
α鎖:TRAV3*01/TRAJ28*01
β鎖:TRBV29-1*01/TRBD2*01/TRBJ2-3*01
(注記:用語「*01」は、IMGT命名法により指定されているように、この配列についての対立遺伝子バリアントを示す。)
この天然型TCRは、本発明の変異配列を誘導する鋳型として用いられた。
本発明は、第1の観点として、SLLMWITQC(配列番号1)-HLA-A*02複合体に対する結合性を有し、TCR α鎖可変ドメイン及び/又はTCR β鎖可変ドメインを含んでなり、
α鎖可変ドメインが配列番号2のアミノ酸1〜117と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含み、及び/又は
β鎖可変ドメインが配列番号3のアミノ酸1〜115と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含む、

TCR α鎖可変ドメインは配列番号12と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含んでもよい。
TCR β鎖可変ドメインは配列番号15と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含んでもよい。
α鎖可変ドメインは配列番号6〜12又は51のアミノ酸配列から選択されてもよく、β鎖可変ドメインは配列番号13〜23又は52のアミノ酸配列から選択されてもよい。
本発明のTCRは、α鎖TRAC定常ドメイン配列及びβ鎖TRBC1又はTRBC2定常ドメイン配列を有するα-βヘテロダイマーであってもよい。
本発明のTCRは、Vα-L-Vβ、Vβ-L-Vα、Vα-Cα-L-Vβ、Vα-L-Vβ-Cβ型(式中、Vα及びVβはそれぞれTCR α及びβ可変領域であり、Cα及びCβはそれぞれTCR α及びβ定常領域であり、Lはリンカー配列である)の単鎖形式であってもよい。
本発明また、本発明のTCRと、該TCRのα鎖又はβ鎖のC末端又はN末端に共有結合した抗CD3抗体とを含んでなるTCR-抗CD3融合体を提供し、このTCR-抗CD3融合体は、配列番号6〜12又は51から選択されるα鎖可変ドメイン及び配列番号13〜23又は52から選択されるβ鎖可変ドメインを含んでなっていてもよい。β鎖は抗CD3抗体配列にリンカー配列を介して連結されていてもよく;リンカー配列は、GGGGS(配列番号24)、GGGSG(配列番号25)、GGSGG(配列番号26)、GSGGG(配列番号27)、GSGGGP(配列番号28)、GGEPS(配列番号29)、GGEGGGP(配列番号30)及びGGEGGGSEGGGS(配列番号31)からなる群より選択されてもよい。
本発明はまた、本発明のTCR又はTCR-抗CD3融合体をコードする核酸を提供する。
TCR若しくはTCR-抗CD3融合体を提示する細胞又は発現ベクターを有する細胞は、T細胞であってもよい。
医薬に用いるための、本発明のTCR、TCR-抗CD3融合体、核酸又は細胞もまた提供される。TCR、TCR-抗CD3融合体、細胞又は核酸は、ヒト対象におけるガン治療法に用いられるものであってもよい。
ヒト対象はNY-ESO-1及び/又はLAGE-1Aを発現する腫瘍を有していてもよく、腫瘍は固形/充実腫瘍であってもよく、滑膜肉腫、非小細胞肺ガン(NSCLC)、膀胱の腫瘍、胃の腫瘍、前立腺の腫瘍、直腸結腸の腫瘍、***の腫瘍、卵巣の腫瘍、食道の腫瘍、メラノーマ、多発性骨髄腫、肝細胞ガン及び頭部頸部の腫瘍から選択されてもよい。ヒト対象は、HLA-A*02サブタイプの対象であってもよい。
第1の観点において、本発明は、SLLMWITQC(配列番号1)-HLA-A*02複合体に対する結合性を有し、TCR α鎖可変ドメイン及び/又はTCR β鎖可変ドメインを含んでなり、α鎖可変ドメインが配列番号2のアミノ酸1〜117と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含み、及び/又は、β鎖可変ドメインが配列番号3のアミノ酸1〜115と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含む、T細胞レセプター(TCR)を提供する。TCRは、単離され、細胞フリーであり及び/又は可溶性であってもよい。すなわち、TCRは、人体においてT細胞内に天然の状態で生じるTCRでなくてもよい。

本発明の範囲内のアミノ酸置換は、天然に存在するアミノ酸又は天然に存在しないアミノ酸を用いて行い得ることを理解すべきである。例えば、本明細書では、アラニンのメチル基はエチル基で置換されてもよく及び/又は軽微な変化がペプチド骨格になされてもよい。天然又は合成のアミノ酸が用いられているか否かにかかわらず、L-アミノ酸のみが存在することが好ましい。
2つのアミノ酸配列又は2つの核酸配列のパーセント同一性は、最適な比較目的のために配列を整列させ(例えば、最良整列のために第1の配列にギャップを導入することができる)、対応する位置でアミノ酸残基又はヌクレオチドを比較することにより決定する。「最良整列」は、最も高いパーセント同一性をもたらす2つの配列の整列である。パーセント同一性は、比較する配列中の同一アミノ酸残基又はヌクレオチドの数により決定する(すなわち、%同一性 = 同一位置の数/位置の総数×100)。
第2の観点のTCRは、β鎖FM2領域及びβ鎖FM3領域を含んでなってもよく、前記FM2領域及びFM3領域はそれぞれ配列番号49及び50を含み並びに/又は1若しくは2以上の保存的置換及び/若しくは3つまでの寛容される置換を含有する。
第2の観点のTCRは、配列番号2のアミノ酸1〜117及び/又は配列番号3のアミノ酸1〜115を含んでなってもよく、前記アミノ酸の各々は1若しくは2以上の保存的置換及び/若しくは3つまでの寛容される変異並びに/又は表1若しくは表2に規定される変異の1若しくは2以上を含有してもよい。
結合親和性(平衡定数KDに反比例する)及び結合半減期(T1/2と表す)は、本願実施例3の表面プラズモン共鳴(BIAcore)及び/又はOctet法を用いて決定することができる。TCRの親和性が二倍になればKDは1/2になると理解される。T1/2はln2/解離速度(koff)として算出される。したがって、T1/2が二倍になればkoffは1/2になる。TCRのKD値及びkoff値は、通常、可溶形態のTCR(すなわち、細胞質ドメイン及び膜貫通ドメインの残基を除去するように短縮された形態のもの)について測定する。好ましくは、所与のTCRの結合親和性又は結合半減期は、同じアッセイプロトコルを用いて数回、例えば3回又は4回以上測定し、その結果の平均をとる。
単鎖形式としては、Vα-L-Vβ、Vβ-L-Vα、Vα-Cα-L-Vβ又はVα-L-Vβ-Cβタイプ(ここで、Vα及びVβはそれぞれTCR α及びβ可変領域であり、Cα及びCβはそれぞれTCR α及びβ定常領域であり、Lはリンカー配列である)のαβ TCRポリペプチドが挙げられる。特定の実施形態において、本発明の単鎖TCRは、WO 2004/033685に記載されるように、それぞれの定常ドメインの残基間に導入されたジスルフィド結合を有してもよい。単鎖TCRは、WO2004/033685;WO98/39482;WO01/62908;Weidanzら(1998) J Immunol Methods 221(1-2):59-76;Hooら(1992) Proc Natl Acad Sci U S A 89(10):4759-4763;Schodin(1996) Mol Immunol 33(9):819-829)に更に記載されている。
本発明のα-βヘテロダイマーTCRは、通常、α鎖TRAC定常ドメイン配列及び/又はβ鎖TRBC1若しくはTRBC2定常ドメイン配列を含んでなる。α及びβ鎖の定常ドメイン配列は、TRACのエキソン2のCys4とTRBC1又はTRBC2のエキソン2のCys2との間の天然型ジスルフィド結合が欠失するように短縮化又は置換により改変されていてもよい。α鎖及び/又はβ鎖の定常ドメイン配列も、TRACのThr 48及びTRBC1又はTRBC2のSer 57からシステイン残基への置換により改変され、該システインがTCRのαβ定常ドメインとβ定常ドメインと間のジスルフィド結合を形成していてもよい。
別の1つの観点において、本発明は、本発明の核酸を含んでなるベクターを提供する。好ましくは、ベクターはTCR発現ベクターである。
本発明の可溶性TCRは、検出可能な標識又は治療剤の、抗原提示細胞及び抗原提示細胞を含有する組織への送達に有用である。したがって、これらは、検出可能な標識(TCRを用いてSLLMWITQC-HLA-A*02複合体を提示する細胞の存在を検出する診断目的のため);治療剤;又は(例えばPEG化による)PK調整成分と(共有結合又はその他の様式で)結合していてもよい。
本発明のTCRと結合され得る治療剤としては、免疫調整剤、放射性化合物、酵素(例えば、パーフォリン)又は化学療法剤(例えば、シスプラチン)が挙げられる。毒性効果が確実に所望の位置で発揮されるために、毒物は、緩徐に放出されるようにTCRに連結されたリポソームの内部に存在し得る。このことにより、人体における輸送の間の損傷効果が防止され、該当する抗原提示細胞へのTCRの結合後に、毒物が最大効果を有することが保証される。
・小分子細胞傷害性物質、すなわち、哺乳動物細胞を殺傷する能力を有する分子量700ダルトン未満の化合物。このような化合物はまた、細胞傷害性効果を有することができる毒性金属を含有し得る。更に、これら小分子細胞傷害性物質にはまた、プロドラッグ、すなわち、生理学的条件下で崩壊又は変換して細胞傷害性物質を放出する化合物が含まれると理解される。このような物質の例として、シスプラチン、メイタンシン(maytansine)誘導体、ラケルマイシン(rachelmycin)、カリケアマイシン(calicheamicin)、ドセタキセル、エトポシド、ゲムシタビン、イホスファミド、イリノテカン、メルファラン、ミトキサントロン、ソルフィマーソディウムホトフィリンII(sorfimer sodiumphotofrin II)、テモゾロマイド(temozolmide)、トポテカン、トリメトレキサート(trimetreate)、グルクロナート、オーリスタチンE(auristatin E)、ビンクリスチン及びドキソルビシンが挙げられる;
・ペプチド細胞毒素、すなわち、哺乳動物細胞を殺傷する能力を有するタンパク質又はそのフラグメント。例えば、リシン、ジフテリア毒素、シュードモナス細菌外毒素A、DNAアーゼ及びRNAアーゼ;

特に好適な抗CD3-TCR融合体は配列番号37のα鎖及び配列番号38のβ鎖を含んでなる。
本発明の物質の投薬量は、治療すべき疾患又は異常、治療すべき個体の年齢及び状態などに依存して広範囲に変化し得る。抗CD3抗体に結合した本発明の可溶性TCRに適切な用量範囲は、25ng/kg〜50μg/kgであり得る。最終的には医師が使用すべき適切な投薬量を決定する。
本発明のTCR、医薬組成物、ベクター、核酸及び細胞は、実質的に純粋な形態、例えば、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも91%、少なくとも92%、少なくとも93%、少なくとも94%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%又は100%純粋で提供されてもよい。
・医薬における使用のため、好ましくはガン治療法における使用のための、本発明のTCR、TCR-抗CD3融合体、核酸又は細胞

本発明の各々の観点の好適な特徴は、他の観点の各々についてのとおりである(ただし、必要な変更は加える)。本明細書において言及した先行技術文献は、法が許容する最大範囲まで組み込まれる。
下記では、本発明を以下の非限定的な図面及び実施例を参照して説明する。
実施例1−可溶性TCRの発現、リフォールディング及び精製
本発明の可溶性TCRのα及びβ細胞外領域をコードするDNA配列を、(Sambrookら,Molecular cloning. Vol. 2. (1989) New York:Cold spring harbor laboratory pressに記載のような)標準的方法を用いて、別々にpGMT7ベースの発現プラスミドにクローニングした。発現プラスミドを別々にE. coli株Rosetta(BL21pLysS)に形質転換し、単一アンピシリン抵抗性コロニーを、TYP(+アンピシリン100μg/ml)培地中で37℃にて、約0.6〜0.8のOD600まで増殖させた後、0.5mM IPTGでタンパク質発現を誘導した。誘導の3時間後に細胞を遠心分離により採集した。BugBusterタンパク質抽出試薬(Merck Millipore)を製造業者の指示に従って用いて細胞ペレットを溶解させた。封入体ペレットを遠心分離により回収した。ペレットをTriton緩衝液(50mM Tris-HCl pH8.1、0.5% Triton-X100、100mM NaCl、10mM NaEDTA)中で2回洗浄し、最後に界面活性剤フリーの緩衝液(50mM Tris-HCl pH8.1、100mM NaCl、10mM NaEDTA)中に再懸濁した。6Mグアニジン-HClで可溶化し、OD280を測定することによって、封入体タンパク質収量を定量した。次いで、消光係数を用いてタンパク質濃度を算出した。8M尿素で可溶化し、約2μgを還元条件下の4〜20% SDS-PAGEにロードして封入体の純度を測定した。次いで、デンシトメトリーソフトウェア(Chemidoc, Biorad)を用いて純度を推定又は算出した。封入体を、短期保存については+4℃にて、長期保存については−20℃又は−70℃にて保存した。
透析したリフォールド物をPOROS(登録商標) 50HQアニオン交換カラムに適用し、Akta(登録商標)精製装置(GE Healthcare)を用いて50カラム容量にわたり20mM Tris pH8.1中0〜500mM NaClのグラジエントで結合タンパク質を溶出させることにより可溶性TCRの精製を開始した。ピークTCR画分をSDS PAGEにより同定した後、プールし、濃縮した。次いで、濃縮サンプルを、ダルベッコのPBS緩衝液で予め平衡化させたSuperdex(登録商標) 75HRゲル濾過カラム(GE Healthcare)に適用した。ピークTCR画分をプールし、濃縮し、精製物質の最終収量を計算した。
ImmTACの調製は、TCR β鎖を、抗CD3単鎖抗体にリンカーを介して融合させたこと以外は、実施例1の記載のとおりに行った。加えて、カチオン交換工程を、アニオン交換後の精製の間に行った。この場合、アニオン交換からのピーク画分を20mM MES(pH6.5)中で20倍希釈し、POROS(登録商標) 50HSカチオン交換カラムに適用した。結合タンパク質を、20mM MES中0〜500mM NaClのグラジエントで溶出させた。ピークImmTAC画分をプールし、50mM Tris pH8.1に調整した後、濃縮し、実施例1に記載のとおりにゲル濾過マトリクスに直接適用した。
精製した可溶性TCR及びImmTAC分子の該当のペプチド-HLA複合体に対する結合の分析を、BIAcore 3000若しくはBIAcore T200装置を用いる表面プラズモン共鳴又はForteBio Octet装置を用いる二重層干渉分光法により行った。ビオチン化クラスI HLA-A*02分子を、目的のペプチドと共にリフォールディングし、当業者に公知の方法を用いて精製した(O'Callaghanら(1999),Anal Biochem 266(1):9-15;Garbocziら(1992),Proc Natl Acad Sci USA 89(8):3429-3433)。全ての測定は、0.005% P20を補充したダルベッコのPBS緩衝液中で25℃にて行った。
ビオチン化ペプチド-HLAモノマーをストレプトアビジン共役CM-5センサチップに固定した。平衡結合定数は、約200応答単位(RU)のペプチド-HLA-A*02複合体を被覆したフローセル上に30μl/分の定流速で注入した可溶性TCR/ImmTACの系列希釈物を用いて決定した。平衡応答は、各TCR濃度について、無関係のペプチド-HLAを含有するコントロールフローセルでのバルク緩衝液応答を減算することにより規格化した。KD値は、Prismソフトウェア及びLangmuir結合等温式 結合 = C×Max/(C+KD) (式中、「結合」は注入したTCR濃度Cでの平衡結合(RU)であり、Maxは最大結合である)を用いる非線形曲線フィッティングにより得た。
高親和性相互作用については、結合パラメータは、単一サイクル速度論分析により決定した。5つの異なる濃度の可溶性TCR/ImmTACを、約100〜200RUのペプチド-HLA複合体を被覆したフローセル上に、50〜60μl/分の流速で注入した。代表的には、60〜120μlの可溶性TCR/ImmTACを100〜200nMの最高濃度で注入し、他の4回の注入については2倍系列希釈物を用いた。最低濃度を先ず注入した。解離相を測定するため、その後、≧10%の解離が生じるまで、代表的には1〜3時間後まで、緩衝液を注入した。速度論パラメータはBIAevaluation(登録商標)ソフトウェアを用いて算出した。半減期の算出が可能とするため、解離相を一次指数関数的減衰式にフィッティングした。平衡定数KDはkoff/konから算出した。
ビオチン化ペプチド-HLAモノマーを、ストレプトアビジンを予め固定した(SA)ストレプトアビジンバイオセンサ(Pall ForteBio)に1nmに捕捉した。センサを遊離ビオチン(2μM)で2分間ブロックした。平衡結合定数は、ロードしたバイオセンサを、96ウェル又は384ウェルサンプルプレート内で系列希釈した可溶性TCR/ImmTAC中に浸漬することにより決定した。プレート振盪は1000rpmに設定した。低親和性相互作用(μM範囲)については、短い結合時間(約2分間)及び短い解離時間(約2分間)を用いた。Octetデータ分析ソフトウェア(Pall ForteBio)を用いて無関係のpHLAをロードした参照バイオセンサの二重参照減算により結合曲線を加工処理した。平衡時の応答(nm)を用いて、等式 応答 = Rmax×conc/(KD + conc) (式中、「応答」は各TCR濃度(conc)での平衡結合(nm)であり、RmaxはpHLA飽和時の最大結合応答である)にフィッティングさせた定常状態のプロットからKD値を推定した。
高親和性相互作用(nM〜pM範囲)については、速度論パラメータは、≧3 TCR/ImmTAC濃度のとき、代表的には10nM、5nM及び2.5nMの結合曲線から決定した。結合時間は30分であり、解離時間は1〜2時間であった。無関係のpHLAをロードし、ビオチンでブロックした参照バイオセンサの二重参照減算により結合曲線を加工処理した。速度論パラメータkon及びkoffは、Octetデータ分析ソフトウェア(Pall ForteBio)を用いて、結合曲線への直接の全体フィッティングにより算出した。KDはkoff/konから算出し、解離半減期はt1/2 = 0.693/koffから算出した。
可溶性野生型TCRを、実施例1に記載の方法に従って調製し、pHLAに対する結合を実施例3に従って分析した。α鎖及びβ鎖のアミノ酸配列は、図2に示したものに対応するものであった。可溶性ビオチン化HLA-A*02を、野生型NY-ESO-1ペプチド(SLLMWITQC)又はヘテロクリティック(heteroclitic)NY-ESO-1ペプチド(SLLMWITQV:配列番号34)のいずれかを用いて調製し、BIAcoreセンサチップに固定した。KD値はそれぞれ5.2μM及び4.3μMと決定された。15の無関係なペプチド-HLA-A*02複合体に対して有意な結合は検出されなかった。
これらデータは、TCRが標的に適切な親和性及び特異性で結合することを示している。したがって、これらTCR鎖は、本発明の更なるTCRの同定に有用な足場を提供する。
可溶性変異TCR及びImmTAC分子を実施例1及び2に記載のように調製し、結合特性を実施例3に従って決定した。TCR α鎖及び/又はβ鎖は、少なくとも1つのCDR領域に、図2に示したCDR配列に対する変異を含有していた(配列番号41〜46)。本発明の特定の変異TCR α鎖及びβ鎖可変領域のアミノ酸配列をそれぞれ図4及び5に示す。下記の表は、示したα及びβ可変領域を含んでなる可溶性TCR及び/又はImmTAC分子の結合特性を提供する。結合は、ヘテロクリティックNY-ESO-1ペプチド(SLLMWITQV)を用いて測定した。
本発明のα及びβ可変鎖配列を含有するImmTAC分子を、CD3+ T細胞の強力で特異的な再指向化を仲介する能力について、T細胞活性化の指標としてインターフェロン-γ(IFN-γ)分泌を利用するELISPOTアッセイにより試験した。
試験した4つのImmTAC分子のα鎖及びβ鎖の配列を図5〜8に示す。
アッセイはヒトIFN-γ ELISPOTキット(BD Biosciences)を用いて行った。標的細胞をアッセイ培地(10%熱不活化FBS及び1%ペニシリン-ストレプトマイシン-L-グルタミンを含むRPMI 1640)中に1×106/mlの密度で準備し、50,000細胞/ウェルにて50μl容量でプレートした。本実施例では次の標的細胞株を用いた:
・NCI-H1755(NY-ESO-1+ve;HLA-A*02+ve)ヒト肺ガン細胞株(ATCCから供給;cat. no:CRL-5892)
・HAo5(NY-ESO-1-ve;HLA-A*02+ve)ヒト心臓細胞(Promocellから供給;cat. no:C-12271)
プレートを製造業者の指示に従って準備した。標的細胞、エフェクター細胞及びImmTAC分子を該当するウェルに加え、アッセイ培地で最終容量を200μlとした。全ての反応を三連で行った。ImmTAC、エフェクター細胞又は標的細胞のいずれかを省略したコントロールウェルも準備した。次いで、プレートを一晩インキュベートした(37℃/5%CO2)。翌日、プレートを洗浄緩衝液(脱イオン水中に作製した0.05% P20を含む1×PBS)で3回洗浄した。次いで、一次検出抗体を各ウェルに容量50μlで加えた。プレートを室温にて2時間インキュベートした後、再び3回洗浄した。二次検出は、50μlの希釈ストレプトアビジン-HRPを各ウェルに加え、室温にて1時間インキュベートし、洗浄工程を繰り返すことにより行った。次いで、プレートを200μlのPBS(pH7.4)で2回洗浄した。使用前15分以内に1滴(20μl)のAEC色原体を各1mlのAEC基質に加えて混合し、50μlを各ウェルに加えた。スポット発色を定期的にモニターし、プレートを水道水で洗浄して発色反応を停止させた。次いで、プレートを室温にて少なくとも2時間乾燥させた後、Immunospotソフトウェアを備えたCTL分析装置(Cellular Technology Limited)を用いてスポットを計数した。
ImmTAC分子3及び4について効力の更なる評価を行い、EC50値を決定した。ELISpotアッセイを、10-13M〜10-8Mの範囲のImmTAC濃度で、上記のとおりに行った。Prism 5.0ソフトウェア(GraphPad)を用いてデータを分析し、EC50値を算出した。その値は、ImmTAC分子3及び4についてそれぞれ95pM及び121pMと決定された(図10)。このデータから、これらImmTAC分子が強力な再指向化T細胞応答を仲介する能力が確証される。
ImmTAC分子の更なる特異性試験を、正常細胞のパネルに対して行った。本実施例ではImmTAC分子2〜4(図6〜8)を用いた。インターフェロン-γ(IFN-γ)分泌をT細胞活性化の指標として用いた。
アッセイは、IFN-γ DuoSet ELISAキット(R&D Systems, Cat No:DY285)を用いて行い、製造業者の指示のとおりに行った。簡潔には、IFN-γを10,000pg/mlに希釈し、2倍希釈物を作製して標準曲線を作成した。標的細胞を計数し、アッセイ培地に10μl中10,000細胞/ウェルでプレートした。ImmTAC分子を、10μl/ウェルで最終濃度が2nM及び1nMとなるように希釈した。ImmTACを含まないコントロールサンプルも準備した。エフェクターPBMCを解凍し、10μl中10,000細胞/ウェルでプレートした。プレートを48時間インキュベートした後、発色させて読み取った。
図11に示すデータは、治療上妥当なImmTAC濃度範囲内で、正常組織に由来する細胞株の存在下でのIFN-γ産生は、抗原ポジティブガン細胞株の存在下と比して、極めて低いことを証明する。このデータは、本発明のImmTAC分子が高レベルの特異性を有し、したがって治療用途に特に適切であることを示している。
本発明のImmTAC分子が再指向化T細胞による抗原ポジティブ腫瘍細胞の強力な殺傷を仲介する能力を、IncuCyteプラットフォーム(Essen BioScience)を用いて調べた。このアッセイは、アポトーシスのマーカーであるカスパーゼ-3/7の放出の顕微鏡観察によるリアルタイム検出を可能とする。
アッセイは、CellPlayer 96ウェルカスパーゼ-3/7アポトーシスアッセイキット(Essen BioScience, Cat. No.4440)を用いて行い、製造業者のプロトコルに従って行った。簡潔には、標的細胞(NCI-H1755 - NYESO+ve HLA A*02+ve又はHCT-116 - NYESO-ve HLA A*02+ve)を5000細胞/ウェルでプレートし、一晩インキュベートして細胞を接着させた。ImmTAC溶液を2.16nM〜8.8pMの濃度で調製した。25μlの各濃度溶液を該当するウェルに加えた。PBMCをエフェクター細胞として用い、50μl中50,000/ウェルでプレートした。ImmTACを含まないコントロールサンプルも調製した。NucViewアッセイ試薬を30μMで作製し、25μlを各ウェルに加えた(最終濃度5μM)。プレートをIncuCyte装置内に配置し、2時間ごとに(ウェルあたり1画像)3日間にわたって撮像した。各画像中のアポトーシス細胞の数を決定し、mm2あたりの対象物の数として記録した。アッセイは3連で行った。
このデータから、ImmTAC3及びimmTAC4が、治療上妥当な濃度範囲で、再指向化T細胞による抗原ポジティブ腫瘍細胞の強力な殺傷を仲介することが確証される。
Claims (34)
- SLLMWITQC(配列番号1)-HLA-A*02複合体に対する結合性を有し、TCR α鎖可変ドメイン及びTCR β鎖可変ドメインを含んでなり、
α鎖可変ドメインが配列番号12のアミノ酸1〜117と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含み、
β鎖可変ドメインが配列番号15のアミノ酸1〜115と少なくとも90%の同一性、例えば90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又は100%の同一性を有するアミノ酸配列を含み、
アミノ酸残基27〜32、50〜57及び91〜107のα鎖可変ドメイン配列並びにアミノ酸残基27〜31、49〜55及び93〜106のβ鎖可変ドメイン配列が次の表:
T細胞レセプター(TCR)。 - SLLMWITQC(配列番号1)-HLA-A*02複合体に50μMより大きい親和性で結合する請求項1に記載のTCR。
- α鎖TRAC定常ドメイン配列及びβ鎖TRBC1又はTRBC2定常ドメイン配列を有するα-βヘテロダイマーである請求項1〜3のいずれか1項に記載のTCR。
- α鎖及びβ鎖の定常ドメイン配列がTRACのエキソン2のCys4と、TRBC1又はTRBC2のエキソン2のCys2との間の天然型ジスルフィド結合が欠失するように短縮化又は置換により改変されている、請求項4に記載のTCR。
- α鎖及び/又はβ鎖の定常ドメイン配列がTRACのThr 48及びTRBC1又はTRBC2のSer 57からシステイン残基への置換により改変されており、該システイン同士がTCRのα定常ドメインとβ定常ドメインと間で非天然型ジスルフィド結合を形成している、請求項4又は5に記載のTCR。
- Vα-L-Vβ、Vβ-L-Vα、Vα-Cα-L-Vβ、Vα-L-Vβ-Cβ型(式中、Vα及びVβはそれぞれTCR α及びβ可変領域であり、Cα及びCβはそれぞれTCR α及びβ定常領域であり、Lはリンカー配列である)の単鎖形式である請求項1〜6のいずれか1項に記載のTCR。
- 検出可能な標識、治療剤又はPK調整成分と結合した請求項1〜7のいずれか1項に記載のTCR。
- 請求項1〜8のいずれか1項に記載のTCRと、該TCRのα鎖又はβ鎖のC末端又はN末端に共有結合した抗CD3抗体とを含んでなるTCR-抗CD3融合体。
- 配列番号6〜12又は51から選択されるα鎖可変ドメイン及び配列番号13〜23又は52から選択されるβ鎖可変ドメインを含んでなる請求項9に記載のTCR-抗CD3融合体。
- β鎖が抗CD3抗体配列にリンカー配列を介して連結されている、請求項10に記載のTCR-抗CD3融合体。
- リンカー配列が、GGGGS(配列番号24)、GGGSG(配列番号25)、GGSGG(配列番号26)、GSGGG(配列番号27)、GSGGGP(配列番号28)、GGEPS(配列番号29)、GGEGGGP(配列番号30)及びGGEGGGSEGGGS(配列番号31)からなる群より選択される、請求項11に記載のTCR-抗CD3融合体。
- 配列番号37からなるα鎖及び配列番号38からなるβ鎖を含んでなる請求項13に記載のTCR-抗CD3融合体。
- 配列番号32からなるα鎖及び配列番号33からなるβ鎖を含んでなる請求項13に記載のTCR-抗CD3融合体。
- 配列番号12のアミノ酸配列からなるα鎖可変ドメイン及び配列番号15のアミノ酸配列からなるβ鎖可変ドメインを含んでなり、ここで、β鎖が抗CD3抗体にリンカー配列を介して連結されている、請求項9〜12のいずれか1項に記載のTCR-抗CD3融合体。
- 配列番号12のアミノ酸配列からなるα鎖可変ドメイン及び配列番号15のアミノ酸配列からなるβ鎖可変ドメインを含んでなり、ここで、α鎖が抗CD3抗体にリンカー配列を介して連結されている、請求項9〜12のいずれか1項に記載のTCR-抗CD3融合体。
- α鎖可変ドメインが配列番号12のアミノ酸配列からなり、β鎖可変ドメインが配列番号15のアミノ酸配列からなり、ここで、β鎖が抗CD3抗体に配列番号24のリンカー配列を介して連結されている、請求項17に記載のTCR-抗CD3融合体。
- 請求項1〜8のいずれか1項に記載のTCR又は請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体をコードする核酸。
- 請求項19に記載の核酸を含んでなる発現ベクター。
- 請求項1〜8のいずれか1項に記載のTCR又は請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体を提示する天然に存在しない及び/又は精製された及び/又は工学的に操作された細胞。
- (a)請求項20に記載のTCR発現ベクターを、単一オープンリーディングフレーム中又はα鎖及びβ鎖をそれぞれコードする2つの別個のオープンリーディングフレーム中に有するか;又は
(b)請求項1〜8のいずれか1項に記載のTCR又は請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体のα鎖をコードする核酸を含んでなる第1の発現ベクターと、請求項1〜8のいずれか1項に記載のTCR又は請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体のβ鎖をコードする核酸を含んでなる第2の発現ベクターとを有する、
細胞。 - T細胞である請求項21又は22に記載の細胞。
- 請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸又は請求項21〜23のいずれか1項に記載の細胞を、1又は2以上の医薬的に許容可能な担体又は賦形剤と共に含んでなる医薬組成物。
- ヒト対象における医薬に用いるための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸又は請求項21〜23のいずれか1項に記載の細胞。
- ヒト対象におけるガン治療法に用いるための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- ヒト対象がNY-ESO-1及び/又はLAGE-1Aを発現する腫瘍を有する、請求項26に記載の使用のための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項20〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- 腫瘍が固形腫瘍である、請求項27に記載の使用のための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- 腫瘍が、滑膜肉腫、非小細胞肺ガン(NSCLC)、膀胱の腫瘍、胃の腫瘍、前立腺の腫瘍、直腸結腸の腫瘍、***の腫瘍、卵巣の腫瘍、食道の腫瘍、メラノーマ、多発性骨髄腫、肝細胞ガン及び頭部頸部の腫瘍から選択される、請求項27又は28に記載の使用のための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- ヒト対象がHLA-A*02サブタイプの対象である、請求項26〜29のいずれか1項に記載の使用のための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- 抗腫瘍剤と別々に、組み合わせて又は逐次に投与することに用いる請求項24に記載の医薬組成物。
- 請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物を含んでなる、ヒト対象投与用の注射可能な製剤。
- TCR、TCR-抗CD3融合体、核酸、細胞又は医薬組成物が注射により、例えば静脈内注射又は腫瘍内直接注射により投与される、請求項25〜30のいずれか1項に記載の使用のための、請求項1〜8のいずれか1項に記載のTCR、請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体、請求項19に記載の核酸、請求項21〜23のいずれか1項に記載の細胞又は請求項24に記載の医薬組成物。
- a)請求項22に記載の宿主細胞を、核酸の発現に最適な条件下に維持し、b)核酸によりコードされるTCR又はTCR-抗CD3融合体を単離することを含んでなる、請求項1〜8のいずれか1項に記載のTCR又は請求項9〜18のいずれか1項に記載のTCR-抗CD3融合体の製造方法。
