Description
The present invention relates to a synthetic polymer film having a surface having a bactericidal action, a sterilization method using the surface of the synthetic polymer film, a mold for producing the synthetic polymer film, and a method for producing the mold. The term “mold” as used herein includes molds used for various processing methods (stamping and casting), and may be referred to as a stamper. It can also be used for printing (including nanoprints).
Recently, it has been announced that the nano-surface structure of black silicon, semi and dragonfly wings has a bactericidal action (Non-Patent Document 1). It is said that the physical structure of the nanopillars of black silicon, semi-finished and dragonfly wings exerts a bactericidal action.
According to Non-Patent
From the results described in
Further, even if the bactericidal action of black silicon is used, black silicon is poor in mass productivity, and is hard and brittle, so that there is a problem that shape processing is low.

"Microorganism" includes viruses, bacteria (bacteria), and fungi (mold).
"Antimicrobial" broadly includes controlling and preventing the growth of microorganisms, and also includes suppressing darkening and slimming caused by microorganisms.

By using an anodized porous alumina layer, a mold having an inverted moth-eye structure can be manufactured with high mass productivity (for example,
The present inventor has developed a synthetic polymer membrane whose surface has a bactericidal effect by applying the above-described technology.
The structure of the synthetic polymer film according to the embodiment of the present invention will be described with reference to FIGS.
FIGS. 1A and 1B show schematic cross-sectional views of
The
The
The
When viewed from the normal direction of the
Note that, in this specification, the “moth-eye structure” refers to a convex shape having an increased cross-sectional area (a cross section parallel to the film surface), like the convex portion 34Ap of the
A mold for forming a moth-eye structure on the surface as illustrated in FIGS. 1A and 1B (hereinafter referred to as a “moth-eye mold”) is obtained by reversing the moth-eye structure. Have. If the anodized porous alumina layer having the inverted moth-eye structure is used as it is as a mold, the moth-eye structure can be manufactured at low cost. In particular, when a cylindrical moth-eye mold is used, a moth-eye structure can be efficiently manufactured by a roll-to-roll method. Such moth-eye molds can be manufactured by the methods described in

First, as shown in FIG. 2A, as a mold base, an
As the

As the material of the
It is preferable that the thickness of the

When the aluminum alloy film is used, a mirror surface having a reflectance of 80% or more can be obtained. The average grain size of the crystal grains constituting the aluminum alloy film when viewed from the normal direction of the aluminum alloy film is, for example, 100 nm or less, and the maximum surface roughness Rmax of the aluminum alloy film is 60 nm or less. The content of nitrogen contained in the aluminum alloy film is, for example, 0.5 mass% or more and 5.7 mass% or less. The absolute value of the difference between the standard electrode potential of a metal element other than aluminum contained in the aluminum alloy film and the standard electrode potential of aluminum is 0.64 V or less, and the content of the metal element in the aluminum alloy film is 1.0 mass % Or more and 1.9 mass% or less. The metal element is, for example, Ti or Nd. However, the metal element is not limited to this, and other metal elements (for example, Mn, Mg, Zr, V, and the like) whose absolute value of the difference between the standard electrode potential of the metal element and the standard electrode potential of aluminum is 0.64 V or less. The aluminum alloy film is formed by, for example, a DC magnetron sputtering method. The thickness of the aluminum alloy film is also preferably in the range of about 500 nm or more and about 1500 nm or less, for example, about 1 μm.
Next, as shown in FIG. 2B, the
The
Next, as shown in FIG. 2C, the opening of the
Next, as shown in FIG. 2 (d), the
Thereafter, if necessary, the
As described above, the above-described anodizing step and etching step are alternately repeated a plurality of times (for example, 5 times: 5 times of anodic oxidation and 4 times of etching), thereby inverting as shown in FIG. A moth-
The
In addition, under the
The method for manufacturing a moth-eye mold illustrated here can manufacture a mold for manufacturing an antireflection film described in
Further, according to the above-described mold manufacturing method, a mold having a low regularity of arrangement of concave portions suitable for manufacturing an antireflection film can be manufactured. When the bactericidal property of the moth-eye structure is used, it is considered that the regularity of the arrangement of the convex portions does not affect. A mold for forming a moth-eye structure having regularly arranged projections can be manufactured, for example, as follows.
For example, after forming a porous alumina layer having a thickness of about 10 μm, the generated porous alumina layer may be removed by etching, and then anodic oxidation may be performed under the conditions for forming the porous alumina layer. The porous alumina layer having a thickness of 10 μm is formed by increasing the anodic oxidation time. When a relatively thick porous alumina layer is generated in this way and the porous alumina layer is removed, the porous alumina layer is regularly arranged without being affected by irregularities and processing strain due to grains existing on the surface of the aluminum film or the aluminum base material. A porous alumina layer having a concave portion can be formed. For removing the porous alumina layer, it is preferable to use a mixed solution of chromic acid and phosphoric acid. When etching is performed for a long time, galvanic corrosion may occur. However, a mixed solution of chromic acid and phosphoric acid has an effect of suppressing galvanic corrosion.
The moth-eye mold for forming the
First, in the same manner as described with reference to FIGS. 2A and 2B, the
Next, as shown in FIG. 3A, the opening of the
Next, as shown in FIG. 3B, the
(Dp<Dint)。Thereafter, as described with reference to FIG. 2E, the etching step and the anodic oxidation step are alternately repeated a plurality of times. For example, by repeating the etching step three times and the anodic oxidation step three times, a moth-
The size of the microorganism depends on its type. For example, Pseudomonas aeruginosa has a size of about 1 μm, but some bacteria have a size of several 100 nm to about 5 μm, and fungi are several μm or more. For example, a projection having a two-dimensional size of about 200 nm is considered to have a bactericidal action on microorganisms having a size of about 0.5 μm or more, but has a bactericidal effect on bacteria having a size of several 100 nm. In addition, there is a possibility that a sufficient bactericidal action is not exhibited because the convex portion is too large. In addition, the size of the virus is several tens nm to several hundreds nm, and is often 100 nm or less. The virus does not have a cell membrane, but has a protein shell called capsid surrounding the virus nucleic acid. Viruses are classified into viruses having a membrane envelope outside the shell and viruses not having an envelope. In a virus having an envelope, it is considered that the convex portion acts on the envelope in the same manner, since the envelope is mainly composed of lipid. Examples of viruses having an envelope include influenza virus and Ebola virus. In a virus without an envelope, it is considered that a convex portion acts similarly on a shell of a protein called capsid. When the projection has a nitrogen element, the affinity with a protein composed of amino acids can be increased.
Therefore, the structure of a synthetic polymer membrane having a convex portion capable of exhibiting a bactericidal action even for microorganisms of several hundred nm or less and a method for producing the same will be described below.
Hereinafter, a convex portion having a two-dimensional size of more than 20 nm and less than 500 nm included in the synthetic polymer film exemplified above is referred to as a first convex portion. Further, a convex portion formed so as to overlap with the first convex portion is referred to as a second convex portion, and the two-dimensional size of the second convex portion is two-dimensional size of the first convex portion. And not more than 100 nm. Note that when the two-dimensional size of the first protrusion is less than 100 nm, particularly less than 50 nm, it is not necessary to provide the second protrusion. The concave portion of the mold corresponding to the first convex portion is referred to as a first concave portion, and the concave portion of the mold corresponding to the second convex portion is referred to as a second concave portion.
By alternately performing the anodic oxidation step and the etching step, the second recess cannot be formed even if the method of forming the first recess having a predetermined size and shape is applied as it is.
FIG. 4A shows a SEM image of the surface of the aluminum base material (
Therefore, the method for manufacturing a mold according to the embodiment of the present invention comprises the steps of (a) preparing an aluminum film deposited on an aluminum substrate or a support, and (b) electrolyzing the surface of the aluminum substrate or the aluminum film. An anodic oxidation step of forming a porous alumina layer having a first concave portion by applying a first level of voltage in a state where the porous alumina layer is in contact with the liquid; Contacting the porous alumina layer with the electrolytic solution after the etching step of enlarging the first concave portion by contacting the porous alumina layer with the electrolytic solution, and (d) step (c). Forming a second recess in the first recess by applying a voltage at a second level. For example, the first level is above 40V and the second level is below 20V.
That is, in the anodic oxidation step at a first level of voltage, a first recess having a size not affected by the grains of the aluminum substrate or the aluminum film is formed, and then the thickness of the barrier layer is reduced by etching. After the reduction, the second recess is formed in the first recess by an anodizing process at a voltage of a second level lower than the first level. When the second recess is formed in such a manner, the influence of the grains is eliminated.
With reference to FIG. 5, a mold having a first concave portion 14pa and a second concave portion 14pb formed in the first concave portion 14pa will be described. FIG. 5A is a schematic plan view of a porous alumina layer of a mold, FIG. 5B is a schematic cross-sectional view, and FIG. 5C shows an SEM image of a prototype mold.
As shown in FIGS. 5A and 5B, the surface of the mold according to the present embodiment has a plurality of first concave portions 14pa having a two-dimensional size in a range of more than 20 nm and less than 500 nm, and a plurality of first concave portions 14pa. It further has a plurality of second concave portions 14pb formed so as to overlap with the first concave portions 14pa. The two-dimensional size of the plurality of second recesses 14pb is smaller than the two-dimensional size of the plurality of first recesses 14pa and does not exceed 100 nm. The height of the second concave portion 14pb is, for example, more than 20 nm and 100 nm or less. The second recess 14pb also preferably includes a substantially conical portion, like the first recess 14pa.
The porous alumina layer shown in FIG. 5C was manufactured as follows.
As the aluminum film, an aluminum film containing 1 mass% of Ti was used. An oxalic acid aqueous solution (concentration 0.3 mass%,
Figure 5 (c) As can be seen from, among D p is in the first recess of about 200 nm, a second recess of D p is about 50nm is formed. In the above manufacturing method, the first level of the voltage change from 80V to 45V, was formed a porous alumina layer, in D p is in the first recess of about 100 nm, a D p is about 50nm Two recesses were formed.
When a synthetic polymer film is manufactured using such a mold, a synthetic polymer having a convex portion obtained by inverting the structure of the first concave portion 14pa and the second concave portion 14pb shown in FIGS. A film is obtained. That is, a synthetic polymer film further having a plurality of second protrusions formed to overlap the plurality of first protrusions is obtained.
As described above, the synthetic polymer film having the first convex portion and the second convex portion formed so as to overlap the first convex portion can be formed by a relatively small microorganism having a size of about 100 nm and a relatively large size of 5 μm or more. It can have a bactericidal effect on microorganisms.
Of course, depending on the size of the target microorganism, only the concave portions whose two-dimensional size is in the range of more than 20 nm and less than 100 nm may be formed. A mold for forming such a convex portion can be manufactured, for example, as follows.
Anodizing is performed using neutral salt aqueous solution such as ammonium tartrate aqueous solution (ammonium borate, ammonium citrate, etc.) or organic acid with low ion dissociation degree (maleic acid, malonic acid, phthalic acid, citric acid, tartaric acid, etc.). Then, a barrier-type anodic oxide film is formed, the barrier-type anodic oxide film is removed by etching, and then anodized at a predetermined voltage (the voltage of the second level) to reduce the two-dimensional size. Concave portions in the range of more than 20 nm and less than 100 nm can be formed.
For example, an aluminum film containing 1 mass% of Ti is used as an aluminum film, and a barrier-type anodic oxide film is formed by performing anodization at 100 V for 2 minutes using a tartaric acid aqueous solution (concentration: 0.1 mol / l, temperature: 23 ° C.). To form Thereafter, the barrier type anodic oxide film is removed by etching with a phosphoric acid aqueous solution (concentration: 10 mass%, temperature: 30 ° C.) for 25 minutes. Thereafter, in the same manner as above, an oxalic acid aqueous solution (concentration: 0.3 mass%, temperature: 10 ° C.) is used as an anodizing solution, and anodizing at 20 V is performed for 52 seconds, and etching using the etching solution is performed alternately for 5 minutes. By repeating the anodic oxidation five times and the etching four times, it is possible to uniformly form a recess having a two-dimensional size of about 50 nm.

The bactericidal properties of a synthetic polymer film having a moth-eye structure on the surface have a correlation with not only the physical structure of the synthetic polymer film but also the chemical properties of the synthetic polymer film. For example, the applicant of the present application has reported that the chemical properties include the contact angle of the surface of a synthetic polymer film (Patent Publication 1: Japanese Patent No. 5788128) and the concentration of a nitrogen element contained in the surface (International Publication No. 2: 2016/080245). FIG. 7 shows an SEM image shown in International Publication 2 (FIG. 8). FIGS. 7A and 7B are views showing SEM images of Pseudomonas aeruginosa that has died on the surface having the moth-eye structure shown in FIG. 1A, observed by SEM (scanning electron microscope).
Looking at these SEM images, it can be seen that the tips of the protrusions have penetrated into the cell wall (outer membrane) of Pseudomonas aeruginosa. Also, when looking at FIGS. 7A and 7B, it does not appear that the protruding portion has penetrated the cell wall, but it looks as if the protruding portion has been taken into the cell wall. This may be explained by the mechanism suggested in Supplemental Information of

7. An NB medium (Normal broth medium E-MC35, manufactured by Eiken Chemical Co., Ltd.) was added as a nutrient to the bacterial diluent A to a concentration of 1/500. (The number is on the order of 1E + 05 CFU / mL) (based on JIS Z2801 5.4a))
Bacterial diluent B: Bacterial diluent A1 mL + sterile water 8.98 mL + NB medium 20 μL
8. 400 μL of the bacterial diluent B (the number of bacteria in the bacterial diluent B at this time may be referred to as “initial bacterial count”) is dropped on each sample film, and a cover (for example, a cover glass) is placed on the bacterial diluent B And adjust the amount of the diluent B per unit area. Here, the initial number of bacteria was set to 3.5E + 05 CFU / mL.
9. Leave for a certain period of time in an environment of 37 ° C and 100% relative humidity (Leaving time: 4 hours, 24 hours)
10. The entire sample film with the bacteria diluent B and 9.6 mL of sterile water are put into a filter bag, and the filter bag is rubbed by hand to thoroughly wash off the bacteria on the sample film. The washing solution in the filter bag is obtained by diluting the bacteria diluent B by 25 times. This washing solution may be referred to as a bacteria dilution B2. When there is no increase or decrease in the number of bacteria in the bacteria dilution B, the bacteria dilution B2 is on the order of 1E + 04 CFU / mL.
11. The diluent B2 is diluted 10 times to prepare the diluent C. Specifically, it is prepared by adding 120 μL of the washing solution (bacterial diluent B2) to 1.08 mL of sterilized water. When there is no increase or decrease in the number of bacteria in the bacteria diluent B, the bacteria diluent C is on the order of 1E + 03 CFU / mL.
12. In the same manner as in the preparation of the diluent C, the diluent C is diluted 10-fold to prepare the diluent D. When there is no increase or decrease in the number of bacteria in the diluent B, the diluent D is on the order of 1E + 02 CFU / mL. Further, the bacterial dilution D is diluted 10 times to prepare a bacterial dilution E. When there is no increase or decrease in the number of bacteria in the bacteria dilution B, the bacteria dilution E is on the order of 1E + 01 CFU / mL.
13. 1 mL of the bacterial diluent B2 and the bacterial diluents CE are dropped on Petrifilm (registered trademark) medium (manufactured by 3M, product name: AC plate for measuring the number of viable bacteria) at 37 ° C. and 100% relative humidity. After 48 hours of the culture, the number of bacteria in the bacterial dilution B2 is counted.
In addition, in 5.6h) of JISZ2801, phosphate buffered saline is used when preparing a diluent, but sterile water was used here. It has been confirmed that the sterilization effect due to the physical structure and chemical properties of the surface of the sample film can be examined even when using sterile water.
[Evaluation of antibacterial properties]
According to JIS Z 2801, the antibacterial activity value determined from the number of bacteria after culturing for 24 hours was 2.0 or more (99% or more mortality), indicating that there was an antibacterial effect on P. aeruginosa. A base film (PET film) was used as a reference film. The antibacterial activity value is a logarithmic value of the number obtained by dividing the number of bacteria after culturing the PET film for 24 hours by the number of bacteria after culturing the sample film for 24 hours.
FIG. 8 is a graph showing the evaluation results of the bactericidal properties. In FIG. 8, the horizontal axis represents the standing time (hour), and the vertical axis represents the number of bacteria (CFU / mL) in the diluent B2. Note that, in FIG. 8, for ease of viewing, the plot is plotted as 0.1 when the number of bacteria is 0 (ND). Table 2 below shows the number of bacteria and the antibacterial activity value after culturing. In addition, PET2 data was used for the calculation of the antibacterial activity value of the sample film C1, and PET1 data was used for the other sample films.
As can be seen from FIG. 8 and Table 2, the samples other than the sample film C1 have an antibacterial activity value of 2.0 or more and have antibacterial properties. The antibacterial activity values of the sample films C2 and B are 2.6 and 3.2, respectively. The antibacterial activity values of the sample films A1, A2 and A3 are 6.2, which means that they have bactericidal properties. Here, when the antibacterial activity value is 6.0 or more, it is assumed to have bactericidal properties. The results of evaluating the antibacterial properties and the bactericidal properties in this way are shown in Table 1 by // ×. ○ indicates antibacterial or bactericidal, and X indicates no antibacterial or bactericidal.
Regarding antibacterial properties and bactericidal properties, it has at least antibacterial properties except for the film using the resin C1. From the viewpoint of antibacterial properties, it is considered preferable to contain more than 0.0020 EO units. In addition, the resins A4, A5, D and E were not evaluated this time, but from the evaluation results of the resins having the same or similar composition, all of these resins have antibacterial properties and bactericidal properties. it seems to do.
Table 1 shows the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine, and the total nitrogen element concentration (that is, the nitrogen element forming the tertiary amine). The calculated nitrogen element concentration is also shown. The evaluation results for antibacterial and bactericidal properties correlate with the nitrogen element concentration of the primary or secondary amine, as compared to the nitrogen element concentration including the nitrogen element forming the tertiary amine. Looks to have a relationship. The reason is considered as follows. Since the nitrogen element forming the tertiary amine has low basicity, it is considered that the contribution of the synthetic polymer membrane to the bactericidal property is low. In the acrylic resins I and II containing a nitrogen element, the nitrogen element forming the tertiary amine forms a ring. Since the nitrogen element forming the ring exists at a position relatively far from the surface of the synthetic polymer film and has a large distance from the microorganism, it is considered that the contribution of the synthetic polymer film to the bactericidal property is low.
In addition, in the above International Publication No. 2, from the viewpoint of sterilization, it is preferable that the concentration of the nitrogen element on the surface is 0.7 at% or more, but this time, a resin having a nitrogen element concentration lower than this is used. It was also found that germicidal properties could be obtained. At least when the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine is 0.293 at% or more (resin C2), it can be said that the composition can have antibacterial properties. If the third digit after the decimal point is rounded off, if the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine is 0.29 at% or more, it can have antibacterial properties. It can be said. In order to have bactericidal properties, the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine is preferably 0.327 at% or more (resin A1). When rounded to the third decimal place, in order to have bactericidal properties, the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine is 0.33 at% or more. Is preferred. At this time, it is considered that the number of moles of the EO unit contained in 1 g of the resin is preferably 0.0040 or more.
Since the resin containing the EO unit at an appropriate ratio has hydrophilicity, it is possible to wipe off dirt with water. Moreover, since it has flexibility, it has excellent scratch resistance.
Further, as can be seen from the SEM image of FIG. 7, the convex portion to which Pseudomonas aeruginosa does not adhere is almost parallel to the normal direction of the synthetic polymer film, whereas the convex portion to which Pseudomonas aeruginosa adheres. Some parts are leaning toward the Pseudomonas aeruginosa. By tilting (bending) the convex portions, more convex portions can come into contact with the microorganism. It is considered that a synthetic polymer membrane having a convex portion on its surface capable of inclining (bending) in the direction of microorganisms can have a better bactericidal effect. It is also conceivable that the resin containing an appropriate amount of EO units may exhibit a bactericidal effect due to this bending.
Next, sample films F, G, and H having the same structure as the
The chemical structural formula of the acrylic resin I ′ is shown in [Chem. 6]. The chemical formula shown in [Formula 6] is estimated. The acrylic resin I ′ contains a repeating structure of EO units (the number of repetitions is 4 or 5). The acrylic resin I ′ differs from the acrylic resin I in that the number of EO units is about half that of the acrylic resin I. The acrylic resin I ′ is urethane acrylate (manufactured by Shin-Nakamura Chemical Co., Ltd.) and contains a nitrogen element. Acrylic resin I 'is a trifunctional urethane acrylate. The acrylic resin I ′ contains a cyanuric ring that is a heterocyclic ring containing a nitrogen element (heterocyclic ring).
Table 3 shows the molecular weight (MW) of each acrylic resin and the number of EO units contained in one molecule, as in Table 1. Regarding the molecular weight (MW) of the acrylic resin I ′ and the number of EO units contained in one molecule, the number of repetitions of the repeating structure of the EO units was all 4 (1 = m = n = 4 in the chemical structural formula). The case (upper) and the case where the number of repetitions of the repeating structure of the EO unit is all 5 (l = m = n = 5 in the chemical structural formula) (lower) are also described. Table 3 shows the number of moles of EO unit contained in 1 g of each resin calculated based on the composition and the chemical formula for each of resins F, G, and H. The number of moles of the EO unit contained in 1 g of the resin G containing the acrylic resin I ′ is described in both the case where l = m = n = 4 (upper row) and the case where l = m = n = 5 (lower row). ing. As shown in Table 3, the number of moles of the EO unit contained in 1 g of the resin G containing the acrylic resin I 'is considered to be 0.0095 or more and 0.0108 or less.
In Table 3, for each of the resins F, G and H, the nitrogen element at% calculated based on the composition and the chemical formula is shown. Table 3 shows the total nitrogen element concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine, and the total nitrogen element concentration (that is, the nitrogen element forming the tertiary amine). And the calculated nitrogen element concentration (including the nitrogen element that does not form an amine). As shown in Table 3, the concentration of the nitrogen element that forms the primary amine or the secondary amine of the resin G containing the acrylic resin I ′ (that is, the nitrogen element and the secondary amine that form the primary amine are It is considered that the total nitrogen element concentration of the formed nitrogen element) is 1.461 at% or more and 1.629 at% or less. By rounding off the third digit after the decimal point, the concentration of the nitrogen element forming the primary amine or the secondary amine of the resin G containing the acrylic resin I ′ is considered to be 1.46 at% or more and 1.63 at% or less. .
Each of the resins F, G, and H is dissolved in MEK (manufactured by Maruzen Petrochemical Co., Ltd.) to form a solution having a solid content of 70% by mass, applied to the
The bactericidal and antibacterial properties of each of the thus obtained sample films F, G and H were evaluated by the same method as that performed for the sample films A to E. However, the method for evaluating the sterility of the sample films F, G, and H differs from the method for evaluating the sterility of the sample films A to E in the following points.
In the method for evaluating the bactericidal properties of the sample films F, G, and H, a 4 cm square PET film was used as the cover on the diluent B. The bacteria of the sample films F, G and H were washed out using 10 mL of the SCDLP medium. Therefore, the wash solution in the filter bag is the bacteria diluent B diluted 26 times. This washing solution may be referred to as a diluent B3. The SCDLP medium was prepared as follows.
-Add 38 g of SCDLP agar medium "Digo" (manufactured by Nippon Pharmaceutical Co., Ltd.) to 1 L of purified water, shake well, and heat and dissolve at 90 ° C or higher.
・ After dispensing in a container, sterilize in an autoclave at 121 ° C for 15 minutes.
-Immediately after sterilization, shake well to make the polysorbate layer uniform.
In the step of diluting the washing solution (bacteria diluent B3), phosphate buffered saline (PBS) was used. For example, 1 mL of the washing solution (bacterial diluent B3) was added to 9 mL of phosphate buffered saline (PBS) to dilute the washing solution (bacterial diluent B3) 10-fold.
For the evaluation of the antibacterial properties of the sample films F, G and H against Pseudomonas aeruginosa, the “quasi-bacterial activity value (6h)” is defined according to the antibacterial activity value. As already described, the antibacterial activity value is a logarithmic value of the number obtained by dividing the number of bacteria after culturing the PET film for 24 hours by the number of bacteria after culturing the sample film for 24 hours. According to this, the logarithmic value of the number obtained by dividing the number of bacteria after culturing the PET film for 6 hours by the number of bacteria after culturing the sample film for 6 hours is defined as “quasi-antibacterial activity value (6h)”.
9 to 11 are graphs showing the evaluation results of the bactericidal properties. 9 to 11, the horizontal axis represents the standing time (hour), and the vertical axis represents the number of bacteria (CFU / mL) in the bacterial dilution B3. 9 to 11, for ease of viewing, the plot is plotted as 0.1 when the number of bacteria is 0 (ND). Table 4 below shows the number of bacteria, the antibacterial activity value, and the quasi-antibacterial activity value (6h) after the culture. The antibacterial activity value and the quasi-bacterial activity value (6h) of the sample films F and G were calculated using the data of PET3, and the antibacterial activity value and the quasi-bacterial activity value (6h) of the sample film H were calculated. PET4 data was used.
As described above, the liquid can be sterilized by bringing the liquid into contact with the surface of the synthetic polymer membrane according to the embodiment of the present invention. Similarly, the gas can be sterilized by bringing the gas into contact with the surface of the synthetic polymer film. Microorganisms generally have a surface structure that easily adheres to the surface of an object in order to increase the probability of contact with organic matter that is a nutrient source. Therefore, when a gas or liquid containing microorganisms is brought into contact with the bactericidal surface of the synthetic polymer film according to the embodiment of the present invention, the microorganisms tend to adhere to the surface of the synthetic polymer film. It will be bactericidal.
Here, the bactericidal action of the synthetic polymer membrane according to the embodiment of the present invention has been described for Pseudomonas aeruginosa, which is a gram-negative bacterium, but it is not limited to gram-negative bacteria, and it is also bactericidal for gram-positive bacteria and other microorganisms. It is considered to have an effect. Gram-negative bacteria have one feature in that they have a cell wall that includes an outer membrane, but gram-positive bacteria and other microorganisms (including those that do not have a cell wall) also have a cell membrane, and the cell membrane is also outside of Gram-negative bacteria. Like a membrane, it is composed of a lipid bilayer. Therefore, it is considered that the interaction between the projection on the surface of the synthetic polymer membrane according to the embodiment of the present invention and the cell membrane is basically the same as the interaction with the outer membrane.
Next, as a method for producing a synthetic polymer film, a production method using no solvent was examined. When a solvent is used, equipment and time are required to remove the solvent, so that mass productivity is poor. A production method using a reactive monomer (monofunctional acrylate) instead of the solvent was studied.
For evaluation of bactericidal and antibacterial properties, Staphylococcus aureus, which is a gram-positive bacterium, was used. Since Staphylococcus aureus is infected by contact, the stamp method was employed to evaluate whether or not the synthetic polymer membrane according to the embodiment of the present invention has an effect of suppressing contact infection. In the stamp method, after culturing a bacterial solution on the surface of a synthetic polymer membrane for a certain period of time, the bacteria are copied to a medium (here, Petan check), and evaluated by the number of bacteria after culturing under a certain condition.
As a sample film, as a reference sample, a resin I having the same composition as the resin G, a resin J using a reactive diluent, and a resin K were prepared, and the
The compositions of Resin I, Resin J and Resin K are shown in Table 5 below. As the reactive diluent, acryloylmorpholine (ACMO (registered trademark) manufactured by KJ Chemicals Corporation) shown in the following [Chemical Formula 7] was used. As the resin K, the above acrylic resin V and 1,9-nonanediol diacrylate shown by the following [Chemical Formula 8] (ND-DA, manufactured by Daiichi Kogyo Seiyaku Co., Ltd., referred to as “acrylic resin VI”). And were used. The acrylic resin VI is a low-viscosity bifunctional acrylate monomer, and the resin K was prepared in the same manner as the resin J without using a solvent (MEK).
As in the case of the sample films A to H, the sample film I was dissolved in MEK to form a solution having a solid content of 70% by mass, applied to the
The exposure amount was about 1200 mJ / cm 2 (200 mW × 6 sec). For UV irradiation, irradiation was performed for 30 seconds at an output level of 45% (50 mW / cm 2 ) using a UV lamp (Fusion UV Systems: Light Hammer6 J6P3, maximum output 200 W / cm). D p is about 200nm in each sample film, D int of about 200nm, D h is about 150 nm. The size of each sample film was a square (5.1 cm square) with the length of each side being 5.1 cm.
Evaluation of bactericidal and antibacterial properties was performed according to the following procedure.
1. A bacterial solution is prepared so as to have an initial bacterial count of 1E + 04 CFU / mL order, and the bacterial solution is dropped onto each sample film at 9 points of 3 × 3 at 1-cm intervals using a pipette. The bacterial solution was dissolved in Phosphate buffered saline (PBS). For sample films I and K, the initial bacterial count was 4.3E + 04 CFU / mL, and for sample film J, the initial bacterial count was 6.3E + 03 CFU / mL.
2. The sample film to which the bacterial solution has been dropped is placed in a closed container maintained at 100% relative humidity, and left at room temperature for 24 hours.
3. The bacteria on the surface of the sample film are attached to a standard agar medium by stamping with Petan Check (registered trademark, product name: PT1025, manufactured by Eiken Chemical Co., Ltd.).
4. Bacteria attached to the standard agar medium are cultured at 37 ° C for 24 hours, and then the presence of colonies is confirmed.
As a result of the above evaluation, no colonies were observed in the sample films I and J. On the other hand, in the sample film K, the number of colonies was 21. Similar experiments were performed on E. coli, and similar results were obtained. This indicates that the synthetic polymer membrane according to the embodiment of the present invention has bactericidal and antibacterial properties against not only Gram-negative bacteria but also Gram-positive bacteria. On the other hand, the sample film K having no ethylene oxide unit and no nitrogen element has no bactericidal or antibacterial properties. Therefore, as described above, the total concentration of the nitrogen element forming the primary amine and the nitrogen element forming the secondary amine is 0.29 at% or more, and the number of moles of ethylene oxide units contained in 1 g is If it exceeds 0.0020, it is considered that a synthetic polymer film having bactericidal and antibacterial properties can be obtained even if it is produced using a reactive diluent. It may be preferable that the number of moles of ethylene oxide units contained in 1 g is 0.0070 or more.
Like the sample film J, according to the embodiment of the present invention, by using the reactive diluent, it is possible to produce a synthetic polymer membrane having a bactericidal effect on the surface without using a solvent. Since equipment and time can be omitted for removal, mass productivity can be improved.
In addition, the synthetic polymer membrane according to the embodiment of the present invention has an effect of preventing contact infection. Therefore, for example, by covering the surface of a part touched by many people with a synthetic polymer film according to the embodiment of the present invention, such as a door knob, a handrail, and a hanging leather, contact infection can be prevented or suppressed.
The synthetic polymer membrane having a bactericidal surface according to the embodiment of the present invention can be used for various uses such as sterilizing the surface, for example, suppressing or preventing contact infection. Synthetic polymer membranes having a germicidal surface according to embodiments of the present invention can be manufactured at low cost.
