この実施例では、Brunenders親株をPER.C6細胞を用い、4mMのL−グルタミンを添加したPermexcisTM培地(化学的に定義された無血清培地、例えば、Lonzaから入手可能、cat.#BE02−039Q)において、低温(≦30℃)、かつ低MOI(0.01)で34代連続継代した。得られたウイルス集団を分析し、その集団の中で生理的温度(37℃)で増殖障害を示し、低温(30℃)で野生型の増殖を示す温度感受性ウイルスクローンを同定した。約1000個のクローンをスクリーニングし、37℃で増殖障害を示す温度感受性を示した3クローン(G12P5、F9P4、およびG11P3と命名)を見出した。増殖障害を、最大力価の親ウイルスと比べて100〜1000倍の低下と定義した。3クローンは、生理的温度でまだ増殖することができたが、力価は低下した。低温(30℃)での3クローンの増殖動態は出発親株より速かった。温度感受性クローンの配列を決定し、3つの異なるクローン全体で、合計31の変異があった。各クローンは18のヌクレオチド変異を有し、そのうちのいくつかは、異なるクローンで共通し、いくつかは各クローンに特有であった。
浮遊PER.C6(sPER.C6)細胞、生産細胞株、における30および37℃でのCAVA−PVBackbone の増殖動態を他の1型PV(PV1)株と比較した。他のPV1株はBrunhilde、Brunenders、Mahoney、およびSabin1であった。Brunhildeは、次にCAVA−PVBackboneの親株となるBrunendersの親株である。Mahoneyは、神経毒性のある野生型PV株であり、一般にソークIPVの1型成分のワクチン株として使用されている。Sabin1は弱毒化経口生ポリオウイルスワクチン(OPV)用として使用されている弱毒化株である。浮遊PER.C6細胞の細胞密度は、感染時、4mMのL−グルタミンを添加したPermexcis培地において10×106個/mlであった。細胞をMOI2で感染させ、感染は1回行った(N=1)。ウイルス採集物をTCID50アッセイにより30℃で滴定し、1ml当たりの組織培養感染量50%(TCID50)である、試料の50%が感染を示す感染量(CPE)を得た。sPER.C6細胞における複製動態の折れ線グラフを図2に示す。30℃における他のPV1株との比較で、CAVA−PVBackboneは、野生型増殖動態、またはより速い増殖を示した。それどころか、驚いたことに、37℃では、CAVA−PVBackboneは、3つの異なる温度感受性クローンの変異を組み合わせたことによる相乗累積効果を示す有意な複製は示さなかった。他のPV1株は全て正常に増殖し、37℃で、sPER.C6細胞において同様の増殖挙動を示した。
成功する感染は、ウイルスと宿主細胞との複雑な相互作用であり、したがって、温度感受性は宿主細胞の因子によって影響を受ける可能性がある。したがって、CAVA−PVBackboneが生理的条件で複製する能力がないことを確認するため、様々な哺乳動物の細胞株の集団でウイルスの増殖を調べた。
インビトロの温度感受性表現型が神経弱毒化につながるか否かを調べるために、インビボのCD155遺伝子導入マウスモデルを使用した(Koike,Taya et al.1991)。遺伝子導入マウスを、PV感染に対する感受性を高めるポリオウイルス受容体(PVRまたはCD155)が発現するように、CD155遺伝子導入マウスを遺伝子学的に改変する。CD155マウスにCAVA−PV株および他の選択したPV株を感染させた。マウスの脳内(i.c.)、筋肉内(i.m.)または腹腔内(i.p.)に量を変えて感染させ、所与の試験群のマウスの50%が麻痺を引き起こすか、致死させるのに必要な感染単位数(TCID50で表される)に相当する、(麻痺または)致死量50((P)LD50)を求めた。(P)LD50が小さいほど、ウイルスの神経毒性はより大きい。表2は、インビボでの神経毒性試験の結果を示す。CAVA−PV(活性なCAVA−PVBackbone、CAVA−PVMahoney、CAVA−PVMEF−1およびCAVA−PVSaukett)では、マウスに投与されたウイルスの最大量でも、注入したどのマウスにも麻痺の兆候は見られず、したがって、(P)LD50は投与し得る最大量を超える(>)。神経毒性を示すものとして、他のPVウイルス株の(P)LD50も調べられた。BrunendersおよびBrunhildeはCAVA−PVの親株であり、一方、Mahoney、MEF−1、SaukettおよびSabinウイルスはそれぞれIPVおよびOPVのワクチン株である。Mahoneyは最も毒性が高く、Brunhilde、Saukett、Brunenders、MEF−1およびSabin株と続く。これは、これらの株の文献による神経毒性のデータと同じである。CAVA−PVは野生型のどの株よりも神経毒性が少ない。CAVA−PVは、脳内投与ルート(神経細胞を破壊し、麻痺を発症させるウイルスの能力を測定するために、ここで使用されている最も感受性の高いルート)のMahoneyの少なくとも100万倍弱毒化されている。CAVA−PVは、一部弱毒化されている株であるBrunendersの少なくとも100倍弱毒化されている。このモデルはSabinとCAVA株を十分に区別することができるだけの感受性を有さないため、CAVA株がSabinより弱毒化されていることを確認するためには、神経毒性を測定するためのより感受性の高いモデルを使用する必要があるであろう。しかしながら、Sabin株を最大用量投与したマウスの中に麻痺を発症したものがあり、一方、CAVA株ではなかった。
CAVA−PVBackbone株を、想定される生産条件下、小規模で継代し(Permexcis培地中、細胞密度10×106vc/mlのsPER.C6細胞、MOI1、30℃)、24hpiで採集した。継代は、理論的な商業生産バッチを5代超える8回行った。これらの継代後、全ゲノムの配列を決定した。ウイルスに導入された31の変異のどれも元に戻らなかった。継代後の全ゲノムは、3A遺伝子中の5206位のヌクレオチドで、アミノ酸の置換を引き起こす混合集団を示した1つのヌクレオチドを除いて出発ストックと同じであった。ポリオウイルスのRNAポリメラーゼのエラー率が大きいため、この変異はランダムであり、温度感受性(および弱毒化)表現型の転換を引き起こすことはないと思われる。その後、本発明者は、複製動態を行うことによって温度感受性表現型を確認し、8継代したCAVA−PVが30℃および37℃の両方でCAVA−PV出発ストックと似た増殖曲線を示すことを観察した(図10)。このように、CAVA−PVBackboneの温度感受性表現型、インビトロでの神経弱毒化は、生産条件下での複数回の継代後も安定である。このことは、CAVA−PVを商業利用のバッチ生産の製造方法における使用に適したものとするであろう。
従来のIPVと同じ免疫プロファイルを示すことができるCAVA−PVベースのIPVを生産するために、数例のウイルスのカプシドをコードする配列を、元のCAVA−PVBackboneカプシドをコードする配列と置き換えることによって、CAVA−PVゲノムのバックグラウンドに挿入した。このカプシドの交換はCAVA−PVに意図的に遺伝子操作した31の変異のうちの7つを除くものである。カプシドの交換はまずコンピューターで設計し、その結果として、新規のゲノムを化学的に合成してDNAを作る。プラスミドDNAが合成されたなら(Genscriptに外部委託)、これを使用して、インビトロでの転写によりウイルスRNAを作製し、その結果として、トランスフェクションにより異なる新規の組換えCAVA−PVをレスキューする。得られたワクチン株をCAVA−PVMahoney、CAVA−PVMEF−1およびCAVA−PVSaukettと名付けた。これらはそれぞれ、Mahoney、MEF−1およびSaukettのカプシドを含む。PV1型株Mahoney(配列番号2)、PV2型株MEF−1(配列番号:3)およびPV3型株Saukett(配列番号:4)の代表的なカプシドのアミノ酸配列を本明細書に示す。図12にこれらの異なるCAVA−PVワクチン株の概観図を示す。
30および37℃の生産細胞種の浮遊PER.C6(sPER.C6)細胞におけるCAVA−PVMahoney、CAVA−PVMEF−1およびCAVA−PVSaukettの増殖動態を、PV1Brunendersの増殖と比較した。感染時の浮遊PER.C6細胞の細胞密度は、4mMのL−グルタミンを添加したPermexcis培地において10×106個/mlであった。細胞をMOI1で感染させ、感染は、CAVA−PVSaukettおよびBrunendersで2回(N=2)、CAVA−PVMahoneyおよびCAVA−PVMEF−1で3回(N=3)行った。ウイルス採集物をTCID50アッセイにより30℃で滴定した。sPER.C6細胞中の複製動態の折れ線グラフを図13に示す。30℃における親Brunenders株との比較で、CAVA−PVは野生型の増殖動態を示した。エラーバーで平均値からの標準偏差を示す。それどころか、37℃では、CAVA−PVは、CAVA−PVBackboneでも観察されたように、実質的な複製は示さず、ウイルスの温度感受性がカプシド領域の外の変異に存在することを示した。Brunenders株は正常に複製し、sPER.C6細胞中、37℃で、同様の増殖挙動を示した。
IPV投与は、インビトロでのD−抗原ELISA法(Beale 1961)によって定量されるD−抗原単位(DU)に基づいて、ヨーロッパ薬局方(European Pharmacopeia)モノグラフ0214に記載されるように行われる。 野生型IPVでは、ワクチンの1回の投与量は、不活化Mahoney、MEF−1およびSaukettウイルスでそれぞれ40、8および32DUを含むことになっており、それらはワクチン接種者に防御免疫反応を誘発する用量に相当する(Grassly 2014)。活性なCAVA−PVMahoney、CAVA−PVMEF−1およびCAVA−PVSaukettウイルスのD−抗原量、およびしたがって間接的に免疫原性効力をD−抗原ELISAアッセイにより測定し、従来のIPV株、Mahoney、MEF−1およびSaukettと比較した。感染は、10×106個/mlの細胞密度の浮遊PER.C6細胞で、MOI1で行った。感染温度はCAVA−PV株で30℃、野生型株で35℃とした。これは従来のIPVの生産温度である。
CAVAワクチン株:CAVA−PVMahoney、CAVA−PVMEF−1およびCAVA−PVSaukettを精製し不活化した。精製は、想定される生産条件下(Permexcis培地中、細胞密度10×106vc/mlのsPER.C6細胞を含むローラーボトル内の2×250ml、MOI1、30℃)での感染からの浄化した粗採集物を使用し、2回の続くクロマトグラフィー工程によって行われた。陽イオン交換クロマトグラフィー(CEX)をSartobind Sカチオン膜を使用して行い、その後、抽出物をその結果としてサイズ排除クロマトグラフィー工程に使用し、さらなる精製(純化)と緩衝剤の交換を行った。不活化の前に、SEC溶出液をM199およびグリシンを用いてコンディショニングした。不活化は、0.009%ホルマリン(または3.3mMホルミアルデヒド)を添加することによって行い、37℃で13日間インキュベートした。不活化6日目および13日目に濾過した。不活化バルクを、ラットによるインビボでの免疫原性試験に使用した。メスのWistarラット(n=10)からなる4つの群に、各不活化CAVAワクチン株の1:1(ヒトの全用量)、1:2、1:4および1:16の希釈液で免疫を付与した。ヒトの全用量は、1型、2型および3型でそれぞれ40、8または32D−抗原単位であり、それは従来のIPVの用量である。ラットを21日間放置し、その後、血液を採り、血清を、Sabinウイルス(攻撃ウイルスとして)とHep2C細胞を用いるウイルス中和アッセイに使用した。図14は、不活化CAVA−PVMahoney、CAVA−PVMEF−1またはCAVA−PVSaukettによる免疫付与後に、ラットで生起された中和抗体力価を示す。3種のワクチン株全てが、ワクチン総量の希釈による用量に応じて高力価の中和抗体の産生を誘発したことから、それらは免疫原性を有することを示した。
3つの独立したクローンで生じた31の変異(実施例1、表1を参照)を分析し、PV株の集団間での変異の保存性を調べた。本発明者は、CAVA−PVで観察された温度感受性表現型の提示に大きな役割を果たしていると見られる14の変異を同定した。これらを表4に示す。この表において、1つのアスタリスクはCAVA−PVに特有で、その位置のヌクレオチドがアライメントで使用されている、Brunenders、ならびにBrunhilde、Mahoney、Sabin1、Sabin2、Sabin3、MEF−1、およびSaukettの他のPV株で保存されている変異を示している。この表において、2つのアスタリスクはCAVA−PVに特有で、その位置のヌクレオチドがアライメントで使用されている他の1型PV株、Brunenders、Brunhilde、MahoneyおよびSabin1の全てで保存されている変異を示している。CAVA−PVの4120の位置でのヌクレオチドの変異はSabin1と共通しているが、アライメントで使用されている他のPV株の全てでその位置のヌクレオチドは保存されている。
以上の実施例はいずれも、種々のCAVA−PVを生成するための出発株もしくは親株として、Brunenders株(一部弱毒化1型PV株)を使用することを記載している。本実施例では、2つの大きく異なる親株、すなわち、2型の神経毒性のある野生型MEF−1株と、3型の神経毒性のあるSabin3株をバックグラウンド(親)株として使用した。MEF−1は、通常、従来のIPV製剤の2型免疫原として使用され、一方、Sabin3はOPV製剤の成分である。
37℃でのCAVA−PV株の滴定(TCID50)によるウイルス感染の定量化は、細胞変性効果(CPE)の観察をもたらさず、したがって、滴定は30℃で行わなければならない。このことは、37℃で感染中、感染単位の上昇を示さないCAVA−PV株の複製動態曲線により裏付けられる(図2〜11、13、15〜17)。このことは、ウイルスがこの温度で複製できないことを示している。しかしながら37℃での複製の阻害をさらに理解するため、本発明者はPV感染の特性を示す代替法として、電子顕微鏡法(EM)を実施した。図18は、10×106個/mlの細胞密度のsPER.C6細胞に、Mahoney、CAVA−PVMahoneyまたはPBS(モック感染)を使用して、MOI1、30および37℃で感染させた後のEMの結果を示す。
感染時のウイルスRNA濃度を測定するため、EMに使用された試料(実施例12)を定量PCRに供した。QIAampウイルスRNA単離キットを用いて、ウイルスRNAを感染採集物から単離し、その後、RT−qPCRのためにpowerSYBR Green RNA−to−Ct 1 step Kitを使用し、3Dポリメラーゼ遺伝子に結合するプライマーを使用した。図19は、感染採集物の滴定によって観察されたvRNA濃度を示す。CAVA−1 Mahoney感染が37℃で維持されるとウイルスRNA濃度は低下するが、これは感染温度が30℃のときは当てはまらず、vRNAの増加が観察される。したがって、RNA複製に障害があり、37℃では、複製が完全に阻害される可能性があると結論付けることができる。したがって、CAVA−PVの複製阻害はRNA複製レベルで、または感染サイクルの早い時期に起こる。
どの変異がCAVA表現型に含まれるかをさらに理解するために、親Brunenders株のバックグラウンドの5’UTRに7つのCAVA変異か、または非構造タンパク質に17の変異を起こした新規の中間体ウイルスを構築した(配列番号1、これらの2つの領域のCAVA変異は表4を参照されたい)。実施例6の−CAVA−PV株は、カプシドにおけるCAVA変異がCAVA表現型に影響を及ぼさずに除き得ることを既に示した。したがって、ここでは5’UTR領域および非構造タンパク質領域の変異のみを調べた。中間体ウイルスの30および37℃における複製動態を試験し、親Brunenders株、実施例1のCAVA−PVBackboneおよび実施例6のCAVA−PVMahoneyと比較した。

b)前記細胞培養中の前記細胞を培養して前記ポリオウイルスを増殖させる工程と
c)前記細胞または前記細胞培養から前記ポリオウイルスを単離して、前記ポリオウイルスを含む製剤を得る工程と
を含む方法。

IPVを調製する方法であって、[13]に記載の方法、および医薬組成物に前記不活化ポリオウイルスを配合する工程を含む方法。

30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない組換え弱毒化ポリオウイルス株を得る方法であって、
a)親ポリオウイルス株に対し30℃以下で、37℃で増殖障害を示すウイルスを生産するのに十分な継代を行う工程と、
b)37℃で増殖障害を示す2つ以上の異なる温度感受性クローンを単離する工程と、
c)前記温度感受性クローンのゲノムの配列を解析する工程と、
d)前記温度感受性クローンの配列と前記親ポリオウイルス株の配列とを比較して、前記温度感受性クローンのゲノムの配列における変異を同定する工程と、
e)2つ以上の異なる温度感受性クローンの変異をポリオウイルス株のゲノムに組み合わせて前記組換え弱毒化ポリオウイルス株を合成する工程と、
f)30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない前記組換え弱毒化ポリオウイルス株をレスキューする工程と
を含む方法。

g)前記レスキューされた組換え弱毒化ポリオウイルス株のカプシドをコードする配列を、異なるポリオウイルス株のカプシドをコードする配列と置換する工程
をさらに含む[15]に記載の方法。

g)前記レスキューされた組換え弱毒化ポリオウイルス株のカプシドをコードする配列を、Mahoney、MEF−1またはSaukett株のカプシドをコードする配列と置換する工程
をさらに含む[15]または[16]に記載の方法。

[15]〜[17]のいずれか一項に記載の方法によって得られる組換え弱毒化ポリオウイルス。

- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない組換えポリオウイルス株であって、Mahoney、MEF−1またはSaukett株のカプシドを含み、前記組換えポリオウイルス株のゲノムは、Brunenders株のゲノム(配列番号1)のゲノムと比較して、次の変異:5’UTRの133(AからG)、142(UからC)、163(AからG)、597(CからU)、および609(GからA)、ならびに非構造タンパク質の3486(GからA)、3852(AからU)、4120(UからC)、4428(AからG)、4563(AからU)、5436(GからA)、6210(AからG)、6848(GからA)、および7102(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化1型ポリオウイルス株であって、そのゲノムは、Mahoney株(配列番号6)のゲノムと比較して、次の変異:5’UTRの131(AからG)、140(UからC)、161(AからG)、593(CからU)、および605(GからA);ならびに非構造タンパク質の3482(GからA)、3848(AからU)、4116(UからC)、4424(AからG)、4559(AからU)、5432(GからA)、6206(AからG)、6844(GからA)および7098(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化2型ポリオウイルス株であって、そのゲノムは、MEF−1株(配列番号5)のゲノムと比較して、次の変異:5’UTRの134(AからG)、143(UからC)、164(AからG)、598(CからU)、および610(GからA);ならびに非構造タンパク質の3847(AからU)、4115(UからC)、4423(AからG)、4558(AからU)、5431(GからA)、6205(AからG)、6843(GからA)および7097(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化3型ポリオウイルス株であって、そのゲノムは、Saukett株(配列番号7)のゲノムと比較して、次の変異:5’UTRの133(AからG)、142(UからC)、163(AからG)、596(CからU)、および608(GからA);ならびに非構造タンパク質の3839(AからU)、4107(UからC)、4415(AからG)、4550(AからU)、5423(GからA)、6197(AからG)、6835(GからA)および7089(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化1型ポリオウイルス株であって、そのゲノムは、Sabin1株(配列番号9)のゲノムと比較して、次の変異:5’UTRの131(AからG)、140(UからC)、161(AからG)、593(CからU)、および605(GからA);ならびに非構造タンパク質の3482(GからA)、3848(AからU)、4424(AからG)、4559(AからU)、5432(GからA)、6206(AからG)、6844(GからA)および7098(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化2型ポリオウイルス株であって、そのゲノムは、Sabin2株(配列番号10)のゲノムと比較して、次の変異:5’UTRの131(AからG)、140(UからC)、161(AからG)、594(CからU)、および606(GからA);ならびに非構造タンパク質の3481(GからA)、3847(AからU)、4115(UからC)、4423(AからG)、4558(AからU)、5431(GからA)、6205(AからG)、6843(GからA)および7097(UからC)を含む組換えポリオウイルス株。
- 30℃の細胞培養で増殖することができ、かつ37℃の細胞培養では実質的に増殖することができない、温度感受性の組換え弱毒化3型ポリオウイルス株であって、そのゲノムは、Sabin3株(配列番号8)のゲノムと比較して、次の変異:5’UTRの133(AからG)、142(UからC)、596(CからU)、および608(GからA);ならびに非構造タンパク質の3839(AからU)、4107(UからC)、4415(AからG)、4550(AからU)、5423(GからA)、6197(AからG)、6835(GからA)および7089(UからC)を含む組換えポリオウイルス株。
- 請求項1〜7のいずれか一項に記載のポリオウイルス株を含む組成物。
- 第1、第2および第3の組換えポリオウイルス株を含む組成物であって、前記第1、第2および第3の各組換えポリオウイルス株は、請求項1〜7のいずれか一項に記載の組換えポリオウイルス株から選択され、前記第1の組換えポリオウイルス株はMahoney株のカプシドを含み、前記第2の組換えポリオウイルス株はMEF−1株のカプシドを含み、かつ前記第3の組換えポリオウイルス株はSaukett株のカプシドを含む組成物。
- 薬学的に許容される担体または賦形剤をさらに含む請求項8または9に記載の組成物。
- 請求項8〜10のいずれか一項に記載の組成物を含む不活化ポリオウイルスワクチン(IPV)組成物であって、前記組成物中の前記ポリオウイルスが不活化されている組成物。
- 請求項11に記載のIPV 組成物を含み、さらにジフテリア、破傷風、百日咳、ヘモフィルスインフルエンザ菌(Heamophilus influenzae)b型(Hib)、またはB型肝炎ウイルス(HBV)の1種以上の抗原を含む混合ワクチン組成物。

c)前記細胞または前記細胞培養から前記ポリオウイルスを単離して、前記ポリオウイルスを含む製剤を得る工程と
を含む方法。 - 前記ポリオウイルスを不活化する工程をさらに含む請求項15に記載の方法。
- IPVを調製する方法であって、請求項16に記載の方法、および医薬組成物に前記不活化ポリオウイルスを配合する工程を含む方法。
