トリエチレングリコール(Sigma−Aldrich、5620mg、37.4mmol、1当量)を、無水THF(50mL)に溶解し、得られた溶液を乾燥条件で0℃に冷却した。水素化ナトリウム(988mg、1.1当量)をゆっくりと添加し、次いでプロパルギルブロミド(トルエン中80重量%、4360μL、1.1当量)を滴下して添加した。反応物を不活性雰囲気下、室温で12時間、撹拌した。
THFを減圧下で除去し、残留物を塩化メチレン(ジクロロメタン、DCM)に取り、塩水で数回洗浄した。得られた有機層を、硫酸マグネシウム(MgSO4)で脱水し、濾過し、減圧下で濃縮し、真空下で乾燥した。粗生成物をシリカカラムクロマトグラフィー(溶離剤:シクロヘキサン(cHex)/酢酸エチル(AcOEt):8/2)で精製し、3.31gの黄色油を回収した(収率47%)。

製造物1A(2000mg、10.6mmol、1当量)、フタルイミド(2345mg、1.5当量)およびトリフェニルホスフィン(4179mg、1.5当量)を、乾燥条件で、無水THF(50mL)に溶解した。ジイソプロピルアゾジカルボキシレート(DIAD)(3.14mL、1.5当量)をゆっくり添加し、反応物を不活性雰囲気下、室温で48時間撹拌した。
THFを減圧下で除去し、残留物をDCM中に取り、塩水で数回洗浄した。得られた有機層を、MgSO4で脱水し、濾過し、減圧下で濃縮し、真空下で乾燥した。粗生成物をシリカカラムクロマトグラフィー(溶離剤:cHex/AcOEt:8/2)で精製し、3.37gの黄色油を回収した(収率60%)。

アルキントリエチレングリコール(製造物1A、4g、212mmol、1当量)のDCM(60mL)中溶液に、不活性雰囲気下で、触媒量のDMAP、塩化メタンスルホニル(3.3mL、2当量)およびトリエチルアミン(5.9mL、2当量)を滴下して添加した。反応混合物を室温で4時間、撹拌した。
その溶液を塩水で(50mLで3回)洗浄し、次いで水相をDCM(50mL)で抽出し、合わせた有機層をMgSO4で脱水し、濾過し、減圧下で濃縮した。
メシル−アルキントリエチレングリコール(製造物1C、5.1g、19.2mmol、1当量)のDMF(100mL)中溶液に、カリウムフタレート(7.87g、2.2当量)および触媒量のヨウ化ナトリウム(1当量未満、例えば、スパチュラ先1回分)を添加した。溶液を80℃で一晩撹拌し、溶媒を減圧下で除去した。
得られた残留物を、シリカカラムクロマトグラフィー(溶離剤:cHex/AcOEt:2/8から4/6)で精製した。5.7gの黄色油が回収された(収率94%)。

製造物1B(2034mg、6.4mmol、1当量)を、エタノール(EtOH)(200mL)に溶解し、ヒドラジン水和物(3.1mL、10当量)を添加した。反応混合物を、還流条件下で一晩撹拌した。
反応物を室温に冷却し、8mLの濃塩酸を反応物に添加した(pH約2〜3)。沈殿物を濾過によって除去し、pHを、NaOH(2M)を使用して、10より上に上げた。水相をDCMで3回抽出した。得られた有機層をMgSO4で脱水し、濾過し、減圧下で濃縮し、真空下で乾燥した。911mgの黄色油が回収された(収率76%)。

製造物1E(200mg、1.07mmol、1当量)のDCM(20mL)中溶液に、不活性雰囲気下で、PyBOP(780mg、1.4当量)、p−メトキシ安息香酸(229mg、1.4当量)およびDIEA(260μL、1.4当量)の下で添加した。反応物を室温で4時間、撹拌した。
その溶液を塩水(20mLを3回)で洗浄し、次いで水相をDCM(20mL)で抽出し、合わせた有機層を硫酸マグネシウムで脱水し、濾過し、減圧下で濃縮した。粗生成物をシリカカラムクロマトグラフィー(溶離剤:cHex/AcOEt:5/5から7/3へ)で精製し、300mgの黄色油を回収した(収率90%)。

製造物1E(319mg、1.70mmol、1当量)のDMF(70mL)中溶液に、不活性雰囲気下、EDC(393mg、1.2当量)、NHS(236mg、1.2当量)および2、3滴のトリエチルアミン(Et3N)を添加した。反応物を50℃まで加熱し、葉酸(Sigma−Aldrich、754mg、1当量)を反応混合物に添加した。次いで、反応物を、50℃で一晩撹拌した。
その溶液は減圧下で濃縮した。残留物を、DCMおよびアセトンに沈殿させて、濾過し、真空下で乾燥した。
得られた生成物を、逆相カラム(C18、Kromasil 10μm、4.6×250mm)を用いて、例えば、酢酸アンモニウム緩衝液(pH5に調整)の5から95%のアセトニトリル勾配で20分、次に95%アセトニトリルで5分間、分析した。
生成物を、溶離液の20%DMFに溶解させた。精製は、100mmのカラム(1.5kgのフェーズ)に入れられたKromasil C18 10μmを用いて、85%酢酸アンモニウム緩衝液(pH5)20mMと15%アセトニトリルとで溶離して実施した。200mgの未精製生成物を、DMF注入ピークと同時に溶離した。
最終的な精製は、200mgの未精製の生成物を溶解させて、逆相カラムWaters XBridge C18(30×100mm)、5μmを備える分取HPLCを使用して行った。生成物を、最初にDMSO(5mL)と5mLの緩衝液(炭酸アンモニウム10mMを28%アンモニア水溶液でpH9.3に調整した液)に溶解させた。1mlの注入を10回、95:5(緩衝液(炭酸アンモニウム)/アセトニトリル)から5:95の勾配を使用して、12分間、30mL/分で行った。

FP−547−NHS(Interchim、2.5mg、2.55μmol、1当量)のDMSO(356μL)中溶液を、EDC(0.49mg、1当量)、NHS(0.29、1当量)、TEA(0.35μL、1当量)および製造物1E(1.07mg、2.2当量)を含有するDMSO(92μL)溶液に添加した。その溶液を暗所下、室温で12時間撹拌した。
反応物を減圧下で濃縮し、DCMに溶解し、塩水で抽出した。ピンク色の油が得られた。
得られたスペクトルは、市販のフルオロフォア化合物によって得られるスペクトルと類似していた。
PEG2500ベンジル(Polymer Source、Mn=2572g・mol−1、2.43g、0.94mmol、1当量)、DMAP(58mg、0.5当量)およびTEA(747μL、5.6当量)のDCM(65mL)中溶液を、0℃に冷却し、20分間かけてMsCl(326μL、4.4当量)をゆっくりと添加した。反応物を室温で一晩撹拌し、減圧下で濃縮した。残留物をDMF(20mL)へ溶解し、アジ化ナトリウム(330mg、5.3当量)を溶液に添加した。反応物を、50℃で24時間、撹拌した。
反応物を減圧下で濃縮し、残留物をDCM(50mL)に溶解させ、塩水(50mLで3回)洗浄した。有機層を、MgSO4上で脱水し、濾過し、減圧下で濃縮してDCMを最小体積とした。後者をジエチルエーテル中に沈殿させた。2.16gの白色粉末が得られた。

製造物3A(2.16g、0.83mmol、1当量)を濃HCl(20mL)に溶解させ、室温で2日間、撹拌した。
溶液を、濃NaOH溶液でpH1に塩基性化した。水相をDCMで抽出し(50mLで4回)、合わせた有機層を硫酸マグネシウムで脱水し、濾過し、減圧下で濃縮してDCMの体積を最小化した。後者をジエチルエーテルに沈殿させた。1.89gの白色粉末が得られた。

Nicolasら(Macromol 2008、41、8418)の方法に従って製造したPEG2100−ベンジル(Mn=2176g・mol−1,500mg、0.23mmol、1当量)とNBS(50mg、1.2当量)との混合物に、トリフェニルホスフィン(PPh3)(75mg、1.2当量)のDCM(50mL)中***液を添加した。次いで反応物を一晩室温で撹拌した。
反応物を減圧下で25mLへ濃縮し、ヘキサン(100mL)で希釈した。沈殿物(トリフェニルホスフィンオキシド)を濾過によって除去し、重合体をジエチルエーテル(200mL)で2回沈殿させた。

2つの異なる経路を使用して、式(I)の化合物を合成した。最初の化合物(4A)は一工程経路を用い、第2の化合物(4B)は二工程経路を用いた。
アジド−ポリ(エチレングリコール(3B、Mn=2507g・mol−1、293mg、0.12mmol)およびD,L−ラクチド(7,01g、48.62mmol)の混合物に、乾燥条件で、Sn(Oct)2(18.7mg、46.1μmol)の無水トルエン(11.2mL)中溶液を加えた。反応混合物を、アルゴンで20分間バブリングすることによりガス抜きし、次に不活性雰囲気下、予備加熱油浴中120℃で90分間撹拌した。反応はおよそ55%転換して停止した。
トルエンを減圧下で除去し、得られた生成物を最小量のDCMに溶解し、さらにジエチルエーテルに沈殿させた。次いで沈殿物を最小量のTHFに溶解し、さらに水に沈殿させ、次いで一晩凍結乾燥させて白色粉末を得た。

ブロモ−ポリ(エチレングリコール(3D、Mn=2149g・mol−1、49mg、22.8μmol)およびD,L−ラクチド(0.72g、4.97mmol)の混合物に、乾燥条件で、Sn(Oct)2(1mg、2.5μmol)を添加した。反応混合物を、アルゴンで20分間ガス抜きし、次いで不活性雰囲気下、予備加熱油浴中120℃で20時間、バルク条件で撹拌した。反応は、完全に転換して停止した。
得られた生成物を、最小量のDCMに溶解し、さらにジエチルエーテルに沈殿させた。その後、沈殿物を最小量のTHFに溶解し、さらに水に沈殿させ、次いで一晩凍結乾燥させて白色粉末を得た。
製造物4B(1)(200mg、5.9μmol、1当量)のDMF(10mL)中溶液に、不活性条件下で、アジ化ナトリウム(20mg、54当量)を添加した。次いで、反応物を、不活性雰囲気下で、3日間50℃で撹拌した。

製造4Aに記載した方法に従って製造した式(I)PLA−PEG−N3の化合物(200mg、個々の場合のバッチに応じたモル量、6.7μmol、1当量)と、式(XI)のアルキン誘導体(0.12mmol、18当量)との無水DMF(2.5mL)中先にガス抜きされた溶液に、シリンジを用いて、CuBr(5.8mg、6.1当量)およびPMDETA(16.8μL、17.8当量)の無水DMF(400μL)中ガス抜きされた溶液を添加した。反応混合物を不活性雰囲気下、50℃で15時間撹拌した。同じ方法をすべての製造に用いた。
溶液を減圧下で濃縮し、残留物を最小量のTHFに溶解させ、さらに水に沈殿させた。沈殿物を凍結乾燥させ、最小量のTHFに再び溶解させ、さらに水に沈殿させた。沈殿物を凍結乾燥させて白色粉末を得た。
アルキン誘導体が水に不溶の場合、最初の凍結凍乾燥沈殿物を最小量のDCMに溶解させて、さらにジエチエルエーテルに沈殿させる中間工程を加えてもよい。

トルエンを減圧下で除去し、得られた生成物を最小量のDCMに溶解し、さらにジエチルエーテルに沈殿させた。その後、沈殿物を最小量のTHFに溶解し、さらに水に沈殿させ、次いで一晩凍結乾燥させて白色粉末を得た。

ナノ粒子形成
ナノ粒子を、下記一般的プロトコルに従って、下記表で特定された構成成分および量を使用して製造した。
1.共重合体あるいは共重合体の混合物を、有機溶媒(最終水性重合体濃度は1から40mg/mLの間で変動した)に溶解させた。
2.有機相を、安定剤(濃度は0.1から1w/v%の間で変動した)を含有する水相(水相/有機相体積比は2.5から5の間で変動した)と混合した。乳化およびサイズリダクション技術を使用したナノ粒子の製造については、混合物を、ボルテックスシェーカーを使用して1分間激しくかき混ぜ、エマルジョンを得た。エマルジョンを超音波処理した(プローブを使用し、時間は1から10分で変動した)。
3.有機相を、蒸発(減圧下または気流下)によって除去した。
4.ナノ粒子を、30000gで30分間、超遠心分離した。
5.ナノ粒子を、水性媒体中で再懸濁した。
6.ナノ粒子を、1μmのガラス濾過盤(Acrodisc)にかけて濾過した。
7.ナノ粒子を使用時まで4℃で保存した。
1.共重合体または共重合体混合物(全質量:30mg)をAcOEt(1.2mL)に溶解させた。
2.有機相を、1%のPluronic F68を含有する3.3mLの水相に添加した。
3.混合物を、ボルテックスシェーカーで1分間、激しくかき混ぜた。
4.エマルジョンを、(プローブを使用して)3分間超音波処理した。
5.有機相を、ロータリーエバポレーターを使用して、減圧下で除去した。
6.ナノ粒子を、30000gで30分間超遠心分離した。
7.ナノ粒子は、pH7.4のリン酸緩衝生理食塩水(PBS)3mL中で再懸濁した。
8.ナノ粒子を、1μmのガラス濾過盤(Acrodisc)にかけて濾過した。
9.ナノ粒子を使用時まで4℃で保存した。
− DCM/PVAは、230nmの平均径範囲を有する物理的に安定したナノ粒子をもたらした。
− EtOAc/NaChは、結果として160nmの平均径範囲を有する物理的に安定したナノ粒子になった。
− EtOAc/Pluronic(登録商標)は、110nmの平均径範囲を有する物理的に安定したナノ粒子をもたらした。
− アセトンの使用により、30nmの平均径範囲を有するミセルが得られた。

PLA−PEG−葉酸ナノ粒子を使用した表面プラズモン共鳴(SPR)
シリーズSセンサチップCM5(GEヘルスケア)の調製
このセンサチップは、チップの金製表面が共有結合的に結合されたカルボキシメチル化デキストランのマトリックスで被覆されており、4つのチャネルで構成されている。
タンパク質の固定化に使用したプロトコルは、Johnsonにより記載されたプロトコルである(Johnsonら、Anal. Biochem.、1991、198、268〜277頁)。簡単に述べると、pH7.4のPBSで機器を平衡化した後に、以下のサンプルを自動的におよび連続的にBIAcore T100に注入した:(i)カルボキシル化デキストランを活性化するためのNHS/EDCの混合溶液(1:1、v/v)を420秒、(ii)125μg/mLの濃度で酢酸緩衝液(pH5.0)に溶解させたFBPを420秒、(iii)センサチップ上のNHSエステル残基を不活性化するためのエタノールアミンを420秒。各工程は、PBS洗浄で区切った。固定化プロトコル(10μL/分の流量で行った)により、1つのチャネル当たり約6.8ナノグラム/mm2のFBPが結合した。
第1のフローチャネル(Fc1)は、エタノールアミンによってのみ遮蔽されており、参照チャネルとして使用することができ、デキストランがナノ粒子の吸着に役割を果たしているかどうかをチェックした。
固定されたFBPが正しいコンフォメーションにあるかを調べるために、ポリクローナル抗体抗FBP(IgG抗FBP、Thermo Scientific)を使用した。免疫グロブリンG(IgG)を、50μg/mLで120秒間、30μL/分で注入した。
特異的シグナルは、非特異的シグナル(参照チャネルFc1上のIgG)より4.5倍大きく、全体のシグナルのおよそ22%を表す。
グリシンとエチレングリコールの使用から、表面が再生したと考えられた。実際、後でIgG抗FBPの同様の特異的シグナルを得ることができた。
ナノ粒子懸濁液を、新たに用意したタンパク質被覆センサチップチャネルでさらに試験した。
固定化FBP上のPLA−PEG−葉酸ナノ粒子の吸着についての表面プラズモン共鳴分析を、対照として抱合されていないPLA−PEGナノ粒子を使用して行った。これらの実験は、5μL/分の流速で、接触時間500秒で実行した。
ナノ粒子の表面の葉酸濃度を変えるために、様々なナノ粒子懸濁液を、先に記載したプロトコル(安定剤としてpluronic(登録商標)を含有)を使用して製造した。そのようにするために、ナノ粒子をPLA−PEG−OMe(Mn(NMR)=34820g・mol−1)およびPLA−PEG−葉酸(Mn(NMR)=32880g・mol−1)の混合物から製造した。
すべてのナノ粒子懸濁液を、新たに用意したタンパク質被覆センサチップチャネルでさらに試験した。
各サンプルのSPRセンサグラムから得られたシグナルの最終値を、ナノ粒子上の葉酸の濃度に対してグラフ上でプロットした(図4)。したがって、このグラフは、ナノ粒子表面の葉酸の濃度に対する特異的シグナルの増大を示している。12%で最大に到達し、この場合の特異的シグナルは全体のシグナルのおよそ70%を表す。12%を超えると、特異的シグナルは減少する。これは、異なる葉酸部分の間がいくらか重なり合い、良好な相互作用が妨げられるためであり得る。非特異的シグナルは、表面に葉酸を有しないナノ粒子に対する値によって示される。
表の最初の7行に示されるように、いずれのナノ粒子を参照チャネルに注入しても、シグナルは得られない。したがって、もとのコーティング表面(デキストラン)はナノ粒子のどんな吸着も誘導しない。
非特異性は間違いなくFBPを有するナノ粒子のPEG被覆によるものである。
細胞毒性
実施例4a:PLA−PEG−葉酸ナノ粒子の細胞毒性
これらのナノ粒子を試験するために、KB−3−1細胞株(ヒト子宮頸癌、DSMZ(German collection of microorganisms and cell cultures、カタログ番号:ACC158)を、葉酸受容体の発現に使用した。
この細胞株は、単層中で成長する接着細胞株であり、細胞を培養して葉酸受容体の過剰発現を誘導させた。葉酸受容体を過剰発現させるために使用した媒体は、これにL−グルタミン(0.584g/Lで200mM、BioWhittaker、Lonza)および炭酸水素ナトリウム(3.7g/L、Sigma−Aldrich))を添加し、5%CO2湿気雰囲気下37℃で、1%のペニシリン/ストレプトマイシン(Lonza)と10%のウシ胎児血清(FBS、Lonza)を補充したDMEM2429(葉酸を含まない媒体)である。
葉酸ナノ粒子と対照ナノ粒子(葉酸なし)を有するために、2つの異なるナノ粒子懸濁液を、先に記載したプロトコル(安定剤としてpluronic(登録商標)を含有)を使用して製造した。
そのようにするために、ナノ粒子を、PLA−PEG−OMe(Mn(NMR)=34820g・mol−1、70重量%または100重量%)、およびPLA−PEG−葉酸(Mn(NMR)=32880g・mol−1、それぞれ30重量%または0重量%)の混合物から製造した。
96ウェルプレートにおいて、50μLの培地(上記のもの)で希釈した細胞をウェルあたり5×102個で置いた。細胞培養器で24時間後、異なる共重合体濃度(0.5、0.1、0.05、0.01mg/mL)のナノ粒子含有PBSバッファー50μLを添加した。その後、プレートを、細胞培養器(5%CO2、37℃)に48時間、静置した。その後、20μLの(3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホ−フェニル)−2H−テトラゾリウム(MTS、CellTiter96(登録商標)AQueous、非放射性細胞増殖分析(Promega)に含まれたテトラゾリウム化合物)を添加し、プレートを細胞培養器で3時間インキュベートした後、マイクロプレートリーダー(Labsystem Multiscan MS、Type 352)を用いて492nmで分析した。
データは、5×102細胞のみを含有させた50μLの培地および50μLのPBS緩衝液に含むウェルと比較し、20μLのMTSで明らかにした。すべてのデータから、50μLの培地および50μLのPBS緩衝液中の関連する濃度でのナノ粒子からなるバックグラウンドを除去し、20μLのMTSで明らかにした。実験は3連で行った。
プロットは、細胞およびMTSのみを含有するウェルを100%として生細胞の割合(%)の関数として表現した。
CellTiter96(登録商標)AQueous One Solution Cell Proliferation Assay (Promega)を用いて行った実験を図5aに示す。
ナノ粒子は、高濃度であっても、葉酸の有無にかかわらず、葉酸受容体を過剰発現するKB−3−1細胞に何ら細胞毒性を誘導しないことが観察された。
これらのナノ粒子を試験するために、PC−3細胞株(ヒト前立腺腺癌、ATCC(番号:CRL−1435)を使用した。その理由は、アニスアミド部分が結合する可能性のあるシグマ受容体をこの細胞が発現するためである。
この接着細胞株を、1%のペニシリン/ストレプトマイシン(Lonza)と10%のウシ胎児血清(FBS、Lonza)を補充したRPMI 1640(Fisher Scientific)中で、5%CO2湿気雰囲気下37℃で培養した。この細胞株はシグマ受容体を過剰発現する。85〜90%コンフルエンシーの培養物をすべての実験で使用した。細胞をトリプシン処理(トリプシン−EDTA、Invitrogen、Gibco)し、計数し、96ウェルプレートに継代培養し、生存試験を行った。細胞を実験前に24時間、接着させた。
蛍光アニスアミドナノ粒子と、対照としての蛍光ナノ粒子(アニスアミドなし)とを得るために、2つの異なるナノ粒子懸濁液を、先に記載したプロトコル(安定剤としてpluronic(登録商標)含有)を使用して製造した。
そのようにするために、ナノ粒子を、PLA−PEG−FP547(Mn(NMR)=34820g・mol−1;両方の製造物で10重量%)、PLA−PEG−OMe(Mn(NMR)=34820g・mol−1;45重量%または90重量%)およびPLA−PEG−アニスアミド(Mn(NMR)=37060g・mol−1;それぞれ45重量%または0重量%)の混合物から製造した。
96ウェルプレートにおいて、50μLの培地(上記のもの)で希釈した細胞をウェルあたり5×103個で播種した。細胞培養器中で24時間後、異なる共重合体濃度(5、2.5、0.5、0.05mg/mL)でナノ粒子含有PBSバッファー50μLを添加した。その後、プレートを、細胞培養器(5%CO2、37℃)に48時間、静置させた。その後、20μLの(3−(4,5−ジメチルチアゾール−2−イル)−5−(3−カルボキシメトキシフェニル)−2−(4−スルホ−フェニル)−2H−テトラゾリウム(MTS、CellTiter96(登録商標)AQueous非放射性細胞増殖分析(Promega)に含まれたテトラゾリウム化合物)を添加し、プレートを細胞培養器で3時間インキュベートした後、マイクロプレートリーダー(Labsystem Multiscan MS、 Type 352)を用いて492nmで分析した。
データは、5×103細胞のみを含有する50μLの培地と50μLのPBS緩衝液を含むウェルと比較し、20μLのMTSで明らかにした。すべてのデータから、50μLの培地および50μLのPBS緩衝液中の関連する濃度でのナノ粒子からなるバックグラウンドを除去し、20μLのMTSで明らかにした。実験は3連で行った。

蛍光ナノ粒子は、最大少なくとも5mg/mlの高濃度であっても、アニスアミドの有無にかかわらず、シグマ受容体を発現しているPC−3細胞に何ら細胞毒性を誘導しないことが観察された。
PLA−PEG葉酸ナノ粒子の細胞透過分析
これらのナノ粒子を試験するために、KB−3−1細胞株(ヒト子宮頸癌、DSMZ(German collection of microorganisms and cell cultures、カタログ番号 ACC 158)を、葉酸受容体の発現に使用した。
この細胞株は、単層中で成長する接着細胞株であり、細胞を培養して葉酸受容体の過剰発現を誘導させた。葉酸受容体を過剰発現させるために使用した媒体は、これにL−グルタミン(0.584g/Lで200mM、BioWhittaker、Lonza)および炭酸水素ナトリウム(3.7g/L、Sigma−Aldrich)を添加し、5%CO2湿気雰囲気下37℃で、1%のペニシリン/ストレプトマイシン(Lonza)と10%のウシ胎仔血清(FBS、Lonza)を補充したDMEM2429(葉酸を含まない媒体)である。
蛍光葉酸ナノ粒子と対照としての蛍光ナノ粒子(葉酸なし)を有するために、2つの異なるナノ粒子懸濁液の3つのバッチを、先に記載したプロトコル(安定剤としてpluronic(登録商標)を使用)を用いて製造した。そのようにするために、ナノ粒子を、PLA−PEG−FP547(Mn(NMR)=34820g・mol−1)、PLA−PEG−OMe(Mn(NMR)=34820g・mol−1)およびPLA−PEG葉酸(Mn(NMR)=32880g・mol−1)の混合物から製造した。
最初のバッチ(S1’、S2’)を作製してから23日後に第2のバッチ(S3’、S4’)を作製し、34日後に第3のバッチ(S5’、S6’)を作製した。
葉酸受容体を過剰発現させるための媒体(上述の通り)で培養したKB−3−1細胞株を、24ウェルプレートで、ほぼコンフルエンス(約300000細胞/ウェル)になるまで成長させた。次いで、培地を除去し、同じ培地に希釈した1mLのナノ粒子を、最終共重合体濃度約60μg/mLで、細胞と様々な時間、インキュベートした(10分間から24時間)。その後、培地を除去し、各ウェルをPBSバッファー(1mL)で2回洗浄し、細胞をトリプシン(200μLで3分間)で分離した。次いで、トリプシンを培地(800μL)で中和し、細胞を遠心分離した(1000gで5分間)。上清を除去し、細胞をPBS(1mL)に再懸濁し、遠心分離(1000gで5分間)し、最後にはパラホルムアルデヒド(200μL、PBS中1%)で回収した。
FACS実験から得られた結果を、図6aおよび6bに示す。葉酸がナノ粒子の表面に存在する場合(S3’)、後者は、葉酸が存在しないナノ粒子(S4’)よりも115倍、内在化されていることが分かった。6から10時間の間のインキュベートでプラトーに到達することも観察された。
同じ結果が、1か月後、10日後または1日後に得られ得ることも観察された。したがって、葉酸ナノ粒子は時間内に安定化されており、葉酸部分がナノ粒子表面に残ることも結論付けることができる。
PLA−PEG−アニスアミドナノ粒子中へのドセタキセル(DTX)のカプセル化
ドセタキセルをカプセル化するために、ナノ粒子の製造のために先に言及したものと同じプロトコルを使用した。その方法の間にトリチウム化されたドセタキセルを使用して、薬物充填率と捕捉効率を評価した。
上清およびNP溶液を、Beckmanベータカウンタを用いて計数し、4.3%の薬物充填率と46%の捕捉効率の計算結果が得られた。
予め形成されたPLA−PEG−N3ナノ粒子へクリック化学を実施するための水性条件でのHuisgen反応のための典型的手順
ナノ粒子の懸濁液(先に記載されたプロトコルに従って製造したPLA−PEG−N3(20%w/w)とPLA−PEG−OMe(80%w/w)とで作製し、ポリ(ビニルアルコール)(PVA)(1%w/v)で安定化させたもの)に、PEGアルキン(アジド基と比較して2当量)、CuSO4・5H2Oおよびアスコルビン酸ナトリウム(過剰量)を添加した。
反応混合物を、24時間撹拌した。
最後に、その懸濁液を、20000Daカットオフ(SpectrumLabs)のセルロース膜を使用して水に対して透析した。
NP懸濁液のサイズはクリック方法の間、安定している:
Z−平均=198.5nmおよびPdI=0.09
同様の方法で、他のリガンド(例えば、蛍光性リガンド...)を、上記に記載された方法に従ってナノ粒子に結合させることができる。
Claims (31)
- 式(A)の化合物:
PLAは、下記式:
(3)は、PEG部分への結合のアタッチメントであり;および
mは、構成単位の数で、1から500の間である)
のポリ乳酸残基を表し;
PEGは、下記式:
(4)は、−PLAへの結合のアタッチメントであり;
(5)は、
nは、構成単位の数で、1から300の間である)
のポリエチレングリコール残基を表し;
リンカーPEG’は、下記式:
n’は、構成単位の数で、1から10の間であり
(1)は、−(CH2)−トリアゾール基への結合のアタッチメントであり;
(2)は、リガンドへの結合のアタッチメントである)
のポリエチレングリコール残基を表し;および
リガンドは、機能性リガンドの残基を表す)。 - リガンドは、ホーミングデバイス、診断剤、イメージング剤、刺激反応剤、ドッキング剤、細胞透過剤、解毒剤および薬物の残基から選択される、請求項1に記載の式(A)の化合物。
- ホーミングデバイスは、膜認識リガンドである、請求項2に記載の式(A)の化合物。
- リガンドは、下記:
エストロゲン受容体アンタゴニスト、アンドロゲン受容体アンタゴニスト、葉酸、アニスアミド、相当する表面抗原を認識可能な抗体、腫瘍血管新生内皮に過剰発現したαvβ3インテグリンに結合するRGD配列、ヒアルロン酸、トランスフェリン、ペプチド標的遺伝子ベクター、アプタマー、もしくは腫瘍壊死因子から選択される膜認識リガンド、
酸化鉄、ガドリニウム複合体、インドシアニングリーン、近赤外蛍光プローブ、もしくはポジトロン放射体から選択される診断/イメージング剤、
酸化鉄ナノ粒子、金ナノ粒子、もしくは任意の放射活性物質から選択される刺激反応物質、
オリゴペプチドから選択されるドッキング剤、
転写トランス活性化因子(TAT)配列、ペネトラチン、ポリアルギニン配列、もしくはVP22タンパク質配列から選択される細胞透過剤、
コバラミン、コビナミド、ロダネーゼ酵素、有機リン加水分解酵素、ナロキソン、アトロピン、もしくは特定の毒素を認識する抗体/抗体断片から選択される解毒剤;または
抗生物質、抗癌剤、抗ウイルス剤、抗炎症剤、ワクチン抗原、もしくは栄養補助食品から選択される薬物
から選択される、請求項1〜3のいずれか1項に記載の式(A)の化合物。 - 請求項1〜4のいずれか1項に記載の1つまたはそれ以上の式(A)の同一または異なる化合物を含むナノ粒子。
- 1つまたはそれ以上の式(I’)の化合物:
PLA−PEG−OR (I’)
(式中、PLAおよびPEGは、式(A)で定義した通りであり、Rは、HまたはC1〜C6アルキルである)
をさらに含む、請求項5に記載のナノ粒子。 - 薬物を含む、請求項5または6に記載のナノ粒子。
- 薬物は細胞毒性剤である、請求項7に記載のナノ粒子。
- 薬物はタキソイドである、請求項7または8に記載のナノ粒子。
- 薬物はドセタキセルである、請求項7〜9のいずれか1項に記載のナノ粒子。
- 薬物は、ナノ粒子に非共有結合的にカプセル化されている、請求項7〜10のいずれ
か1項に記載のナノ粒子。 - 薬物は、ナノ粒子に共有結合的に抱合されている、請求項7〜11のいずれか1項に記載のナノ粒子。
- 前記カップリングは、クリック化学により生じる、請求項13に記載の方法。
- 前記カップリング反応は、Huisgen反応に従って、有機または水性条件で実行される、請求項13または14に記載の方法。
- 前記クリック化学カップリング反応は、CuBrおよびPMDETA(N,N,N’,N’,N”−ペンタメチルジエチレントリアミン)の存在下で実行される、請求項13〜15のいずれか1項に記載の方法。
- 前記クリック化学カップリング反応は、水、CuSO4・5H2O、およびアスコルビン酸ナトリウムの存在下で実行される、請求項13〜15のいずれか1項に記載の方法。
- 前記カップリングは、ペプチドカップリング試薬の存在下、塩基の存在下で実行される、請求項19に記載の方法。
- ペプチドカップリング試薬は、PyBOP(ベンゾチアゾール−1−イル−オキシトリピロリジノホスホニウムヘキサフルオロホスフェート)から選択され、塩基がN,N−ジイソプロピルエチレンジアミン(DIPEA)である、請求項20に記載の方法。
- 前記ペプチドカップリング試薬はEDC/NHS(1−エチル−3−[3−ジメチルアミノプロピル]カルボジイミド塩酸塩/N−ヒドロキシスルホスクシンイミド)であり、塩基はトリエチルアミンである、請求項20に記載の方法。
- 1つまたはそれ以上の同一または異なる式(I)の化合物:

- 請求項1〜4のいずれか1項に記載の1つまたはそれ以上の式(A)の化合物をナノ沈殿させることを含む、請求項5〜12のいずれか1項に記載のナノ粒子の製造方法。
- 請求項6に記載の式(I’)の化合物の存在下で実施される、請求項25に記載のナノ粒子の製造方法。

- 活性成分として請求項1〜4のいずれか1項に記載の式(A)の化合物を、少なくとも1つの医薬的に許容される賦形剤と共に含む、医薬組成物。
- 活性成分として請求項1〜4のいずれか1項に記載の式(A)の化合物を、請求項5〜12のいずれか1項に記載のナノ粒子の形態で含む、請求項30に記載の医薬組成物。
