Claims (10)
- 上下を2枚の親水性の高い平面基板で挟まれた空間に形成される溶液槽、前記溶液槽内に生体分子および蛍光標識と結合した磁気微粒子を導入する入口、磁力のオンオフ、あるいは磁力の強弱を切り替える機能を有し、前記溶液槽を形成する一方の平面基板上に、生体分子および蛍光標識と結合した磁気微粒子を固定可能な磁場発生手段、を有する生体分子分析デバイスと、
前記溶液槽を形成する一方の平面基板上に固定された磁気微粒子に結合された蛍光標識を励起する励起手段と、
前記蛍光標識が発光する画像を取得する撮像手段と、
前記撮像手段で取得された画像から蛍光標識の輝点数をカウントすることで生体分子の濃度を得る手段を備え、
前記平面基板間の距離は100μm以下であり、
前記磁気微粒子は粒径0.02〜1μmの常磁性磁気微粒子である生体分子分析装置。
- 前記生体分子分析用デバイスの平面基板の表面が蒸留水に対する接触角が30°以下であることを特徴とする請求項1記載の生体分子分析装置。
- 前記生体分子分析用デバイスは完全には密封されておらず、入口と出口を有しており、
前記入口に配置された液体は、2枚の濡れ性の高い前記平面基板から得られる毛細管現象によって、溶液槽内に溶液を導入されることを特徴とする請求項1〜2のいずれか1項記載の生体分子分析装置。
- 前記磁場発生装置は、電磁石、可動式の永久磁石、可動式の磁場遮蔽物が組み合わさった電磁石、可動式の磁場遮蔽物が組み合わさった永久磁石のうちいずれかであることを特徴とする請求項1〜3のいずれか1項記載の生体分子分析装置。
- 溶液槽の導入口と排出口にそれぞれ接続された疎水性表面を有する送液管を有する送液手段を有する請求項1〜4のいずれか1項記載の生体分子分析装置。
- 前記生体分子が抗原分子であり、磁気微粒子と生体分子または蛍光標識の結合が抗原抗体反応によって実施されることを特徴とする請求項1〜5のいずれか1項記載の生体分子分析装置。
- 前記生体分子が核酸分子であり、磁気微粒子と生体分子または蛍光標識の結合がハイブリダイゼーションによって実施されることを特徴とする請求項1〜6のいずれか1項記載の生体分子分析装置。
- 前記撮像手段と一体化された蛍光標識を励起する手段を有する、請求項1〜7のいずれか1項記載の生体分子分析装置。
- 検出器側の平面基板は、ガラス、あるいは親水化処理を行った光学用ポリマーであることを特徴とした請求項8記載の生体分子分析装置。
- 前記蛍光色素を励起して、蛍光を検出する手段は、落射型光学顕微鏡であることを特徴とする請求項8または請求項9に記載の生体分子分析装置。
