Description

本発明は、抗原結合分子(ABM)に関する。特定の実施態様において、本発明は、ヒト癌胎児性抗原に結合するキメラ抗体、霊長類化抗体、又はヒト化抗体を含む、組換えモノクローナル抗体に関する。
癌胎児性抗原(CEA)及び抗CEA抗体
癌胎児性抗原(CEA、CEACAM−5又はCD66eとしても知られている)は約180kDaの分子量を有する糖タンパク質である。CEAは免疫グロブリンスーパーファミリーのメンバーであり、グリコシルホスファチジルイノシトール(GPI)アンカーを介して細胞膜にリンクされている7つのドメインを含む(Thompson J.A., J Clin Lab Anal. 5:344-366, 1991)。7つのドメインは、単一のN末端Ig可変ドメイン及びIg定常領域に相同な6つのドメイン(A1−B1−A2−B2−A3−B3)を含む(Hefta L J,ら、 Cancer Res. 52:5647-5655, 1992)。
オリゴ糖成分は、物理的安定性、プロテアーゼ攻撃に対する抵抗性、免疫系との相互作用、薬物動態学、及び特異的生物学的活性を含む、治療用糖タンパク質の効力に関連する特性に有意に影響を与える。このような特性は、オリゴ糖の存在又は非存在に依存するのみならず、オリゴ糖の特異的構造にもまた依存する可能性がある。オリゴ糖構造と糖タンパク質機能との間のある程度の一般化がなされ得る。例えば、特定のオリゴ糖構造は、特異的炭水化物結合タンパク質との相互作用を通して血流からの糖タンパク質の急速なクリアランスを媒介するが、他の構造は、抗体によって結合され得、そして望ましくない免疫反応を誘発し得るJenkinsら、 Nature Biotechnol. 14:975-81, 1996)。
定義
本明細書において用いられる用語は、下記に別に定義されていなければ、一般的に当該技術分野で用いられている。
1)このアッセイは、抗体の抗原結合領域によって認識される標的抗原を発現することが知られている標的細胞を使用する;
2)このアッセイは、エフェクター細胞として、ランダムに選択された健常ドナーの血液から単離されたヒト末梢血単球細胞(PBMC)を使用する;
3)このアッセイは以下のプロトコールに従って使用される。
i)PBMCを、標準的な密度遠心分離手順を使用して単離し、RPMI細胞培養培地中、5×106細胞/mlで懸濁する;
ii)標的細胞を、標準的な培養方法によって増殖させ、90%よりも高い生存度を有する指数増殖期から収集し、RPMI細胞培養培地中で洗浄し、100マイクロキュリーの51Crで標識し、細胞培養培地で2回洗浄し、そして105細胞/mlの密度で細胞培養培地中に再懸濁する;
iii)100マイクロリットルの上記の最終的な標的細胞懸濁物を、96ウェルマイクロタイタープレートの各ウェルに移す;
iv)抗体を、細胞培養培地中で4000ng/mlから0.04ng/mlまで段階希釈し、得られる抗体溶液の50マイクロリットルを96ウェルマイクロタイタープレート中の標的細胞に加えて、上記の全体の濃度範囲を網羅する種々な抗体濃度を3通りで試験する;
v)最大放出(MR)コントロールのために、標識された標的細胞を含む、プレート中の3つのさらなるウェルは、抗体溶液(上記の要点iv)の代わりに、50マイクロリットルの2%(V/V)非イオン性界面活性剤(Nonidet,Sigma,St.Louis)の水溶液を受容する;
vi)自発性放出(SR)コントロールのために、標識された標的細胞を含む、プレート中の3つのさらなるウェルに、抗体溶液(上記の要点iv)の代わりに、50マイクロリットルのRPMI細胞培養培地を受容する;

4)「ADCCの増加」は、上記の試験された抗体濃度範囲内で観察される特異的溶解の最大パーセンテージの増加、及び/又は上記の試験された抗体濃度範囲内で観察される特異的溶解の最大パーセンテージの半分を達成するために必要とされる抗体の濃度の減少のいずれかとして定義される。ADCCの増加は、上記のアッセイを用いて測定される、当業者に公知である同じ標準的な産生、精製、製剤、及び保存の方法を使用して、同じ型の宿主細胞によって産生される、同じ型の抗体によって媒介されるADCCに比例するが、これは、GnTIIIを過剰発現するように操作された宿主細胞によって産生されなかった。
CEAは、長い間診断目的のために癌マーカーとして使用されている。それは同一の細胞型の非腫瘍組織に比べて多くの腫瘍組織内において異常に発現される(例えば、細胞内で異なるパターンで過剰発現されるか及び/又は分布している)。しかしながら、CEAは、一般的に腫瘍細胞表面から切断され、使用可能な抗CEA抗体のほとんどはまた、可溶性CEAに結合されているので、CEAに対する非抱合型抗体は一般に、治療目的のために使用されてはいない。たとえば、現在パイロット試験にある抗CEA抗体は放射抱合体として投与される (Wongら、2004; Lierschら、 2007)。いくつかのメカニズムは、抗体依存性細胞傷害(ADCC)及び補体依存性細胞傷害(CDC)を含む抗CEA抗体の治療効果に関与している。CEAの発現の増大は癌細胞の転移につながる細胞間接着の増加を促進する(Marshall J., Semin Oncol. 30(3) 補足 8:30-36)。したがって、抗CEA抗原結合分子はまた、CEA介在細胞接着及び癌細胞の転移の阻害に重要な役割を果たす可能性がある。

変異体抗CEA ABM(例えばヒト化、親和性成熟型及び/又は安定性成熟型ABM)のコード配列を含み、かつ生物学的に活性な遺伝子産物を発現する宿主細胞は、少なくとも4つの一般的アプローチ:(a)DNA−DNA又はDNA−RNAのハイブリダイゼーション;(b)「マーカー」遺伝子機能の存在又は非存在;(c)宿主細胞中のそれぞれのmRNA転写物の発現によって測定されるような転写物のレベルを評価すること;及び(d)免疫アッセイによって、又はその生物学的活性によって測定されるような遺伝子産物の検出によって同定されてもよい。
本発明はまた、CEAを発現する細胞をインビボ又はインビトロで標的とするための方法に向けらている。CEAを発現する細胞を治療目的のために標的とすることができる(例えば、免疫系による破壊のためCEA発現細胞を標的とすることで疾患を治療するために)。一実施態様において、本発明は、被験体に、本発明のABMを含む組成物を投与することを含む、CEAを発現する細胞を標的とするための方法に向けられている。CEAを発現する細胞また、診断目的のために標的とされ得る(例えば、それらがCEAを正常又は異常に発現しているかどうかを決定することなど)。従って、本発明はまた、CEAの存在又はCEAを発現する細胞を検出するための方法に向けられている。本発明によればCEAの発現を検出する一つの方法は、ABM及びCEAとの複合体の形成を可能にする条件下で、テストする試料を必要に応じてコントロール試料と、本発明のABMと、接触させることを含む。複合体形成は、その後(例えば、ELISA又は当技術分野で公知の他の方法により)検出される。試験試料とともにコントロール試料を用い、試験試料及びコントロール試料を比較する場合、ABM−CEA複合体の形成における任意の統計的な有意差は、被験試料中のCEAの存在を示している。
本発明はまた、一つ以上の細胞傷害性薬物、例えば、化学療法剤又は薬物、成長阻害剤、毒素(例えば、タンパク質毒素、細菌、真菌、植物、又は動物由来の酵素的に活性な毒素、又はその断片)又は放射性同位体などに結合した本明細書の抗CEA ABM又は変異体ABMを含む、免疫複合体を提供する。
一態様において、本発明は、本発明の抗CEA ABM又は変異体ABMを含む薬学的組成物及び薬学的に許容される担体に向けられている。本発明は更に、癌などの疾患の治療法、及び癌などの疾患の治療のための医薬の製造における、そうした薬学的組成物の使用に向けられている。具体的には、本発明は、疾患の治療のための方法、より具体的には、癌の治療のための方法に向けられており、該方法は本発明の薬学的組成物の治療有効量を投与することを含む。

実施例1
親和性成熟ライブラリの生成
H1/H2ライブラリー

HCDR3領域内の無作為化された親和性成熟ライブラリーを生成するために、位置W95、D96、F97、Y98、D99、Y100、V100a、E100b、A100c、及びM100dをコードするトリプレットは、2つの異なるアプローチに無作為化された。:(1)セグメント全体(H3完全ライブラリー)の無作為化、又は(2)各位置の個々の無作為化は10個のサブライブラリ−を与えた。個別に無作為化された位置を持つクローンを含むサブライブラリ−は、細菌(H3プールライブラリ−)への形質転換の後にプールされた。HCDR3領域の無作為化のために、断片は、CLの3’末端でアニールされたプライマー及びHCDR3の無作為化配列(図13)を含むプライマーを用いてPCR増幅された。次いで重複伸長PCRが、断片1の3’末端と重なり、VHの終端及びCH1の5’領域を含む第二の断片を用いて行われた。集合断片はSacI/NheI消化後pMS22へ連結された。H3プールライブラリを生成するために、10個のDNA断片は、プライマーEAB−749(配列番号146)と組み合わせてプライマーAC7−AC16(配列番号135;配列番号144)の各々を用いて別個にPCR増幅された。L3完全ライブラリを生成するために、プライマーAC17(配列番号145)及びEAB−749(配列番号146)が使用された。プラスミドpMS22が、テンプレートとして使用された。増幅条件は最初の5分間94℃インキュベーション工程を含み、その後、1分間94℃の変性、1分間55℃のアニーリング、及び36秒間72℃の伸長工程を含む各サイクルが25サイクル続き、その後最終の10分間72℃のインキュベーション工程が続いた。これによりアガロースゲル上で精製した約580bp長の断片を得た。重複伸長PCRのために、第二の断片はEAB−752(配列番号149)と組み合わせて、EAB−750(配列番号147)又はEAB−751(配列番号148)の何れかのプライマーを用いて増幅された。プライマーEAB−750(配列番号147)は、無作為化プライマーAC7−11(配列番号139)と重複配列を有し、一方EAB−751(配列番号148)は無作為化プライマーAC12−17(配列番号140−145)と配列相同性を共有していた。増幅条件は最初の5分間94℃インキュベーション工程を含み、その後、1分間94℃の変性、1分間55℃のアニーリング、及び12秒間72℃の伸長工程を含む各サイクルが25サイクル続き、その後最終の10分間72℃のインキュベーション工程が続いた。得られた断片はおよそ180bp長であった。両方の断片の集合に対して、断片1及びそれに対応する断片2の等モル量が使用された。増幅条件は最初の5分間94℃インキュベーション工程を含み、その後、1分間94℃の変性、1分間55℃のアニーリング、及び60秒間72℃の伸長工程を含む各サイクルが5サイクル続いた。外側のプライマーEAB−749(配列番号146)及びEAB−752(配列番号149)を添加した後、更に20サイクルが同じパラメータを用いて実施された。最後に、最後の10分間72℃でのインキュベーション工程が実施された。ゲル精製断片は次いでSacI/NheI消化後にpMS22へ連結され、精製されたライゲーションが電気穿孔法によりTG1細菌に形質転換された。
軽鎖のCDR3領域内の無作為化された親和性成熟ライブラリーを生成するために、位置Y91、Y92、T93、Y94、及びL95aをコードするトリプレットは、セグメント全体(L3完全ライブラリー)又は各々の何れかで無作為化され、5個のサブライブラリ−を得た。個別に無作為化された位置を持つクローンを含むサブライブラリ−は、細菌(L3プールライブラリ−)への形質転換の後にプールされた。5個のサブライブラリ−の生成のために、5個のDNA断片が、プライマーMS43(配列番号123)との組み合わせでプライマーAC1−AC5(配列番号150−154)のそれぞれを用いてPCR増幅された。L3完全ライブラリ−を生成するために、AC6(配列番号155)及びMS43(配列番号123)のプライマーの組み合わせが使用された(図14)。プラスミドpMS22が、テンプレートとして使用された。増幅条件は最初の5分間94℃インキュベーション工程を含み、その後、1分間94℃の変性、1分間55℃のアニーリング、及び25秒間72℃の伸長工程を含む各サイクルが25サイクル続き、その後最終の10分間72℃のインキュベーション工程が続いた。VLドメインの位置1から104までを包含する得られた断片はアガロースゲル上で精製され、追加のPCR増幅のテンプレートとして使用された。全ての反応は上記と同じ条件を用いて無作為化プライマー及びMS43(配列番号123)との重複配列を有するプライマーEAB−746(配列番号156)で実施された。精製断片並びにpMS22はNcoI/XhoIで消化された。全5個のサブライブラリについて、0.5μgの挿入は、0.5μgのpAC16と連結された。L3完全ライブラリ−について、ライゲーションは9.8μgの挿入及び9.8μgのpMS22により実施された。精製されたライゲーションは電気穿孔法によってTG1細菌に形質転換された。
マウス及びヒト化PR1A3抗体の両方が膜結合CEAのみを認識するが、離脱した可溶性ヒトCEAを与えないので、PR1A3のエピトープを含む組換えキメラタンパク質はヒト化PR1A3のインビトロ親和性成熟のために生成された(配列番号7及び8)。このハイブリッドタンパク質の生成はSteward ら(1999)に記載の通り行われた。簡潔には、ヒトの胆汁糖タンパク質(BGP)のBドメインのDNA配列が、PR1A3のエピトープを含むヒトCEA−B3ドメインの配列に置き換えられた。その結果、配列は、BGPのN及びA1ドメイン、CEAのB3ドメイン及びBGPのA2ドメインを含むハイブリッドタンパク質をコードしている(N−A1−B3−A2、huNABA)。この融合産物は、次いでヒトIgG1のFc部分にリンクされるか(huNABA−Fc) (Stewardら、Cancer Immunol Immunother, 47:299-306, 1999)、又は正確なプロテアーゼ切断部位、aviタグ及び(His)6タグをコードする配列に融合するか(huNABA−avi−his) (配列番号158)の何れかであった。huNABA−Fcは、プロテインAカラムを用いて安定的にトランスフェクトしたCHO細胞株の培養上清から精製された。huNABA−avi−his(配列番号158)は、安定してEBV由来タンパク質EBNAを発現するHEK293細胞に一時的にトランスフェクトした。同時にビオチンリガーゼをコードする同時形質移入されたプラスミドは、インビトロでaviタグ固有のビオチン化を可能とした。タンパク質は、その後、固定化金属アフィニティークロマトグラフィー(IMAC)、続いてゲル濾過により精製された。
親和性成熟ヒト化PR1A3 Fabの生成は、標準プロトコルを使用して、ファージディスプレイによって行われた (Silacci ら、Proteomics, 5(9):2340-2350, 2005)。親和性成熟ライブラリーによる選択は以下の手順に従って溶液中で実施された:[1].各親和性成熟ライブラリーのおよそ1012ファージミド粒子を100nMのビオチン化huNABA−avi−hisに1ミリリットルの総体積で0.5時間結合させ、[2].ビオチン化huNABA−avi−his及び特異的に結合したファージ粒子を、5.4×107のストレプトアビジン被覆磁気ビーズの3分間の添加により、捕獲し、[3].ビーズを5−10×1mlPBS/Tween20及び5−10×1mlPBSを用いて洗浄し、[4].1mlの100mMのTEA(トリエチルアミン)を10分間の添加によりファージ粒子を溶出し、及び500ulの1Mトリス/HCl pH7.4を加えることによる中和し、及び、[5].指数関数的に増殖する大腸菌TG1細菌を再感染させ、ヘルパーファージVCSM13により感染させ、続いて、その後の選択のラウンドで使用されるファージミド粒子をPEG/NaCl沈殿させる。選択は一定又は減少する抗原濃度(10−7Mから2x10−9M)のいずれかを使用して、3から5回のラウンドで行われた。ラウンド2において、抗原:ファージ複合体の捕捉が、ストレプトアビジンビーズの代わりにニュートラアビジンプレートを用いて行われた。特異的バインダーは、次のようにELISAにより同定した:ウエルあたり10nMのビオチン化huNABA−avi−hisの100μlはニュートラビジンプレート上に被覆された。Fabを含む細菌の上清が添加され、結合したFabは抗フラグ/HRP二次抗体を使用して、フラグ-タグを介して検出された。ELISA陽性クローンを96穴フォーマットに可溶性Fab断片として細菌に発現させ、BIACORE T100を使用して、SPR分析により、上清は動力学的スクリーニング実験にかけられた。最も高い親和性定数を持つFabを発現するクローンを同定し、対応するファージミドが配列決定された。
動力学パラメータの正確な分析のために、Fabを細菌培養から精製した。500mlの培養は播種され、OD600が0.9にて1mMのIPTGで誘導した。細菌は25℃で一晩インキュベートし、遠心分離によって回収した。再懸濁したペレットを25mlのPPB緩衝液(30mMトリス−HCl PH8、1mMのEDTA、20%ショ糖)で20分間のインキュベーションした後、細菌は再び遠心分離し、上清を回収した。このインキュベーション工程は、5mMのMgSO4溶液25mlで一回繰り返した。両方のインキュベーション工程の上清をプールし、ろ過し、IMACカラム(His gravitrap、GE Healthcare)にロードされた。その後、カラムを40体積で洗浄した。溶出した後に(500mMのNaCl、500mMのImidazole、20mMのNaH2PO4 、pH7.4) 、溶出液はPD10カラム(GE Healthcare)を使用して再バッファリングされた。精製されたFabの速度論的パラメータは、200nmから6.25nMの範囲にあった希釈列にてSPR分析により調べられた。
PR1A3抗体は、ヒトIgG1/kappa定常領域を持たせるようにキメラ化され、及びFcにおいて高度にフコシル化された糖を持たせるためにGylcoMab技術を用いて発現された。糖鎖を操作された抗体及び糖鎖は操作されてない抗体は、エフェクターとターゲットの比率が25:1で比較された。抗体依存性標的細胞死滅の最大量は、Fc領域の糖鎖操作により倍増した(図2)。細胞死滅の更なる増加はエフェクターとターゲットの比を増加させることにより達成された(図2)。
実施例2に記載の親和性成熟型抗CEA抗体の生産に使用されるアクセプターフレームワークは、ヒトVH7クラスのものであった。安定性を増加させるために、より安定なフレームワーク配列が、抗体の安定性操作のための基礎として使用された。マウス抗体PR1A3の配列相同性、及びVH1由来配列は、VH7、又はヒトVH族の偶数よりもより高い内因性安定性を有するはずであるという仮説に基づいて、配列IGHV−1−18; 受託番号M99641が新しいアクセプターフレームワークとして使用された(Ewert, S., Huber, T., Honegger, A. and Pluckthun, A. (2003) J. Mol.Biol., 325, 531-553)。PR1A3抗体の通常のCDRループ移植がCH1A(配列番号279)の構築につながる。残念なことに、この分子はCEA抗原に対する有意な結合活性を示さなかった。この構築物の結合活性は、様々な濃度で、マウス由来の可変ドメインを保有するキメラ抗体PR1A3の結合活性に対して比較した。BxPC3細胞が、CEAに対する抗体の特異的結合のために使用され、結合強度はFACS解析により測定された(図17)。
親和性成熟の過程で選択されたCDR−H3の残基が、抗体の安定性の増加を試験するためにPR1A3配列に個別に導入された(図36)。
ヒト化PR1A3誘導体CH7Aの二重変異t(Y98A/D99Y)が、VH1の基づくヒト化−構築物のCH1A1A及びCH1A1Bの最終変異体に導入された。
CH1A1A構築物及びCH1A1B構築物はADCC活性を示した。CH1A1A及びCH1A1B変異体、それらの前駆体変異体CH1A1、及び親CH7A変異体により媒介されるADCCが、4時間後に、MKN45細胞を標的細胞(T)として、ヒトPBMCをエフェクター細胞(E)として、E:Tの比を25:1で使用して、乳酸脱水素酵素の放出によって測定された。乳酸脱水素酵素の放出は、標的細胞の溶解に比例し、パーセント細胞毒性として示される。図26これらの変異体のADCCは、MKN45細胞を標的細胞(T)として、ヒトPBMCをエフェクター細胞(E)として、E:Tの比を25:1で使用して、カルセインの放出によって測定されて確かめられた。カルセイン放出は、標的細胞の溶解に比例し、パーセント細胞毒性として、平均値±標準偏差値により示される(図27)。
CH1A1A(98/99)の重鎖と2F1軽鎖を含む抗CEA抗体の糖鎖を操作した型は、ヒトCD16の遺伝子導入SCIDマウスにおける結腸直腸癌の異種移植モデルで、有効性について試験した。モデルのアッセイ条件を以下に記載する。アッセイの結果は、この抗CEA抗体は、ビヒクルコントロールに比べて、延命効果を提供することを示している(図30)。
CH1A1A(Y98A/D99Y)の重鎖と2F1軽鎖を含む抗CEA抗体の糖鎖を操作した型は、ヒトCD16の遺伝子導入SCIDマウスにおけるA549肺癌の異種移植モデルで、有効性について試験した。モデルのアッセイ条件を以下に記載する。アッセイの結果は、この抗CEA抗体は、ビヒクルコントロールに比べて、用量依存性の延命効果を提供することを示している(図31)。

配列番号56の軽鎖CDR3を含む軽鎖可変領域、を含む抗体。 - CH1A1A(配列番号261)又はCH1A1B(配列番号262)のフレームワーク残基を含む、請求項1に記載の抗体。
- 重鎖可変領域が、配列番号239及び配列番号247からなる群から選択される配列に対して少なくとも95%同一であるアミノ酸配列を含み、軽鎖可変領域が、配列番号209の配列に対して少なくとも95%同一であるアミノ酸配列を含む、請求項1又は2に記載の抗体。
