Description

変異体を提示する、生理学的溶液中に溶解性のニューレグリン−1アイソフォームを参照

ニューレグリン(NRG)は、シナプス性シグナリングの鍵となる制御因子として出現

ないことが必須である。動物は、足場を介した場合にのみ救助がくるように訓練され、こ
れは足場を見いだせない全ての動物は足場に案内され、かつ、このセットアップから取り
除かれる前に休息させることを意味する。したがって、マウスにとって最も重要な参照点
の1つは、人間の操作者である。

れていく想起を支持する。
・隠された足場獲得訓練。足場はマウスに可視可能でなく、かつ、先行する水泳の間に同
じ配置に位置される。この課題は、隠された足場の正確な配置を想起することについての
てマウスの進歩をモニターすることを可能にする(「空間的学習」)。
・検査試験実験。この課題において、オンデマンド足場を表面下に最大限低下させ、マウ
スをこれを自由に探索させながら泳がせる。検査試験実験は、動物の絶対的な想起を評価
し、これは、この文脈においては、足場配置に関する信念、忍耐又は確信とも解釈される
ことができる。実験を解釈するための慣用的アプローチとは、足場の固く固定された場所
を有する動物が、限定された場所においてより忍耐強く探索し、したがって足場の隣の区
域においてより時間を費やすであろうことである。
・検査試験実験において、足場を見出すことについての不能性が足場区域へと泳ぐための

ハロペリドール(125μg/kg)について使用したものよりも約200〜1000倍
より低い。
・ハロペリドール、クロザピン、オランザピンその他とは対照的に、NRG−1β−EC
Dはビヒクル対照レベル未満に試験動物の活動を減少させないので、不利な作用は観察さ
れない。
精神***病:プレパルス阻害
NRG1ノックアウトを有する齧歯類は、顕著に損なわれたプレパルス阻害(PPI)
を示し、これはNRG1を精神***病につなげる。動物モデル中の精神病の広く使用され
る代理測定、PPIは、精神***病エンドフェノタイプと考えられる。精神***病及び健
康な対照集団の両者において、広範なジェノタイピング後に、PPIに対して、NRG1
(rs3924999)上に配置されている非同義単一ヌクレオチド多形のミスセンス突然変異の
神経生理学的作用があることが報告された(Hong et al. 2007)。PPIに対するNRG
−1β−ECDの作用を試験した。これまでの結果は以下のように要約されてよい:
この作用は、115dBで、統計学的有意には達せず、観察されなかったが、105d
Bで、NRG−1β−ECDは、PPIの再確立に対して一般的な傾向を示した(+26

かったが、600ng/kgで刺激の非存在において自発的な動きを顕著に減少させた(

アリピラゾールに類似である)。NRG−1β−ECDは、プレパルス単独に対する反応
に対して作用を有しなかった。

クマウスモデルに繰り返した(APPPSマウス(Meyer-Luehmann et al. 2006;Radde e
t al. 2006))。

−4−フェニル−1,2,3,6−テトラヒドロピリジン)モデルにおいて使用した。

P傷害(insult)の間にこのパーキンソン病のためのモデルにおいて、NRG−1β−E
CDの投与の明白な作用を明らかにする。

HRG−α2B、HRG−α3、HRG−β1、HRG−β2、HRG−β3(GGF)
、GGF2及びSMDFのウェスタンブロット(開始希釈1:200、希釈範囲1:10
0〜1:1000)、免疫沈降[全タンパク質100〜500μgにつき1〜2μg(細
胞溶解物1ml)]及び免疫蛍光(開始希釈1:500、希釈範囲1:50〜1:500
)による検出のために推奨される。

RG−1β−ECDの濃度は、約pl5.0で、同時により良好な学習者である処理され
たAPPPSマウスにおいて顕著により高い。

材料を用いたウェスタンブロット実験の結果を示す。NRG−1β−ECD断片が、アル
ツハイマーの場合に豊富さが顕著により少ないことを明白に示す。内部対照として、NR
G−12の豊富さを測定し、これは疾病に関連する記憶喪失により作用を受けないように
見える。
ムの更なる検査を、図9について使用した同じ死後のヒトの脳材料の2次元ゲル(2D−
PAGE)のウェスタンブロットにより行い、これは、図10に示される代表的な例によ
り示されるとおりに、確かに、アルツハイマー状態において減少しているNRG−1β−
ECDの酸性のアイソフォームであることを明らかにする。
我々はここで初めて、nrg−1遺伝子の転写産物の翻訳後修飾、特にタンパク質加水
分解的開裂により産生された切断された形態であって、MW15〜35、pl4〜10を
有するNRG−1βの細胞外ドメインを含有するもののin vivo作用の機能的な証
拠を提示する;より特異的には、我々は、精神***病のための動物モデルにおける抗精神
病薬活性を、5〜600ng/kg(i.v.)の濃度で、ことによるとMAO−B及び
COMTの制御に基づいて見出した。100〜1000倍より高い濃度で使用される対照
神経遮断薬とは対照的に、不利な作用は観察されなかった。

前記組み換え溶解性タイプIニューレグリン−1βアイソフォームがSDS−PAGEにより測定して15〜35kDの分子量を有し、
前記神経学的状態が、精神***病、双極性障害、うつ病、パーキンソン病、アルツハイマー病、てんかん、MS、ALS、卒中、外傷性脳損傷及び脊髄損傷からなる群から選択され、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが、250アミノ酸までの長さを有し、ならびに、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが、処置すべき被験体の体重1kgあたり0.1〜5000ngの量で投与される、前記医薬組成物。 - 前記神経学的状態が、精神***病の認知に関連した観点である請求項1記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが組み換え溶解性タイプIヒトニューレグリン−1βアイソフォームである請求項1または2記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームがこの細胞外ドメインの少なくとも一部を含有する請求項1から3までのいずれか1項記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームがSDS−PAGEにより測定して25〜32kDの分子量を有する請求項1から4までのいずれか1項記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが4〜10の等電点(pI)を有する請求項1から5までのいずれか1項記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが4〜6の等電点(pI)を有する請求項1から5までのいずれか1項記載の医薬組成物。
- 前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが修飾したポリペプチドであり、その際この修飾がリン酸化、グリコシル化、メチル化、ミリストイル化、酸化及びこの任意の組み合わせから選択される請求項1から7までのいずれか1項記載の医薬組成物。
- 更なる医薬品と組み合わせて用いる請求項1から8までのいずれか1項記載の医薬組成物。
- 更なる医薬品が神経学的状態の治療のための医薬品である請求項9記載の医薬組成物。
- 更なる医薬品が、カテコールアミン代謝に作用する化合物、アセチルコリンエステラーゼ阻害剤、MAO−B−又はCOMT−阻害剤、メマンチン型チャネル遮断剤、ドーパミン又はセロトニンレセプターアゴニスト又はアンタゴニスト、カテコールアミン又はセロトニン再取り込み阻害剤又は任意の種類の抗精神病薬、ガバペンチン様薬剤であってアルツハイマー病及びパーキンソン病、精神***病、双極性障害、うつ病又はその他の神経学的状態の治療におけるものから選択される、請求項10記載の医薬組成物。
- 更なる医薬品が、クロザピン又はオランザピンである、請求項10記載の医薬組成物。
- 更なる医薬品が、精神***病、双極性障害又はうつ病の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品がパーキンソン病の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品がアルツハイマー病の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品が多発性硬化症(MS)の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品が筋萎縮性側索硬化症(ALS)の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品がてんかんの治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品が卒中の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品が外傷性脳損傷の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 更なる医薬品が脊髄損傷の治療のための医薬品である請求項9又は10記載の医薬組成物。
- 記憶及び認知促進のための全身投与用の医薬組成物であって、
組み換え溶解性ニューレグリン−1アイソフォームを含み、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームがSDS−PAGEにより測定して15〜35kDの分子量を有し、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが、250アミノ酸までの長さを有し、ならびに、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが、処置すべき被験体の体重1kgあたり0.1〜5000ngの量で投与される、前記医薬組成物。 - 神経学的状態に関連した記憶及び認知喪失を減少化及び/又は抑制するための請求項21記載の医薬組成物。
- 神経学的状態が、アルツハイマー病である請求項23記載の医薬組成物。
- 組み換え溶解性タイプIニューレグリン−1βアイソフォームが請求項1から8までのいずれか1項に定義されているとおりである請求項22から24までのいずれか1項に記載の医薬組成物。
