Description
本発明は、認知に関連した神経学的状態、例えば精神***病、アルツハイマー病及びパーキンソン病における薬物療法としての、翻訳後ニューレグリン−1修飾又はスプライス変異体を提示する、生理学的溶液中に溶解性のニューレグリン−1アイソフォームを参照する。
ニューレグリン(NRG)は、シナプス性シグナリングの鍵となる制御因子として出現した。この膜貫通型タンパク質は、4個の遺伝子(NRG−1、−2、−3、及び−4)によりコードされ、この多様性は更に代替的RNAスプライスング及びプロモーターの使用により増加され、特にタンパク質加水分解性プロセッシングなどの翻訳後修飾により増加され、これは膜結合ホロタンパク質からの溶解性アイソフォームの放出を引き起こす。更に、リン酸化及びグリコシル化の証拠が存在する(Buonanno and Fischbach 2001)。これらは、異なる細胞外ドメインにより特徴付けられ、かつ、ErbBレセプターチロシンキナーゼのリガンドであり、これは、神経炎症及び遺伝子転写に対する下流の暗示的意味合いを有する(Holbro and Hynes 2004)。特にNRG−1の溶解性アイソフォームは、NRGの膜貫通型形態から、電気的刺激の間のタンパク質加水分解性開裂を通じて産生され、引き続き活性依存性シナプス性調節因子として分泌される(Ozaki et al. 2004)。
精神***病は、症状、例えば幻聴、障害された思考及び妄想、意欲消失、快感消失、鈍化された情動及び無関心を伴う深刻かつ不能化にする精神障害である。疫学的、臨床的、神経心理学的かつ神経生理学的研究は、脳の発達における異常性及び進行する神経可塑性が、前記疾患の病原論において重要な役割を果たすことについての実質的な証拠を提供している(Arnold et al. 2005)。
本発明者らによる初期の研究は、ニューレグリン1βがアルツハイマー患者の脳の海馬の死後の切片において、年齢適合対照に比較して減少していること(Sommer et al., 2004)を、ニューレグリン1の溶解性の断片と放射状迷路試験における学習能力との明白な正の相関と一緒に示した(Sommer et al., 2004)。
USにおける独立した外部研究による一連の卒中に関連したin vivo実験は、自体で抗アポトーシス性であるニューレグリン1の神経保護を実証する(Xu et al., 2004, 2005 and 2006; Guo et al., 2006)。
一般:
以下の全実験においては、ニューレグリン−1βの断片を使用しており、これは、ヒトのnrg−1遺伝子の全転写産物の細胞外ドメイン(ECD)のみを含有する。これは、リン酸化及び/又はグリコシル化状態に依存して約25〜32kDの分子量及び約5〜9.5の等電点を有した。
初期の毒物学的データは、NRG1β(ECD)が急性毒物学及びin vitro突然変異性試験において不利な作用を有しないことを示す。
・ラットにおいて急性の静脈性毒性は存在しなかった:全ての動物は研究期間の終わりまで生存した。研究の経過の間に臨床徴候は観察されなかった。動物の体重は、この株及び年齢について一般的に記録される範囲内にあった。屍検では顕微鏡的知見は記録されなかった。14日間の期間にわたり観察された、メスのラットへの単一静脈投与後のNRGβ1(ECD)の中央致死量は:LD50(メスのラット):5000ng/kg体重より多い。
・用量レベル50、200及び600ng/kg体重/dでの7日間の期間にわたるニューレグリンの毎日の静脈投与は、いかなる未成熟な死も生じなかった。臨床徴候は観察されなかった。この処置は、餌消費及び体重発育に作用しなかった。この無影響レベル(NOEL)を600ng/kg体重/dで確立した。
・細胞系L5178Yを使用した、OECD Guideline for the Testing of Chemicals, No. 476 "In vitro Mammalian Cell Gene Mutation Test"によるマウスリンパ腫チミジンキナーゼ遺伝子座(locus)アッセイにおいて、NRG1B(ECD)は非突然変異誘発性であった。
・OECD Guideline for the Testing of Chemicals, No. 473によるチャイニーズハムスターV79細胞中での染色体異常試験において、NRG1β(ECD)は構造的な染色体異常を誘発しなかった。
学習及び記憶:NRG−1β−ECD適用あり及び無しでの空間学習
方法:
モリス水迷路は、空間学習を評価する。これは、動物を水で満たしたプール中で泳がせ、かつ、この表面のすぐ下に沈められた救助足場(rescue platform)を見つけさせる。この足場が迷路の壁から離れて配置され、かつ、動物が位置の推定を可能にする水表面から可視できる参照点を有し、但し、これが連想学習を可能にするために標的に十分近すぎないことが必須である。動物は、足場を介した場合にのみ救助がくるように訓練され、これは足場を見いだせない全ての動物は足場に案内され、かつ、このセットアップから取り除かれる前に休息させることを意味する。したがって、マウスにとって最も重要な参照点の1つは、人間の操作者である。
・マウスがこの足場を配置し直す(relocate)ことを学習する速度、
・短期間でこの情報を保持する能力(訓練期間内又はオーバーナイト)。
この研究をAPP/PSマウス(Meyer-Luehmann et al. 2006;Radde et al. 2006)の2つの群を用いて実施し、このうちの一方をNRG−1β−ECDの毎日の用量により処置し、かつ、もう一方は対照として偽処置されている。それぞれの群は、最初の実験系列の開始時に9週齢の8匹の雄からなる。
この処理及び未処理APP/PSマウスの学習適性を、円形のモリス水迷路を用いて評価し、これはマウスを消耗させることなく探索空間を提供するために十分大きい必要がある。最大限に注意を払うべきは、全ての実験を通じて実験のセットアップの各詳細を可能な限り変動しないように維持することである。
各セッションにおいて、マウスを予め定義された部位でプール中に配置し、60秒間水泳させる。動物の運動路をビデオ追跡システムにより記録し、パラメーターをコンピューター計算し、ここから動物の学習適性に関する結論が導き出される(最も明らかであるのは、マウスがこの足場に最初にぶつかるまでの時間=「逃走潜時」;更なる詳細については以下参照のこと)。マウスが足場の発見に成功すると、短期間(〜15s)そこに休息させたままにする。さもなければ、水泳60秒後、マウスを操作者により足場に案内し、〜15秒間休息させる。その後、操作者により持ち上げ、優しく乾燥させ、その小屋に戻すか又は次の水泳のために準備させる。
・合図された配置ナビゲーション。プラットフォームを手がかりでマークし、マウスを足場を見つけるまで泳がせる。この手順は連想的学習を試験し、マウスを可能な限り類似している学習適性である2つの実験群にわけるのに役立つ。更に、第2の及び更なる系列の実験において、合図された場所ナビゲーションは、先行する系列における足場の位置の薄れていく想起を支持する。
・隠された足場獲得訓練。足場はマウスに可視可能でなく、かつ、先行する水泳の間に同じ配置に位置される。この課題は、隠された足場の正確な配置を想起することについてのてマウスの進歩をモニターすることを可能にする(「空間的学習」)。
・検査試験実験。この課題において、オンデマンド足場を表面下に最大限低下させ、マウスをこれを自由に探索させながら泳がせる。検査試験実験は、動物の絶対的な想起を評価し、これは、この文脈においては、足場配置に関する信念、忍耐又は確信とも解釈されることができる。実験を解釈するための慣用的アプローチとは、足場の固く固定された場所を有する動物が、限定された場所においてより忍耐強く探索し、したがって足場の隣の区域においてより時間を費やすであろうことである。
・検査試験実験において、足場を見出すことについての不能性が足場区域へと泳ぐための刺激を減少させる可能性があるという危険が存在する。これらのいらだちを可能な限り小さく維持するために、ヒトの救助の調節は同じに維持されなくてはならず、この結果、足場の非存在にもかかわらず幾ばくかの空間的な不変性が存在する。したがって、水泳60s後足場を表面のちょうど下におろし、このマウスをここに操作者により案内し、この装置から取り出される前に〜15sにわたり休息させる。
検査試験実験の全ての日に、ただ一回の水泳が実施される。

動物のビデオ記録から、それぞれのマウスの動き経路を抽出し、x、yの系列及び時間軸として更なるプロセス処理のためエクスポートする。
評価を可能な限り柔軟に維持するために、5.5〜30cmの直径の4つの同心標的区域(足場に関して中心にある)を利用する。

一日量3ng/kgのNRG−1β−ECDでi.v.で訓練の30分前に処理したこれらの動物は、ビヒクル処理群に比較して学習関連パラメーターにおいて顕著により良好であった。
精神***病:ラット中でアンフェタミン誘発された活動過剰
方法:
抗精神病及び抗パーキンソン病活性を検出する方法は、Costall et al. 1978により説明された方法にならい、そしてBoissier and Simon 1966により説明されたものに類似する活動メーターを使用する。
図3に示されるとおり、NRG−1β−ECDは用量依存的にアンフェタミンにより誘発された過剰活動を精神***病のための動物モデル中で抑制した。著しく、この実験はNRG−1β−ECDの傑出した特性を明らかにする:
・図3中に示される作用は、実験の第2の半分(20〜40分)において最強である。最初の20分間においてはより小さな作用のみが見出されることができ、これはこのタンパク質の更なるプロセッシングへと作用点を遅延させた。
・ここで使用したNRG−1β−ECDの有効濃度は、典型的な対照神経遮断薬、例えばハロペリドール(125μg/kg)について使用したものよりも約200〜1000倍より低い。
・ハロペリドール、クロザピン、オランザピンその他とは対照的に、NRG−1β−ECDはビヒクル対照レベル未満に試験動物の活動を減少させないので、不利な作用は観察されない。
精神***病:プレパルス阻害
NRG1ノックアウトを有する齧歯類は、顕著に損なわれたプレパルス阻害(PPI)を示し、これはNRG1を精神***病につなげる。動物モデル中の精神病の広く使用される代理測定、PPIは、精神***病エンドフェノタイプと考えられる。精神***病及び健康な対照集団の両者において、広範なジェノタイピング後に、PPIに対して、NRG1(rs3924999)上に配置されている非同義単一ヌクレオチド多形のミスセンス突然変異の神経生理学的作用があることが報告された(Hong et al. 2007)。PPIに対するNRG−1β−ECDの作用を試験した。これまでの結果は以下のように要約されてよい:
この作用は、115dBで、統計学的有意には達せず、観察されなかったが、105dBで、NRG−1β−ECDは、PPIの再確立に対して一般的な傾向を示した(+26%、+23%及び+36%、150、300及び600ng/kg、それぞれ)。これは150又は300ng/kgで刺激の非存在において自発的な動きに対する作用を有しなかったが、600ng/kgで刺激の非存在において自発的な動きを顕著に減少させた(−20%及び−29%、平均及びピーク強度に対して、それぞれ、p<0.05、これはアリピラゾールに類似である)。NRG−1β−ECDは、プレパルス単独に対する反応に対して作用を有しなかった。
アルツハイマー病のための動物モデル(APPPS dtマウス)における学習及び記憶
通常マウスについて上述したとおりのモリス水迷路セットアップにおけるニューレグリン1β(NRG−1β−ECD)の溶解性細胞外ドメインの適用あり又はなしでの学習及び記憶を試験する動物実験を、大脳アミロイドーシスについてのダブルトランスジェニックマウスモデルに繰り返した(APPPSマウス(Meyer-Luehmann et al. 2006;Radde et al. 2006))。
パーキンソン病のニューレグリン1−βMPTPマウスモデル
方法:
10週齢の雄のC57Bl/6マウスをパーキンソン病のためのMPTP(1−メチル−4−フェニル−1,2,3,6−テトラヒドロピリジン)モデルにおいて使用した。
最後のMPTP投与1日後:マイクログリア活性化の開始
最後のMPTP投与3日後:最大限のマイクログリア活性化
最後のMPTP投与7日後:最大限のアストロサイト活性化
最後のMPTP投与21日後:最大限の細胞死。
図5に示されるとおり、ドーパミン及びその代謝産物のHPLC測定の結果は、MPTP傷害(insult)の間にこのパーキンソン病のためのモデルにおいて、NRG−1β−ECDの投与の明白な作用を明らかにする。
有効成分(active principle)としてのNRG−1β−ECDの酸性翻訳後アイソフォームの同定
学習及び記憶においてNRG−1β−ECDの特定の翻訳後酸性アイソフォームが活性のある形態であるという証拠を刊行した(Schillo et al. 2005a)。ここでは、同様のパターンがアルツハイマー病の動物モデル及びアルツハイマー病及びパーキンソン病患者からの死後の脳組織において観察されることを示す。我々は、この酸性アイソフォームが有効成分であることを結論づける。
ウェスタンブロット染色のために以下の抗体を使用した:抗NRG1−ECD、ウサギポリクローナル(sc-28916 ロット:I 2905 Santa Cruz; H-210)
ニューレグリン−1(H−210)は、ヒト由来のニューレグリン−1アイソフォームHRG−αのN末端細胞外ドメイン内のアミノ酸21−230マッピングに対して産生されたウサギポリクローナル抗体である。ニューレグリン−1(H−210)は、マウス、ラット及びヒト由来のニューレグリン−1アイソフォームHRG−α、HRG−α1A、HRG−α2B、HRG−α3、HRG−β1、HRG−β2、HRG−β3(GGF)、GGF2及びSMDFのウェスタンブロット(開始希釈1:200、希釈範囲1:100〜1:1000)、免疫沈降[全タンパク質100〜500μgにつき1〜2μg(細胞溶解物1ml)]及び免疫蛍光(開始希釈1:500、希釈範囲1:50〜1:500)による検出のために推奨される。

- 神経学的状態の治療のための全身投与による医薬品の製造のための組み換え溶解性タイプIニューレグリン−1βアイソフォームの使用であって、
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームがSDS−PAGEにより測定して15〜35kDの分子量を有し、
前記神経学的状態が、精神***病、双極性障害、うつ病、パーキンソン病、アルツハイマー病、てんかん、MS、ALS、卒中、外傷性脳損傷及び脊髄損傷からなる群から選択され、ならびに
前記組み換え溶解性タイプIニューレグリン−1βアイソフォームが、250アミノ酸までの長さを有する、前記使用。 - 前記神経学的状態が、精神***病の認知に関連した観点である請求項1記載の使用。
