本願明細書において使用する場合、「B細胞リンパ腫を有する患者」及び「B細胞リンパ腫患者」なる用語は、ある種のB細胞リンパ腫と診断されている患者又はある種のB細胞リンパ腫の可能性があると診断されている患者を指す。
「プライマーの対」は、特異的な標的遺伝子の部分を増幅するために使用することができる5’プライマー及び3’プライマーを指す。
治療剤の「治療的有効量」は、例えば個体の疾病ステージ、年齢、性別、及び個体の体重、並びに個体に所望の応答を誘発する抗体の能力のような因子に従って変わりうる。また、治療的有効量は治療剤の任意の毒性又は有害な効果よりも治療的に恩恵のある効果が上回るものである。「予防的有効量」とは所望の予防結果を達成するために必要な用量及び時間での効果的な量を意味する。典型的には、予防的用量は疾患の初期ステージ又はその前の患者に用いるので、予防的有効量は治療的有効量よりも少ないであろう。
本発明は、抗CD40抗体による治療に対するB細胞リンパ腫を有する患者の応答を評価するか又は応答を補助する方法を提供する。更に、本発明はB細胞リンパ腫を有する患者における抗CD40抗体治療への応答を予測するか又は治療/応答を監視する方法提供する。本発明は、抗CD40抗体による治療に適しているB細胞リンパ腫を有する患者を選択し、抗CD40抗体治療によってフォーローアップする方法を提供する。いくつかの実施態様において、方法は、患者から得られたBリンパ腫細胞を含む試料において表2−4、6、7及び13の何れかの1又は複数のマーカー遺伝子の発現レベルを決定すること;及び前記1又は複数のマーカー遺伝子の発現レベルの測定値に基づいて抗CD40抗体治療に対する患者の応答を予測するか、評価するかまたは評価の補助することを含む。いくつかの実施態様において、方法は、患者由来のBリンパ腫細胞を含む試料において表2−4、6、7及び13の何れかの1又は複数のマーカー遺伝子の測定された発現レベルをそれぞれのマーカー遺伝子の対照標準レベルと比較することを含む。

本願明細書に記載の方法は、マーカー遺伝子の測定された発現レベルと対照標準レベルとを比較する手順を含む。対照標準レベルは、マーカー遺伝子と異なる対照標準遺伝子の測定された発現レベル又は異なる試料の同じマーカー遺伝子の測定された発現レベルであってもよい。
ここで、表13に示す正の相関値を有する少なくとも1つのマーカー遺伝子及び負の相関値を有する少なくとも1つのマーカー遺伝子の発現レベルが測定され;

本発明で同定されるマーカー遺伝子が、1又は複数の抗CD40抗体による治療に対するB細胞リンパ腫の応答性を予測するか、評価するかまたは、評価の補助するために使われることができる。抗CD40抗体は、1又は複数のアゴニスト抗体(すなわち、CD40に結合して刺激する)であってもよい。例えば、刺激性抗体は、以下のような異なる種類のものでもよい:(1)CD40による刺激性のシグナルを送るが、CD40とCD40Lとの間の相互作用を増大させないもの(例えば、米国特許第5182368号に記載されているG28−5抗体及びG28−5に由来する抗体;及びPCT WO 96/18413)または、CD40とCD40Lとの間の相互作用を減少させるもの(例えば、米国特許第5674492号に記載されている抗体HuCD40−M2とHuCD40−M3とヒト化抗体);及び(2)CD40による刺激性シグナルを送り、CD40とCD40Lの間の相互作用を増加させることができるもの、例えばS2C6(Francisco et al., 2000, Cancer Res. 60: 3225-31)及びS2C6由来の抗体。また、アゴニスト抗体は米国特許第7288251号に記載されている。抗CD40抗体は、1又は複数のアンタゴニスト抗体(すなわち、CD40に結合しCD40Lによって誘発される活性を阻害する)であってもよい。アンタゴニスト抗CD40抗体の例は、米国特許公開番号2007/0110754に記載されているヒト抗体CHIR-12.12及びWO 97/31025に記載されている抗CD40抗体を含む。
上に記載又は示唆された適用における使用のために、キット又は製造品が本発明により提供される。当該キットは、本願明細書に記載のマーカー遺伝子の発現レベルを検出するために特異的な少なくとも1の試薬を含んでもよく、更に本願明細書に記載の方法を実施するための指示書を更に含んでもよい。
以下は、本発明の方法及び組成物の例である。上記の一般的説明を想定すれば、さまざまな他の実施態様が実施されてもよいと理解される。
材料と方法
細胞生存率測定
NHL細胞は、2%のFBSを添加したRPMI1640中の1500−5000細胞/ウェルの50ulを384枚のウェルプレートにまいて、連続的濃度の架橋結合した抗CD40Ab.1または対照抗体(抗−gD5B6で処理された。架橋結合するために、細胞に加える前に室温で30分間培地において1:4の割合で、抗CD40Ab.1または抗gDは、ヤギ抗ヒトFcK断片特異的抗体のF(ab’)2断片(Jackson ImmunoResearch, West Grove, PA)と共にインキュベートした。96時間のインキュベーション後、細胞生存度は、製造業者の指示書に従ってCellTiter-Glo発光細胞生存率測定(プロメガ、マディソン、WI)を使用して数値を求めた。各データポイントは、4重で実施された。
抗CD40Ab.1は、CD40に対するヒト化IgG1 mAbである。それは、遺伝的に改変されたチャイニーズハムスター卵巣(CHO)細胞株によって産生され、分泌される。実施例で使用され、抗CD40Ab.1と呼ばれる抗CD40Ab.1は、以下のアミノ酸配列を有する:

全RNAは、mirVanaTM miRNA単離キット(Ambion、オースティン、TX)で抽出され、Affymetrix HGU133P2全体ゲノム発現マイクロアレイを使用してアッセイされた。生データはAffymetrixスキャナを使用して抽出され、結果として生じるセル・ファイルはR Bioconductorパッケージ(bioconductor.orgのワールドワイド・ウェブ)のデフォルトのgcRMAを使用して処理された。有意に差異的に発現する遺伝子は、抗CD40Ab.1感受性及び生存度クラス全体について相違のための緩和されたt検定を使用して同定された。更なるパラメータはLIMAパッケージ及びt−統計値、p値、調製されたp値及びB−統計値が各遺伝子のために算出された。プローブは、各遺伝子に対してマップされ、遺伝子マッピングに対する1:1プローブが感受性の測定値に最も強く相関するプローブを使用した下流の解析のために選択された。耐性群に対して、感受性又は中間体を分類する場合、アッセイに含める遺伝子の節約的なセットを同定するために、定量的段階的線形モデリングは、標的経路の定性分析と組合せられた。更なる詳細及び結果を結果(表7)に示す。
標的遺伝子がインデックスへの算入のために選択される工程の順序で、各標的遺伝子をその対応する逆相関(反相関)ペア遺伝子(表7)と共に示す。最初の3つのメイン遺伝子(表7のVNN2、RGS13、CD22)は、感受性及び中間体細胞株における差異的過剰発現の優位な構成要素をモデル化するために、表2−4(工程1)から選択された。これらの3つの遺伝子の発現は、+0.77以上の相関係数を有し非常に相関している。それらの類似性のために、単一のペア遺伝子EPDR1は、耐性細胞株における過剰発現と対比して測定するために表2−4から選択された。当該反相関ペア遺伝子をアッセイに含むことは、感受性と耐性の両方がペアの1アームの高い発現を伴うという点で、自己正規化を提供する。この機構によって、アッセイは任意のクラスを定めるために低い全mRNAアッセイ・レベルに依存しないで、むしろ、それらの反相関ペアに対するメイン遺伝子の相対的発現のパターンによってそれぞれ記載される(すなわち、倍率変化推定に対応する符号付きlog2スケールにおける符号付きt−スコアの合計)。工程2−5において、遺伝子の更なるペアは、前工程において同定される遺伝子のための符号付きt−値の累積和の調整の後、有意な相関を有するそれらの新しいリストからIC25まで作用機構に基づいて選択された。このステップワイズ手順は、遺伝子の各々の新規なペアが更なる予測力を感受性インデックスに加えることを必要とする。工程5の後、どんな遺伝子ペアも、IC25予測のために必要とされなかった。工程6において、単一の更なるペアは、遺伝子の前の7ペアに基づいて累積的インデックスの調整後、最大阻害で細胞生存度を予測するその能力のために加えられた。BCL6は、作用機構の論理的根拠に基づく対応するペアのない単体として加えられた:それは現在最終的な感受性インデックスに取り入れられない。それは遺伝子ペア1−8のlog2−スケール発現のための符号付きt値の合計によって与えられる。それは、明白に、臨床経験に基づくインデックスに組み込まれることができる。耐性群に対して、感受性又は中間体を分類する場合、予備的カットオフは、全体的に正しい分類比を最大にするように、感受性インデックスのために選択された。選択されたプローブに基づく代替の分類規則は、臨床応用のために後に最適化されることができる。
抗CD40抗体の作用機序の理解をするために、及び抗CD40抗体療法に対するNHL患者の応答性のための1又は複数の予測マーカーを同定するために、31のNHL細胞株のパネルに渡って抗CD40Ab.1の活性を試験して、抗CD40抗体の力価に応答する細胞生存度を評価した。この実験から表1において強調したIC25値は、抗CD40抗体感受性化の10の細胞株は<0.4μg/mlの濃度において細胞生存度の減少があり(「感受性」細胞株として定義される)、および13の細胞株は1μg/mlの濃度に上げても細胞生存度の減少がなかった(「耐性」細胞株として定義する)ことを明らかにした。8の細胞株は、>0.4と<0.8間とのIC25を有しており、ここについて「中間体」細胞株とする。
臨床試験における抗CD40Ab.1による治療への応答性に関するマーカーの同定
臨床試験001(第II相)
全体の反応速度及び再発したDLBCLを有する患者の抗CD40Ab.1の毒性プロフィールを測定するための多施設、第II相、非盲検試験。腫瘍試料は、病理学確認とCD40発現のために中央研究室によって評価された。適格患者は診断でデノボまたは転換されたDLBCLを有しており、緩慢性リンパ腫の病歴がある場合は除外された。要求される前治療は、リツキシマブによる組合せ化学療法、適格であるならば自己由来の幹細胞移植からなる。患者は、5週(サイクル1)にわたって、抗CD40Ab.1の6回のIV注入を、投与量(日1に1mg/kg;日4に2mg/kg;日8に4 mg/kg)及びその後8mg/kg/wkにより受けた。応答している患者及びSD(安定した疾患)を有する患者は、疾患の進行または多くとも最高12サイクルまで治療を続ける資格があった。患者が抗CD40Ab.1による治療を受ける前に、腫瘍組織は患者から取り出された。例えば、試料は、ルーチンのリンパ腫診断の部分として取り出された。
多施設、マルチドース、第I相試験を、CD40Ab.1の再発したNHLを有する患者の静脈内抗安全性、薬物動態学的特性、免疫原性と抗腫瘍活性をテストするために実施した。NHLの複数の組織学的亜型を有する患者が本試験に含まれ、びまん性大B細胞(DLBCL;14)、濾胞性(FCL;9)、マントル細胞(MCL;9)、周縁帯(MZL;2)及び小リンパ球で(SLL;1)を含む。日1と日4に1mg/kgの抗CD40Ab.1、続いて週2−5の間に4つのコホートに対して最高投与量3、4、6、又は8mg/kgの漸増投与の投与スケジュールにより治療された。続いて、急速投与スケジュールは1つのコホートで試験された(サイクル1の間、投与されるトータル抗CD40Ab.1は40%増加)。応答患者及びSD(安定した疾患)患者は、抗CD40Ab.1のコホート特異的な最大量の4回の連続的な週ごとの注入からなる、第2サイクルに適格である。8人のDLBCL患者はサイクル1を終えて、37.5%の客観的な反応速度及び2SDで少なくとも3mg/kgの抗CD40Ab.1の最高投与量を受けた(すなわち、1CR及び2PR)。
更なる客観的反応は、1人のMCL(CR)患者と1人のMZL(PR)患者において見られた。これらの5人の患者の応答期間の中央値は、まだ到達されなかった(範囲8−37週)。抗Cd40Ab.1による治療を受ける前に、腫瘍組織は患者から取り出された。例えば、試料は、ルーチンのリンパ腫診断の一部として取り出された。
上記第I相及び第II相臨床試験由来のホルマリン固定されてパラフィン包埋された(FFPE)アーカイブの腫瘍組織は、適切なIRB承認と患者の同意を得て臨床検査所から得られた。腫瘍組織に由来する4−6ミクロン切片はガラススライドにマウントし、各ケースにつき1つのスライドは標準病理学実験室プロトコルを使用してH&E染色に供した。委員会が証明した病理学者が腫瘍内容についてH&Eスライドに印をつけ、FFPE組織のためのAmbion RecoverAllTM全核酸隔離キットを使用してRNA抽出のために残りの腫瘍含有領域を切り出すためのガイドとして使用した(Cat. No. AM1975; Applied Biosystems/Ambion, Austin, TX)。
qRT−PCRの生の結果は、正規化、変換及びインピュテーションにより、下記の説明に従って、予め前処理され、感受性インデックスは感受性インデックス及び分類子の下で記載されているように計算された。スピアマンの順位相関係数が、相関推定及び対応するP値のために使われた。多変量の感受性インデックスのために、プローブを選択し、係数はZhou et al.,Statist.Soc.B.67:301-320,2005によって記載されて、Friedman, Hastie and Tibshirani, Regularization Paths for Generalized Linear Models via Coordinate Descent.(スタンフォード大学統計学部テクニカル・レポート(www-stat.stanford.edu/~hastie/Papers/glmnet.pdf.))に実装されているように、ラッソ(L1)とリッジ(L2)の罰則付き回帰のelastic net blendを使用して推定した。Χ2テストが、分類変数の中の関連のためのテストに使われた。
以下は、アッセイ・データとモデル・パラメータのための定義である:
試料の対照標準(例えばインデックス係数及び分類子カットオフに適合させるために用いる)について、平均及び標準偏差モデル・パラメータは、(下の対照標準セットモデルパラメータの式に関連する)対照標準セットデータを使用して計算される。新規な試料(例えばインデックスとクラスが計算されることになっている単一の新規な試料)について、モデルパラメータは対照標準セット、l、から取り出されなければならず、それは新規な試料が抽出された集団の代表例となるように選択される。例えば、アッセイが使用される治療の各指標及びラインに対する臨床対照標準セットが維持されてもよい。対照標準セットモデルパラメーター及び変換され正規化されたアッセイ値のための式を以下にしめす。

下記の表11は、臨床試験001由来の臨床サンプル及びアッセイ化標本を説明する試料を提供する。24人のDLBCL患者由来の29の保存FFPE腫瘍標本が、qRT−PCR処理のために提出された。3人の患者は複数の標本を有しており、全24人の患者には少なくとも一つの標本に対し有用なqRT−PCR結果があった。これら24の中で、21が治験期間開始前及び最終観察時までの少なくとも1回の観察の両方の、腫瘍の生産物直径の合計(SPD)を有していた。
生のqRT−PCR結果は、アーカイブ標本を有する10人患者について成功裏に作成された。それらの10人の患者について、診断、治療群、多変量感受性インデックス、臨床応答及び治験開始前からのSPD変化を表16に示す。多変量の感受性インデックス加重は21人の臨床試験001患者(表14)から取られ、その結果これらの患者はセットされる非常に小さい検証セットを構成する。 感受性インデックス≧0を有する4人患者のうちの2人は抗CD40Ab.1暴露後腫瘍収縮を示し、感受性インデックス<0を有する6人の患者のうちの4人は腫瘍増加又はPDの最大応答を示した(SPDは2人の患者について入手出来なかったが、最高の臨床効果の結果がこの患者について入手できた)。
qRT−PCRアッセイは、BCL6遺伝子の第15プローブを含む。現在、多変量の感受性インデックスで使われていないが、それは以前に抗CD40Ab.1に対する応答性の潜在的予測因子として同定された。図8に示すように、合わせたDLBCL患者試料(P=0.25、N=26)のSPD変化は有意に相関しないが、腫瘍増加があった患者においてBCL6が低下する傾向がある(rho=−0.23)。
Claims (52)
- 抗CD40抗体治療に対するB細胞リンパ腫を有する被検体の応答性を予測するための方法であって、
(a)前記被検体から得られたBリンパ腫細胞を含む試料における1又は複数のマーカー遺伝子の発現レベルを測定する工程であって、前記1又は複数のマーカー遺伝子がIFITM1、CD40、RGS13、VNN2、LMO2、CD79B、CD22、BTG2、IGF1R、CD44、CTSC、EPDR1、UAP1及びPUS7からなる群から選択される工程;及び
(b)工程(a)から前記1又は複数のマーカー遺伝子の測定された発現レベルに基づいて抗CD40抗体治療に対して被検体が応答する可能性があるかについて予測する工程であって、
(i)抗CD40抗体治療はアゴニスト抗CD40抗体による治療であり、対照標準レベルと比較した、IFITM1、CD79B、IGF1R、CD44、CTSC、EPDR1及びPUS7の1又は複数の増大した発現は、前記被検体がアゴニスト抗CD40抗体治療に応答する可能性が低いことを示し、あるいは、
(ii)抗CD40抗体治療はアゴニスト抗CD40抗体による治療であり、対照標準レベルと比較した、CD40、RGS13、VNN2、LMO2、CD22、BTG2及びUAP1の1又は複数の増大した発現は、前記被検体がアゴニスト抗CD40抗体治療に応答する可能性が高いことを示す、工程
を含む方法。 - 測定された発現レベルが正規化される、請求項1に記載の方法。
- 請求項1の(i)に記載の対照標準レベルは、抗CD40抗体治療後に増大した腫瘍体積を有する被検体由来のBリンパ腫細胞を含む試料における、対応するマーカー遺伝子の発現レベルに基づいて決定される、請求項1に記載の方法。
- 対照標準レベル決定用の被検体由来の試料は、抗CD40抗体治療に対する応答性が予測される被検体由来の試料と同タイプのBリンパ腫細胞を含む、請求項3に記載の方法。
- 請求項1の(ii)に記載の対照標準レベルは、抗CD40抗体治療後に減少した腫瘍体積を有する被検体由来のBリンパ腫細胞を含む試料における、対応するマーカー遺伝子の発現レベルに基づいて決定される、請求項1に記載の方法。
- 対照標準レベル決定用の被検体由来の試料は、抗CD40抗体治療に対する応答性が予測される被検体由来の試料と同タイプのBリンパ腫細胞を含む、請求項5に記載の方法。
- アゴニスト抗CD40抗体はCD40を刺激し、CD40とCD40リガンドとの間の相互作用を促進する、請求項1から6の何れか一項に記載の方法。
- アゴニスト抗CD40抗体は、配列番号1に示す重鎖アミノ酸配列と配列番号2に示す軽鎖アミノ酸配列とを含む、請求項7に記載の方法。
- アゴニスト抗CD40抗体はCD40を刺激し、CD40とCD40リガンドとの間の相互作用を促進も阻害もしない、請求項1から6の何れか一項に記載の方法。
- 少なくとも2、少なくとも3、少なくとも4、少なくとも5、少なくとも6、少なくとも7、少なくとも8、少なくとも9、少なくとも10、少なくとも11、少なくとも12、少なくとも13又は14のマーカー遺伝子の発現レベルが測定される、請求項1から9の何れか一項に記載の方法。
- IFITM1、RGS13、CD79B、CD22、BTG2、CD44、EPDR1及びUAP1の発現レベルが測定される、請求項10に記載の方法。
- IFITM1、CD22、IGF1R、CD44、EPDR1及びUAP1の発現レベルが測定される、請求項10に記載の方法。
- B細胞リンパ腫がびまん性大細胞型B細胞リンパ腫(DLBCL)である、請求項1から12の何れか一項に記載の方法。
- B細胞リンパ腫が非ホジキンリンパ腫である、請求項1から12の何れか一項に記載の方法。
- 非ホジキンリンパ腫が、ろ胞性リンパ腫、マントル細胞リンパ腫、周縁帯リンパ腫又は小リンパ球性リンパ腫である、請求項14に記載の方法。
- Bリンパ腫細胞を含む試料はホルマリン固定されてパラフィン包埋された生検試料である、請求項1から15の何れか一項に記載の方法。
- 1又は複数のマーカー遺伝子の発現レベルが1又は複数のマーカー遺伝子のRNA転写産物のレベルによって測定される、請求項1から16の何れか一項に記載の方法。
- RNA転写産物はqRT-PCRによって測定される、請求項17に記載の方法。
- 1又は複数のマーカー遺伝子の発現レベルは、1又は複数のマーカー遺伝子のタンパク質発現レベルによって測定される、請求項1から16の何れか一項に記載の方法。
- 前記方法はBCL6の発現レベルを測定することを更に含んでなり、前記1又は複数のマーカー遺伝子の測定された発現レベル及びBCL6の測定された発現レベルに基づいて、被検体が抗CD40抗体治療に対して応答する可能性が高いかどうかが予測される、請求項1から19の何れか一項に記載の方法。
- 対照標準レベルと比較したBCL6のより高い発現レベルは、被検体が抗CD40抗体治療に対して応答する可能性が高いことを示す、請求項20に記載の方法。
- 前記方法はBCL6の発現レベルを測定することを更に含んでなり、対照標準レベルと比較したBCL6のより高い発現レベルは、被検体が抗CD40抗体治療に対して応答する可能性が高いことを示す、請求項1から21の何れか一項に記載の方法。
- 対照標準レベルは、抗CD40抗体治療後に減少した腫瘍体積を有する被検体由来のBリンパ腫細胞を含む試料における、BCL6の発現レベルに基づいて決定される、請求項21又は22に記載の方法。
- 抗CD40抗体治療に対するB細胞リンパ腫を有する被検体の応答性を予測するための個人用のゲノミクス・プロフィールを調製する方法であって、
(a)IFITM1、CD40、RGS13、VNN2、LMO2、CD79B、CD22、BTG2、IGF1R、CD44、CTSC、EPDR1、UAP1、PUS7及びBCL6からなる群から選択される1又は複数のマーカー遺伝子の発現レベルを前記被検体から得られたBリンパ腫細胞を含む試料において測定する工程;および
(b)工程(a)から得られた1又は複数のマーカー遺伝子の発現レベルをまとめた報告書であって、被検体用の抗CD40抗体治療の推奨事項を含んでなる報告書を作成する工程であって、
(i)抗CD40抗体治療はアゴニスト抗CD40抗体による治療であり、対照標準レベルと比較した、IFITM1、CD79B、IGF1R、CD44、CTSC、EPDR1及びPUS7の1又は複数の増大した発現は、前記被検体がアゴニスト抗CD40抗体治療に応答する可能性が低いことを示し、あるいは、
(ii)抗CD40抗体治療はアゴニスト抗CD40抗体による治療であり、対照標準レベルと比較した、CD40、RGS13、VNN2、LMO2、CD22、BTG2及びUAP1の1又は複数の増大した発現は、前記被検体がアゴニスト抗CD40抗体治療に応答する可能性が高いことを示す、工程
を含む方法。 - 発現レベルが正規化される、請求項24に記載の方法。
- 少なくとも2、少なくとも3、少なくとも4、少なくとも5、少なくとも6、少なくとも7、少なくとも8、少なくとも9、少なくとも10、少なくとも11、少なくとも12、少なくとも13、少なくとも14又は15のマーカー遺伝子の発現レベルが測定される、請求項24又は25に記載の方法。
- IFITM1、RGS13、CD79B、CD22、BTG2、CD44、EPDR1及びUAP1の発現レベルが測定される、請求項26に記載の方法。
- IFITM1、CD22、IGF1R、CD44、EPDR1及びUAP1の発現レベルが測定される、請求項26に記載の方法。

- 対照標準レベル決定用の被検体由来の試料は、個人用ゲノミクス・プロフィールが調整される被検体由来の試料と同タイプのBリンパ腫細胞を含む、請求項31に記載の方法。

(a)被検体由来のBリンパ腫細胞を含む試料において、IFITM1、CD40、RGS13、VNN2、LMO2、CD79B、CD22、BTG2、IGF1R、CD44、CTSC、EPDR1、UAP1及びPUS7から成る群から選択される少なくとも2つのマーカー遺伝子の発現レベルを測定する工程、
(b)以下の式
によって、工程(a)におけるマーカー遺伝子の測定された発現レベルに基づいて感受性インデックス値(SI)を計算する工程であって、
IFITM1、CD79B、IGF1R、CD44、CTSC、EPDR1及びPUS7から選択される正の相関値を有する少なくとも1つのマーカー遺伝子及び、CD40、RGS13、VNN2、LMO2、CD22、BTG2及びUAP1から選択される負の相関値を有する少なくとも1つのマーカー遺伝子の発現レベルが測定される工程;
を含んでなり、
(i)βjは測定された各マーカー遺伝子のための係数値であり;(ii)pは測定されたマーカー遺伝子の数であり;(iii)χjは被検体由来の試料の、測定された各マーカーの発現レベルについて、変換され正規化された発現レベルであり;及び(iv)μj及びσjは測定された各マーカー遺伝子の平均及び標準偏差であり;ここで、βj、μj及びσjは臨床試験由来のBリンパ腫細胞を含む患者試料から測定されたものであり;
感受性インデックスのゼロ以上の値は被検体が抗CD40抗体治療に対し応答する可能性が高いことを示し、又は感受性インデックスのゼロ未満の値は被検体が抗CD40抗体治療に対し応答する可能性が低いことを示す、方法。 - 少なくとも3、少なくとも4、少なくとも5、少なくとも6、少なくとも7、少なくとも8、少なくとも9、少なくとも10、少なくとも11、少なくとも12、少なくとも13又は14のマーカー遺伝子の発現レベルが測定され、感受性インデックス計算のために使用される、請求項43に記載の方法。
- IFITM1、RGS13、CD79B、CD22、BTG2、CD44、EPDR1及びUAP1の発現レベルが測定され、感受性インデックス計算のために使用される、請求項43に記載の方法。
- βj、μj及びσjは抗CD40抗体治療に対する応答性を予測される被検体由来の試料と同タイプのBリンパ腫細胞を有する患者試料から測定された、請求項43に記載の方法。
- 被検体由来のBリンパ腫細胞を含む試料において、IFITM1、CD79B、IGF1R、CD44、CTSC、EPDR1、PUS7、CD40、RGS13、VNN2、LMO2、CD22、BTG2及びUAP1からなる群から選択される少なくとも1つのマーカー遺伝子の発現レベルを測定するための試薬、及び
測定される1又は複数のマーカー遺伝子の発現レベルに基づいて、B細胞リンパ腫を有するヒト被検体が抗CD40抗体治療に応答する可能性が高いかどうかを評価するための指示書を含む、キット。 - 試薬が、qRT-PCRによって各マーカー遺伝子の発現レベルを検出するための少なくとも1対のプライマー及びプローブを含む、請求項47に記載のキット。
