Description
ここで、「駆動時間」には、駆動タイミングをも含む。
なお、「磁力部」は、前記チップ脱落防止部と共通のモータで駆動することで、装置構造を簡単化し、装置規模の拡大を抑制し、その製造費用を削減することができる。
ステップS1において、12人の被験者から全血を採取し、予め、前記担体・流体封入分注チップ処理装置のノズルに分注チップを装着してまたは他の分注装置を用いて20μリットルを吸引して、前記担体・流体封入分注チップ処理装置10の収容部群127の液収容部178の中のY軸方向に沿って一列状に配列された12個のウェルw1に分注しておく。次に、希釈用バッファ、例えば、Tris バッファで処理された食塩水、Ph7.4(TBS)を該ウェルw1に分注して全体として500μリットルに希釈する。

Claims (15)
- 気体の吸引吐出が行われるとともに、種々の液体を収容しまたは収容可能な液収容部を有する収容部群に相対的に移動可能なノズルに装着しまたはノズルに装着可能な連結管に装着して前記ノズルと連通可能な装着用開口部、および前記気体の吸引吐出によって液体の流入および流出が可能な口部を有する分注チップと、該分注チップ内に封入され、前記液体中の生体物質を吸着もしくは捕獲しまたは該生体物質と反応もしくは結合することが可能な担体と、前記分注チップ内に開封可能な状態で封入され該担体と接触する所定の液体または所定の気体からなる担体活性化維持用流体とを有するとともに、
前記装着用開口部は、穿孔可能な被穿孔部材または着脱可能な蓋部材によって閉塞されることによって、または、前記被穿孔部材をその軸方向を横断するように設けた前記連結管に装着されることで閉塞され、前記口部は、着脱可能なキャップによって閉塞されることで、分注チップ内に前記担体活性化維持用流体が開封可能な状態で封入された担体活性化維持機能をもった担体・流体封入分注チップ。
- 前記分注チップは、太管と、前記太管と連通し該太管の下側に設けられ、前記太管より細く形成された細管と、太管と細管との間の移行部とからなり、前記装着用開口部は前記太管の上側に設けられ、前記口部は前記細管の先端に形成された請求項1に記載の担体・流体封入分注チップ。
- 前記担体を該分注チップ内に封入する封入部を該分注チップに設け、該封入部は、前記装着用開口部に設けた前記被穿孔部材若しくは前記蓋部材または装着した前記連結管に設けた前記被穿孔部材と、前記口部に設けた前記キャップとの間に挟まれた領域内に設けられ、該分注チップ内に流入した液体および封入された前記担体活性化維持用流体が通過可能である請求項1または請求項2のいずれかに記載の担体・流体封入分注チップ。
- 前記封入部は、流入した前記液体または封入した前記担体活性化維持用流体と接触可能となるように前記分注チップの前記口部と前記装着用開口部との間を仕切るように分注チップとは別体に設けた1または2以上の担体通過阻止部材を有する請求項3に記載の担体・流体封入分注チップ。
- 前記封入部は、前記装着用開口部と前記口部との間で前記分注チップの内壁面を仕切るように、該内壁面に、内側方向に突出する突出部、口部に向かって先細りの傾斜面または口部に向かって内側方向に突設する段差を有する請求項4に記載の担体・流体封入分注チップ。
- 気体の吸引吐出を行う1または複数連のノズルを有するノズルヘッドと、該ノズルを介して気体の吸引吐出を行う吸引吐出機構と、前記ノズルまたはノズルと装着可能な連結管に装着してノズルと連通しまたは連通可能であって、液体中の生体物質の吸着もしくは捕獲、または該生体物質との反応もしくは結合が可能な担体を封入し、かつ該担体と接触する所定の液体または所定の気体からなる担体活性化維持用流体を開封可能状態で封入した担体活性維持機能をもった1または2以上の担体・流体封入分注チップと、前記担体・流体封入分注チップを収容しまたは収容可能なチップ収容部および種々の液体を収容しまたは収容可能な液収容部を設けた収容部群と、前記ノズルヘッドを前記収容部群に相対的に移動させる移動手段とを有するとともに、
前記担体・流体封入分注チップは、前記ノズルに装着しまたは前記連結管に装着して前記ノズルと連通可能な装着用開口部、および前記気体の吸引吐出によって液体の流入および流出が可能な口部を有する分注チップと、該分注チップ内に封入された前記担体と、前記分注チップ内に開封可能状態で封入され、前記担体と接触する前記所定の液体または前記所定の気体からなる担体活性化維持用流体とを有し、前記担体・流体封入チップの装着用開口部は、穿孔可能な被穿孔部材または着脱可能な蓋部材によって閉塞され、または、穿孔可能な被穿孔部材によって軸方向を横断するように閉塞された前記連結管に装着され、前記口部が着脱可能なキャップによって閉塞されることで、分注チップ内に前記担体活性化維持用流体が開封可能状態で封入された担体・流体封入分注チップ処理装置。
- 前記ノズルヘッドには、前記被穿孔部材を穿孔するための1または複数の穿孔用針を有し、該穿孔用針は、該ノズルヘッドに設けられた1または複数連の前記ノズルの連数に等しい連数およびノズル間の相互の配列間隔に等しい相互の配列間隔をもつように設けられた請求項6に記載の担体・流体封入分注チップ処理装置。
- 前記ノズルヘッドに設けられた担体・流体封入分注チップに対しノズルを通して所定圧力で加圧する際に、該担体・流体封入分注チップと係合して担体・流体封入分注チップを支持しノズルからの担体・流体封入分注チップの脱落を防止するチップ脱落防止部を前記ノズルヘッドに設けた請求項6または請求項7のいずれかに記載の担体・流体封入分注チップ処理装置。
- 前記ノズルヘッドに設けた前記担体・流体封入分注チップを前記ノズルから脱着するためのチップ脱着部をノズルヘッドに設けた請求項6、請求項7または請求項8のいずれかに記載の担体・流体封入分注チップ処理装置。
- 前記吸引吐出機構、および前記チップ脱着部または/および穿孔用針駆動部は、同一のモータを共用して駆動される請求項9に記載の担体・流体封入分注チップ処理装置。
- 前記担体・流体封入分注チップ処理装置は、穿孔用針の穿孔動作、チップ脱着部またはチップ脱落防止部を有するとともに、前記穿孔用針の穿孔動作、前記チップ脱着部または前記チップ脱落防止部の駆動時間または位置を物質条件、及び、処理内容に基づいて制御する請求項6乃至請求項10のいずれかに記載の担体・流体封入分注チップ処理装置。
- 前記担体・流体封入分注チップ処理装置は、前記ノズルの下方において、ノズルの軸方向に垂直方向に沿って磁場を及ぼしかつ除去することが可能な磁力部が設けられた請求項6乃至請求項11のいずれかに記載の担体・流体封入分注チップ処理装置。
- 気体の吸引吐出を行うとともに種々の液体を収容しまたは収容可能な液収容部を有する収容部群に相対的に移動可能な1または2以上のノズルに装着しまたは該ノズルに装着可能な連結管に装着して前記ノズルと連通しまたは連通可能な装着用開口部、および前記気体の吸引吐出によって液体の流入および流出が可能な口部を有する分注チップ内に、前記液体中の生体物質の吸着もしくは捕獲、または該生体物質との反応もしくは結合が可能な担体が封入され、かつ該担体と接触する所定の液体または所定の気体からなる担体活性化維持用流体が開封可能な状態で封入された担体活性化維持機能をもった1または2以上の担体・流体封入分注チップについて、前記装着用開口部をまたは該装着用開口部に装着された前記連結管を前記ノズルに装着する工程と、装着した前記担体・流体封入分注チップを所定の液収容部に移動し、液収容部に収容されている液体の吸引または吐出によって前記担体と接触させて、該液体中の前記生体物質の吸着若しくは捕獲、または該生体物質との反応若しくは結合を行わせる接触工程と、を有するとともに、
前記装着工程は、穿孔可能な被穿孔部材または着脱可能な蓋部材によって閉塞され、または穿孔可能な被穿孔部材によって軸方向に横断するように閉塞された連結管に装着され、前記口部が着脱可能なキャップによって閉塞された前記担体・流体封入分注チップに対して、前記担体活性化維持用流体について開封状態にする工程を含む担体・流体封入分注チップ処理方法。
- 前記担体に結合した物質を前記担体から解離する解離工程をさらに有する請求項13に記載の担体・流体封入分注チップ処理方法。
- 前記担体・流体封入分注チップを前記ノズルから脱着した後、または担体・流体封入分注チップを前記ノズルに装着する前に該ノズルに分注チップを装着する工程と、磁性粒子が懸濁する液体を分注する工程と、該分注チップ内に外部から磁場を及ぼすことによって、該分注チップの内壁に磁性粒子を吸着させる工程をさらに有する請求項13または請求項14のいずれかに記載の担体・流体封入分注チップ処理方法。
