Description
The present invention relates to a needle sheet and a method for manufacturing the needle sheet, and more particularly to a needle sheet having a high aspect ratio needle used as a transdermal absorption system in the medical technical field and a method for manufacturing the needle sheet.
In recent years, functional films having a high aspect ratio structure formed on the surface have been applied in various fields. For example, in the medical technology field, a percutaneous absorption system that efficiently administers a drug to a patient via the patient's skin surface or skin stratum corneum by using a functional membrane having a high aspect ratio structure attracts attention. ing.
As a high aspect ratio structure for a transdermal absorption system, sheets having various outer shapes such as a needle shape, a tip knife shape, and an arrowhead shape have been proposed. These sheets are conventionally manufactured by injection molding (see, for example, Patent Document 1). However, when a drug is kneaded into a high aspect ratio structure, it is manufactured by cast molding capable of low temperature molding from the viewpoint of preventing its deterioration. In particular, in order to produce a sheet having a high aspect ratio structure having no defects up to the tip, a cast imprint in which a cast material is applied to a mold (mold) having an inverted structure, filled in the mold, and dried and peeled off is preferable (Patent Document) 2).
However, it is very difficult to manufacture a sheet having a constricted needle, and it is particularly difficult to manufacture a sheet in which needles are arranged three-dimensionally. Specifically, when a sheet having a constricted needle is manufactured by injection molding, a mold having an inverted constricted shape is required, but the needle molded in the mold cannot be removed from the mold. The problem occurs. For this reason, in the case of manufacturing a sheet having a constricted needle, it is necessary to manufacture only the constricted needle in advance and arrange the needles on the sheet, resulting in poor productivity.
Further, the constricted needle can prevent the needle from being pulled back due to the elasticity of the skin, but there is also a problem that the needle is easily broken.
This invention is made | formed in view of such a situation, and it aims at providing the needle seat provided with the needle which has a constriction and is hard to bend. Another object of the present invention is to provide a method for manufacturing a needle sheet that can easily manufacture a sheet having a needle having a constriction.
In order to achieve the above object, the present invention is applied in the application step of applying a polymer solution having a gelatin concentration of 10 % or more and 20% or less to a mold having a needle-like recess having a width of 400 μm, and the application step. Further, by constricting the polymer solution to dry shrinkage, a constricted shape is formed inside the acicular recess so that a polymer sheet having a base film thickness of 170 ± 20 (μm) or more and 300 ± 20 (μm) or less is formed. The needle sheet is manufactured through a drying shrinkage step for forming the needle and a peeling step for peeling the sheet having the needle formed in the drying shrinkage step from the mold. I will provide a.
The inventor of the present invention has obtained knowledge that a constricted needle is formed inside a needle-like recess by drying and shrinking a polymer solution applied to the needle-like recess of the mold. The present invention has been made on the basis of such knowledge, and by applying a polymer solution to the needle-like concave portion of the mold and drying and shrinking it, a needle sheet having a curved constricted shape can be easily and highly accurate. Can be manufactured.
In the present invention, the mold preferably contains silicon. According to the present invention, due to the property of silicon having good peelability, the sheet can be peeled from the mold without damaging the sheet in the peeling step.
In this invention, it is preferable that the said provision process adjusts the film thickness of the said polymer solution according to the shape of the said acicular recessed part. According to the present invention, a needle having a curved constricted shape can be reliably formed by adjusting the film thickness of the polymer solution in accordance with the shape of the needle-like recess.
In the present invention, the polymer solution preferably contains a drug. According to the present invention, a needle sheet containing a drug can be produced as a transdermal absorption system.
In the present invention, the polymer solution preferably includes a polymer of a biodegradable material.
According to the present invention, it is possible to easily and accurately manufacture a needle sheet having a curved constriction that does not easily peel off or shift while leaving a needle when it is attached to the skin or the like.
Embodiments of the present invention will be described below with reference to the accompanying drawings. First, a needle seat manufacturing method according to an embodiment of the present invention will be described.
Fig.1 (a)-FIG.1 (d) have shown the manufacture process of the needle seat of this Embodiment. 1A shows a state before the applying step, FIG. 1B shows a state of the applying step, FIG. 1C shows a state of the drying shrinkage step, and FIG. d) shows the state of the peeling process.
When manufacturing a needle sheet, first, a
The arrangement of the needle-
The
First, a
As the
The polymer of the
In the application step of FIG. 1B, the film thickness t1 formed on the surface of the
After the application step, a drying shrinkage step is performed as shown in FIG. The drying shrinkage step is a step of shrinking the
By drying the
By this drying shrinkage process, a
By the manufacturing method described above, the
As shown in the figure, the
The diameter of the
As an example of a preferable shape of the
On the other hand, the
In order to form the
When the
Furthermore, in the present embodiment, since the
Note that at least one
Further, the shape of the
Further, in the above-described embodiment, the needle-
Further, the needle-like
In the above-described embodiment, all the
The
Next, an example of a method for producing the
The
A
As mentioned above, although the example of the manufacturing method of the
For example, after applying the
FIG. 6 is a view showing a pressure filling device that promotes filling of the polymer solution into the needle-like concave portion. A
In a state where the
As the pressurized fluid, gas or liquid can be used. Examples of the gas that can be used as the pressurized fluid include air. From the viewpoint of preventing dissolution of the pressurized fluid in the
From the viewpoint of preventing an increase in viscosity due to volatilization of the
In this way, by promoting the filling of the

[Example 1]
<Mold production>
First, a wire-like metal having a diameter of 400 μm was used, and a 1000 μm tip portion thereof was ground into a conical shape having a tip curvature radius of 5 μm. And after making a hole in the central part of a smooth Cu plate of 40 mm x 40 mm, the tip of the wire-like metal machined at the tip is protruded 2000 μm from the hole, and 5 × 5 is arranged three-dimensionally at a pitch of 1000 μm. Fixed and used as the master. By using this master, a transfer product inverted with silicon rubber (RTV rubber for Shin-Etsu silicone mold making) is produced, and the outside of the 40 mm flat part including the 5 × 5 array hole is cut off at the center, and the thickness is 5 mm. What was done was used as a mold.

[Comparison study of Examples and Comparative Examples]
The obtained needle seat was evaluated. The results are shown in [Table 1].
As can be seen from Table 1, the film thickness of the polymer sheet varies depending on the gelatin concentration and the amount of the polymer solution dropped. When the film thickness of the polymer sheet is 250 ± 20 μm (Example 1) or 300 ± 20 μm (Examples 2 and 3), the constricted needle can be formed, while the film thickness of the polymer sheet is 150 ± 20 μm. In the case of 100 ± 20 μm, it is understood that a constricted needle cannot be formed. It can also be seen that the diameter φ2 of the
[Conditions for forming a constricted needle]
The conditions under which a constricted needle was formed were determined. According to the method of Example 1, four types of polymer solutions having gelatin concentrations of 5% by weight, 10% by weight, 15% by weight and 20% by weight, respectively, were prepared and poured into molds with various dropping amounts. As in Example 1, a mold having a conical columnar concave pattern in which a tapered portion is a cone having a bottom diameter of 400 μm and a height of 1000 μm, and a root portion is a cylinder having a diameter of 400 μm and a height of 1000 μm is used. It was.
The film thickness (base film thickness) of the produced polymer sheet and the width of the needle (maximum width in the case of a constricted shape) were measured. The result is shown in FIG. FIG. 7 shows the relationship between the base film thickness and the needle width. In the drawing, “non-hollow” means a constricted
As can be seen from FIG. 7, when the base film thickness is smaller than the predetermined value (about 170 μm), the needle width is substantially the same as the width of the conical columnar recess (400 μm), so that a hollow needle is formed. I understand. On the other hand, when the basal film thickness is larger than a predetermined value (about 170 μm), the needle width is smaller than the width of the conical columnar recess, so that it is understood that a constricted needle is formed by contraction. When the base film thickness was around 170 μm, hollow needles and constricted needles were mixed.
