アデノウイルス感染細胞の活動を制御している主な免疫担当細胞は細胞傷害性T細胞(Cytotoxic T cell ; CTL、以下CTLと称する)である。CTLはアデノウイルス感染細胞を発見し、それを破壊する能力を持っているため、その機能を有効に活用すれば、アデノウイルス関連疾患の新しい診断と治療法の開発につながる可能性が高いと考え本発明技術の開発の糸口とした。

以下に、エピトープの候補ペプチドの選択方法についてその一例を記載する。
本発明のアデノウイルスに特異的なCTLエピトープペプチドは、アデノウイルスタンパク質のアミノ酸配列について、目的とするHLA分子の各結合モチーフを有する8〜10個のアミノ酸よりなるエピトープペプチドを検索し得る、インターネット上に公開されている複数のソフトウエアー(Pingping Guan, Irini A. Doytchinova, Christianna Zygouri, and Darren R. Flower, MHCPred: a server for quantitative prediction of peptide-MHC binding, Nucleic Acids Res., 2003;31:3621-3624)に照合してCTLエピトープの候補ペプチドを選択する事ができる。
HLAクラスI分子は、主としてHLA-A、HLA-B、HLA-Cがあり、これらに結合して提示されるエピトープペプチドは、8〜10個のアミノ酸からなる。エピトープペプチドのN末端側から2番目と、9あるいは10番目のアミノ酸はHLA分子との結合に対して最も重要なアミノ酸であり、アンカーモチーフと呼ばれている。このアンカーモチーフは、各々のHLA分子の種類によって異なることが報告されている。例えば、世界的に最も頻度の高いHLA-A2分子に結合するペプチドとしては、N末端より2番目の位置にLeu が配置され、9あるいは10番目の位置にLeu又はValが配置されたペプチドであって、9〜10個のアミノ酸残基からなるペプチドが最も良く知られている(T Sudo, N Kamikawaji, A Kimura, Y Date, CJ Savoie, H Nakashima, E Furuichi, S Kuhara, and T Sasazuki, Differences in MHC class I self peptide repertoires among HLA-A2 subtypes, J. Immunol., 1995;155:4749-4756)。また、日本人を含むアジアの人種に多いHLA-A24に結合するペプチドとしては、N末端より2番目の位置にTyr、Phe、Met又はTrpのいずれかが配置され、9あるいは10番目の位置にLeu、Ile、Trp又はPheのいずれかが配置されたペプチドであって、9〜10個のアミノ酸からなるペプチドが最もよく知られている(A Kondo, J Sidney, S Southwood, MF del Guercio, E Appella, H Sakamoto, E Celis, HM Grey, RW Chesnut, and RT Kubo, Prominent roles of secondary anchor residues in peptide binding to HLA- A24 human class I molecules, J. Immunol., 1995;155:4307-4312)。蛋白質のアミノ酸配列中からこのアンカーモチーフを有する配列を検索し、CTLエピトープの候補ペプチドを選択する事ができる。
アデノウイルス蛋白質の蛋白質全体を網羅する20個前後のアミノ酸よりなるペプチドライブラリーを合成する。20個前後のアミノ酸のうち、前後10個程度のアミノ酸は、前後のペプチド配列と重複するようにライブラリーを作製する。これによって、蛋白質全体を網羅的に検索する事ができ、一度ライブラリーを作製すればHLA拘束性も網羅的に検討する事が可能になる。
アデノウイルス感染歴がある人から分離した末梢血単核球(peripheral blood mononuclear cells;PBMC)、あるいはPBMCから分離したT細胞を適切な培地に0.1〜2×106/mL の細胞濃度で浮遊させる。これに同じ人からあらかじめ分離培養しておいたアデノウイルス感染細胞1×105/mLを加え、5% 炭酸ガス(CO2)恒温槽にて37℃で7日間培養する。培養7日後にアデノウイルス感染細胞とインターロイキン2(IL-2)を添加し、以後、アデノウイルス感染細胞とIL-2による刺激を毎週繰返すことによりCTLを誘導する。このようにして誘導したCTLがエピトープ候補ペプチドに対して特異性があるかどうかの検討は、MHC−テトラマー法、エリスポットアッセイ、クロムリリースアッセイ、細胞内サイトカイン染色法等で判定する(Current Protocols in Immunology, Edited by: John E. アデノウイルス感染歴がある人から分離したPBMCを適切な培地に0.1〜2×106/mL の細胞濃度で浮遊させ、これにエピトープ候補ペプチドの任意の1種を0.01〜100μg/mLの濃度で加える。5% CO2恒温槽にて37℃で培養し、2日後にIL-2を添加する。以後、前記ペプチドとIL-2による刺激を週に1度あるいは2週間に1度繰返すことによりCTLを誘導する。このようにして誘導したCTLがエピトープ候補ペプチドに対して特異性があるかどうかの検討は、MHC−テトラマー法、エリスポットアッセイ、クロムリリースアッセイ、細胞内サイトカイン染色法等で判定する。
アデノウイルス感染歴がある人から分離したPBMCを適切な培地に0.1〜2×106/mL の細胞濃度で浮遊させ、これに合成したペプチドライブラリーを適当な数(例えば10種類ずつ)にプールした物を加える。5% CO2恒温槽にて37℃で培養し、2日後にIL-2を添加する。以後、プールペプチドとIL-2による刺激を週に1度あるいは2週間に1度繰返すことにより、CTLを誘導する。このようにして誘導したCTLがエピトープ候補ペプチドに対して特異性があるかどうかの検討は、エリスポットアッセイ、クロムリリースアッセイ、細胞内サイトカイン染色法等で判定する。良好な結果を示したプールペプチドに対しては、1種類ずつペプチドを加えて上記実験を繰返せば、CTL誘導能を有するペプチドを選択する事が可能である。反応したペプチドについて順次短くし、基幹のペプチドを得て本発明のエピトープペプチドとする。一般的に、エピトープペプチドは最終的に8〜10個のアミノ酸までに短くすることができる。
また本発明のペプチドは、配列番号:1〜6のいずれかに記載のアミノ酸領域を含むペプチドであってもよい。即ち、本発明のペプチドの好ましい態様としては、アデノウイルスに特異的な細胞傷害性T細胞エピトープペプチドが配列番号:1〜6からなる群から選択される少なくとも1つのアミノ酸配列を含むペプチドである。
即ち本発明の好ましい態様としては、配列番号:1〜6のいずれかに記載のアミノ酸配列において、1もしくは複数のアミノ酸が置換、欠失、挿入及び/又は付加されたアミノ酸配列からなるペプチドであって、改変前の配列番号:1〜6のいずれかに記載のペプチドと同等の機能を有するペプチドを挙げることができる。

本発明のペプチドをコードする上記核酸もしくはベクターもまた、本発明に含まれる。
1. 適当な培養液中で、抗原提示細胞とCTLエピトープペプチドを30分から1時間混合したエピトープペプチドパルス抗原提示細胞、
2. CTLエピトープペプチドをコードする核酸を用い、遺伝子導入等で抗原提示細胞にCTLエピトープペプチドを提示された細胞、
3. 人工的に作製した抗原提示能を有する人工抗原提示細胞、等を意味する。
以下にCTLの調製方法の具体例を記載するが、必ずしもこれらの方法に制限されない。
PBMCと、適当な濃度のアデノウイルス特異的MHC−テトラマー試薬を反応させる。MHC−テトラマー試薬と結合したアデノウイルス特異的CTLは標識色素により染色されるので、セルソーター、顕微鏡などを用いて染色されたCTLのみを単離する。このようにして単離されたアデノウイルスに特異的なCTLは、抗CD3抗体、PHA、IL-2等のT細胞刺激薬剤や、X線照射あるいはマイトマイシン処理等で増殖能を損失させた抗原提示細胞で刺激増殖させ、受動免疫療法に必要な細胞数を確保する。
アデノウイルス特異的MHC−モノマー及び/又はMHC−テトラマー試薬を無菌プレートなどに固相化し、PBMCを固相化プレートで培養する。プレートに固相化されたMHC−モノマー及び/又はMHC−テトラマーに結合したアデノウイルス特異的CTLを単離するためには、結合せずに浮遊している他の細胞を洗い流した後に、プレート上に残った抗原特異的CTLだけを新しい培地に懸濁する。このようにして単離されたアデノウイルス特異的CTLは、抗CD3抗体、PHA、IL-2等のT細胞刺激薬剤や、X線照射あるいはマイトマイシン処理等で増殖能を損失させた抗原提示細胞で刺激増殖させ、受動免疫療法に必要な細胞数を確保する。
アデノウイルス特異的MHC−モノマー及び/又はMHC−テトラマー試薬と、CD80、CD83、CD86等の共刺激分子か、もしくは、共刺激分子と結合するT細胞側のリガンドであるCD28に対してアゴニスティックに作用する抗体等を無菌プレートなどに固相化し、PBMCを固相化プレートで培養する。2日後にIL-2を培地に添加し5% CO2恒温槽にて37℃で7〜10日培養する。培養した細胞を回収し新たな固相化プレート上で培養を続ける。この操作を繰り返す事で受動免疫療法に必要な細胞数のCTLを確保する。
PBMCあるいはT細胞を本発明のCTLエピトープペプチドで直接刺激するか、該ペプチドをパルスした抗原提示細胞、遺伝子導入した抗原提示細胞、又は人工的に作製した抗原提示能を有する人工抗原提示細胞で刺激する。刺激によって誘導されたCTLを5% CO2恒温槽にて37℃で7〜10日培養する。CTLエピトープペプチドとIL-2、又は抗原提示細胞とIL-2による刺激を週に1度繰り返す事で受動免疫療法に必要な細胞数のCTLを確保する。
アデノウイルス特異的MHC−テトラマー試薬と、CTL調製方法にて誘導されたCTLを反応させ、MHC−テトラマーを標識している標識色素に対する抗体等を磁気標識した2次抗体を用いて分離する事が可能である。このような磁気標識した2次抗体と、磁気細胞分離装置は例えば、Dynal社やMiltenyi Biotec GmbH社から入手可能である。このようにして単離されたアデノウイルス特異的CTLは、抗CD3抗体、PHA、IL-2等のT細胞刺激薬剤で刺激増殖させ、受動免疫療法に必要な細胞数を確保する。
アデノウイルス特異的CTLが、放出するサイトカイン等を利用して、アデノウイルスに特異的なCTLを精製する事ができる。例えば、Miltenyi Biotec GmbH社から入手可能なキットを用いる事で、CTLから放出されるサイトカインを細胞表面で特異抗体により補足し、サイトカイン特異的な標識抗体で染色し、続いて磁気標識した標識物質特異的な抗体で染色後、磁気標識細胞分離装置を用いて精製する事も可能である。このようにして単離されたアデノウイルス特異的CTLは、抗CD3抗体、PHA、IL-2等のT細胞刺激薬剤で刺激増殖させ、受動免疫療法に必要な細胞数を確保する。
特異的CTLの細胞表面では、特異的なペプチドの刺激により発現が増強する細胞表面蛋白質(例えばCD107a、CD107b、CD63、CD69など)が報告されている(Betts MR, Brenchley JM, Price DA, De Rosa SC, Douek DC, Roederer M, Koup RA., Sensitive and viable identification of antigen-specific CD8+ T cells by a flow cytometric assay for degranulation., J Immunol Methods., 2003;281:65-78, Trimble LA, Shankar P, Patterson M, Daily JP, Lieberman J., Human immunodeficiency virus-specific circulating CD8 T lymphocytes have down-modulated CD3zeta and CD28, key signaling molecules for T-cell activation., J Virol., 2000;74:7320-7330)。このような蛋白質の特異抗体を磁気標識する事で、磁気分離装置等を用いてCTLを精製する事が可能である。また、このような特異抗体に対する抗IgG抗体等を磁気標識する事でも同様にCTLの精製が可能である。あるいは、これら抗体を培養用のプラスティックプレートにコートし、このプレートを用いて刺激を加えたPBMCを培養し、プレートに結合しなかった細胞集団を洗い流す事で特異的なCTLを精製することも可能である。このようにして単離されたアデノウイルス特異的CTLは、抗CD3抗体、PHA、IL-2等のT細胞刺激薬剤で刺激増殖させ、受動免疫療法に必要な細胞数を確保する。
なお、本発明の「受動免疫療法剤」は、「免疫治療剤」、「アデノウイルス関連疾患治療剤」、又は「CTL誘導剤」と表現することも可能である。
本発明のCTLエピトープペプチドを使用して作製したMHC−テトラマー試薬を用いて、末梢血中のアデノウイルスに特異的なCTLを定量することができる。定量は、例えば、以下のようにして実施することができる。末梢血あるいはPBMCを、適当な濃度のMHC−テトラマー試薬と反応させる。該MHC−テトラマー試薬と結合したCTLは標識色素により染色されるので、フローサイトメーター、顕微鏡等を用いてカウントする。MHC−テトラマー試薬と反応させる時に、MHC−テトラマー試薬と異なる色素で標識された抗CD3抗体、抗CD4抗体、抗CD8抗体等を反応させる事で、アデノウイルスに特異的なCTLのT細胞サブセットも同時に判定できる。
PBMCを本発明のCTLエピトープペプチドで刺激することによってCTLが産生するIFNγ(interferon gamma)、TNF(tumor necrosis factor)、インターロイキン等のサイトカイン及び/又はケモカインを定量する方法である。以下にIFNγを例にとり具体的に方法を示す。
PBMCを適当な培地におよそ2×106/mLの細胞濃度で浮遊させ、本発明のCTLエピトープペプチドを加える。さらに細胞内蛋白輸送阻止剤(例えば、Brefeldin AやMonensin等)を加え、5% CO2恒温槽にて37℃で5〜16時間培養する。培養後、T細胞マーカー抗体(抗CD3抗体、抗CD4抗体、抗CD8抗体)あるいは/または、MHC−テトラマー試薬と反応させ、細胞を固定後、膜透過処理を行い、色素標識抗IFNγ抗体を反応させる。フローサイトメーター等を用いて解析し、全細胞中、T細胞中あるいはMHC−テトラマー陽性細胞中のIFNγ陽性細胞率を定量する。
抗IFNγ抗体を固相化した96穴MultiScreen-HAプレート (Millipore社)にPBMCをまく。エピトープペプチドを各穴に入れ37℃の5% CO2恒温槽培養器にて20時間培養する。翌日、プレートを洗浄し、抗IFNγ抗体、ペルオキシダーゼ標識抗IgG抗体の順で反応させる。さらにペルオキシダーゼの基質を加え、発色によりIFNγスポットを可視化し、実体顕微鏡でカウントする。
PBMCを適当な培地におよそ2×106/mLの細胞濃度で浮遊させ、本発明のCTLエピトープペプチドを加える。5% CO2恒温槽にて37℃で24〜48時間培養する。培養後、上清を回収し、その中に含まれるIFNγ濃度を市販のELISA キット(例えばMBL社のHUMAN IFN gamma ELISA)を使用して定量する。
細胞表面蛋白質特異的抗体を用いて定量を行う。特異的CTLは、特異的なペプチドの刺激により細胞表面に発現が増強する細胞表面蛋白質(例えばCD107a、CD107b、CD63、CD69など)が報告されている。このような蛋白質を特異的に認識する標識抗体と、ペプチド刺激したPBMC等を混合する事で、標識抗体と結合したCTLは標識色素により染色されるので、フローサイトメーター、顕微鏡等を用いてカウントする。標識抗体と反応させる時に、同時にあるいは、順番に、標識抗体と異なる色素で標識された抗CD3抗体、抗CD4抗体、抗CD8抗体等を反応させる事で、アデノウイルスに特異的なCTLのT細胞サブセットも同時に判定できる。
上記方法においては、被検者から予め取得・単離された末梢血試料を用いることができる。
また、上記方法によって誘導されたアデノウイルス特異的CTLを検出及び定量する方法もまた本発明に含まれる。
また、上述の方法によって誘導されたアデノウイルス特異的な細胞傷害性T細胞は、アデノウイルスに対する受動免疫療法剤の成分となり得る。従って、上述の方法によってアデノウイルス特異的な細胞傷害性T細胞を誘導し、該T細胞を取得することによって、受動免疫療法剤を製造することが可能である。
上記製造方法には、上述の工程に加えて、必要に応じて、「取得される細胞傷害性T細胞へ薬学的に許容される担体を混合する工程」を含んでいてもよい。
アデノウイルスは、51タイプの血清型が報告されており、6種類のサブグループ(A〜F)に分類される。移植後の感染症で特に問題となるサブグループBの11型を特異的に認識するエピトープペプチド、及びサブグループBだけでなくアデノウイルス全般に特異的なエピトープペプチドを発明する為に、51タイプの血清型の中で、最も相同性の高い蛋白質であるヘキソンに関して解析を行った。アデノウイルス粒子は直径80〜90 nmのエンベロープをもたない正20面体構造をしている。正20面体の20個の三角形の面と稜を形づくるのがヘキソンである。

アデノウイルス特異的CTLエピトープ候補ペプチドは、HLA-A*24分子に対して結合モチーフを有する。したがって、HLA-A*24分子保持者由来の末梢血を用いてエピトープペプチドを同定する事が望ましい。最初に、供血者がHLA-A*24あるいはHLA-A*2分子保持者であるかどうかの判定を血清法にて実施した。具体的には、健康成人末梢血100μL、あるいは末梢血から分離したPBMC 2〜10×105細胞に対して抗HLA-A*24抗体(クローン名 22E1、MBL社)あるいは抗HLA-A*2抗体(クローン名 BB7.2、MBL社)を1μgあるいは10μg/mLの濃度でそれぞれ加えた。それぞれの陰性コントロールとしてアイソタイプ抗体を同様に加え、室温にて15〜30分反応後、FITC(fluorescein isothiocyanate)標識抗マウスIgG抗体を反応させた。末梢血を用いた場合は、溶血固定試薬(OptiLyse B、Beckman Coulter社)の手法に従い、PBMCの場合は洗浄後、それぞれフローサイトメーターFACSCalibur(BECTON DICKINSON社)にて反応性を検討した。図1にHLA型の判定例を示す。以降の検討は、抗HLA-A*24抗体に反応性を示した10名の健康成人のPBMCを用いて行った。
(1)EBV感染B細胞株の調製
定法(Kuzushima K, Yamamoto M, Kimura H, Ando Y, Kudo T, Tsuge I, Morishima T. Establishment of anti-Epstein-Barr virus (EBV) cellular immunity by adoptive transfer of virus-specific cytotoxic T lymphocytes from an HLA-matched sibling to a patient with severe chronic active EBV infection. Clin Exp Immunol. 1996;103:192-198)に従い、EBV産生細胞株であるB95-8細胞(JCRB Cell Bankより入手)の上清(生EBVウイルスを含む) とPBMCを混合培養し、EBV感染B細胞株(Lymphoblastoid cell line、以下、EBV感染LCLと称する)を樹立した。約2週間後にHLA分子の発現、CD80、CD83とCD86の発現を確認した。
ヒトCD40Lを遺伝子導入し、安定的に発現させたNIH3T3細胞(NIH-CD40L)とPBMCを、IL-4存在下で共培養した。NIH-CD40Lは96 GyのX線照射により増殖阻害し、3〜4日毎に共培養を繰り返した(Kondo E, Topp MS, Kiem HP, Obata Y, Morishima Y, Kuzushima K, Tanimoto M, Harada M, Takahashi T, Akatsuka Y. Efficient generation of antigen-specific cytotoxic T cells using retrovirally transduced CD40-activated B cells. J Immunol. 2002;169:2164-2171)。約2週間後にHLA分子の発現、CD80、CD83とCD86の発現を確認した。
PBMCをプラスティック製の培養皿で37℃、5% CO2 恒温槽内にて2時間培養し、培養皿に接着しなかった細胞を軽く洗い流した。これにGM-CSFとIL-4を加え24時間培養後、続けてTNFαとIL-1βおよびPGE2 (Prostaglandin E2)を加え、24〜48時間培養した。適当な培地等で軽く洗い流して回収できた細胞を樹状細胞とした(Dauer M, Obermaier B, Herten J, Haerle C, Pohl K, Rothenfusser S, Schnurr M, Endres S, Eigler A. Mature dendritic cells derived from human monocytes within 48 hours: a novel strategy for dendritic cell differentiation from blood precursors. J Immunol. 2003;170:4069-4076)。48時間後にHLA分子の発現、CD80、CD83とCD86の発現を確認した。
(1)抗原提示細胞を利用した誘導
抗HLA-A*24抗体に反応性を示した健康成人のPBMCより、前述の抗原提示細胞(EBV感染LCL、CD40-B細胞、樹状細胞)を予め調製した。抗原提示細胞をパルス用培地(0.1% 人血清アルブミン/55μM 2-メルカプトエタノール/RPMI 1640)あるいは、AIM-V medium (Invitrogen社)に浮遊させ、10μg/mLの濃度でCTLエピトープ候補ペプチドを加え、およそ15分間隔で穏やかに混合させながら室温にて30〜60分間放置後、過剰量の洗浄液(2% ウシ胎児血清(FCS)/PBS)にて3回洗浄し、HLA分子に未結合のペプチドを洗い流した。この操作を行う事で、抗原提示細胞上のHLA分子にCTLエピトープ候補ペプチドが結合すると考えられる。この操作を行った抗原提示細胞をペプチドパルス抗原提示細胞と呼ぶ。ペプチドパルス抗原提示細胞は、増殖能を失わせる為に、致死量のX線照射、またはマイトマイシンC処理を行った。これを同一人物から分離したPBMC、あるいはCD8陽性またはCD4陽性のT細胞と混和し37℃、5% CO2恒温槽にて培養を行った。用いる培地は、10%FCS含有RPMI1640培地、あるいは10%ヒト血清含有RPMI1640培地、または、1〜10%のヒト血漿含有RPMI1640培地等の検討を行ったが、本方法においては、10%ヒト血清含有RPMI1640培地で良好な結果が得られた。T細胞の生存の維持と、増殖を補助する目的でIL-2(シオノギ製薬社)を添加したが、そのタイミングは通常混合培養開始後7〜10日目とする報告が多く、本発明ではIL-2は培養開始時、培養開始2日後、6日後等の検討を行ったが培養開始2日後に投与した場合に良好な結果が得られた為、培養開始2日後に50 U/mLの濃度で加えた。培養開始7日後に再度ペプチドパルス抗原提示細胞を用いて刺激を加えた。1週間毎にペプチドパルス抗原提示細胞を用いて刺激を加え、CTL誘導の評価は、およそ2週間後と4週間後に実施した。アデノウイルス特異的CTLの誘導が確認できた場合は、更にペプチドパルス抗原提示細胞を用いて刺激を加えCTLラインを樹立した。
本誘導法は、PBMC培養液中にペプチドを投入してCTLを誘導する方法である。PBMC中に存在する抗原提示細胞、例えば、樹状細胞、B細胞、マクロファージ、ある種のT細胞にペプチドが提示され、PBMCに含まれるCTL前駆細胞が刺激を受け増殖すると考えられる。前述の抗原提示細胞を利用した誘導法と異なり、前もって抗原提示細胞を調製する必要が無い点で区別され、簡便に実施できる事が利点である。
前述の方法で培養した細胞集団にアデノウイルス特異的なCTLが存在するかどうかの検討は、細胞内IFNγ産生細胞定量法、MHC−テトラマー法、エリスポットアッセイ、膜表面蛋白質の発現検討により実施した。
前述の方法で誘導した細胞集団の約1/10〜全量を96穴U底細胞培養用マイクロテストプレートに移し、誘導に用いたペプチドを最終0.01〜0.05μg/mLの濃度で加えた。さらに細胞内蛋白輸送阻止剤(例えば、Brefeldin AやMonensin等)を加え、5% CO2恒温槽にて37℃で5〜16時間培養した。培養後、細胞を洗浄し、PE(phycoerythrin)標識MHC−テトラマー試薬とPC5(phycoerythrin-Cy5)標識CD8抗体(Beckman Coulter社)を加え、室温にて15〜30分放置した。洗浄後、4% ホルムアルデヒドにて、4℃、15分固定後、過剰量の洗浄液にて洗った。0.1%サポニンにて膜透過処理後、FITC標識抗IFNγ抗体(Beckman Coulter社)を加え、室温にて15〜30分反応させた。洗浄後、フローサイトメーターを用いて、T細胞中のIFNγ陽性細胞率あるいはMHC−テトラマー試薬陽性細胞中のIFNγ陽性細胞率を定量した。
エリスポットアッセイは市販のキット(例えばMABTECH社等)を購入し、その能書に従い実施する事ができる。また以下のように実施する事もできる。96穴MultiScreen-HAプレート(Millipore社)を抗IFNγモノクローナル抗体 (R&D Systems社)で一晩、4℃にてコーティングした。各穴をPBSで洗浄後、標的細胞を各穴にまいた。標的細胞はHLA-A*24分子を細胞表面に発現している細胞であれば使用する事ができ、例えば174CEM.T2細胞にHLA-A*24分子の遺伝子を導入した培養細胞T2-A24を使用する事ができる。またHLA-A*24分子を細胞表面に発現しているEBV感染LCLも標的細胞として用いる事ができる。これら標的細胞と、アデノウイルス特異的なCTLエピトープ候補ペプチド、あるいは陰性コントロール(HIV)ペプチドを各穴に入れて、30分室温で静置した後、適当な数のCTLを添加し、37℃、5% CO2培養器にて20時間培養した。エピトープ候補ペプチドに反応したCTLはこの培養期間にIFNγを分泌し、このIFNγはプレートにコーティングした抗IFNγモノクローナル抗体と結合した。培養後、0.05% Tween-20添加PBSでプレートを洗浄し、抗IFNγポリクローナル抗体(Genzyme社)、ペルオキシダーゼ標識抗ウサギIgGヤギ血清の順で各々90分ずつ室温にて反応させた。さらにペルオキシダーゼの基質である3-amino-9-ethylcarbasole (Sigma社)と0.015%のH2O2を含む0.1M sodium acetate buffer (pH 5.0)を各穴に入れ、室温で40分反応させ、IFNγスポットを可視化した。スポットは実体顕微鏡でカウントした。
CTLは、その特異的刺激により細胞表面蛋白質(例えばCD107a、CD107b、CD63、CD69など)の発現が増強する事が報告されている。前述の方法で誘導した細胞集団の約1/10〜全量を96穴U底細胞培養用マイクロテストプレートに移し、誘導に用いたペプチドを最終0.01〜0.05 μg/mLの濃度で加え、5% CO2恒温槽にて37℃で5〜16時間培養した。培養後、細胞を洗浄し、各種標識された細胞表面蛋白質抗体等とMHC−テトラマー試薬あるいはCD8抗体を加え穏やかに攪拌後、室温にて15〜30分間放置した。洗浄後、フローサイトメーターを用いて、CD8陽性細胞あるいはMHC−テトラマー陽性細胞中の各種膜表面蛋白質発現細胞数を検討した。
[MHC−テトラマー試薬の合成]
本発明者らの所属するMBL社は、MHC−テトラマー試薬に関する特許(US Patent No. 5,635,363とFrench Application No. FR9911133および、U.S. Patent Numbers. 5,723,584; 5,874,239; 5,932,433; 6,265,552.)を有するBeckman Coulter社より、日本国内における独占的な特許使用許諾権を得ている。これら特許に基づきアデノウイルス特異的なCTLエピトープペプチドとHLA-A*2402分子を用いてPE標識MHC−テトラマー試薬を作製した。本発明で作製したMHC−テトラマー試薬は例えば、TYF-Tetと略号で示すが、これは、HLA-A*2402分子とペプチドTYF(TYFNLGNKF/配列番号:4)とβ2−ミクログロブリンの3者複合体を用いて合成されたものを示す。
本発明のCTLエピトープペプチドを使用して作製したMHC−テトラマー試薬を用いて、末梢血、末梢血から分離したPBMC、CTLエピトープペプチドを使用して誘導したCTLラインを試料として、アデノウイルス特異的なCTLの定量を行った。以下にPE標識MHC−テトラマー試薬を用いた場合を例に実施例を示す。用いるフローサイトメーターの機種に合わせ標識色素は適切な組合せで用いれば良く、下記に限定される物ではない。
採血した末梢血200μLに対して、10μLのPE標識MHC−テトラマー試薬と、20μLのFITC標識T細胞表面抗体(例えばCD8, CD4, CD3)等を加えた。さらに、混入した赤血球による非特異的な蛍光を除外するために、PC5等で標識されたCD45抗体を加えても良い。穏やかに混合し室温にて30分間放置した。OptiLyse Bを加え能書に従い溶血固定処理を行った。2 mLのPBSを加え攪拌後、400 x gで5分間遠心分離した。上澄みを吸引廃棄後、細胞を500 μLのPBSに再懸濁し、24時間以内にフローサイトメーターにて解析した。
適量のPBMC(105〜106個)または、CTLエピトープペプチドを使用して誘導した適量のCTLラインに対して10μLのPE標識MHC−テトラマー試薬と、20μLのFITC標識T細胞表面抗体(例えばCD8, CD4, CD3)等を加えた。さらに、混入した赤血球による非特異的な蛍光を除外するために、PC5等で標識されたCD45抗体を加えても良い。穏やかに混合し室温にて30分放置した。3 mLのPBSを加え攪拌後、400 x gで5分間遠心分離した。上澄みを吸引廃棄後、細胞を500μLのPBSに再懸濁した。CTLラインの場合は、死細胞による非特異的な蛍光を除外するために、7-AAD viability Dye(死細胞検出試薬、MBL社)を加えてもよい。24時間以内にフローサイトメーターにて解析した。
TYF-Tetは、Donor ID*24-2と*24-8で明らかなCD8陽性TYF-Tet陽性の細胞集団がURに検出された。この事は、TYFがHLA-A*2402拘束性を示すアデノウイルス ヘキソン特異的なCTLエピトープペプチドである事を証明している。
VYS-Tetは、Donor ID*24-2と*24-8で明らかなCD8陽性VYS-Tet陽性の細胞集団がURに検出された。この事は、VYSがHLA-A*2402拘束性を示すアデノウイルス ヘキソン特異的なCTLエピトープペプチドである事を証明している。またVYS-TetではULにCD8陰性VYS-Tet陽性の細胞集団がDonor ID *24-1、*24-3、*24-11、*24-12、*24-14で検出された。これは、VYSがCD8陽性のCTLだけでなく、CD8陰性のCTLも誘導する事が可能なエピトープペプチドである事を示している。
LYA-Tetでは、ULにCD8陰性テトラマー試薬陽性の細胞集団が検出された。この事は、CD8陰性LYA-Tet陽性のCTLが存在する事を示唆しており、更に詳細な検討結果を図6に示す。Donor ID*24-14のPBMCを24日間LYAにて刺激後、LYA-TetおよびCD4、CD3、CD8にて染色した。また陰性コントロールとしてNegativeテトラマー試薬(MBL社)と反応性を比較した。その結果、CD4陽性LYA-Tet陽性細胞が0.09%、CD8陽性LYA-Tet陽性細胞が0.18%存在し、かつCD3陽性細胞LYA-Tet陽性細胞の比率(1.13%)が最も高い事が判明した。従って、CD4陰性CD8陰性CD3陽性のHLAクラスI拘束性のCTLが存在し、アデノウイルス感染細胞の排除に関与している可能性が示された。
LYSでは、表2に示した通り、検討した10名中2名で、ペプチド特異的なIFNγの産生が確認され、図3e-hに示した通りDonor ID *24-12のLYSでは2.95%ものIFNγ産生細胞が存在していたにも関わらず、相応のLYS-Tet陽性の細胞像は得られなかった。またDYLでも、表2に示した通り、検討した10名中4名で、ペプチド特異的なIFNγの産生が確認され、図3e-hに示した通り、DYLでも0.49%のIFNγ産生細胞存在していたにも関わらず、相応のDYL-Tet陽性の細胞像は得られなかった。この事は、LYS とDYLがHLA-A*2402拘束性を受けず、別のHLA分子の拘束性を受けている事を強く示唆している。LYSの場合、Donor ID*24-12と*24-13の2名のHLAジェノタイプを解析し、その2名にHLA-A*24以外で共通のHLAを検索する事でHLA拘束性は明らかにできると考えられる。DYLの場合は、Donor ID*24-1、*24-2、*24-10と*24-12の4名のHLAジェノタイプを解析し、その4名にHLA-A*24以外で共通のHLAを検索する事でHLA拘束性は明らかにできると考えられる。最近報告された論文では、HCMVのpp65蛋白質のHLA-A*2402拘束性のエピトープペプチドであるQYDPVAALF(配列番号:12、アミノ酸番号341-349)は同時にHLA-Cw*0401の拘束性を受ける事が報告された(Kondo E, Akatsuka Y, Kuzushima K, Tsujimura K, Asakura S, Tajima K, Kagami Y, Kodera Y, Tanimoto M, Morishima Y, Takahashi T.Identification of novel CTL epitopes of CMV-pp65 presented by a variety of HLA alleles. Blood. 2004;103:630-638.)。HLA-A*24拘束性ペプチドは、N末端より2番目の位置にTyr、Phe、Met又はTrpのいずれかが配置され、9あるいは10番目の位置にLeu、Ile、Trp又はPheのいずれかが配置されたペプチドである。HLA-Cw*0401拘束性ペプチドは、N末端より2番目の位置にTyr、Phe又はProのいずれかが配置され、9あるいは10番目の位置にLeu又はPheのいずれかが配置されたペプチドであり、両者は良く似ている。実際BIMASにて算出されたスコアは、LYSはA*24の場合は280で、Cw*0401で200であった。また同様にDYLはA*24の場合は360で、Cw*0401で220であった。LYSとDYLに関して日本人で頻度の高いHLA-A、BおよびCに関して同様の検索を実施したが、このような高いスコアを示すHLAはHLA-Cw*0401以外には存在しなかった。これらのことから、LYS、およびDYLのHLA拘束性に関してHLA-Cw*0401の可能性が示唆された。
何らかの原因により免疫能が低下した人、先天性免疫不全症患者、または骨髄移植、造血幹細胞移植、臍帯血移植、固形臓器移植を受けて拒絶予防のために免疫抑制剤の投与を受けている患者、慢性ウイルス感染症患者、エイズ患者、高齢者、幼小児、妊婦等のハイリスクの患者、または移植ドナーの末梢血中にアデノウイルス特異的CTLが存在するか否かを知ることは、抗ウイルス剤や免疫抑制剤の適正な使用を含めた感染症管理の上で重要である。アデノウイルス特異的MHC−テトラマー試薬を用いれば、採血から1時間程度でアデノウイルス特異的CTLの存在を判定できる。
MHC−テトラマー試薬は、特異的なCTLの定量だけでなく細胞内サイトカイン産生細胞定量法や細胞表面蛋白質に対する特異抗体と組み合わせることで、特異的CTLの分化段階や機能性も同時に判定する事が可能である。本発明で提供されるCTLエピトープペプチドを用いて合成されたMHC−テトラマー試薬がアデノウイルス特異的CTLの定量と機能解析に有効であるか検討した。その結果を図8に示した。
PE標識したアデノウイルス特異的MHC−テトラマー試薬と、CTL調製方法にて誘導されたCTLを混合し、室温にて30分間放置した。過剰量の洗浄液で1回洗い、磁気標識抗PEマイクロビーズ(Miltenyi Biotec GmbH社)を加え、4〜8℃で15分間静置した。過剰量の洗浄液で1回洗い、細胞を1 mLの洗浄液に希釈後、自動磁気細胞分離装置(AutoMACS、Miltenyi Biotec GmbH社)を用いて、MHC−テトラマー陽性細胞、すなわちアデノウイルス特異的CTLを精製した。
[CTLエピトープTYFに関するサブグループCとの反応性検討]
本発明にて同定されたTYFは、図11−4に示すようにサブグループC特異的なCTLエピトープと4番目と6番目の2箇所でアミノ酸変異が存在する。そこで、TYFにて誘導したCTLラインを用いてサブグループC特異的なCTLエピトープ(TYFSLNNKF(配列番号:14)、下線は変異アミノ酸を示す)(Leen AM, Sili U, Vanin EF, Jewell AM, Xie W, Vignali D, Piedra PA, Brenner MK, Rooney CM.Conserved CTL epitopes on the adenovirus hexon protein expand subgroup cross-reactive and subgroup-specific CD8+ T cells. Blood. 2004;104:2432-2440.)との交差反応性を検討した。
アデノウイルスのサブグループ間で、本発明で得られたCTLエピトープペプチドの反応性を検討するためにNCBI (National Center for Biotechnology Information, http://www.ncbi.nlm.nih.gov/)より50タイプのアデノウイルス ヘキソンのアミノ酸配列を入手し(Accession Numberの一覧を表3に示す)、アミノ酸相同配列解析を行った。最近、CTLエピトープC末端側のアンカーモチーフ直後のアミノ酸に変異が生じる事で、細胞内蛋白質分解経路にて分解される場所が変化し、結果としてCTLエピトープペプチドが生じなくなる事が報告されている(Beekman NJ, van Veelen PA, van Hall T, Neisig A, Sijts A, Camps M, Kloetzel PM, Neefjes JJ, Melief CJ, Ossendorp F. Abrogation of CTL epitope processing by single amino acid substitution flanking the C-terminal proteasome cleavage site. J Immunol. 2000;164:1898-1905.)。これは、HIVやHCVのような慢性ウイルス疾患において、ウイルス感染細胞がCTLの標的からエスケープし、持続感染が継続する原因の1つと考えられている(Furutsuki T, Hosoya N, Kawana-Tachikawa A, Tomizawa M, Odawara T, Goto M, Kitamura Y, Nakamura T, Kelleher AD, Cooper DA, Iwamoto A.Frequent transmission of cytotoxic-T-lymphocyte escape mutants of human immunodeficiency virus type 1 in the highly HLA-A24-positive Japanese population. J Virol. 2004;78:8437-8445.)。
DMSOに、配列番号:1〜6のペプチドを最終濃度20 mg/mlとなるように各々溶解し、ろ過滅菌した。得られたペプチド含有溶液を滅菌バイアル瓶に1mLずつ分注密栓し、ワクチン注射剤とした。
(1)CTLはウイルス粒子そのものを直接認識することはできない。
(2)CTLはウイルスタンパク質中の8〜10個のアミノ酸からなるペプチド(以下、エピトープペプチドと称する)を認識して感染細胞を破壊する。
(3)このエピトープペプチドは、ウイルス感染細胞表面にあるヒト白血球抗原(human leucocyte antigen、以下、HLAと称する)とβ2-ミクログロブリンの複合体に結合して提示され、これにCTL細胞表面上のT細胞受容体(T cell receptor;TCR)が結合する事でCTLはウイルス感染細胞を認識し破壊する。
(4)HLAは人種間、個人間によって異なり、HLAが異なると同じウイルスでもエピトープペプチドが異なる(これをエピトープペプチドのHLA拘束性と称する)。
Claims (14)
- アデノウイルスサブグループBに対する細胞傷害性T細胞を誘導するための、HLA-A24分子拘束性のペプチドであって、該ペプチドはアデノウイルスサブグループCと交差反応せず、さらに該ペプチドが、配列番号:4または配列番号:3に記載のアミノ酸配列を含むものである、ペプチド。
- 請求項1に記載のペプチドをコードする核酸。
- 請求項1に記載のペプチドを有効成分として含む、アデノウイルスの感染を治療又は予防するためのワクチン。
- 請求項2に記載の核酸を有効成分として含む、アデノウイルスの感染を治療又は予防するためのワクチン。
- 請求項1に記載のペプチドをHLAに提示した抗原提示細胞を有効成分として含む、アデノウイルスの感染を治療又は予防するためのワクチン。
- 請求項1に記載のペプチドもしくは該ペプチドをHLAに提示した抗原提示細胞により末梢血リンパ球を刺激して得られるアデノウイルス特異的な細胞傷害性T細胞を有効成分として含む、アデノウイルスに対する受動免疫療法剤。
- 請求項1に記載のペプチドから調製した主要組織適合性抗原複合体及び/又は主要組織適合性抗原複合体-テトラマーと末梢血リンパ球とを反応させ、該主要組織適合性抗原複合体及び/又は主要組織適合性抗原複合体-テトラマーに細胞傷害性T細胞が結合した結合体を形成させ、該結合体から単離して得られる細胞傷害性T細胞を有効成分として含む、アデノウイルスに対する受動免疫療法剤。
- 請求項1に記載のペプチドで単離された末梢血を刺激し、該ウイルスに特異的な細胞傷害性T細胞を取得し、単離された細胞傷害性T細胞が産生するサイトカイン及び/又はケモカイン及び/又は細胞表面分子を測定することを特徴とする、アデノウイルスに特異的な細胞傷害性T細胞の定量方法。
- 請求項1に記載のペプチドから主要組織適合性抗原複合体-テトラマーを調製し、主要組織適合性抗原複合体-テトラマーと単離された末梢血とを反応させることを特徴とする、該末梢血中のアデノウイルスに特異的な細胞傷害性T細胞の定量方法。
- 請求項1に記載のペプチドを用いて被検者から単離された末梢血において細胞傷害性T細胞を誘導することを特徴とする、細胞傷害性T細胞の誘導方法。
- 請求項1に記載のペプチドと、末梢血単核球を血漿を含む培地中で接触させることにより、アデノウイルス特異的細胞傷害性T細胞を誘導する、請求項10に記載の誘導方法。
- 請求項1に記載のペプチドもしくは該ペプチドをHLAに提示した抗原提示細胞により単離された末梢血リンパ球を刺激してアデノウイルス特異的な細胞傷害性T細胞を取得する工程を含む、アデノウイルスに対する受動免疫療法剤の製造方法。
- 請求項1に記載のペプチドから調製した主要組織適合性抗原複合体及び/又は主要組織適合性抗原複合体-テトラマーと単離された末梢血リンパ球とを反応させ、該主要組織適合性抗原複合体及び/又は主要組織適合性抗原複合体-テトラマーに細胞傷害性T細胞が結合した結合体を形成させ、該結合体から単離して得られる細胞傷害性T細胞を取得する工程を含む、アデノウイルスに対する受動免疫療法剤の製造方法。
- 請求項1に記載のペプチドを使用し作製される主要組織適合性抗原複合体-テトラマー。
