Description
(a)骨髄間質細胞(ただし、該骨髄間質細胞は脱メチル化剤で処理されていない細胞である)の培養物に、サイクリックAMP(以下、「cAMP」と略す場合がある。)上昇作用性薬剤、cAMPアナログ、及び細胞分化生存作用性因子からなる群から選ばれる1以上の物質を添加して培養する工程;
(b)上記工程(a)で得られた細胞にNotch遺伝子及び/又はNotchシグナリング関連遺伝子を導入して培養し筋芽細胞培養物を得る工程(ただし、上記培養は、遺伝子導入された細胞と未導入細胞との共培養を含まない);及び、
(c)上記工程(b)で得られた筋芽細胞培養物にNotchリガンドを添加して培養する工程
を含む方法
が提供される。
上記発明の好ましい態様によれば、上記工程(c)においてNotchリガンドとしてJagged1蛋白を用いる上記の方法;及び上記工程(c)を無血清培地で行なう上記の方法が提供される。
(a)骨髄間質細胞(ただし、該骨髄間質細胞は脱メチル化剤で処理されていない細胞である)の培養物に、cAMP上昇作用性薬剤、cAMPアナログ、及び細胞分化生存作用性因子からなる群から選ばれる1以上の物質を添加して培養する工程;及び
(b)上記工程(a)で得られた細胞にNotch遺伝子及び/又はNotchシグナリング関連遺伝子を導入して培養する工程(ただし、上記培養は、遺伝子導入された細胞と未導入細胞との共培養を含まない)
を含む方法が提供される。
さらに別の観点からは、本発明により、筋芽細胞から骨格筋細胞を分化誘導する方法であって、筋芽細胞培養物にNotchリガンドを添加して培養する工程を含む方法が提供される。
(a)骨髄間質細胞(ただし、該骨髄間質細胞は脱メチル化剤で処理されていない細胞である)の培養物に、cAMP上昇作用性薬剤、cAMPアナログ、及び細胞分化生存作用性因子からなる群から選ばれる1以上の物質を添加して培養する工程;
(b)上記工程(a)で得られた細胞にNotch遺伝子及び/又はNotchシグナリング関連遺伝子を導入して培養し筋芽細胞培養物を得る工程(ただし、上記培養は、遺伝子導入された細胞と未導入細胞との共培養を含まない);及び、
(c)上記工程(b)で得られた筋芽細胞培養物にNotchリガンドを添加して培養する工程
を含んでいる。
例1
特開2003-144155号公報に記載の方法に従って、骨格筋誘導成体ラット(Wister種)及びヒトの骨髄より間質細胞を採取して培養した。培地としてはMinimum Essential Medium (MEM)Eagle Modification (Sigma社、M4526)に15%のウシ胎児血清 (Biowhittaker社、14-501F、Lot#61-1012)を加えたものを用いた。4代まで継代培養し、80-90%の細胞密度に達した時点で1800〜1900 cells/cm2の密度に調節して細胞をまき、その翌日よりForskolin 5μM (Calbiochem、344273)、basic-Fibroblast growth factor 10ng/ml (Peprotech EC LTD、100-18B)、Platelet-derived growth factor-AA 5ng/ml(Peprotech EC LTD、396-HB)、Heregulin 200ng/ml(R&D Systems、396-HB)を添加した培地に切り替えて3日間培養した。その後、特開2003-144155号公報の実施例1の方法に従ってNotch細胞内ドメインの遺伝子を細胞に導入した。
上記例1と同様の方法により骨格筋細胞の分化誘導を行なった。ただし、フュージョンインダクションにおいて無血清培地を用いた。無血清培地としては、インスリン 10 μg/ml、トランスフェリン 5 μg/ml、sodium selenite 10 nmol、BSA 1mg/ml、カナマイシン 60 μg/mlを含むDMEM(この培地を「ITS-serum free medium」と呼ぶ)にJagged1蛋白 (recombinant Rat Jagged1/Fc Chimera、GT TECHNE社、3599)を最終濃度が5μg/mlとなるように添加したものを用いた。この結果、培養1日目には多核の骨格筋細胞の数が35ミリ培養皿1枚あたり16となり、培養5日目には664となった。一方、対照として上記無血清培地のみ(Jagged1蛋白無添加)で培養した場合には、培養1日目に13、培養5日目には487であった。この結果から、無血清培地にJagged1蛋白を添加することによっても成熟した骨格筋細胞を分化誘導できることが示された。
Claims (3)
- 骨髄間質細胞から骨格筋細胞を分化誘導する方法であって、以下の工程:
(a)骨髄間質細胞(ただし、該骨髄間質細胞は脱メチル化剤で処理されていない細胞である)の培養物に、フォルスコリン、並びに細胞分化生存作用性因子である塩基性線維芽細胞成長因子、血小板由来成長因子、及びニューレグリンを添加して培養する工程;
(b)上記工程(a)で得られた細胞にNotch細胞内ドメインの遺伝子を導入して培養し筋芽細胞培養物を得る工程(ただし、上記培養は、遺伝子導入された細胞と未導入細胞との共培養を含まない培養である);及び、
(c)上記工程(b)で得られた筋芽細胞培養物にJagged1蛋白を添加して培養する工程
を含む方法。 - 上記工程(c)を無血清培地で行なう請求項1に記載の方法。
- 骨髄間質細胞から筋芽細胞を分化誘導する方法であって、以下の工程:
(a)骨髄間質細胞(ただし、該骨髄間質細胞は脱メチル化剤で処理されていない細胞である)の培養物に、フォルスコリン、並びに細胞分化生存作用性因子である塩基性線維芽細胞成長因子、血小板由来成長因子、及びニューレグリンを添加して培養する工程;及び
(b)上記工程(a)で得られた細胞にNotch細胞内ドメインの遺伝子を導入して培養し筋芽細胞培養物を得る工程(ただし、上記培養は、遺伝子導入された細胞と未導入細胞との共培養を含まない培養である)
を含む方法。
