Description
【発明の属する技術分野】
本発明は、ドラッグ・デリバリー・システムやアジュバントに使用しうる生体内可溶性複合体粒子に関する。

【従来の技術】
病巣に直接薬剤を供給するドラッグ・デリバリー・システムの一方法として、経皮カテーテル法が知られている。この方法は、副作用が強くても局所に高濃度で薬剤を供給しうる点で癌のような悪性腫瘍の治療に有望視されている。この目的で、リン酸カルシウム系化合物粒子に抗腫瘍剤を吸着させることが提案されている(例えば、特公平1−51266号公報、特開平2−200628号公報参照)。
しかしながら、リン酸カルシウム系化合物粒子に薬剤を吸着させて血管内に投与すると、徐々に薬剤を放出することができ、効果をある程度持続することができるものの、比重が大きく、血管閉塞の恐れがある。
また、薬剤を含浸したゼラチンスポンジを用いる方法も知られているが、この方法では、薬剤の担持量が少ないばかりか、ゼラチンの溶解が速いため、薬効の持続が望めないという欠点があった。

【発明が解決しようとする課題】
本発明は、薬剤、抗原などに対して高い吸着能を有するとともに、生体内で溶解あるいは分解されて体内残留物を生じない生体内可溶性複合体粒子を提供することを目的とする。

【課題を解決するための手段】
本発明は、生体内可溶性高分子物質粒子をコアとし、その表面に吸着性を有するリン酸カルシウム系化合物を被覆することによって上記課題を達成したものである。
すなわち、本発明の生体内可溶性複合体粒子は、生体内可溶性高分子物質粒子の表面がCa/P比1.0〜2.0のリン酸カルシウム系化合物で被覆されていることを特徴とする。

【発明の実施の形態】
本発明の複合体粒子のコアとして用いる生体内可溶性高分子物質としては、生体内で体液に溶解するものであれば、特に制限はなく、生理学的に許容しうるものが好ましく、例えば、ゼラチン、ポリ乳酸、キチン、ポリビニルアルコール、ポリアクリル酸、コラーゲンなどが挙げられる。なお、キチンは、生体内で酵素リゾチームによって最終的に分解され、可溶化される。
本発明において、生体内可溶性とは、キチンのように生体内の酵素によって分解されて可溶性となるものも包含するものとする。

本発明の生体内可溶性複合体粒子は、生体内可溶性高分子物質粒子の表面をリン酸カルシウム系化合物で被覆することによって製造することができる。被覆方法は、任意の方法で行なうことができるが、生体内可溶性高分子物質粒子にリン酸カルシウム系化合物粒子を物理的に衝突させることにより高分子物質粒子中にリン酸カルシウム系化合物粒子の一部分を貫入させて該高分子物質粒子の表面をリン酸カルシウム系化合物で被覆するのが好ましい。

使用する生体内可溶性高分子物質の粒子の大きさは、通常、d75/d25≦2の粒度分布の範囲内の平均粒径が1〜20μmの粒子であるのが好ましい。なお、d25は、粒子の積算ふるい下25%の粒径を意味し、d75は、粒子の積算ふるい下75%の粒径を意味する。この平均粒径が1μm未満であると、リン酸カルシウム系化合物の貫入率が著しく低下し、20μmを超えると、複合体の密度が不安定になり高密度のものが出来にくくなる。

また、生体内可溶性高分子物質粒子は、密度が0.9〜1.2g/cm3 のものが好ましい。複合体を液中で使用する場合に、その複合体が水の比重より重いのが好ましいので、高分子物質粒子としては、密度が0.9g/cm3 以上であるのが好ましく、物理的衝突工程や複合体の実用面から1.2g/cm3 以下であるのが好ましい。

本発明に用いる生体内可溶性複合体粒子は、前記のように、生体内可溶性高分子物質中にリン酸カルシウム系化合物粒子の一部分が貫入して被覆層を形成しているのが好ましい。ここで、リン酸カルシウム系化合物としては、特に制限はなく、Ca/P比1.0〜2.0の各種の化合物を使用することができ、例えば、Ca10(PO4)6 (OH)2、Ca10(PO4)6 F2 、Ca10(PO4)6 Cl2、Ca3(PO4)2 、Ca2 P2 O7 、Ca4O(PO4)2 のうちから選ばれた1種又は2種以上を使用することができる。これらのうちハイドロキシアパタイトを主成分とするものが最も好ましい。フッ素アパタイトを用いる場合、全リン酸カルシウム系化合物中のフッ素含有率が5重量%以下であるのが好ましい。フッ素含有率が5重量%を超えると、フッ素の溶出が起こり好ましくない。これらのリン酸カルシウム系化合物は、公知の湿式合成法、乾式合成法などによって合成することができる。

リン酸カルシウム系化合物の粒子は、例えばリン酸カルシウム系化合物のスラリーを噴霧乾燥することによって造粒し、これを焼成することによって調製することができるが、この方法に限らず他の造粒法によって調製することも可能である。なお、ふるい分けなどの手段により、粒子の粒度を目的に応じて所定の範囲に選定して用いることがより好ましい。

本発明に使用するリン酸カルシウム系化合物粒子は、みかけ密度が1.5〜2.5g/cm3 であるのが好ましい。みかけ密度が1.5g/cm3 未満であると、複合体の密度が水の密度より軽くなり、また2.5g/cm3 を超えると、吸着特性が著しく低下する。なお、リン酸カルシウム系化合物粒子のみかけ密度は、含水みかけ密度、すなわち水を媒体として沈降法により粒子のstokes径を求めるときに適用される密度であり、下記の式によって算出される。

また、リン酸カルシウム系化合物粒子は、リン酸カルシウム系化合物粒子が比表面積10m2 /g以上で、細孔径500〜1000Åの、一次粒子が凝集結合した多孔質粒子であるのが好ましい。比表面積が10m2 /g未満では充分な吸着能が望めない。また、蛋白質などが吸着されて気孔内へ入り込むためには、細孔径500〜1000Å程度の気孔を有するのが好ましい。

多孔質粒子は、公知方法で製造することができる。例えば、公知の方法で湿式合成したリン酸カルシウム系化合物の結晶粒子を原料粒子とし、この原料粒子を懸濁したスラリーを直接噴霧乾燥などにより二次粒子に造粒するかあるいはこのスラリーに粘度調整剤、加熱により消失する有機化合物粒子又は繊維等の添加物を加えて噴霧乾燥などにより二次粒子に造粒する。この二次粒子自体、多孔質粒子となっており、この二次粒子をそのまま原料として用いることもでき、さらに高気孔率のものが好ましい場合には、次の方法で多孔質顆粒を作製する。この二次粒子を再びスラリー状に懸濁して湿式成形するか又は加圧による乾式成形等によりブロック体に成形する。その際、焼成の過程で消散して気孔を形成するための有機化合物を添加してもよい。無添加でも、焼成温度など、他の条件を調節することにより気孔径を制御することもできる。得られたブロック体を500℃〜1300℃の温度範囲で焼成する。焼成温度が500℃未満では、有機化合物の熱消失やブロック体の焼結が充分に行われない。また、焼成を1300℃を超える高温で行うと、焼結体が緻密化しすぎたり、リン酸カルシウムが分解を起こすおそれがある。このように焼成したブロック体を粉砕後、分級して必要な粒径の顆粒を得ることができる。この顆粒の気孔径は、二次粒子造粒用の原料スラリー中の結晶粒子の大きさ、スラリーの粘度、添加物などを適切に調節することによって調整することができる。

本発明に用いる生体内可溶性高分子物質粒子にリン酸カルシウム系化合物粒子を物理的に衝突させる方法としては、市販のハイブリダイゼーションシステム、例えば、(株)奈良機械製作所製の奈良ハイブリダイゼーションシステムNHS−1、日清エンジニアリング(株)製Hi−X200などを用いて、乾式で行う方法がある。このような装置を用いて、通常、リン酸カルシウム系化合物粒子/可溶性高分子物質粒子の重量比で0.05〜0.50の範囲となるように混合し、装置内の温度が使用した可溶性高分子物質の軟化温度以下、通常、80℃以下となるような温度条件で行うのが好ましい。

生体内可溶性高分子物質粒子に衝突させるリン酸カルシウム系化合物粒子は、平均粒径100μm以下であることが好ましい。リン酸カルシウム系化合物粒子は、物理的衝突により破砕されるので、高分子物質粒子に比べてかなり大きくてもかまわないが、平均粒径が100μmを超えると衝突時の速度が低くなり、リン酸カルシウム系化合物粒子が破砕されにくくなり、破砕されてもリン酸カルシウム系化合物粒子が高分子物質粒子中に貫入しにくくなる。

可溶性高分子物質粒子に平均粒径100μm以下のリン酸カルシウム系化合物粒子を衝突させる際、リン酸カルシウム系化合物が生体内でマクロファージなどによって貪食可能な粒径7μm以下の粒子となって貫入するように装置を制御することが好ましい。
また、衝突によって高分子物質粒子中に貫入するリン酸カルシウム系化合物の平均粒径は、コアとなる生体内可溶性高分子物質粒子の平均粒径の1/5以下、好ましくは1/10以下とすることが好ましい。リン酸カルシウム系化合物粒子の平均粒径がコア粒子の平均粒径の1/5より大きいと、リン酸カルシウム系化合物の脱離安定性が極めて悪化する傾向がある。

上記のような物理的衝突によって、リン酸カルシウム系化合物粒子が高分子物質粒子の中心部まで達する必要はなく、高分子物質粒子中にリン酸カルシウム系化合物粒子の一部分がくい込んで貫入していればよい。高分子物質粒子が一般に弾性を有するため、リン酸カルシウム系化合物粒子の一部分がくい込むと、その周囲からそれを押さえる力が働き、リン酸カルシウム系化合物粒子をしっかりとつかんで固定することが可能となる。
物理的衝突によって高分子物質粒子の表面上にリン酸カルシウム系化合物の層を0.1〜5.0μmの厚さで形成するのが好ましい。この厚さが0.1μm未満では、吸着能が充分でなく、5.0μmを超えて厚くしても、それに見合った吸着能の増加は得られない。

上記のようにして、d75/d25≦2の粒度分布の範囲内の平均粒径が1.2〜30.0μm、密度が1.05〜1.35g/cm3 、細孔径が500〜1000Åで、リン酸カルシウム系化合物層の厚さが0.1〜5.0μmである生体内可溶性複合体粒子が得られる。また、この生体内可溶性複合体粒子により0.2〜4.0mgリゾチーム/gの蛋白質吸着量を達成することができる。

本発明の生体内可溶性複合体粒子は、その製造に当たって予め染色された生体内可溶性高分子物質粒子を用いるか、あるいは複合体を製造後に染色することによって着色したものとすることができる。
このように着色した生体内可溶性複合体粒子は、その後に吸着させる薬剤の種類や量の区別に利用することができる。

本発明の生体内可溶性複合体粒子は、表面に微細なリン酸カルシウム系化合物粒子の層を有するため、高い吸着能を有し、薬剤を吸着させてドラッグ・デリバリー・システムにより薬剤を病巣に投与したり、抗原を吸着させてアジュバントとして使用することができる。

次に、実施例に基づいて本発明をさらに詳述するが、本発明はこれによって制限されるものではない。

実施例1
平均粒径5μm、見かけ密度1.07g/cm3 のポリ−L−乳酸(以下、単にP−L−LAと略す)ビーズ50gと、Ca/P比1.67、平均粒径20μm、比表面積24m2 /g、かさ密度2.2g/cm3 、細孔径800Åのハイドロキシアパタイト粒子5.0gを奈良機械ハイブリダイゼーションシステムNHS−1(奈良機械製作所、定格動力5.5kw、定格電流23A)を8000回転/分で36〜64℃で5分稼動させ、P−L−LAビーズ表面をハイドロキシアパタイトで被覆した。得られたハイドロキシアパタイト被覆層の厚さは、平均0.75μmであり、得られた複合体粒子は、平均粒径5.4μm、密度1.15g/cm3 、細孔径800Åであった。
上述した複合体粒子と卵白リゾチームをpH7.2のリン酸塩緩衝液中で混合した。卵白リゾチームを複合体粒子に吸着させた後、pH7.2のリン酸塩緩衝液で洗浄し、非吸着蛋白を除去した。複合体粒子を0.5mg/mlに調製し、4週齢のメスのddyマウスの腹腔に投与した。リゾチームに対する抗体価は1週間後で顕著に上昇が認められた。

実施例2
平均粒径7μmのキチン粒子ビーズ50gと、Ca/P比1.67、平均粒径5μm、比表面積45m2 /g、かさ密度1.8g/cm3 、細孔径600Åのハイドロキシアパタイト粒子7.5gを奈良機械ハイブリダイゼーションシステムNHS−1(奈良機械製作所、定格動力5.5kw、定格電流23A)を8000回転/分で36〜47℃で5分稼動させ、キチン粒子ビーズ表面をハイドロキシアパタイトで被覆した。得られたハイドロキシアパタイト被覆層の厚さは0.8μmであり、得られた複合体粒子は、平均粒径5.8μm、細孔径600Åであった。
上述した複合体粒子とA型インフルエンザ抗原をpH7.2のリン酸塩緩衝液中で混合し、抗原を吸着させた。非吸着抗原はpH7.2のリン酸塩緩衝液で洗浄し、除去した。複合体粒子を0.5mg/mlに調製し、4週齢のメスのddyマウスの腹腔に投与した。A型インフルエンザに対する抗体価は1週間後で顕著に上昇が認められた。

【発明の効果】
本発明の生体内可溶性複合体粒子は、その表面に吸着性の高いリン酸カルシウム系化合物層を有するので、薬剤を吸着させてドラッグ・デリバリー・システムに用いて直接病巣に投与することができ、したがって、病巣付近で薬剤を高濃度に維持することができる。また、表面のリン酸カルシウム系化合物粒子は、微細であるため、マクロファージによって徐々に貪食され、それに伴ってコアの生体内可溶性高分子物質が露出するが、これは体液によって溶解されるため、薬剤を放出した後、コアも被覆層も消失し、体内に残留物を残さない。
また、本発明の生体内可溶性複合体粒子に抗原を吸着させて体内に投与することができ、これにより高い抗体価を生じさせることができる。
Claims (6)
- 生体内可溶性高分子物質粒子の表面がCa/P比1.0〜2.0のリン酸カルシウム系化合物で被覆されており、密度が1.05〜1.35g/cm 3 であることを特徴とする生体内可溶性複合体粒子。
- 生体内可溶性高分子物質がゼラチン、ポリ乳酸、キチン、ポリビニルアルコール、ポリアクリル酸又はコラーゲンである請求項1記載の生体内可溶性複合体粒子。
- 生体内可溶性高分子物質粒子中にリン酸カルシウム系化合物粒子の一部分が貫入している請求項1記載の生体内可溶性複合体粒子。
- 生体内可溶性高分子物質粒子がd75/d25≦2の粒度分布の範囲内の平均粒径が1〜20μmの粒子である請求項1記載の生体内可溶性複合体粒子。
- d75/d25≦2の粒度分布の範囲内の平均粒径が1.2〜30.0μm、密度が1.05〜1.35g/cm3 、細孔径500〜1000Åであり、リン酸カルシウム系化合物層の厚さが0.1〜5.0μmのものである請求項1又は4記載の生体内可溶性複合体粒子。
- リン酸カルシウム系化合物がCa/P比1.65〜1.67のハイドロキシアパタイトである請求項1記載の生体内可溶性複合体粒子。
