のセグメントのためのコンセンサスプライマー(consen
sus primers)である。

結果、最終の免疫グロブリン又はT−リセプタの分子は
事実上他には見られない独特なものとなる。しかし、本
発明に関して特に重要な2つの特徴がある。

つの理由から作用する。第一に、それらプライマーは免

って全DNA又はRNAにおけるそのような領域だけを認識す
ることである。第二に、PCR領域は2つのプライマーに
よって認識される配列が短かい場合に作用するだけであ
る、ということである。この状態はV、D及びJ(又は
C)領域が一緒に結合されており、成熟した免疫グロブ
リン又はT−リセプタの分子が形成されているDNA又はR
NAのそのようなセグメントについて達成されるに過ぎな
い。これら2つの性質の結果として、プライマーは最終
の成熟免疫グロブリン又はT−リセプタ分子だけを認識

ローチは9V−領域と5J−領域のためのプライマー混合物

モノクローナリティーについての試験結果 第1表及び第2表に免疫グロブリン遺伝子だけを用い
てモノクローナリティーについて各種試料を試験した結
果を示す。

大されたフラグメントを2%の寒天中での電気泳動で分
離し、そしてエチジウムブロマイドで染色して紫外光で
の可視化を可能にした。同様に結果が最後の5サイクル
の拡大のための放射性ヌクレオチド(CTP)を加え、電
気泳動を行い、そしてフラグメントをオートラジオグラ
フィーで可視化することによって得られた。
イティについての試験結果 コンセンサンプライマーの1つの対又は他の対を用
い、上記と同じPCR法を使用すると、個別の再配置が検
出された。この検出においては8つの正常T−細胞クロ
ーンのうち6つに1つのプライマー対又は他のプライマ
ー対が用いられた。

Claims (17)
- 【請求項1】組織の試料中の免疫グロブリン遺伝子及び
/又はT−リセプタ遺伝子のセグメント長の均一性又は
不均一性を判定して該試料中のB−及び/又はT−リン
パ球集団のモノクローナリティーの存否を表示すること
を含んで成る、組織試料におけるリンパ系白血病及び/
又はリンパ腫の検出法。 - 【請求項2】(a)組織の試料中の免疫グロブリン遺伝
子及び/又はT−リセプタ遺伝子のセグメントをそれら
セグメントのための特異性プライマー又は特異性プライ
マー混合物を使用するポリメラーゼの連鎖反応で拡大
し、そして (b)拡大されたセグメントを大きさに基づいて分離し
てそれら拡大セグメントの長さの均一性又は不均一性を
判定する 工程を含んで成る、組織試料におけるモノクローナリテ
ィー及び推定悪性腫瘍を検出し、及び/又は腫瘍のB−
リンパ球又はT−リンパ球の起源を判定する方法。 - 【請求項3】該プライマーが免疫グロブリン遺伝子及び
/又はT−リセプタ遺伝子のセグメントのためのコンセ
ンサスプライマーである、請求の範囲第2項に記載の方
法。 - 【請求項4】該組織試料が血液、節又は骨髄における腫
瘍組織、ホルマリン固定され、包埋された組織材料、ア
スピレートされた細胞材料及びスライドガラス上の細胞
から選択されたものである、請求の範囲第1項又は第2
項に記載の方法。 - 【請求項5】大きさに基づいて分離する該工程が電気泳
動法及びクロマトグラフィーから選択されたものであ
る、請求の範囲第2項に記載の方法。 - 【請求項6】大きさに基づく該分離工程がアガロースゲ
ル又はポリアクリルアミドゲルによる電気泳動法から成
る、請求の範囲第5項に記載の方法。 - 【請求項7】大きさに基づいて分離され、拡大された該
遺伝子のセグメントを非同位体手段、放射性同位体の組
み込み、又は内部プローブへのサウザン転移又はハイブ
リダイゼーションによって検出する、請求の範囲第2項
に記載の方法。 - 【請求項8】非同位体手段による該検出がエチジウムブ
ロマイド染色法を含んで成る、請求の範囲第7項に記載
の方法。 - 【請求項9】免疫グロブリン遺伝子のセグメントのため
の該プライマーが免疫グロブリンのV領域のフレームワ
ーク部に対するプライマー及び該遺伝子のD.J及び/又
はC領域の一部分に対するプライマーから成る、請求の
範囲第2項に記載の方法。 - 【請求項10】免疫グロブリン遺伝子のセグメントのた
めの該プライマーが (ただし、Nはヌクレオチド塩基のうちのいずれかを表
わす) から選択されたものである、請求の範囲第9項に記載の
方法。 - 【請求項11】T−リセプタ遺伝子のセグメントのため
の該プライマーがT−リセプタ遺伝子の保存V領域に対
するプライマー及び該遺伝子のD.J及び/又はC領域の
一部分に対するプライマーから成る、請求の範囲第2項
に記載の方法。 - 【請求項12】T−リセプタ遺伝子のセグメントのため
の該プライマーが (a)T−リセプタのα−鎖プライマー: (b)T−リセプタのβ−鎖プライマー: (c)T−リセプタのγ−鎖プライマー: から選択されたものである、請求の範囲第11項に記載の
方法。 - 【請求項13】T−リセプタ遺伝子のセグメントのため
の該プ から選択された、V−領域及びJ−領域のためのT−リ
セプタ遺伝子のγ−鎖プライマーの混合物から成る、請
求の範囲第11項に記載の方法。 - 【請求項14】組織の試料中の免疫グロブリン遺伝子及
び/又はT−リセプタ遺伝子のセグメントをポリメラー
ゼの連鎖反応で拡大するための、免疫グロブリン遺伝子
及び/又はT−リセプタ遺伝子のセグメント用の1組の
特異性プライマー又は特異性プライマー混合物を含んで
成る、請求の範囲第2項に記載の方法を実施するための
キット。 - 【請求項15】免疫グロブリン遺伝子及び/又はT−リ
セプタ遺伝子のセグメントのための1組のコンセンサス
プライマーを含んで成る、請求の範囲第14項に記載のキ
ット。 - 【請求項16】請求の範囲第9項または第10項に記載
の、免疫グロブリン遺伝子のセグメントのためのプライ
マーを含んで成る、請求の範囲第14項に記載のキット。 - 【請求項17】請求の範囲第11項若しくは第12項又は第
13項に記載の、T−リセプタ遺伝子のセグメントのため
のプライマーを含んで成る、請求の範囲第14項に記載の
キット。
