Description

[0155] フローサイトメトリーアッセイを使用して、MATa、MATalphaと二倍体細胞とを区別することができる。天然酵母性的(sexual)凝集素は、表面提示バインダー(SAP)で置換されており、接合効率はフローサイトメトリーを用いて測定された。次世代シーケンシングを用いて、大きなライブラリーでバインダーの特定の対の接合頻度を評価することができるように、単一染色体上に両方のバインダーの遺伝子を組み合わせるために二倍体染色体転座システムが開発された。

[0166] 必須の性的凝集素タンパク質を欠くサッカロミセス・セレビジエ(S. cerevisiae)一倍体細胞に関して、結合は、液体培養物における凝集及び接合の回復に十分である。Sag1、一次MATalpha性的凝集素タンパク質は、凝集に必須である。Sag1がノックアウトされた場合に、MATalpha細胞は、乱流(turbulent)液体培養物中で野生型MATa細胞と接合することができない。しかしながら、相補的SAPがディスプレイ対上で発現され、接合が回復される。非相補的SAPは、接合を回復することができない。
Claims (21)
- 2つのタンパク質結合パートナー間の代償性変異を同定する方法であって、
第1野生型ポリペプチド及び第1の複数の変異ポリペプチドを含む、第1タンパク質結合パートナーのライブラリーを提供すること;
第2野生型ポリペプチド及び第2の複数の変異ポリペプチドを含む、第2タンパク質結合パートナーのライブラリーを提供すること;
第1タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーと、第2タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーとの親和性測定値を測定すること;
第1タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーと、第2タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーとのそれぞれの前記親和性測定値に基づいて、
(i)前記第1の複数の変異ポリペプチドの1つのポリペプチド、及び
(ii)前記第2の複数の変異ポリペプチドの1つのポリペプチド、
を含むタンパク質結合パートナーの1つ又は複数の対を同定すること;
を含み、
タンパク質結合パートナーの前記1つ又は複数の対の各対の前記親和性測定値が、タンパク質結合パートナーのそれぞれの前記対間のそれぞれの親和性期待値と実質的に異なり、
タンパク質結合パートナーの所定の対の親和性期待値が、
a)前記所定の対の前記第1野生型ポリペプチドと、前記所定の対の前記第2の複数の変異ポリペプチドの前記1つのポリペプチドとの親和性測定値、及び
b)前記所定の対の前記第1の複数の変異ポリペプチドの前記1つのポリペプチドと、前記所定の対の前記第2野生型ポリペプチドとの親和性測定値に基づいて算出される、方法。 - 第1タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーが、第1の複数の酵母細胞の1つの表面上に発現され、且つ第2タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーが、第2の複数の酵母細胞の1つの表面上に発現される、請求項1に記載の方法。
- 第1タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーと第2タンパク質結合パートナーの前記ライブラリーの各タンパク質結合パートナーとの親和性測定値が、前記第1の複数の酵母細胞と、前記第2の複数の酵母細胞との合成凝集によって測定される、請求項2に記載の方法。
- タンパク質結合パートナーの前記第1ライブラリーの各タンパク質結合パートナーが、抗体、scFv、Fab、又はVHH種である、請求項1に記載の方法。
- タンパク質結合パートナーの前記第2ライブラリーの各タンパク質結合パートナーが、抗原種である、請求項4に記載の方法。
- タンパク質結合パートナーの前記第1ライブラリーの各タンパク質結合パートナーが、受容体種である、請求項1に記載の方法。
- タンパク質結合パートナーの前記第2ライブラリーの各タンパク質結合パートナーが、リガンド種である、請求項6に記載の方法。
- 前記第1の複数の変異ポリペプチドのそれぞれと、前記第2の複数の変異ポリペプチドのそれぞれが、ユーザー指定変異誘発によって産生される、請求項1に記載の方法。
- タンパク質結合パートナーの前記1つ又は複数の対の各対の親和性測定値が、タンパク質結合パートナーの前記対の親和性期待値よりも実質的に高い、請求項1に記載の方法。
- タンパク質結合パートナーの前記1つ又は複数の対の各対の親和性測定値が、タンパク質結合パートナーの前記対の親和性期待値の2倍を超えて高い、請求項9に記載の方法。
- タンパク質結合パートナーの前記1つ又は複数の対の各対の親和性測定値が、タンパク質結合パートナーの前記対の親和性期待値より実質的に低い、請求項1に記載の方法。
- タンパク質結合パートナーの前記1つ又は複数の対の各対の親和性測定値が、タンパク質結合パートナーの前記対の親和性期待値の2倍超低い、請求項11に記載の方法。
- 2つのタンパク質結合パートナー間の代償性変異を同定する方法であって:
第1野生型ポリペプチド及び第1の複数の変異ポリペプチドを含む、タンパク質結合パートナーの第1ライブラリーを提供すること;
第2野生型ポリペプチド及び第2の複数の変異ポリペプチドを含む、タンパク質結合パートナーの第2ライブラリーを提供すること;
タンパク質結合パートナーの前記第1ライブラリーの各タンパク質結合パートナーと、タンパク質結合パートナーの前記第2ライブラリーの各タンパク質結合パートナーとの親和性測定値を測定すること;
タンパク質結合パートナーの前記第1ライブラリーの各タンパク質結合パートナーと、タンパク質結合パートナーの前記第2ライブラリーの各タンパク質結合パートナーとの親和性測定値に基づいて、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に異なる、それぞれの親和性測定値を有するタンパク質結合パートナーの1つ又は複数の対を同定すること;
を含む方法。 - タンパク質結合パートナーの前記1つ又は複数の対が以下の条件:
a.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;
b.前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;及び
c.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;
を満たす、請求項13に記載の方法。 - タンパク質結合パートナーの前記1つ又は複数の対が以下の条件:
a.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;
b.前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;及び
c.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じであること;
を満たす、請求項13に記載の方法。 - タンパク質結合パートナーの前記1つ又は複数の対が以下の条件:
a.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;
b.前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;及び
c.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;
を満たす、請求項13に記載の方法。 - タンパク質結合パートナーの前記1つ又は複数の対が以下の条件:
a.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;
b.前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値と実質的に同じ、又は実質的に高いこと;及び
c.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2の複数のポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;
を満たす、請求項13に記載の方法。 - タンパク質結合パートナーの前記1つ又は複数の対が以下の条件:
a.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値、又は前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1野生型ポリペプチドと前記第2野生型ポリペプチドとの親和性測定値よりも実質的に低いこと;
b.前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値が、前記第1の複数の変異ポリペプチドの1つのポリペプチドと、前記第2野生型ポリペプチドとの親和性測定値、又は前記第1野生型ポリペプチドと、前記第2の複数の変異ポリペプチドの1つのポリペプチドとの親和性測定値よりも実質的に高いこと;
を満たす、請求項13に記載の方法。 - 前記第1の複数の変異ポリペプチドの前記1つのポリペプチドの変異が、前記抗体、scFv、Fab、又はVHH種のパラトープを定義し、及び/又は前記第2の複数の変異ポリペプチドの前記1つのポリペプチドの変異が、前記抗原種のエピトープを定義する、請求項5に記載の方法。
- 前記第1の複数の変異ポリペプチドの前記1つのポリペプチドの変異及び前記第2の複数の変異ポリペプチドの前記1つのポリペプチドの変異によって、
a.前記第1の複数の変異ポリペプチドの前記1つのポリペプチドが、前記第2野生型ポリペプチド及び前記第2の複数の変異ポリペプチドの前記1つのポリペプチドに結合し;
b.前記第2の複数の変異ポリペプチの前記1つのポリペプチドが、前記第1の複数の変異ポリペプチドの前記1つのポリペプチドに結合し、前記第2野生型ポリペプチドには結合しない;ような、
前記第1の複数の変異ポリペプチドの前記1つのポリペプチドと、前記第2の複数の変異ポリペプチドの前記1つのポリペプチドとの間の直交結合関係が生じる、請求項1に記載の方法。 - 前記第1の複数の変異ポリペプチドの前記1つのポリペプチドの変異及び前記第2の複数の変異ポリペプチドの前記1つのポリペプチドの変異によって、
a.前記第1の複数の変異ポリペプチドの前記1つのポリペプチドが、前記第2の複数の変異ポリペプチドの前記1つのポリペプチドに結合し、前記第2野生型ポリペプチドには結合しない;及び
b.前記第2の複数の変異ポリペプチの前記1つのポリペプチドが、前記第1の複数の変異ポリペプチドの前記1つのポリペプチドに結合し、前記第1野生型ポリペプチドには結合しない;ような、
前記第1の複数の変異ポリペプチドの前記1つのポリペプチドと、前記第2の複数の変異ポリペプチドの前記1つのポリペプチドとの間の直交結合関係が生じる、請求項1に記載の方法。
