Abstract
Description

動物におけるアタキシン2(ATXN2)mRNA及びタンパク質の発現を低下させるための組成物及び方法が提供される。このような方法は、脊髄小脳失調症2型(SCA2)、筋萎縮性側索硬化症(ALS)、及びパーキンソン症を含む神経変性疾患を、動物におけるアタキシン2の発現を阻害することによってアタキシン2の発現を阻害することによって、治療、予防、または寛解させるのに有用である。

直列アンチセンスオリゴヌクレオチドの2個または3個の配列からそれぞれ構成された28個及び42個の核酸塩基アンチセンスオリゴヌクレオチドを、ウサギ網状赤血球アッセイにおけるヒトDHFRの翻訳を捕捉する能力について検査した。当該3つの14核酸塩基アンチセンスオリゴヌクレオチド単独の各々は、28個または42個の核酸塩基アンチセンスオリゴヌクレオチドよりも中程度のレベルにあるにもかかわらず、翻訳を阻害することができた。
ある実施形態において、アタキシン2核酸を標的としたアンチセンス化合物は、阻害活性の亢進、標的核酸に対する結合親和性の亢進、またはインビボでのヌクレアーゼによる分解に対する耐性などの特性を当該アンチ化合物へ与えるために、パターン、またはモチーフにおいて配置された化学的に修飾されたサブユニットを有する。
アタキシン2をコードするヌクレオチド配列には、以下、すなわちGENBANK受託番号NM_002973.3(配列番号1として本明細書に組み込まれる)、ヌクレオチド2465000~2616000を切り詰められたGENBANK受託番号NT_009775.17の相補体(配列番号2として本明細書に組み込まれる)及びGENBANK受託番号BX410018.2(配列番号3として本明細書に組み込まれる)が含まれるが、これらに限定しない。

アンチセンスオリゴヌクレオチドは、医薬組成物または製剤の調製のための医薬として許容され得る活性物質または不活性物質と混合され得る。医薬組成物の製剤のための組成物及び方法は、投与経路、疾患の程度、または投与されるべき用量を含むがこれらに限定しないいくつかの基準による。
化合物及び医薬として許容され得る希釈剤を含む医薬組成物が本明細書で説明する方法において採用される。ある実施形態において、当該医薬として許容され得る希釈剤はPBSである。ある実施形態において、当該アンチセンス化合物は、アンチセンスオリゴヌクレオチドである。
アンチセンス化合物は、結果として生じるアンチセンスオリゴヌクレオチドの活性、細胞内分布または細胞内取り込みを亢進する1つ以上の部分または複合体へ共有結合し得る。典型的な複合体基には、コレステロール部分及び脂質部分が含まれる。追加の複合体基には、炭水化物、リン脂質、ビオチン、フェナジン、葉酸、フェナントリジン、アントラキノン、アクリジン、フルオレセイン、ローダミン、クマリン、及び色素が含まれる。
アタキシン核酸のレベル、活性または発現に及ぼすアンチセンス化合物の効果は、種々の細胞型においてインビトロで検査することができる。このような分析に使用する細胞型は、商業的売主(例えば、米国培養細胞系統保存機関,バージニア州マナサス市、Zen-Bio,Inc.,ノースカロライナ州リサーチトライアングルパーク地区、Clonetics Corporation,メリーランド州ウォーカーズビル町)から入手可能であり、市販の試薬(例えば、Invitrogen Life Technologies,カリフォルニア州カールスバッド市)を用いて売主の説明書により培養される。実例となる細胞型には、HepG2細胞、Hep3B細胞、及び初代肝細胞が含まれるが、これらに限定しない。
他のアンチセンス化合物を用いた処置のために適切に修正することのできる、アンチセンスオリゴヌクレオチドを用いた細胞の処置のための方法が、本明細書で説明される。
RNA分析は、全細胞RNAまたはポリ(A)+mRNAに関して実施することができる。RNA単離の方法は、当該技術分野で周知である。RNAは、例えば、製造元の推奨するプロトコルによりTRIZOL試薬(Invitrogen,カリフォルニア州カールスバッド市)を用いて、当該技術分野で周知の方法を用いて調製する。
アタキシン2核酸のレベルまたは発現の阻害は、当該技術分野で公知の種々の方法においてアッセイすることができる。例えば、標的核酸レベルは、例えば、ノザンブロット分析、競合的ポリメラーゼ連鎖反応(PCR)、または定量的リアルタイムPCRによって定量化することができる。RNA分析は、全細胞RNAまたはポリ(A)+mRNAに関して実施することができる。RNA単離方法は当該技術分野で周知である。ノザンブロット分析も当該技術分野で所定である。定量的リアルタイムPCRは、PE-Applied Biosystems(カリフォルニア州フォスター市)から入手可能なかつ製造元
の説明書により使用される市販のABI PRISM 7600、7700、または7900を用いて簡便に達成することができる。
標的RNAレベルの定量化は、製造元の説明書により、ABI PRISM 7600、7700、または7900配列検出システム(PE-Applied Biosystems、カリフォルニア州フォスター市)を用いた定量的リアルタイムPCRによって達成され得る。定量的リアルタイムPCRの方法は、当該技術分野で周知である。
アタキシン2核酸のアンチセンス阻害は、アタキシン2タンパク質レベルを測定することによって評価することができる。アタキシン2のタンパク質レベルは、免疫沈降、ウェスタンブロット分析(イムノブロッティング)、酵素結合免疫吸着測定法(ELISA)、定量的タンパク質アッセイ、タンパク質活性アッセイ(例えば、カスパーゼ活性アッセイ)、免疫組織化学、免疫細胞化学または蛍光標識細胞分取法(FACS)など、当該技術分野で周知の種々の方法において評価または定量化することができる。標的へ向かう抗体は、抗体のMSRSカタログ(Aerie Corporation,ミシガン州バーミンガム市)など、種々の源から識別及び入手することができ、または当該技術分野で周知の従来のモノクローナル抗体もしくはポリクローナル抗体作製方法を介して調製することができる。
アンチセンス化合物、例えば、アンチセンスオリゴヌクレオチドは、運動機能及び認知の改善など、アタキシン2の発現を阻害しかつ表現型の変化を生じる当該化合物の能力を評価するために、動物において検査する。ある実施形態において、当該動物における運動機能は、歩行開始分析、ロータロッド、握力、登り棒、屋外作業成績、平均台、後足足跡
検査によって測定される。
ある実施形態において、本明細書で説明する1つ以上の医薬組成物を投与することを含む、個体を治療する方法、化合物、及び組成物が本明細書で提供される。ある実施形態において、当該個体は神経変性疾患を有している。ある実施形態において、当該個体は、脊髄小脳失調症2型(SCA2)、筋萎縮性側索硬化症(ALS)、及びパーキンソン症を含むがこれらに限定しない神経変性疾患を発達させることについての危険がある。ある実施形態において、当該個体は、アタキシン2関係疾患を有しているものと識別されている。ある実施形態において、個体におけるアタキシン2発現を予防的に低下させるための方法が本明細書で提供される。ある実施形態には、個体へ治療有効量の、アタキシン2核酸を標的としたアンチセンス化合物を投与することによって、治療を必要とする個体を治療することが含まれる。
本明細書で説明するある化合物、組成物、及び方法は、ある実施形態に従って具体的に説明されてきたが、以下の実施例は、本明細書で説明する化合物を説明するためにのみ供されており、当該化合物を制限するよう意図するものではない。本出願において列挙される参考文献の各々は、その内容が全体として参照により本明細書に組み込まれる。
アタキシン2核酸を標的とするアンチセンスオリゴヌクレオチドを設計し、インビトロでのアタキシン2mRNAに及ぼす当該アンチセンスオリゴヌクレオチドの効果について検査した。当該アンチセンスオリゴヌクレオチドは、類似の培養条件を有する一連の実験において検査した。各実験についての結果は、下記に示す個別の表において呈される。ウェルあたり20,000個の密度の培養HepG2細胞に4,500nMアンチセンスオリゴヌクレオチドを、電気穿孔法を用いてトランスフェクトした。およそ24時間の処置期間ののち、RNAを細胞から単離し、アタキシン2mRNAレベルを定量的リアルタイムPCRによって測定した。ヒトプライマープローブセットRTS3642(配列番号5として本明細書で指定の順方向配列ACCAAAGAGTAGTTAATGGAGGTGTTC、配列番号6として本明細書で指定の逆方向配列AGAAGGTGGGCGAGAGGAA、配列番号7として本明細書で指定のプローブ配列CTGGCCATCGCCTTGCCCA)を用いてmRNAレベルを測定した。アタキシン2mRNAレベルは、RIBOGREEN(登録商標)によって測定するように、全RNA含有量により調整した。結果は、未処置の対照細胞に対するアタキシン2の%阻害として呈する。
アタキシン2mRNAの有意なインビトロでの阻害を呈する実施例1由来のギャップマーを選択し、HepG2細胞において種々の用量で検査した。細胞をウェルあたり20,000個の密度で播種し、下記の表に指定の通り、0.625μM、1.250μM、2.500μM、5.000μM及び10.000μMの濃度のアンチセンスオリゴヌクレオチドを、電気穿孔法を用いてトランスフェクトした。およそ16時間の処置期間後、RNAを細胞から単離し、アタキシン2mRNAレベルを定量的リアルタイムPCRによって測定した。ヒトプライマープローブセットRTS3642を用いてmRNAレベルを測定した。アタキシン2mRNAレベルをRIBOGREEN(登録商標)によって測定するように、全RNAにより調整した。結果は、未処置の対照細胞に対するアタキシン2の%阻害として呈する。
アタキシン2mRNAの有意なインビトロでの阻害を呈する実施例1由来のギャップマーを選択し、SCA2[Q22]-BACマウスモデルにおいてインビボで検査した。このマウスモデルは、脊髄小脳失調症2型(SCA2)の研究用に、FVB/B6複合背景のマウスを用いてPulst研究室(ユタ大学、ソルトレイクシティ)において作出した。これらのマウスは、16kbの上流配列及び2.5kbの下流配列を含む全176kbのヒトATXN2遺伝子領域を有している。
各3匹のマウスからなる複数の群に正常塩類溶液(0.9%)またはアンチセンスオリゴヌクレオチドを、脳室内注射を介して投与した。5~7週齢のマウスに酸素及び3%イソフルランからなる混合物を3~4分間個々に注入して鎮静を生じた。次に、頭皮上の毛髪を剪断器具で除去した。当該マウスを定位固定機(マウス用Stoelting Just)に配置した。頭皮をまずヨウ素スクラブで、次に70%エタノールで清浄化した。切開部を両眼の間の箇所の直後の領域から1.5cm後ろの領域まで10番メスで作製した。滅菌綿棒を用いて骨膜を除去した。26ゲージ針のハミルトン注射器を定位固定機の針ホルダーに配置し、正常塩類溶液(0.9%)または塩類溶液(0.9%)中のアンチセンスオリゴヌクレオチド(250μg)のいずれかで10μLの指標まで満たした。針を頭蓋上の十字縫合上に位置取った後、右へ1mmかつ0.46mm後部に位置取った。次に、針の先端を頭蓋のみを通過させて挿入した後、右側脳室へと2.5mm下に位置取った。次に、注射器のプランジャーを押下して、5~7μLの所望の容積を送達した。室内圧を等化させておくために4分間待機した後、針を除去し、頭皮を縫合した。次に、切開部をポビドン溶液で処置し、マウスをケージへ回復のため仰向けで戻した。マウスは毎日モニターした。
7日後、マウスをもはや呼吸しなくなるまでイソフルラン中に置いた。次に、脳を摘出した。RNA分析用の3mm切片1つを含む脳の3つの部分を冠状切片で収集した。RNAをRNeasyキット(Qiagen)を用いて30mg組織から単離した。QuantiTect逆転写キット(Qiagen)を用いてcDNAを生成した。リアルタイムPCR(qPCR)を4つ組で96ウェルプレート中でiCycler(Bio-Rad)において標準曲線を用いてSYBR Green法によって実施した。反応物は20μLであり、15ngのcDNA、2μLの各プライマー(終濃度0.3μM)、及び10μLのSYBR Greem Master Mix(Bio-Rad)からなった。周期パラメータには、10秒間の950変性工程、20秒間のアニーリング温度でのインキュベーション及び720で40秒間の第二のインキュベーションが含まれていた。各プレートは、複数のpGL2-5A3トランスジェニックマウスから調製した小脳RNAを用いた標準曲線を含んでいた。単一のアンプリコンは、変性分析及びゲル電気泳動法によって確認した。
アタキシン2mRNAの有意なインビトロでの阻害を呈する実施例1由来のギャップマーを選択し、ATXN2-Q127マウスモデルにおいてインビボで検査した。このマウスモデル(Hansen,S.T.ら,Human.Molecular Genetics 2012,1~13)は、プルキンエ細胞タンパク質2型(Pcp2)プロモーターの調節下で全長の突然変異体ATXN2Q127相補的DNAを発現する。このモデルは、小脳プルキンエ細胞における散在性細胞質凝集体の形成を付随する早期発症型進行性運動障害表現型を示す。

Claims (24)
- 12~30の連結したヌクレオシドからなるかつ、配列番号11~165の核酸塩基配列のいずれかの少なくとも8、少なくとも9、少なくとも10、少なくとも11、少なくとも12、少なくとも13、少なくとも14、少なくとも15、少なくとも16、少なくとも17、少なくとも18、少なくとも19、または少なくとも20の連続した核酸塩基を含む核酸塩基配列を有する修飾されたオリゴヌクレオチドを含む、化合物。
- 前記修飾されたオリゴヌクレオチドの前記核酸配列は、配列番号1、配列番号2、または配列番号3と少なくとも80%、少なくとも81%、少なくとも82%、少なくとも83%、少なくとも84%、少なくとも85%、少なくとも86%、少なくとも87%、少なくとも88%、少なくとも89%、少なくとも90%、少なくとも91%、少なくとも92%、少なくとも93%、少なくとも94%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、少なくとも99%、または100%相補的である、請求項2に記載の化合物。
- 一本鎖の修飾されたオリゴヌクレオチドからなる、請求項1~2のいずれか一項に記載の化合物。
- 少なくとも1つのヌクレオシド間連結は、修飾されたヌクレオシド間連結である、請求項1~3のいずれか一項に記載の化合物。
- 少なくとも1つの修飾されたヌクレオシド間連結は、ホスホロチオアートヌクレオシド間連結である、請求項4に記載の化合物。
- 各修飾されたヌクレオシド間連結は、ホスホロチオアートヌクレオシド間連結である、請求項4に記載の化合物。
- 少なくとも1つのヌクレオシド間連結は、ホスホジエステルヌクレオシド間連結である、請求項1~6のいずれか一項に記載の化合物。
- 少なくとも1つのヌクレオシド間連結は、ホスホロチオアート連結であり、及び少なくとも1つのヌクレオシド間連結はホスホジエステル連結である、請求項1~7のいずれか
一項に記載の化合物。 - 少なくとも1つのヌクレオシドは、修飾された核酸塩基を含む、請求項1~8のいずれか一項に記載の化合物。
- 前記修飾された核酸塩基は、5-メチルシトシンである、請求項9に記載の化合物。
- 前記修飾されたオリゴヌクレオチドのうちの少なくとも1つのヌクレオシドは、修飾さ
れた糖を含む、請求項1~10のいずれか一項に記載の化合物。 - 前記少なくとも1つの修飾された糖は、二環式糖である、請求項11に記載の化合物。
- 前記二環式糖は、4’-CH(R)-O-2’架橋を含み、式中Rは独立して、H、C1~C12アルキル、または保護基である、請求項12に記載の化合物。
- Rはメチルである、請求項13に記載の化合物。
- RはHである、請求項13に記載の化合物。
- 前記少なくとも1つの修飾された糖は、2’-O-メトキシエチル基を含む、請求項11に記載の化合物。
- 前記修飾されたオリゴヌクレオチドは、
10個の連結したデオキシヌクレオシドからなるギャップセグメント、
5個の連結したヌクレオシドからなる5’ウィングセグメント、及び
5個の連結したヌクレオシドからなる3’ウィングセグメント
を含み、この中で前記ギャップセグメントは、前記5’ウィングセグメントと前記3’ウィングセグメントの間に位置し、かつこの中で各ウィングセグメントの各ヌクレオシドは、修飾された糖を含む、請求項1~16のいずれか一項に記載の化合物。 - 前記修飾されたオリゴヌクレオチドは、20個の連結したヌクレオシドからなる、請求項1~17のいずれか一項に記載の化合物。
- 請求項1~18のいずれか一項に記載の化合物及び医薬として許容され得る担体または希釈剤のうちの少なくとも1つを含む、組成物。
- 請求項1~19のいずれか一項に記載の化合物または組成物を動物へ投与することを含む、方法・
- 前記動物はヒトである、請求項20に記載の方法。
- 前記化合物を投与することは、アタキシン2関係疾患、障害または容態の進行を予防、治療、寛解、または遅延させる、請求項20及び21に記載の方法。
- 前記疾患、障害または容態は、脊髄小脳失調症2型(SCA2)、筋萎縮性側索硬化症(ALS)、またはパーキンソン症である、請求項22に記載の方法。
- 神経変性障害を治療するための医薬剤の製造のための、請求項1~23のいずれか一項に記載の化合物または組成物の使用。
