一部の実施形態では、本文で提供される抗体はヒト抗体である。ヒト抗体は、本分野で既知の様々な技術によって生じることができる。
上記の形態のいずれかにおいて、本文で提供される抗LAG−3抗体は、二重特異性抗体などの多重特異性抗体である。多重特異性抗体は、少なくとも二つの異なるサイトに結合特異性を有するモノクローナル抗体である。一部の実施形態では、一つの結合特異性はLAG−3に対するものであり、もう一つの結合特異性はいずれかその他の抗原(例えば、第2の生体分子、細胞表面抗原、腫瘍抗原)に対するものである。それ相応に、二重特異性抗LAG−3抗体は、LAG−3、および、CD3、CD20、FcRH5、HER2、LYPD1、LY6G6D、PMEL17、LY6E、CD19、CD33、CD22、CD79A、CD79B、EDAR、GFRA1、MRP4、RET、Steap1またはTenB2などの腫瘍抗原に対して結合特異性を有し得る。二重特異性抗体は、完全長抗体または抗体フラグメントとして調製することもできる。
本発明の抗体は、本発明の抗LAG−3抗体のアミノ酸配列変異体を包含する。例えば、抗体の結合親和性および/またはその他の生物学的特性をさらに改善するために調製される抗体変異体が必要になる場合がある。抗体のアミノ酸配列変異体は、その抗体をコードするヌクレオチド配列に適切な修飾を導入することにより、またはペプチド合成により調製できる。このような修飾は、例えば、アミノ酸配列内の残基の欠失および/または挿入および/または置換を含む。欠失、挿入、および置換を任意に組み合わせて最終構築体を得ることができ、その条件として、最終構築体が所望の特徴、例えば抗原結合、を備える必要がある。
一部の実施形態では、一つ以上のアミノ酸置換を有する抗体変異体が提供される。置換型突然変異誘発の関連サイトはHVRとFRを含む。保存的な置換は、下記の「好ましい置換」という見出しの下に表示される。より実質的な変化は、下記の「例示的置換」という見出しの下で提供され、且つ後文でアミノ酸側鎖のタイプを参照しながらさらに説明される。アミノ酸置換を関連抗体に導入し、所望の活性(例えば、抗原結合の保持/改善、またはADCCまたはCDCの改善)について生成物をスクリーニングすることができる。

本発明の抗LAG−3抗体は、例えば米国特許第4,816,567号に記載されているように、組み換え方法で調製することができる。一実施形態では、本文に記載の抗LAG−3抗体をコードする単離された核酸が提供される。このような核酸は、該抗体(例えば、該抗体の軽鎖および/または重鎖)のVLを含むアミノ酸配列および/またはそのVHを含むアミノ酸配列をコードすることができる。別の一実施形態では、このような核酸を含む1つ以上のベクター(例:発現ベクター)が提供される。別の一実施形態では、このような核酸を含む宿主細胞が提供される。このような一実施形態では、宿主細胞は、(1)該抗体のVLを含むアミノ酸配列および該抗体のVHを含むアミノ酸配列をコードする核酸のベクター;または、(2)該抗体のVLを含むアミノ酸配列をコードする核酸を含む第1のベクター、および該抗体のVHを含むアミノ酸配列をコードする核酸を含む第2のベクターを含む(例えば、改質転換によってそれを有する)。一実施形態では、該宿主細胞は、チャイニーズハムスター卵巣(CHO)細胞またはリンパ球様細胞(例えば、Y0、NS0、Sp20細胞)などの真核細胞である。一実施形態では、抗LAG−3抗体を製造する方法が提供され、該方法は、該抗体の発現に適した条件下で前文で提供された該抗体をコードする核酸を含む宿主細胞を培養すること、および該宿主細胞(または宿主細胞の培地)から該抗体を回収してもよいことを含む。
本発明はまた、化学療法剤または化学療法薬剤、成長阻害剤、毒素(例えば、タンパク質毒素、細菌、真菌、植物または動物由来の酵素活性毒素またはそのフラグメント)または放射性同位体などの一つ以上の細胞毒性剤に本文の抗LAG−3抗体が結合した免疫複合体を提供する。
本発明の抗LAG−3抗体の薬物製剤は、所望の純度を有する抗体を一つ以上の選択されてもよい薬学的に許容される担体(Remington‘s Pharmaceutical Science第16版、Osol,A.ed.(1980))と混合して凍結乾燥製剤または水溶液を形成して調製される。薬学的に許容される担体は、使用される用量および濃度下において一般的に被験者に対して毒性がなく、且つ、リン酸塩、クエン酸塩およびその他の有機酸などのバッファー;アスコルビン酸やメチオニンを含む酸化防止剤;防腐剤(例えば、オクタデシルジメチルベンジルアンモニウムクロリド;塩化ヘキサメトニウム;塩化ベンザルコニウム;塩化ベンゼト二ウム;フェノール、ブタノールまたはベンジルアルコール;メチルパラベンまたはプロピルパラベンなどのp−ヒドロキシ安息香酸アルキルエステル;カテコール;レソルシノール;シクロヘキサノール;3−ペンタノール;m−クレゾール);低分子量(約10個より少ない残基を有する)ポリペプチド;血清アルブミン、ゼラチン、または免疫グロブリンなどのタンパク質;ポリビニルピロリドンなどの親水性重合体;グリシン、グルタミン、アスパラギン酸、ヒスチジン、アルギニンまたはリシンなどのアミノ酸;グルコース、マンノース、またはデキストリンを含む、単糖、二糖及びその他の炭水化物;EDTAなどのキレート剤;ショ糖、マンニトール、フコースまたはソルビトールなどの糖類;ナトリウムのような塩形成対イオン;金属錯体(例えば、亜鉛−タンパク錯体);および/またはポリエチレングリコール(PEG)のような非イオン界面活性剤を含むが、これらに限られていない。
本発明のもう一態様において、本発明の抗体または医薬組成物を含む製品が提供される。該製品は、容器と、その容器上にあるまたはその容器に関連付けられたラベルまたはプロトコルを含む。適切な容器はボトル、バイアル、注射器、IV溶液バッグなどを含む。このような容器は、ガラスまたはプラスチックなどの様々な材料から作られることができる。該容器は、本発明の組成物自体または該組成物と別の組成物との組み合わせを収納し、しかも無菌の入口を有してもよい(例えば、該容器は静脈内溶液バッグ、または皮下注射針によって貫通可能な栓を含むバイアルであってもよい)。該組成物中の少なくとも一種類の活性化剤は本発明の抗体である。前記ラベルまたはプロトコルは、選択された腫瘍またはウイルス感染の治療に前記組成物を使用することを説明する。また、前記製品は、(a)本発明の抗体を含む組成物を含有する第1の容器と、(b)別の細胞毒性剤またはその他の治療剤を含む組成物を含有する第2の容器とを含んでもよい。本発明のこの実施形態における製品は、このような組成物を腫瘍またはウイルス感染の治療に使用可能であることを説明するためのプロトコルをさらに含んでもよい。選択されてもよいが、若しくはまた、該製品はさらに第2(または第3)の容器を含んでもよく、その容器は、静菌注射用水(BWFI)、リン酸塩緩衝生理食塩水、リンゲル液、デキストロース溶液などの薬学的に許容されるバッファーを含む。さらには、その他のバッファー、希釈剤、フィルター、針および注射器を含む、商業および使用者の角度から見ると必要とされるその他の材料を含んでもよい。
1.1 ヒトLAG−3組み換え融合タンパク質に高親和性で結合する単鎖抗体のパニング
本実施例は、完全なヒト単鎖scFvファージディスプレイライブラリーからヒトLAG−3組み換え融合タンパク質に高い親和性で結合する単鎖抗体をパニングする方法について説明する。この方法は、後述の完全ヒト単鎖scFvファージディスプレイライブラリーの使用に係り、市販のヒトLAG−3−6*His組み換え融合タンパク質(Novoprotein、cat#CJ91)、大腸菌TG1(Lucigen、cat#60502−1)、およびM13KO7ヘルパーファージ(Thermo Fisher、cat#18311019)の使用に係る。本実施例に係る全ヒト単鎖scFvファージディスプレイライブラリーは、pBR3.22プロモーターとM13ファージpIIIタンパク質ディスプレイシステムを含むファージミドベクターpCLS1.0(配列1)から構築され、そのC末端にはMyc−Hisタグが含まれている。ライブラリーの構築時に大腸菌TG1が使用された。当該全ヒト単鎖scFvファージディスプレイライブラリーは、文献でよく知られている通常の方法によって構築され、下記のように簡単に説明する。すなわち、RNA抽出キット(TaKaRa、cat#9767)を利用して市販の健康人PBMCs(Allcells、cat#PB−003F−S、50サンプル)からRNAを獲得し、各全ヒトscFv単鎖ファージディスプレイサブライブラリーは、5または10のサンプルのRNAを混合して構築された。逆転写キット(Thermo Fisher、cat#4368814)を使用してRNAを逆転写してcDNAの第一鎖を合成し、ヒト抗体可変領域遺伝子を増幅するための特異性プライマーを文献(CaiXH,GarenA.PNAS,1995.92(14):6537−6541)およびV−baseデータベースに従って設計して合成した。cDNA第一鎖をテンプレートとして、ヒト抗体重鎖可変領域と軽鎖可変領域をそれぞれPCRで増幅した。分子クローニング技術を利用してヒト抗体重鎖可変領域および軽鎖可変領域遺伝子フラグメントをファージミドベクターpCLS1.0にクローニングした。具体的には、ヒト抗体軽鎖可変領域をNhe I/Not I二重消化によってベクターpCLS1.0にクローニングし、大腸菌TG1にエレクトロポレーションにより形質転換して、ファージ軽鎖サブライブラリーを得てベクターpCLS1.0−VLを得た。続いてSfi I/Xhо I二重消化を利用してヒト抗体重鎖可変領域をベクターpCLS1.0−VLにクローニングした。同じ方法を使用して重鎖可変領域と軽鎖可変領域の組み合わせを備えた全ヒトscFvファージディスプレイサブライブラリーを得た。各サブライブラリーの合計ストレージ容量は9×109である。
パニング実験におけるヒトLAG−3−6*Hisタグ融合タンパク質はいずれもバイオチンラベリングキット(Biotin Labeling Kit−NH2(Dojindo,cat#LK03))を使用してバイオチンを標識した。パニング実験を以下のように簡単に説明する。すなわち、大腸菌TG1を使用してM13KO7ヘルパーファージを調製し、測定力価は約1.0×1013であり、それをOD600値が0.5〜0.6である上記全ヒトscFv単鎖ファージディスプレイライブラリーに感染させて一回目の入力(input)ファージを取得した。液相パニングストラテジーを利用し、事前にDynabeads(登録商標)M−280 Streptavidin(Thermo,cat#11206D)とプレインキュベーションしたビオチン化ヒトLG−3−6*Hisタグ組み換え融合タンパク質によりヒトLAG−3のscFv単鎖抗体に結合するファージを濃縮した。濃縮された前記単鎖抗体ファージをpH2.2、0.1Mグリシン‐塩酸溶液で溶出し、一回目の出力(оutput)ファージを取得した。一回目の出力ファージをOD600値が0.5〜0.6であるTG1大腸菌に感染させて二回目の入力ファージを取得し、続いて上記生物学的パニング方法を使用してヒトLAG−3−6*Hisタグ組み換え融合タンパク質に結合する二回目の出力ファージを得た。二回目の生物学的パニング方法を繰り返して三回目または四回目のパニングを行い、三回目または四回目のパニングを経て濃縮された出力ファージを大腸菌TG1に感染させてモノクローンを得て、ヒトLAG−3−6*Hisタグ組み換え融合タンパク質と高い親和性を有するモノクローナル大腸菌をファージELISAにより同定、選択して配列決定の分析をし、19個の単一ヌクレオチド配列を得た。具体的な配列を表1に示す。
1.2 フローサイトメトリーによりLAG−3タンパク質に結合する単鎖抗体を選択してIgG転換を行った
単一ヌクレオチド配列を持つ19個のモノクローナルscFv単鎖抗体が細胞表面LAG−3タンパク質に対する結合能力をテストするために、これらのscFv単鎖抗体を持つ大腸菌モノクローンをIPTGで誘導して可溶性scFv単鎖抗体タンパク質を発現させ、ろ過された上清と活性化されたヒトCD4+T細胞を取って結合活性を検出した。ヒトCD4+T細胞濃縮キット(STEMCELL、cat#19052)を使用してヒト末梢血PBMCsからCD4+T細胞を単離した。そして、単離されたCD4+T細胞をCD3/CD28 Dynabeads(Gibco、cat#11131D)と共インキュベーションし、5%CO2インキュベーターで37℃で48時間活性化した。前記IPTGにより誘導発現された19個の可溶性scFv単鎖抗体タンパク質、2YT培地(Sangon Biotech (Shanghai) Co., Ltd.、cat#SD7019)およびブランク対照PBS(Hyclone、cat#SH30256.01)をそれぞれ活性化されたヒトCD4+T細胞と4℃で1時間共インキュベーションした。次いで、細胞を冷1×PBSバッファーで二回洗浄した。1:50希釈のanti−His−PEタグ抗体(Miltenyi Biotec、cat#130−092−691)を入れて混合物を4℃で30分間インキュベーションした後、冷1×PBSバッファーで二回洗浄し、Guava easyCyte HTフローサイトメーター(MERCK MILLIPORE)により、活性化されたCD4陽性T細胞に対する前記誘導発現されたscFv単鎖抗体の結合活性を分析した。フローサイトメトリー中にLAG−3を発現するCD4陽性T細胞はゲーティングにより選択された。結果は表2に示されるように、モノクローンにより誘導発現された19個の可溶性scFv単鎖抗体タンパク質のうち6個の抗体タンパク質(11446、11452、11454、11455、11471、11487)は、活性化されたヒトCD4陽性T細胞に対する最高の結合活性を有する。
11446、11452、11454、11455、11471、11487の6つのscFv単鎖抗体遺伝子を選択して分子クローニングし、IgG4に転換した。一般的には、リーダーペプチド、Nhe I/Not I消化サイト、重鎖遺伝子定常領域およびヒトIgG4−Fcを含む配列をpCDNA3.3+にクローニングして構築し、ベクターpCDNA3.3−IgG4が得られた。同様に、リーダーペプチド、Nhe I/BsiW I消化サイト、軽鎖Kappa遺伝子定常領域を含む配列をpCDNA3.3+にクローニングして構築し、ベクターpCDNA3.3−VKappaが得られ、または、リーダーペプチド、BamH I/Hind III消化サイト、軽鎖lambda遺伝子定常領域を含む配列をpCDNA3.3+にクローニングして構築し、ベクターpCDNA3.3−VLambdaが得られた。前記scFv単鎖抗体の重鎖可変領域遺伝子ヌクレオチド配列の両端にNhe I/Not Iサイトを加えてpCDNA3.3−IgG4プラスミドの対応するサイトに挿入し、前記scFv単鎖抗体の軽鎖可変領域ヌクレオチド配列の両端にNhe I/BsiW Iサイトを加えてpCDNA3.3−Vkappaプラスミドの対応するサイトに挿入し、またはBamH I/Hind III消化サイトを利用してscFv単鎖抗体の軽鎖可変領域ヌクレオチド配列をpCDNA3.3−VLambdaベクターに挿入した。前記モノクローナル抗体を発現する組み換えプラスミドを取得した。表3に示されるように、その中で、すべての重鎖はIgG4である(Padlan EA Mol Immunol.1994 Feb;31(3):169−217. Anatomy of the antibody molecule)。
1.3 IgG4モノクローナル抗体の発現、精製、同定および結合活性
上記表3に記載されている重鎖可変領域と軽鎖可変領域を含む組み換えプラスミドを、ExpiFectamineTM CHO Transfection Kit(Thermo Fisher,cat#A29129)リポソーム法を使用して懸濁培養されたExpiCHO−S細胞(Thermo Fisher,cat#A29127)を一過性トランスフェクションした。
30ml ExpiCHO Expression培地(Thermo Fisher,cat#A29100−01)において、37℃、8%CO2、120rpmの条件下で前記取得したトランスフェクションされたExpiCHO−S細胞を培養した。
10〜14日間培養されたトランスフェクション細胞を二次遠心分離処理(一次遠心分離の条件は、20分間、400gである;二次遠心分離の条件は、20分間、10000gである)し、細胞と細胞破片を除去して上清を得た。清澄化された上清をプロテインAアフィニティークロマトグラフィーカラム(GE Healthcare,cat#GE−17−5438−04)に載せ、三段階で溶出(溶出バッファーは順に、150mM Nacl、pH5.0のリン酸塩バッファー;20mMクエン酸ナトリウム‐1M 塩化ナトリウムpH5.0;20mMクエン酸ナトリウムpH5.0の溶出バッファーである)して不純物を除去した後、pH3.0の20mMクエン酸ナトリウム溶液により目標の抗体を分離して捕獲した。最後に、限外ろ過濃縮ステップにより、目標の抗体をpH7.4の1×PBSバッファーに置き換えた。
精製された6つのIgG抗体(11446、11452、11454、11455、11471、11487)について、ELISA法によりヒトLAG−3−6*Hisタグ組み換え融合タンパク質抗原に対するインビトロ結合活性を検出した。このテストは、市販のヒトLAG−3−6*Hisタグ融合タンパク質(Novoprotein、cat#CJ91)に係る。具体的には、ヒトLAG−3−6*Hisタグ融合タンパク質をコーティングバッファー(炭酸塩バッファー)で1μg/mlの濃度に希釈し、4℃で96ウェルプレート(CORNING、cat#9018)に一晩コーティングした。pH7.4の1×PBSバッファーで三回洗浄した後、5%スキムミルクで2時間ブロックした。精製された6つのIgG抗体をpH7.4の1×PBSバッファーにより100μg/mlから3倍勾配で希釈し、コーティング抗体とともに25℃で2時間共インキュベーションした。そして、抗‐hIgGタグが付けられているHRP検出抗体を使用してヒトLAG−3−6*Hisタグ組み換え融合タンパク質抗原に対する前記抗体の結合を検出した。図1と表4に示すように、11452クローンは、ヒトLAG−3−6*Hisタグ融合タンパク質抗原に対する最高の親和性を有し、該クローン11452はMV705−3と命名され、その重鎖CDR1(HCDR1)アミノ酸配列はSYGIS(配列88)であり、重鎖CDR2(HCDR2)アミノ酸配列はWISAYNGNTNYAQKLQG(配列89)であり、重鎖CDR3(HCDR3)アミノ酸配列はDGWWELLRPDDAFDI(配列90)であり、軽鎖CDR1(LCDR1)アミノ酸配列はSGDKLGDKYAY(配列91)であり、軽鎖CDR2(LCDR2)アミノ酸配列はYDSDRPS(配列92)であり、軽鎖CDR3(LCDR3)アミノ酸配列はQVWDSSSDQVV(配列93)である。
好ましいクローンMV705−3の重鎖および軽鎖遺伝子を含む組み換えプラスミドを、ExpiFectamineTM CHO Transfection Kit(Thermo Fisher,cat#A29129)リポソーム法を使用して大規模(200〜600ml)懸濁培養されたExpiCHO−S細胞(Thermo Fisher,cat#A29127)を一過性トランスフェクションし、実施例1.3に記載のトランスフェクション、培養、精製方法を用いて目標の抗体を調製した。最後に、後続の各インビトロおよびインビボ活性検出および生物学的分析実験のために、限外ろ過濃縮ステップにより、目標の抗体をpH7.4の1×PBSバッファーに置き換えた。
3.1 実施例2で得られたLAG−3モノクローナル抗体MV705−3の純度をキャピラリー電気泳動(CE−SDS)で分析した
SDS−MW Analysisキット(Beijing Biosmart Tech. Institute、cat#BSYK018)を使用して下記の方法に従ってモノクローナル抗体MV705−3サンプルに対して還元および非還元処理を行った。
非還元:100μgの抗ヒトLAG−3全ヒトモノクローナルMV705−3抗体サンプルを75μlの1%濃度のSDSバッファーに入れて、0.1M Tris−HClで容量を95μlにメスアップし、5μlのヨードアセトアミドを加え、均一にボルテックスした後、70℃で5分間インキュベーションし、8℃で1分間6000g遠心分離した。
還元:100μgの抗ヒトLAG−3全ヒトモノクローナルMV705−3抗体サンプルを75μlの1%濃度のSDSバッファーに入れて、0.1M Tris−HClで容量を95μlにメスアップし、5μlのβ−メルカプトエタノールを加え、均一にボルテックスした後、70℃で5分間インキュベーションし、8℃で1分間6000g遠心分離した。
そして、キャピラリー電気泳動装置(Beckman、型番:PA800plus)を用いて純度分析をした。その結果、還元(R−CE−SDS)と非還元(NR−CE−SDS)条件下で、本発明の、ExpiCHO−Sの一過性トランスフェクションにより発現精製された抗ヒトLAG−3全ヒトモノクローナルMV705−3抗体サンプルは、重鎖(HC)、軽鎖(LC)、およびメインピークのピーク純度のパーセンテージ(%)は表5に示される。
3.2 実施例2で得られたLAG−3モノクローナル抗体MV705−3の純度をサイズ排除クロマトグラフィー(SEC−HPLC)により分析した
20μgの抗ヒトLAG−3全ヒトモノクローナルMV705−3抗体(濃度を1mg/mlに調整した)をHPLCクロマトグラフィー(Agilent Technologies)のカラム(TOSOH、型番:TSKgel G3000SWXL)に入れた。移動相は50mMリン酸塩バッファーと300mM NaClであり、pH7.0であり、流速は0.8ml/minであり、検出波長は280nmである。その結果は図2に示される。ピークの計算は、ChemStationソフトウェアを用いて分析し、本発明の抗ヒトLAG−3全ヒトモノクローナルMV705−3抗体のメインピークの割合(%)は99.2%を超え、多量体ピークの割合(%)は1%未満である。
4.1 ELISA法によりヒトLAG−3−6*Hisタグ組み換え融合タンパク質に対するMV705−3モノクローナル抗体の親和性を検出した
本実施例は、ヒトLAG−3−6*Hisタグ組み換え融合タンパク質抗原に対するMV705−3モノクローナル抗体の結合力をインビトロ活性実験で検出することに係る。このテストは、市販のヒトLAG−3−6*Hisタグ融合タンパク質(Novoprotein、cat#CJ91)および陽性対照抗体を使用した。その陽性対照抗体は、抗体25F7(US2011/0150892A1、PCT/US2009/053405)であり、その重鎖ヌクレオチド配列、重鎖アミノ酸配列、軽鎖ヌクレオチド配列、軽鎖アミノ酸配列はそれぞれ、配列64、配列65、配列66、配列67である。そのIgG4 kappaベクターの構築方法について、実施例1.2を参照することができ、しかも実施例1.3に記載の方法でExpiCHO−S細胞をトランスフェクションして発現し精製し、pH7.4の1×PBSバッファーに保存した。抗ヒトLAG−3全ヒトモノクローナル抗体MV705−3、陽性対照抗体をそれぞれコーティングバッファー(炭酸塩バッファー)で1μg/mlの濃度に希釈し、4℃で96ウェルプレート(CORNING、cat#9018)に一晩コーティングした。pH7.4の1×PBSバッファーで三回洗浄した後、5%スキムミルクで2時間ブロックした。ヒトLAG−3−6*Hisタグ融合タンパク質をpH7.4の1×PBSバッファーにより20μg/mlから3倍勾配で希釈し、コーティング抗体とともに25℃で2時間共インキュベーションした。そして、抗‐Hisタグが付けられているHRP検出抗体を使用してヒトLAG−3−6*Hisタグ融合タンパク質抗原に対する前記抗体の結合を検出した。一回のELISA実験結果は図3に示されるように、MV705−3モノクローナル抗体はヒトLAG−3組み換え融合タンパク質に対して高い親和性を有し、ELISAで検出された親和性EC50値は40 ng/mlである。表6は、三回の独立ELISA実験における、ヒトLAG−3−6*Hisタグ組み換え融合タンパク質に対するMV705−3モノクローナル抗体の結合のEC50値の範囲(40−100 ng/ml)_をまとめている。
4.2 MV705−3モノクローナル抗体の親和平衡解離定数(KD)の検出
ヒトLAG−3−6*Hisタグ組み換え融合タンパク質に対するMV705−3モノクローナル抗体の結合能力を、Octet Red96生体分子間相互作用システム(Octet Red96、ForteBio)を使用して検出した。抗ヒトIgG Fc(AHC)動力学的バイオセンサー(Fortebio、#18−5063)をpH1.7のグリシンで前処理してから検出用PBSバッファーに浸した。MV705−3を10μg/mlの濃度でAHCバイオセンサーに固定した。次に、MV705−3がロードされたAHCバイオセンサーを異なる濃度のヒトLAG−3−6*His抗原とバッファーに浸した。検体カラムの最終希釈点は、バッファーとロードされたバイオセンサー間の非特異的な結合をテストするように検出用バッファーのみを含む。0〜300秒で抗原と抗体との結合が検出され、そして300〜900秒で解離が発生した。検出用バッファーで60秒のベースラインが確定された。抗LAG−3モノクローナル抗体の親和性曲線は、1:1の結合の動力学センシング一価結合モデルでフィッティングさせられた。結合動力学分析は表7に示す。
4.3 BIAcоreによるMV705−3モノクローナル抗体の親和平衡解離定数KDの測定
本実施例は、BIAcоre SPR技術を用いて、ヒトLAG−3−6*Hisタグ組み換え融合タンパク質に対するMV705−3モノクローナル抗体または上記陽性対照抗体の結合を検証した。モノクローナル抗体MV705−3(1.1μg/ml)または陽性対照抗体(12.9μg/ml)を500RUレベルに設定し、製造元が推奨した標準的固定化操作手順に基づいてそれをCM5チップ(GE、ロット番号:BR100530)にコーティングした。異なる濃度(0.352〜45nM)のヒトLAG−3−6*Hisタグ組み換え融合タンパク質を30μl/minの流速で、抗体にカップリングされたチップの表面にそれぞれ120秒注射・分析し、抗原を10分間解離させた。抗原結合の解離が完了した後、pH2.1のGly−HCl再生溶液を用いて30μl/minの流速でチップの表面を30秒再生し、さらにPBSバッファーでチップを60秒洗浄してチップの状態を安定させた。BIAcоre制御ソフトウェア(バージョン2.0.1)を使用し、BIAcоre T200(No.1602831)表面プラズモン共振器ですべての実験を行った。BiaEvaluation 3.0ソフトウェアを使用してデータ分析をした。結合動力学分析は表8に示すように、MV705−3モノクローナル抗体は、陽性対照抗体25F7よりも、ヒトLAG−3−6*Hisタグ組み換え融合タンパク質に結合するKDが高い。
本実施例は、ヒトLAG−3−mFc組み換え融合タンパク質とMHCクラスII分子との結合に対して抗ヒトLAG−3完全ヒトモノクローナル抗体MV705−3がブロッキング効果を有することを証明できるインビトロ結合活性テストを行った。このテストは、ヒトLAG−3−mFc組み換え融合タンパク質と、ヒトMHCクラスII分子を表面に発現するDaudi細胞とを使用した。ヒトLAG−3−mFc融合タンパク質(hLAG−3−mFc)は、ヒトLAG−3細胞外ドメインとマウスFcとを融合してなるものであり、そのヌクレオチド配列は配列68であり、アミノ酸配列は配列69である。LAG−3−mFc融合タンパク質を発現するヌクレオチド配列を発現ベクターpcDNA3.3にクローニングし、Expi293細胞(Thermo Fisher,cat#A14527)にトランスフェクションし、実施例1.3に類似した方法で精製して上記hLAG−3−mFc組み換え融合タンパク質を得た。配列#10161を発現ベクターpcDNA3.3に構築し、Expi293細胞にトランスフェクションし、発現精製によりイソタイプ対照タンパク質を得た。このテストは、陽性対照抗体25F7(実施例4に記載の通り)とこのアイソタイプ対照タンパク質の両方に係る。
ヒトLAG−3−mFc組み換え融合タンパク質とMHCクラスII分子との結合に対する前記抗体のブロッキング効果をテストするために、MV705−3、陽性対照抗体25F7およびアイソタイプ対照タンパク質を4℃に予め冷却したの1×PBSバッファーで100μg/mlから3倍勾配希釈し、それと同時に各勾配希釈液に10μg/mlのhLAG−3−mFc組み換え融合タンパク質溶液を加えた。該混合液を4℃で30分間インキュベーションしてから、4℃に予め冷却したの1×PBSバッファーにより洗浄された2×105個のDaudi細胞を加え、4℃で1時間インキュベーションした。その後、予め冷却した4℃の1×PBSバッファーで一回洗浄し、ウサギ抗マウスIgGのPEタグ抗体(Abcam,cat#ab7000)と、洗浄されたDaudi細胞とを1時間インキュベーションし、Guava easyCyte HTフローサイトメーター(MERCK MILLIPORE)でhLAG−3−mFc組み換えタンパク質とDaudi細胞との結合を分析した。
図4と表9に示す結果のように、モノクローナル抗体MV705−3は、ヒトLAG−3−mFc組み換え融合タンパク質とDaudi細胞表面のMHCクラスII分子との結合を効果的にブロッキングすることができ、そのブロッキングIC50値は2.8μg/mlである。これは、ヒトLAG−3−mFcとDaudi細胞表面のMHCクラスII分子との結合に対する陽性対照抗体25F7のブロッキング能力に相当し、陽性対照抗体25F7のブロッキングIC50値は3.3μg/mlである。
本実施例は、前記抗体が、アカゲザル(Macaca fascicularis)LAG−3抗原およびマウスLAG−3抗原と交差反応を有するか否かを説明するためのインビトロ結合活性テストを行った。このテストは、市販の組み換えアカゲザルLAG−3−6*Hisタグ融合タンパク質(Novoprotein、cat#C998)、および構築発現された組み換えマウスLAG−3−6*Hisタグ融合タンパク質に係る。マウスLAG−3遺伝子完全長cDNA(Sinobiological,cat#MG53069−G)を発現ベクターpcDNA3.3にクローニングし、Expi293細胞にトランスフェクションし、上記組み換えマウスLAG−3−6*Hisタグ融合タンパク質(ヌクレオチド配列は配列70であり、アミノ酸配列は配列71である)を得た。
前記抗体とアカゲザルLAG−3抗原との交差反応をテストするために、抗ヒトLAG−3全ヒトモノクローナル抗体MV705−3と陽性対照抗体25F7をそれぞれコーティングバッファー(炭酸塩バッファー)で1μg/mlの濃度に希釈し、4℃で96ウェルプレート(CORNING、cat#9018)に一晩コーティングした。1×PBSバッファーで三回洗浄した後、5%スキムミルクで2時間ブロックした。アカゲザルLAG−3−6*Hisタグ融合タンパク質を1×PBSバッファーにより20μg/mlから3倍勾配で希釈し、コーティング抗体とともに25℃で2時間共インキュベーションした。そして、抗‐hisタグが付けられているHRP検出抗体を使用してアカゲザルLAG−3−6*Hisタグ融合タンパク質抗原に対する前記抗体の結合を検出した。
図5と表10に示すように、モノクローナル抗体MV705−3は、アカゲザルLAG−3−6*Hisタグ融合タンパク質と良好な交差反応活性を有し、そのEC50値は0.11μg/mlである。前記陽性対照抗体25F7は、アカゲザルLAG−3−6*Hisタグ融合タンパク質との交差反応が弱く、対応するEC50値は7.0μg/mlである。
前記抗体とマウスLAG−3抗原との交差反応をテストするために、上記方法を使用してマウスLAG−3−6*Hisタグ融合タンパク質抗原に対する前記抗体MV705−3の結合活性を検出した。その結果は図6と表11に示すように、マウスLAG−3抗原に対して、前記抗体MV705−3と陽性対照抗体25F7のいずれも弱い交差反応の活性を有する。
本実施例は、活性化されたヒトCD4+T細胞に対する前記抗体の結合活性をテストするためのインビトロ結合活性テストを行い、活性化されたヒトCD4+T細胞の表面に天然ヒトLAG−3が発現される。実施例1.2に記載の方法を使用してCD4+T細胞を活性化した。そして、抗体MV705−3、陽性対照抗体25F7、上記陰性対照、すなわちアイソタイプ対照を、冷1×PBSバッファー(Hyclone,cat#SH30256.01)で10μg/mlから3倍または4倍勾配希釈した。結合活性検出について、1:200希釈のヤギ抗ヒトIgG−PEタグ抗体(Abcam,cat#ab98596)を使用してCD4+T細胞に対する前記抗体の結合を検出し、Guava easyCyte HTフローサイトメーター(MERCK MILLIPORE)により、細胞に対する前記抗体の結合活性を分析した。フローサイトメトリー中にLAG−3を発現するCD4陽性T細胞はゲーティングにより選択された。その結果は図7と表12に示すように、MV705−3モノクローナル抗体は、LAG−3のCD4陽性T細胞を高い親和性で結合・発現することができ、その結合EC50値は1.9ng/mlである。
本実施例はインビトロ生物学的機能分析を実行し、市販の、ヒトLAG−3を安定的に発現する細胞株LAG3/NFAT Reporter−Jurkat (BPS bioscience,cat#71278)、Raji細胞(ATCC,cat#CCL−86)、スーパー抗原試薬(Toxin technology−ET404)、biо−glоルシフェラーゼ試薬(Promega,cat#G7940)をテストに利用した。
前記抗体のインビトロT細胞の活性化生物学的機能をテストするために、前記抗体MV705−3および陽性対照抗体25F7をRPIM1640(Gbico,cat#11875093)を150μg/ml(最終濃度の5倍)から3倍勾配希釈し、濃度ごとにウェルを三つ重複し、一つの抗体のブランク濃度のブランク対照を設置した。各希釈勾配の抗体をそれぞれ4×104の細胞/ウェルのLAG3/NFAT Reporter−Jurkat細胞と混合し、37℃、5%CO2インキュベーターで30分間プレインキュベーションした。そして、順に3×104の細胞/ウェルRaji細胞及び10μlの濃度0.08ng/ml(最終濃度の10倍)のスーパー抗原試薬を加えた。混合物を37℃、5%CO2インキュベーターで5〜6時間共インキュベーションした。100μlのbiо−glоルシフェラーゼ検出試薬をすばやく加えて5〜10分間インキュベーションし、TECAN蛍光検出装置(TECAN infinite M1000 PRO)を使用して生体蛍光のシグナルを測定して前記抗体のインビトロT細胞の活性化機能を分析した。誘導倍数は、各濃度勾配の生体蛍光シグナル値とブランク対照生体蛍光シグナル値との比であり、実験結果はGraphPad Prismにより非線形回帰分析された。
その結果は図8および表13に示すように、前記抗体MV705−3の濃度の増加につれて、生体蛍光のシグナル強度は勾配依存的に上昇した。これは、前記抗体MV705−3がNFATレポーター遺伝子の発現を増加させることができることを説明し、そのEC50値は0.04μg/mlである。前記抗体MV705−3は、陽性対照抗体25F7よりも有意に優れたインビトロT細胞活性化生物学的機能を示し、陽性対照抗体25F7のEC50値は0.64μg/mlである。
本実施例の目的は、MV705−3モノクローナル抗体が、BMS−25F7抗体(US2011/0150892A1及びPCT/US2009/053405のような文献に開示されており、ここで言及して本出願に組み込まれる)またはBMS−986016抗体(BMS−986016は、WO2015116539に記載のSEQ ID NO:1と2に示される重鎖および軽鎖を含み、WO2014/008218にも記載されており、ここで言及して本出願に組み込まれる)と同じ結合エピトープを有するか否かを分析することである。実験は、Octet Red96生体分子間相互作用システム(Octet Red96、ForteBio)を使用して表した。実験設計は図9に示され、以下のように簡単に説明する。すなわち、ビオチンで標識されたhLAG−3−6*hisタグ組み換えタンパク質を10μg/mlの濃度でSAバイオセンサー(Fortebio、ロット番号18−5020)に固定し、300秒平衡した後にPBSバッファーに浸漬し、hLAG−3−6*hisタグ組み換えタンパク質をロードしたSAバイオセンサーを下記各グループのサンプルに浸漬した:(1)100μg/mlのBMS対照モノクローナル抗体(BMS−25F7またはBMS−986016);(2)100μg/mlのMV705−3モノクローナル抗体;(3)100μg/mlのMV705−3モノクローナル抗体と100μg/mlのBMS対照モノクローナル抗体BMS−25F7またはBMS−986016の混合物;(4)50μg/mlのMV705−3モノクローナル抗体と50μg/mlのBMS対照モノクローナル抗体BMS−25F7またはBMS−986016の混合物。PBSバッファーで60秒のベースラインが確定された。0〜300秒で抗原と抗体との結合が検出され、そして300〜900秒で解離が発生した。抗LAG−3モノクローナル抗体の親和性曲線は、1:2の結合の動力学センシング一価結合モデルでフィッティングさせられた。結合動力学分析は図10a〜dに示すように、飽和濃度下(100μg/ml)で、hLAG−3−hisタグ組み換えタンパク質に対するMV705−3モノクローナル抗体の結合およびBMS対照モノクローナル抗体の結合はいずれも最高値(Rmax)に達したが、両方の抗体の混合物は100μg/ml+100μg/mlまたは50μg/ml+50μg/mlの濃度下でいずれもhLAG−3−hisタグ組み換えタンパク質に対する結合シグナル値を増加させることができる。これは、hLAG−3抗原に対するMV705−3モノクローナル抗体の結合エピトープは、それに対するBMSの対照抗体の結合エピトープとは異なることを示している。
10.1 MV705−3モノクローナル抗体の変異体の設計及び構築発現
MV705−3モノクローナル抗体のアミノ酸配列の潜在的な分解サイトを分析して調べた結果、その抗体の重鎖中に二つのサイトが「DD」アミノ酸配列を含み、アスパラギン異性化の可能性のある潜在的な識別サイトであり、それぞれ配列41の第89、90サイト、及び108、109サイトに位置することは発見された。Q5(登録商標) Site−Directed Mutagenesis Kit部位特異的変異誘発キット(New England Biolabs、ロット番号E0552S)を使用し、MV705−3モノクローナル抗体のVH領域の第89、90サイト及び108、109サイトに対する部位特異的変異誘発を実施し、それらをDEまたはEDに変更した。誘発が成功したMV705−3モノクローナル抗体変異体VH領域を、ヒトIgG4 S228P定常領域を含むMV705−3モノクローナル抗体の元ベクターにサブクローニングした(得られたクローン番号を表14に示す;得られたクローン重鎖VHヌクレオチド配列とアミノ酸配列番号について表15参照)。MV705−3モノクローナル抗体の各変異体の重鎖を発現するベクターをそれぞれ、MV705−3モノクローナル抗体の軽鎖を発現するベクターと一緒にExpi−CHO−S細胞にトランスフェクションした。
下記の各分析方法により、得られた上記8つのMV705−3モノクローナル抗体変異体とMV705−3モノクローナル抗体の活性を検出した:(a)ヒトLAG−3組み換えタンパク質に結合するELISA;(b)ヒトLAG−3タンパク質を発現する細胞に結合する実験;(c)市販の、ヒトLAG−3を安定的に発現する細胞株LAG3/NFAT Reporter−Jurkatを利用したインビトロ生物学的機能分析。
10.2 MV705−3モノクローナル抗体変異体の活性試験
(a)ヒトLAG−3組み換えタンパク質にMV705−3モノクローナル抗体の変異体が結合するELISA方法は、実施例4に記載の通りであり、その結果は表16に示すように、2つの変異体タンパク質13380(DE+DD)、13381(ED+DD)が、ヒトLAG−3−hisタグ組み換え融合タンパク質に対するMV705−3モノクローナル抗体の結合能力と類似した結合能力を有し、その他の変異体の結合能力は異なる程度で低下した。
(b)ヒトLAG−3を発現する細胞に対するMV705−3モノクローナル抗体の結合の実験方法を下記のように簡単に説明する:
被験抗体MV705−3モノクローナル抗体とその変異体、陽性対照抗体BMS−25F7及びBMS−986016抗体を冷1×PBSバッファー(Hyclone,cat#SH30256.01)で10μg/mlから3倍または4倍勾配希釈した。結合活性検出について、1:200希釈のヤギ抗ヒトIgG−PE標識した抗体(Abcam,cat#ab98596)を使用してヒトLAG−3を発現する細胞に対する前記抗体の結合を検出し、Guava easyCyte HTフローサイトメーター(MERCK MILLIPORE)により、細胞に対する前記抗体の結合活性を分析した。フローサイトメトリー中にLAG−3を発現する陽性細胞はゲーティングにより選択された。その結果は表17に示される。ELISA結果と類似し、MV705−3モノクローナル抗体は、LAG−3の細胞を高い親和性で結合・発現することができ、その2つの変異体タンパク質13380(DE+DD)、13381(ED+DD)が、ヒトLAG−3を発現する細胞に対するMV705−3モノクローナル抗体タンパク質の結合能力と類似した結合能力を有し、その他の変異体の結合能力は異なる程度で低下した。
(c)市販の、ヒトLAG−3を安定的に発現する細胞株LAG3/NFAT Reporter−Jurkatを利用したインビトロ生物学的機能分析は、実施例8に記載の通りである。LAG−3に対するMV705−3変異体クローン13365(DD+DE)、13565(DE+DE)及び13564(ED+DE)の結合能力は大幅に低下していることはすでに分かった。そのため、MV705−3モノクローナル抗体及び、13380(DE+DD)、13381(ED+DD)、13386(DD+ED)、13563(DE+ED)、及び13616(ED+ED)の5つの変異体のみをテストした。その結果は表18に示すように、変異体タンパク質13380(DE+DD)及び13381(ED+DD)は、MV705−3モノクローナル抗体と類似した、インビトロでT細胞を活性化する能力を持っている。
インビボでの腫瘍細胞増殖の阻害におけるMV705−3モノクローナル抗体の13381変異体の有効性をテストするために、ヒト化LAG−3(Lymphocyte−activation gene 3)マウス(SHANGHAI MODEL ORGANISMS CO., LTD.から購入、完全な株名:B6.129−Lag3tm(hLAG3)/Smоc;カタログ番号NM−KI−00049)を使用した。相同組み換え法を使用してマウス内因性Lag3遺伝子のすべての細胞外領域配列をヒトLAG3配列に置き換え、これによってマウスにヒトキメラLAG3タンパク質を発現させた。マウスMC38結腸癌細胞株をマウスに接種して腫瘍移植モデルを調製した。
上記ヒト化LAG−3マウス(B6.129−Lag3tm(hLAG3)/Smоc)にそれぞれ0日目に1×106のMC38細胞を移植し、腫瘍が平均約123(mm3)に増殖した時に群分けして投薬し始め、3週間連続して週に2回投与し(BIWx3)、投与開始から3〜4週間連続して観察して平均腫瘍体積を測定した。動物を、(1)PBS溶媒対照、BIWx3、ip;(2)mPD−1抗体(BE0146、BioCell)、1mg/kg、BIWx3、ip;(3)MV705−3モノクローナル抗体の13381変異体、10mg/kg、BIWx3、ip;(4)MV705−3モノクローナル抗体の13381変異体、10mg/kg、及びmPD−1抗体(BE0146、BioCell)、1mg/kg、BIWx3、ipの四つの群に分けた(n=8)。
その結果は表19及び図12に示すように、MV705−3モノクローナル抗体の13381変異体は単独で腫瘍増殖を効果的に阻害でき、平均腫瘍増殖阻害率は35.11%であり(
)、TiまたはViは、特定の時点での治療群または対照群の平均腫瘍体積を表し、T0またはV0は、群分けの後、投与前の治療群または対照群の平均腫瘍体積を表す。それに対して、抗マウスPD−1モノクローナル抗体は腫瘍増殖に対して阻害の効果が無く、MV705−3モノクローナル抗体の13381変異体は、抗マウスPD−1モノクローナル抗体と組み合わせて投与する場合、腫瘍増殖に対する阻害効果が最も強く、平均腫瘍増殖阻害率は46.07%である。

- リンパ球活性化遺伝子−3(LAG−3)結合抗体であって、前記抗体は、顕著に改善された、主要組織適合性(MHC)クラスII分子へのLAG−3の結合を阻害する作用を有する、リンパ球活性化遺伝子−3(LAG−3)結合抗体。
- 下記のアミノ酸配列:
配列41、配列45、配列49、配列53、配列57、配列61、配列75または配列77
を含む重鎖可変領域と、
下記のアミノ酸配列:
配列43、配列47、配列51、配列55、配列59または配列63
を含む軽鎖可変領域と
を含む、請求項1に記載の抗体。 - 下記の重鎖可変領域と軽鎖可変領域との組み合わせを含む、請求項1または2に記載の抗体:
(1)アミノ酸配列41と43;
(2)アミノ酸配列45と47;
(3)アミノ酸配列49と51;
(4)アミノ酸配列53と55;
(5)アミノ酸配列57と59;

(1)重鎖可変領域はアミノ酸配列41からなり、軽鎖可変領域はアミノ酸配列43からなり、
(2)重鎖可変領域はアミノ酸配列75からなり、軽鎖可変領域はアミノ酸配列43からなり、および
(3)重鎖可変領域はアミノ酸配列77からなり、軽鎖可変領域はアミノ酸配列43からなる、
