本願は、2018年2月14日に出願された米国特許仮出願第62/630,497号の優先権の恩典を主張するものであり、その全体が参照により本明細書に組み入れられる。
本願は、T細胞系列の細胞を発生させる方法、組成物、およびキット、ならびに該細胞の使用に関する。具体的には、本願は、始原T細胞および成熟T細胞を発生させる方法、組成物、およびキット、ならびに該細胞の使用に関する。
T細胞は適応免疫の重要なメディエーターであり、病原体に対する、およびがん免疫療法における治療剤として利用することができる。造血幹細胞移植(HSCT)は、広範な悪性および非悪性の障害の有効な治療を提供するが、治療前に必要なプレコンディショニングレジメンの結果として、T細胞の回復が大幅に遅れてしまう(Krengerら, 2011)。骨髄(BM)中で発生する大半の他の造血系列と違って、T細胞の発生は、BM由来の始原細胞が胸腺に移動することを要し、胸腺に入ってきたリンパ球始原細胞は、T系列細胞への分化を誘導する重要なシグナルを受ける(Shah and Zuniga-Pflucker, 2014)。
本発明者らは、マウスまたはヒト造血幹細胞/始原細胞(HSPC)および人工多能性幹細胞(iPSC)からT細胞系列の細胞を発生させる、無細胞ビーズベース系を開発した。Notchリガンドの非プレート結合浮遊体(たとえばDL4-μビーズ)が、始原T細胞および成熟T細胞などのT系列細胞の有効な発生を可能にする。
(i)(a)幹細胞または始原細胞を含む試料を、浮遊支持体にコンジュゲートしたNotchリガンドとともに培養すること、および(b)T細胞系列の細胞を単離することを含む、T細胞系列の細胞を発生させる段階、ならびに
(ii)有効量の該T細胞系列の細胞を該対象に投与する段階
を含む、方法を、さらに提供する。
上述したように、本発明者らは、マウスまたはヒト造血幹細胞/始原細胞(HSPC)または人工多能性幹細胞(iPSC)などの幹細胞または始原細胞からT細胞系列の細胞を発生させる、無細胞ビーズベース系を開発した。Notchリガンドの非プレート結合浮遊体(たとえばDL4-μビーズ)が、始原T細胞および成熟T細胞などのT系列細胞の有効な発生を可能にする。
したがって、本開示は、(a)幹細胞または始原細胞を含む試料を、浮遊支持体にコンジュゲートしたNotchリガンドとともに培養する段階、および(b)T細胞系列の細胞を単離する段階を含む、T細胞系列の細胞を発生させる方法を提供する。
本発明者らは、新規の浮遊Notchリガンドも開発した。本明細書で用いられる場合、「浮遊Notchリガンド」という用語は、浮遊細胞培養に使用されるNotchリガンドを指す。
浮遊Notchリガンドは、T細胞系列の細胞を発生させるのに使用されるキットとして調製され、かつ包装されている場合がある。
本開示は、本明細書に記載される方法、系、およびキットにより発生したT細胞系列の細胞、または該細胞の子孫である有糸***もしくは分化細胞をさらに提供する。
インビトロ由来のヒト始原T細胞を発生させ、そしてそれらの安全性をヒト/マウス免疫移植モデルで試験できれば、T系列の免疫関連障害を治療する細胞ベースのアプローチの道が開かれる(Legrandら, 2006; van den Brinkら, 2004)。T細胞は、ウイルス性および細菌性の病原体を認識し除去する、適応免疫系の主要なエフェクターアームである。T細胞性急性リンパ性白血病(T-ALL)などの特定の稀な血液がんでは、T細胞が増殖して健常な免疫細胞を締め出し、正常な免疫機能をかく乱させる(Ferrandoら, 2002; Wengら, 2004)。化学療法はがん患者に治療的恩恵を与え得ることもあるが、往々にして免疫不全および日和見感染感受性をもたらし得る。日和見感染はまた、HIV感染後にCD4+T細胞が枯渇しているAIDS患者にとって重大な懸念をもたらす。免疫不全は依然としてHIV/AIDSおよびがんにおける重大な懸念であるが、適切な調節コントロールが効かないT細胞が自己組織に対し免疫応答を生じる自己免疫疾患では、免疫活動亢進も同等に問題となる。
(i)(a)幹細胞または始原細胞を含む試料を、浮遊支持体、任意で粒子またはマイクロビーズにコンジュゲートしたNotchリガンドとともに培養すること、および(b)T細胞系列の細胞を単離することを含む、T細胞系列の細胞を発生させる段階、ならびに
(ii)有効量の該T細胞系列の細胞を、それを必要とする対象に投与する段階
を含む、方法を、さらに提供する。

材料および方法:
造血幹細胞ソース
Research Ethics Board of Sunnybrook Health Sciences Centreが制定した承認済みガイドラインにしたがい、同意が得られた分娩後の母親からヒト臍帯血(UCB)試料をシリンジで抽出して採取し、抗凝血性クエン酸リン酸デキストロース入り血液パックユニット(イリノイ州ディアフィールドのBaxter Healthcare)に回収した。回収から24時間以内にFicoll密度遠心法によりUCB単核細胞を単離し、EasySep Human CD34 Positive Selection Kit(ブリティッシュコロンビア州バンクーバーのStemcell Technologies)をメーカー取扱説明書にしたがって用いて、系列陰性(Lin-)CD34+細胞を予め濃縮した。ヒトHSPCを単離するために、Lin-細胞を抗ヒトCD38-APCおよび抗ヒトCD34-PE mAbで染色し、そしてBD Biosciences FACSAriaソーター(カリフォルニア州サンホセのBD)を用いてCD34+CD38-/lo細胞をソートした。ソートされたヒトHSPCは、ソート後分析により、純度99%超と決定された。臍帯血由来CD34+細胞の一部、および動員末梢血(mPB)由来CD34+細胞の全部を、Stemcell Technologiesから購入した。mPBについては、ボランティアを、G-CSF(回収3〜5日前に最大10 μg/kg/日のG-CSF)とPlerixafor(回収1日前に最大0.24 mg/kg)との組み合わせで処置した。
ヒトDLL4の細胞外ドメイン(アミノ酸残基[aa]1〜529)のコード配列のC末端に、リンカー配列により離間させて、ヒスチジン(His)タグ、続いてヒトIgG3のFc部分(ヒンジ領域を含む)およびBir1A認識配列(Avitag(商標))を融合させることで、Delta様-4を遺伝子改変してpDL4-Fc-His-Bプラスミドコンストラクトを作製した。これらの方法に用いられ得る他のコンストラクトとしては、i)IgG1のFc部分と融合させたヒトDLL4(aa1〜524)、ii)IgG1のFc部分と融合させたアルカリホスファターゼ(aa1〜17)ヒトDLL4(aa27〜524)のシグナル配列;iii)C末端でStreptagIIおよび6xHisと融合させたヒトDLL4(aa1〜524)、iv)C末端で10xHisと融合させたアルカリホスファターゼ(aa1〜17)ヒトDLL4(aa27〜524)のシグナル配列、およびそれらの任意の組み合わせが挙げられる。このコンストラクトをpIRESpuro2哺乳類発現プラスミド(カリフォルニア州マウンテンビューのClontech)に挿入した。得られたプラスミドを、標準的なCaPO4トランスフェクト法を用いてHEK-293T細胞にトランスフェクトし、プラスミドが安定して統合された細胞を、標準DMEM[10%(v/v)FBS、2mMのGlutamax、ペニシリン(100 U/ml)/ストレプトマイシン(100 mg/ml)(すべてイリノイ州ロックフォードのThermo Fisher Scientificの製品)、2 mMの2-メルカプトエタノール(ミシガン州セントルイスのSigma-Aldrich)を補充]に添加した2 mg/mLのピューロマイシンに対する耐性に基づき選択した。細胞を増殖させ、Freestyle 293発現培地(Thermo Fisher Scientific)に移して成長させた。培地に分泌されたDL4-Fc融合タンパク質を、AKTAprime plus(マサチューセッツ州マールボロのGE Healthcare Life Sciences)自動クロマトグラフィー装置に取り付けたHiTrapプロテインG親和性カラム(GE Healthcare)を用いて精製した。
組み換えVCAM1-Fcは、DL4-Fcと同様、VCAM-1細胞外ドメイン、IgG3 Fcドメイン、およびAvitag(商標)ビオチン化部位を含むように遺伝子改変した。VCAM1-Fc融合タンパク質の製造および精製に用いた材料および方法は、上記のDL4-Fc融合タンパク質のものと同じであった。
1個のDL4-Fc分子につき2個のビオチン分子となる最適モル比で、NHS-活性化ビオチンを精製DL4-Fcに添加した。PBS中の透析またはバッファー交換により、反応しなかったNHS-ビオチンを混合物から除去し、そしてビオチン組込みをHABA(4’-ヒドロキシアゾベンゼン-2-カルボン酸)法(Pierce Biotin Quantitation Kit、製品番号28005)により評価した。その後、ビオチン化DL4-Fcを4℃で保存した。
1 μgのビオチン化DL4-Fcを、直径1 μmから100 μmまでさまざまなサイズのストレプトアビジン(SA)被覆ポリスチレンμビーズ(イリノイ州レイクフォレストのSpherotech)とともにインキュベートした。D4-Fcを、SA被覆Dynabeads(Thermo-Fisher)および50 nmナノゴールド粒子にもコンジュゲートさせ、それぞれ2 mLのPBS中、30分間室温でインキュベートし、10分おきに渦流により混合した。どの場合も、粒子の表面積は、2x105個の25 μm SA-μビーズに等しかった。DL4-Fcコンジュゲートビーズを4 mLのPBSで洗浄し、そして3000 x gで10分間スピンダウンした。結合しなかったリガンドがあれば混合物から除去するために、およびDL4-Fc含量をアッセイしてμビーズとの結合を査定するために、上澄みを慎重に回収した。2回目の洗浄後、DL4-μビーズをさまざまな体積のPBSに再び浮遊させて、テキストに記載の表示の濃度を得た。非コンジュゲートμビーズを並行して調製し、陰性対照とした。
Notch応答性エレメント(8x RBPJコンセンサス結合部位)を、プロモーターを持たないpGL4.17[luc2/Neo]プラスミド(Promega)に挿入した。こうして、ネオマイシン耐性を与えながら、ルシフェラーゼ酵素の発現によりNotch活性化を知らせる、1個のプラスミドを作製した(pGL4.17-N1Repと名付けた)。次に、NIH3T3細胞を、pMIGR-NOTCH1(ペンシルバニア大学Warren Pear博士の寄贈)およびpGL4.17N1Repプラスミドを用いてトランスフェクトした。ネオマイシン処理(1 μg/mL)に耐性のあった細胞を最初に選択してから、GFPを発現した細胞をフローサイトメトリーによりソートした。次に、NIH3T3細胞のクローンを、96ウェルプレートへの単細胞捕集により単離した。次に、各クローンの、プレート結合DL4-Fcに対する活性化に対する応答を測定した(一晩、20 μg/mLで捕集)。最低量のバックグラウンドおよび最高のNotch活性化応答を有する、3T3N1CLucと名付けたクローンを増殖させ、Notch受容体活性化を測定する実験の実施に用いた。
標準組織培養(TC)処理した平底96ウェルプレート中の3x104個の3T3-N1CLuc細胞にDL4-μビーズを添加し、5% FBSを補充したαMEM中で一晩インキュベートした。細胞を溶解させ、Firefly Luciferase Assay Kit 2.0(カリフォルニア州フレモントのBiotium)をメーカー取扱説明書にしたがい用いて、ルシフェラーゼ活性についてアッセイした。簡単に説明すると、増殖培地を除去し、細胞をPBSで洗浄してから、溶解バッファーを添加した。細胞を-80℃で10分間凍結させてから解凍することにより溶解させ、次いでライセートを96ウェル平底不透明ポリスチレンプレート(Corning)に移した。D-ルシフェリンを調製し、各ウェルに自動ディスペンサーで添加し、Synergy H1プレートリーダー(バーモント州ウィヌースキのBioTek Instruments Inc.)を用いて分析した。50 μL/ウェルのタンパク質(5〜20 μg/mL)を平底96ウェルプレートに4℃で一晩吸着させてから翌日洗浄し、3T3-N1CLuc細胞を播種することにより、ヒトIgGおよびDL4-Fcのプレート結合対照を前日に調製した。
異なる数のDL4-μビーズを、3x103個のマウスLin- Sca-1+ cKit+ HSC(C57BL6マウスの骨髄からフローサイトメトリーによりソート)に添加して、細胞対ビーズ比を1:1、1:3、1:9、および1:27とした。各条件で、細胞-ビーズの組み合わせを、丸底96ウェルプレートの単一ウェルで、200 μLのIMDM[20% BIT(STEMCELL Technologies)、1% Glutamax(Thermo)、50 ng/mLのSCF、10 ng/mLのFlt3L、および10 ng/mLのIL-7(ミネソタ州ミネアポリスのR&D Systems)を補充]中、インキュベートした。50 μL/ウェルのタンパク質(20 μg/mL)を平底96ウェルプレートに4℃で一晩吸着させてから翌日洗浄し、細胞を播種することにより、ヒトIgGおよびDL4-Fcのプレート結合対照を前日に調製した。同時培養7日目に細胞を収集し、抗マウス抗体CD45、CD25、CD44、CD90、CD11b、およびCD19で染色し、それからLSR IIサイトメーター(BD Biosciences)で分析した。
初期化hiPSC株を、線維芽細胞(AlStemBio Cat #iPSC11)およびT細胞(Harvard Stem Cell Science Cat # STiPS A3)から誘導した。これらをMatrigel上でmTeSR培地(StemCellTech)で培養した。自己凝集EBを発生させるため、iPSCをコラゲナーゼBで20分処理してから、短時間のトリプシン-EDTA工程を行った。細胞スクレーパーで細胞をそっと掻きとって、小さい凝集体を作った。EBは、BMP-4の存在下でStemPro-34(Invitrogen)中、最初の24時間の培養の間に発生し、次いでBMP-4およびbFGFの存在下でさらに24時間培養し、それからBMP-4、bFGF、およびSBの存在下でさらに48時間(2〜4日目)培養した。4日目に、BMP-4およびSBを除去し、VEGF、IL-6、IL-11、IGF-1、SCF、EPO、TPO、Flt-3、IL-3、およびDKK1(すべて、カリフォルニア州オーバーンのMiltenyi Biotec、またはR&D Systemsのサイトカイン)と置き換えた。5% CO2/5% O2/90% N2の低酸素環境で8日間、培養を維持した。8日目に、記載されている(Kennedyら, 2012)ようにして、細胞をMACSを用いてCD34+細胞について濃縮し、DL4-μビーズに加えてStemSpan(商標) T Cell Progenitor Expansion Supplement(Stemcell Technologies)を補充したStemSpan(商標)SFEM II(Stemcell Technologies)とともに上記のようにインキュベートして、T細胞を発生させた。
免疫不全マウスへの養子移入の準備として、大量のヒトHPSC/DL4-μビーズ培養物を準備した。2x105個のCD34+ HSPCを、1.8x106個のDL4-μビーズとともに、T25フラスコ(Thermo Scientific)内で7〜10日間インキュベートし、このとき、CD34+ CD7+細胞と同定された始原T(proT)細胞を、FACSAria細胞ソーター(カリフォルニア州サンホセBD Biosciences)を用いてソートした。あるいは、HSPCを鉄酸化物で被覆されたDL4-μビーズとともに培養する場合、細胞成分からのDL4-μビーズの磁気分離を、autoMACS-pro細胞ソーター(Miltenyi Biotec)を用いて実施した。proT細胞を、3〜6日齢の免疫不全NOD-Scid/IL2rγnull(NSG)新生仔の肝内に注射した。各マウスに、3〜5x105個のproT細胞とともに、α-MEM中総量50 μlのhIL-7(0.5 μg/マウス)および抗IL-7モノクローナル抗体(mAb)、クローンM25(2.5 μg/マウス)を与えた。マウスは、3〜4日おきにIL-7/M25カクテルでブーストされた。移植後3週または12週でリンパ器官の胸腺、脾臓、および骨髄を収集した。各器官から単細胞浮遊物を調製し、染色し、LSR-IIサイトメーター(BD Biosciences)を用いて分析した。細胞死マーカー4'-6-ジアミジノ-2-フェニルインドール(DAPI)を排除した生細胞、およびヒトCD45を発現している細胞に、電子的にゲートをかけることにより、移植を査定した。
DL4-μビーズは強力なNotchシグナリングを誘導する
本発明者らの実験室での過去の研究で、DL4融合タンパク質、DL4-Fcを標準TC処理ウェル/プレート表面に固定化すると、マウスおよびヒトHSPCにおいて、T系列細胞の発生を誘導するのに十分なNotchシグナリングを誘導できることが示された(Shuklaら, 2017)。しかし、この2Dフォーマットは、拡張性、およびT細胞発生を促進する強力かつ一定したNotchシグナルを送達する能力に、限界がある。
ランダム方向第1世代DL4-μビーズに改良を加えるために、本発明者らは、DL4-Fcの方向をビーズ表面に対し相対的に指定することが、Notchシグナリング伝達を増強するかどうかを調査した。そのために、1個のビオチン分子が酵素的にコンジュゲートできるBirA認識配列(AviTag(商標))をC末端に有する、DL4-Fcを設計した(図4)。3T3N1CLuc細胞ルシフェラーゼ活性により査定すると、これら第2世代DL4-μビーズのインキュベーションにより、Notchシグナリングのレベルが大幅に増加した(図5)。この方向指定コンジュゲーション法を用いて、ビーズサイズがNotch伝達に及ぼす影響を再度査定した。結果は、直径100 μm、25 μm、10 μm、および6.5 μmのビーズすべてがNotchを活性化する能力を有したが、直径25 μmのビーズが最も有効であったことを示した(図6)。ビーズ対細胞の最適比も評価し、ビーズ対細胞の比が3:1のとき、プレート結合対照と比較して、Notch活性化が10倍増加したことが決定された(図6)。
上記の結果に基づき、本発明者らは次に、同じ要因がT細胞発生に影響するかどうかの決定に取り掛かった。これについて、まずはマウスHSPCを用いて試験した。図8に示すように、DL4-μビーズは、PB-DL4よりも有効にマウスT細胞の発生を誘導した。
次に、CD34+ UCB由来HSPCからヒトT細胞発生を誘導するためのDL4-μビーズの使用について検証した。そのために、上記で確定した最適HSPC:DL4-μビーズ比、1:9を用いた。細胞を2日おきに数え、フローサイトメトリー分析を実施した。結果は、培養4日目には、CD34、CD7、およびCD5を同時発現するヒトproT細胞の出現を示した(図10A)。これらの結果は、DL4-μビーズとともにインキュベートされたヒトHSPCでは、ロバストなproT細胞表現型(CD34+CD7+またはCD7+CD5+CD1a-)が達成されたが、非コンジュゲートμビーズまたはPB-DL4-Fcでは達成されなかったことを実証しており、プレート結合DL4-Fcと比較すると、DL4-μビーズのほうが、Notchを活性化する能力が高いことが確認された。
次に、DL4-μビーズがNotchを活性化する増加した能力が、発生中の細胞をT細胞発生の後期のステージへと分化するよう誘導でき、したがって培養後期でより成熟した表現型が発現されるかどうかについて、調査した(図11)。28日目と47日目の両方の培養物の分析から、CD4+CD8+二重陽性(DP)細胞が細胞の約25%を占めたことが示された。興味深いことに、47日目の培養物は、CD4+CD8+CD3+DP細胞およびCD8+CD3+単一陽性(SP)細胞の出現を示した。これらの結果は、DL4-μビーズにより誘導された強力なNotchシグナリングは、PB-DL4-Fcを用いた過去の試みで見られたT細胞成熟の発生障害を克服できることを示している。
成人mPBは、潜在的にUCBよりも容易に入手可能なHSPCのソースであるが、それは一人から得られるHSPCの数がUCBの約100倍だからである。T系列発生の個体発生を比較するために、異なる3人からのmPB由来CD34+ HSPCをDL4-μビーズとともに培養し、UBC由来HSPC培養物と比較した(図12A)。14日目、T細胞発生の進行はCB由来HSPCと非常によく似ており、proT細胞群(CD34+CD7+)の同様のパーセンテージが観測された。しかし14日以降の増殖率は、mPB由来HSPCで110倍だったのに対し、CB由来細胞は190倍近くになった(図12B)。
本明細書に記載される方法により、線維芽細胞に由来するヒトiPSCを、CD34+プレ造血前駆細胞に分化するよう誘導した。CD34+細胞をDL4-μビーズとともにインキュベートして、それらが発生の初期(図13A)および後期(図13B)に多能性細胞をT系列へと誘導する能力を決定した。発生初期は、CD7細胞表面マーカー、続いてCD5の正常な獲得が示される。T細胞発生後期は、28日目のCD4+未成熟単一陽性(iSP)マーカー、そして35日目の単一陽性マーカーCD8細胞の獲得を特徴とする。特筆すべきはT細胞受容体(TCR)成分のCD3という細胞表面マーカーの存在であり、この培養物における成熟T細胞の存在を示唆している。
SA-μビーズが他のビオチン化分子を付加するためのモジュラーベースとして機能できるかどうかを決定するために、VCAM-1の機能的効果を調査した。VCAM-1をプレート結合DLL4に付加すると、HSPCのT細胞への分化が加速されることが、過去に示されている(Shuklaら, 2017)。ここでは、新たな融合タンパク質、VCAM1-Fcを遺伝子設計し、発現させ、ビオチン化した。免疫ブロット分析を用いて、この生成されたVCAM1-Fcは、市販のVCAM1製品と同じサイズであることを決定した(図15、左側のパネル)。また、BirA酵素ビオチン化の標的配列を含むVCAM1-Fcは、ビオチン化されたことが実証された(図15、右側のパネル)。次に、固定量のDL4-Fcとともに、ビオチン化VCAM1-Fcを、SA-μビーズの表面の被覆に用い、そしてUBC由来HSPCとともに培養した。7日目の培養物の分析により、概して、VCAM1-Fc/DL4-Fc比が高いほど、分化速度が加速されたことが実証された(図16)。これによって、VCAM-1の活性と、T細胞発生に関与する共刺激分子を制御下で付加するためのプラットフォームとしてのμビーズのフレキシビリティとが、実証された。
HSPC/DL4-μビーズ培養物に由来するCD34+ CD7+始原T(proT)細胞が胸腺に生着する能力を査定するために、免疫不全NSGマウスモデルを選んだ。proT細胞の生産を段階的に拡大して、96ウェルプレートからT25フラスコに切り替えた。7日目の培養物からProT細胞をソートし、NSG新生仔の肝内に注射した。初期の生着を第3週で調査すると、胸腺内でヒトCD45+細胞の存在が示され、そのほとんどが二重陽性CD4+ CD8+(DP)成熟T細胞ステージにあった(図17)。胸腺でB(CD19)細胞およびミエロイド(CD33)細胞は発生していなかった。
臨床目的でHSPC/DL4-μビーズ同時培養物からproT細胞を大規模生産するのに備えて、鉄酸化物で被覆されたDL4-μビーズを同時培養細胞から分離するAutoMACS(Miltenyi)の能力について査定した(図19A)。臨床承認されているCliniMACS(Miltenyi)と同様に機能するAutoMACSは、μビーズを細胞成分から完全に単離し(図19B)、細胞画分中にμビーズは検出できなかった。これに対し、ビーズ画分では、単離されたμビーズに捕捉された、または付着したままの細胞があった。
本明細書では、マウスとヒト両方のHSPCならびにiPSCからT細胞を発生させる無細胞ビーズベース系の開発について記載している。非プレート結合または浮遊Notchリガンド、たとえばDL4-μビーズは、T系列細胞の有効な発生を可能にする独自のストラテジーを表しており、大規模なバイオリアクターベースの浮遊培養で容易に実現でき、かつプレート結合アプローチに付随する発生障害、ならびに非効率性および拡張面の短所を克服する可能性を有する。
Claims (27)
- (a)幹細胞または始原細胞を含む試料を、浮遊支持体にコンジュゲートしたNotchリガンドとともに培養する段階、および
(b)T細胞系列の細胞を単離する段階
を含む、T細胞系列の細胞を発生させる方法。 - 前記浮遊支持体が、粒子である、請求項1記載の方法。
- 前記浮遊支持体が、マイクロビーズである、請求項1または2記載の方法。
- 前記幹細胞または始原細胞が、前記Notchリガンドとともに浮遊培養される、請求項1〜3のいずれか一項記載の方法。
- 前記幹細胞が、造血幹細胞/始原細胞(HSPC)、胚幹細胞、または人工多能性幹細胞(iPSC)から選択される、請求項1〜4のいずれか一項記載の方法。
- 前記幹細胞が、CD34+またはCD34+CD38-/lo HSPCである、請求項1〜5のいずれか一項記載の方法。
- 前記幹細胞が、CD34+造血前駆細胞であり、任意で、iPSCから分化したCD34+造血前駆細胞である、請求項1〜4のいずれか一項記載の方法。
- 前記Notchリガンドが、DL4である、請求項1〜7のいずれか一項記載の方法。
- 前記T細胞系列の細胞が、始原T(proT)細胞である、請求項1〜8のいずれか一項記載の方法。
- 前記幹細胞または始原細胞が、ヒト細胞であり、前記proT細胞が、表現型CD34+CD7+またはCD7+CD5+CD1a-を有する、請求項9記載の方法。
- 前記幹細胞または始原細胞が、マウス細胞であり、任意で、系列- CD117+ Sca-1+マウス細胞であり、前記proT細胞が、表現型CD25+またはCD25+CD90+を有する、請求項10記載の方法。
- 前記T細胞系列の細胞が、CD4+CD8+二重陽性細胞、CD4+CD8+CD3+二重陽性細胞、CD8+CD3+単一陽性細胞、またはCD4+CD3+単一陽性細胞である、請求項1〜8のいずれか一項記載の方法。
- 前記幹細胞または始原細胞が、間質細胞を含まない培地で培養される、請求項1〜12のいずれか一項記載の方法。
- 前記幹細胞または始原細胞が、浮遊支持体に付着した少なくとも1つのT細胞共刺激分子とともに培養され、該少なくとも1つのT細胞共刺激分子が任意でVCAM1である、請求項1〜13のいずれか一項記載の方法。
- 請求項1〜14のいずれか一項記載の方法により発生した、T細胞系列の細胞。
- 始原T細胞、CD4+CD8+二重陽性細胞、CD4+CD8+CD3+二重陽性細胞、CD8+CD3+単一陽性細胞、またはCD4+CD3+単一陽性細胞である、請求項15記載の細胞。
- (a)Notchリガンドと(b)マイクロビーズとを含み、該Notchリガンドが該マイクロビーズにコンジュゲートしている、浮遊Notchリガンド。
- (i)前記マイクロビーズが6.5〜100 μmの直径を有し、かつ/または(ii)前記NotchリガンドのC末端が前記マイクロビーズにコンジュゲートしている、請求項17記載の浮遊Notchリガンド。
- T細胞系列の細胞を発生させるための、請求項17または18記載の浮遊Notchリガンドの使用。
- (i)(a)Notchリガンドと(b)浮遊支持体とを含み、該Notchリガンドが該浮遊支持体にコンジュゲートしている、浮遊Notchリガンド、および
(ii)T細胞系列の細胞を発生させるための該浮遊Notchリガンドの使用説明書
を含む、キット。 - (i)(a)Notchリガンドと(b)浮遊支持体とを含み、該Notchリガンドが該浮遊支持体にコンジュゲートしている、浮遊Notchリガンド、および
(ii)培養培地
を含む、キット。 - 前記浮遊Notchリガンドが、マイクロビーズにコンジュゲートしたDL4を含む、請求項20または21記載のキット。
- (iii)浮遊支持体に付着した少なくとも1つのT細胞共刺激分子
をさらに含み、該少なくとも1つのT細胞共刺激分子が任意でVCAM1である、請求項20〜22のいずれか一項記載のキット。 - T細胞数の増加を要する状態を有する対象を治療する方法であって、
(i)(a)幹細胞または始原細胞を含む試料を、浮遊支持体にコンジュゲートしたNotchリガンドとともに培養すること、および(b)T細胞系列の細胞を単離することを含む、T細胞系列の細胞を発生させる段階、ならびに
(ii)有効量の該T細胞系列の細胞を該対象に投与する段階
を含む、方法。 - 前記T細胞系列の細胞が、始原T細胞である、請求項24記載の方法。
- 前記T細胞系列の細胞が、CD4+CD8+二重陽性細胞、CD4+CD8+CD3+二重陽性細胞、CD8+CD3+単一陽性細胞、またはCD4+CD3+単一陽性細胞である、請求項23記載の方法。
- 前記T細胞系列の細胞が、成熟T細胞である、請求項23記載の方法。
