Abstract
Description
エンドウマメゲノムは、70,000を超える異なるタンパク質をコードする。本出願人は、これらのタンパク質のうち2種を同定し、その各々は、哺乳動物骨格筋においてグルコース輸送を促進可能な1種または複数のペプチド(本明細書において以下、「グルコース輸送促進性ペプチド」または「グルコース輸送促進性断片」)を含有する。同様に、イネゲノムによってコードされる60,000種を超えるタンパク質のうち、本出願人は、4種のタンパク質を同定し、それらの各々は、哺乳動物骨格筋においてグルコース輸送を促進可能な1種または複数のペプチドを含有する。6種の同定されたタンパク質のうちのグルコース輸送促進性断片および複数のグルコース輸送促進性ペプチドを含有する組成物は、in−vitroでインスリン刺激に応答して細胞表面GLUT4転位置(translocation)の有意な増大を引き起こすとわかった(図1〜6)。天然ペプチドが由来する特定の植物タンパク質は、本明細書において、例えば、配列番号1〜6に開示されている。ペプチドが由来する特定のエンドウマメタンパク質は、配列番号1〜2に提供されており、ペプチドが由来する特定のイネタンパク質は、配列番号3〜6に提供されている。これらのタンパク質の相同体は、配列番号67〜84に記載されている。エンドウマメタンパク質において最初に同定された特定のペプチドは、配列番号7〜46に示されている。イネタンパク質において最初に同定された特定のペプチドは、配列番号47〜66に示されている。本発明のペプチドは、哺乳動物骨格筋においてグルコース取り込みの増大を引き起こすのに主に有用であり、したがって、全体的な筋肉の健康の改善において有用性を有するが、哺乳動物における調節不全のグルコースまたはインスリンレベルを特徴とする代謝状態、例えば、糖尿病の治療または管理、より詳しくは、グルコースホメオスタシスの調節およびインスリン抵抗性の減弱においても有用性を見い出す。

1.ヒト骨格骨芽細胞(Sigma 150−05a)を、骨格筋分化培地中に、ウェルあたり10,000個細胞で96ウェルプレート中に播種し、実験に先立って72時間分化させた。
骨格筋は、インスリン刺激された、または食後条件下でのグルコース処理の主な部位である(80%)。これらの条件下では、骨格筋へのグルコースの輸送は、主に、インスリンまたは収縮性刺激の際に細胞表面に転位置するインスリン応答性グルコース輸送タンパク質GLUT4によって促進される。

以下のペプチドの組成物を、骨格筋グルコース輸送活性について、in−vitro試験において試験した。
E_1_BE(配列番号48、49、50、51、54、58、60、61、62、63のペプチドを含む)
L6−GLUT4myc細胞を、10%FBSおよび2μg/mlのブラストサイジン中で成長させた。細胞を48〜72時間成長させ、その後、2%FBS中、ウェルあたり15,000個細胞で24ウェルプレート中に播種し、実験に先立って6から8日間分化させた。
細胞表面でのmycタグをつけたGLUT4の量を、抗体がカップリングされた比色アッセイによって測定した。手短には、インスリンとともの30分間の、または合成ペプチドもしくはペプチド組成物ととものそれぞれ3時間のいずれかのインキュベーション後に、L6−GLUT4myc細胞を、3%パラホルムアルデヒド(PFA)ととものインキュベーションによって固定した。次いで、0.1Mグリシン溶液を添加してPFAをクエンチし、5%ヤギ血清を用いて細胞をブロッキングした。筋管単層を抗myc抗体に曝露し、次いで、ペルオキシダーゼがコンジュゲートしているロバ抗マウスIgGとともにインキュベートした。各ウェルに1mLのo−フェニレンジアミンジヒドロクロリド(OPD)試薬を添加し、250μl/ウェルの3M HCLを添加することによってこの反応を停止した。細胞表面へのGLUT4転位置を決定するために、各条件の測定されるアリコートを、492nmの吸光度を使用してプレートリーダーで分光光度的に調べた。
ペプチドを適した溶媒でまず希釈した。ジメチルスルホキシド(DMSO)を、予測される水溶解度の低いペプチドの最適の溶媒とした。各合成ペプチドについて200、20、2および.2μΜで、各ウェルのDMSOの最終濃度が、表1に示されている。
1M NaOHまたはHCLを使用してpHを6〜7の間に調製することによってペプチド組成物を調製し、続いて、滅菌濾過した。
未処理対照に加えて、100nMのインスリンを使用して、最大GLUT4転位置反応を刺激した、すなわち、各実験における陽性対照。100nMインスリンに対する反応における細胞表面GLUT4転位置の平均増大は、未処理対照と比較した場合に1.72倍であった(図3)。処理は、すべての条件(未処理、インスリンおよびサンプル)が同一時点で処理されるようにずらして配置した。0.2〜2μΜの範囲の濃度で、SP2はGLUT4転位置を増大する傾向があった。200μΜのSP1は、不十分な細胞生存力によって転位置を減少させる傾向があった。
ペプチド組成物E_1_BEは、0.25〜0.5mg/mlの範囲の濃度でGLUT4転位置を増大する傾向があったが、しかし、1および2mg/mlは、進行性の細胞死を誘導した。
SP2ならびに組成物E_1_BEおよびI_2_BEは、骨格筋GLUT4転位置に対する刺激効果の傾向を示し、骨格筋におけるグルコース輸送を促進するその能力についてのさらなる調査を正当化する。
I_2_BEまたはE_1_BEは、精製水中の溶液または懸濁液として投与される。安定性データによれば、精製水中、10mg/mlの試験項目製剤は、光から保護された+2〜+8℃で10時間安定である。したがって、試験項目製剤は、光から保護された+2〜+8℃で維持され、調製後、10時間以内に使用される。製剤化の特徴および貯蔵の最大期間を以下に詳述する。

種の選択:マウスを、ヒトにおける薬物の薬理学的効果の予測材料としてのその許容性、およびこの種は薬力学的研究に適しているという規制当局による認識のために選択した。
体重:ランダム化の当日に、各群間で2.5gの最大範囲が達成されなければならない。ランダム化の当日の動物の体重は、報告書に記載される。体重に基づく動物の選別を可能にするために約10%過剰の動物を注文し、群に割り当てられない場合には、これらは、予期しない事象の場合において予備動物として利用可能となる。

体重および糖血症に対するI_2_BEおよびE_1_BEの効果を、媒体のものと比較し、D1からD15の各群中の血糖の進展に対応する差分(デルタ)を算出する。D−5からD1の、従って処置前の血中グルコースの進展は、疾患の進行が3群すべてにおいて同一であることを示す。D1およびD15の間の対照と比較して両ペプチド組成物について活性の強い傾向が観察され、これは、両ペプチド組成物が、糖尿病性動物において血糖の進展を制御できることを示す。
