Abstract
【解決手段】
マラリアに感染した赤血球を測定するための第1測定試料を血液試料から調製する第1試料調製部5Aと、赤血球の自家蛍光を測定するための第2測定試料を血液試料から調製する第2試料調製部5Bと、第1測定試料及び第2測定試料に光を照射する光源部62と、光を照射された前記第1測定試料で生じる蛍光及び散乱光を検出し、光を照射された前記第2測定試料で生じる自家蛍光を検出する検出器63、64と、第1測定試料から検出された蛍光及び散乱光に基づきマラリア感染に関する情報を生成し、第2測定試料から検出された赤血球の自家蛍光に基づき鉄欠乏性貧血に関する情報を生成する情報処理部3と、を備える血液分析装置。

Description
実施形態に係る血液分析装置は、図1に示すように、測定部2と、情報処理部3と、を備える。測定部2は、血液試料を取り込み、血液試料から第1及び第2測定試料のそれぞれを調製し、第1及び第2測定試料のそれぞれを光学測定する。情報処理部3は、測定部2の測定により得られた第1測定試料由来の第1測定データ及び第2測定試料由来の第2測定データを処理し、血液試料の分析結果を出力する。
図4を参照し、血液分析装置1の動作について説明する。
C57BL/6マウス(メス、6週齢:日本エスエルシー株式会社より購入)3匹に非致死性のげっ歯類マラリア原虫株P.yoelli 17XNL感染赤血球(3×105感染赤血球/匹)を腹腔内投与した。その後、4、7、10、14、17、21、28、35、42、及び49日目に、マウス末梢血を採取し、採取したマウス末梢血をPBSで50倍希釈後、多項目自動血球分析装置XN−30(シスメックス株式会社製)を用いて、全赤血球数に対するマラリア感染赤血球数の比率、自家蛍光赤血球数、及び赤血球数を測定した。結果を図19に示す。多項目自動血球分析装置XN−30は、光学検出装置6と同等の構成を備えている。
Claims (14)
- マラリアに感染した赤血球を測定するための第1測定試料を血液試料から調製する第1試料調製部と、
赤血球の自家蛍光を測定するための第2測定試料を前記血液試料から調製する第2試料調製部と、
前記第1測定試料及び前記第2測定試料に光を照射する光源部と、
光を照射された前記第1測定試料で生じる蛍光及び散乱光を検出し、光を照射された前記第2測定試料で生じる自家蛍光を検出する検出部と、
前記第1測定試料から検出された蛍光及び散乱光に基づきマラリア感染に関する情報を生成し、前記第2測定試料から検出された自家蛍光に基づき鉄欠乏性貧血に関する情報を生成する情報処理部と、
を備える、血液分析装置。 - 前記第1試料調製部は、前記血液試料と、赤血球を収縮させる溶血剤と、核酸を染色する色素とを混合することにより、前記1測定試料を調製する、請求項1に記載の血液分析装置。
- 前記第2試料調製部は、前記血液試料と、赤血球を収縮させる溶血剤および核酸を染色する色素を含まない希釈液と、を混合することにより、前記2測定試料を調製する、請求項1又は2に記載の血液分析装置。
- 前記血液試料を収容した試料容器から前記血液試料を吸引する吸引部をさらに備え、
前記第1試料調製部および前記第2試料調製部のそれぞれは、前記吸引部により吸引された前記血液試料の一部から、前記1測定試料および前記2測定試料を調製する、請求項1から3のいずれか1項に記載の血液分析装置。 - 前記光源部が発する光の波長帯域が400nm以上435nm以下である、請求項1から4のいずれか1項に記載の血液分析装置。
- 前記第1及び第2測定試料を流すフローセルをさらに備え、
前記光源部が、前記フローセル中を流れる前記第1及び第2測定試料に光を照射するように構成されており、
前記検出部が、前記フローセル中を流れる前記第1測定試料で生じる蛍光及び散乱光を検出し、前記フローセル中を流れる前記第2測定試料で生じる自家蛍光を検出するように構成されている、
請求項1から5のいずれか1項に記載の血液分析装置。 - 前記第1及び第2測定試料の一方が前記フローセルを通過した後、前記第1及び第2測定試料の他方が前記フローセルを通過する前に、前記フローセルを洗浄する洗浄装置をさらに備える、請求項6に記載の血液分析装置。
- 前記情報処理部が、前記マラリア感染に関する情報と前記鉄欠乏性貧血に関する情報を同時に表示装置に出力するための表示情報を出力する、請求項1から7のいずれか1項に記載の血液分析装置。
- 前記マラリア感染に関する情報が、マラリアに感染した赤血球の数、又は、赤血球の数に対するマラリアに感染した赤血球の数の比率を含む、請求項1から8のいずれか1項に記載の血液分析装置。
- 前記マラリア感染に関する情報が、マラリアに感染した赤血球の数、又は、赤血球の数に対するマラリアに感染した赤血球の数の比率と、マラリア感染の可能性判断のための閾値との比較結果に基づく、マラリア感染の可能性を示す情報を含む、請求項1から9のいずれか1項に記載の血液分析装置。
- 前記マラリア感染の可能性を示す情報が、感染したマラリアの種類を示す情報を含む、請求項10に記載の血液分析装置。
- 前記鉄欠乏性貧血に関する情報が、自家蛍光を発する赤血球の数、又は、赤血球の数に対する自家蛍光を発する赤血球の数の比率を含む、請求項1から11のいずれか1項に記載の血液分析装置。
- 前記鉄欠乏性貧血に関する情報が、自家蛍光を発する赤血球の数、又は、赤血球の数に対する自家蛍光を発する赤血球の数の比率と、鉄欠乏性貧血の可能性判断のための閾値との比較結果に基づく、鉄欠乏性貧血の可能性を示す情報を含む、請求項1から12のいずれか1項に記載の血液分析装置。
- 前記マラリア感染に関する情報が、マラリアに感染した赤血球の数、又は、赤血球の数に対するマラリアに感染した赤血球の数の比率と、マラリア感染の可能性判断のための閾値と、の比較結果を含み、前記鉄欠乏性貧血に関する情報が、自家蛍光を発する赤血球の数、又は、赤血球の数に対する自家蛍光を発する赤血球の数の比率と、鉄欠乏性貧血の可能性判断のための閾値と、の比較結果を含み、
前記情報処理部は、マラリア感染の可能性判断のための閾値との比較結果がマラリア感染の可能性を示し、鉄欠乏性貧血の可能性判断のための閾値との比較結果が鉄欠乏性貧血の可能性を示すとき、注意喚起を促すフラグを生成する、
