Description
血液脳関門(BBB)は、末梢から脳へのほとんどの高分子の移行を阻止するため、脳への曝露が必要とされる大分子治療薬の使用を制限する。トランスフェリン受容体(TfR)は、BBBで高度に発現され、受容体媒介性トランスサイトーシスを介してBBBを通過させて上述のような治療薬を輸送するために使用することができる。潜在的な治療薬がBBBを通過する能力を評価する目的で、マウスTfRが完全長のヒトTfR cDNAに置換されたマウスモデルが以前に開発された。しかしながら、これらのトランスジェニックマウスは不健康であり、異常に高いTfR発現、少ない赤血球数、及び高い血清鉄濃度を示した。Yu et al.,Science Trans.Med.,6(261):261ra154(2014)(非特許文献1)。結果として、これらの既存のマウスモデルは、脳疾患を治療するためにBBBを通過することができる治療薬を評価するためのツールとしての使用には適しておらず、内因性TfRの発現及び表現型をより呈するモデルが必要とされる。
一態様では、本開示は、非ヒト哺乳動物トランスフェリン結合部位、及び配列番号1と少なくとも80%同一のアミノ酸配列を有する異種頂端ドメインを含む、キメラトランスフェリン受容体(TfR)ポリペプチドをコードする核酸配列を含むポリヌクレオチドを提供する。いくつかの実施形態では、異種頂端ドメインは、配列番号1のアミノ酸配列を含む。いくつかの実施形態では、異種頂端ドメインは、配列番号7、配列番号8、または配列番号9のアミノ酸配列を含む。

本明細書で使用する場合、文脈上明らかに別段に示されている場合を除き、「1つの(a)」、「1つの(an)」及び「その(the)」という単数形には、複数の言及物が含まれる。したがって、例えば、「1つの抗体」への言及は、2つ以上のかかる分子の組み合わせが任意に含まれる、などである。
トランスフェリン受容体は、リガンドが占有するトランスフェリン受容体の受容体媒介性エンドサイトーシスを介して鉄の細胞への取り込みをもたらす。TfRは、ヒトならびに霊長類及びげっ歯類などの非ヒト種の両方に存在する。天然ヒトTfR(huTfR)、Uniprot P02786、配列番号6は、ホモ二量体II型膜貫通タンパク質であり、細胞質ドメイン、膜貫通領域、及び頂端ドメインとトランスフェリン結合ドメインとを含む細胞外ドメインを持つ。huTfRの各モノマーは、構造的に異なる3つのドメイン、膜に近接したプロテアーゼ様ドメイン、全ての二量体の接触を担うヘリカルドメイン、及び膜遠位頂端ドメイン(Lawrence et al.,Science,286(1999),pp.779−782)を有する。HuTfRダイマーは、約190,000ダルトンの分子量を有する。配列番号1の配列(配列番号2によりコードされる)を有するhuTfRの頂端ドメインは、トランスフェリンとTfRとの間の相互作用に関与しない。このドメインは、他のタンパク質がTfRに結合するための接触面を提供し得ると推測される。天然カニクイザル、天然アカゲザル、及び天然チンパンジーのTfRもまた、例えば、受託番号XP_005545315、NP_001244232.1、及びXP_003310238.1のそれぞれで言及されるように即知である。天然カニクイザル、天然アカゲザル、及び天然チンパンジーのTfRの頂端ドメインは、それぞれ配列番号1の天然ヒトTfRの頂端ドメインとの約96%、95%、及び98%の配列同一性を共有する。
いくつかの実施形態では、キメラTfRのコード配列、特にhuTfR頂端ドメインをコードする配列は、マウスでのキメラTfRの発現を改善するためにコドン最適化されている。コドン最適化の方法は、例えば、http://genomes.urv.es/OPTIMIZERよりアクセスできるオプティマイザ、及びDNA 2.0(Newark,California)のGeneGPS(登録商標)Expression Optimization Technologyなど、容易に利用可能である。好ましい実施形態では、コード配列は、GenScript(Piscataway,New Jersey)のOptimumGene(商標)アルゴリズムを使用して、マウスでの発現のためにコドン最適化される。
本明細書に開示される異種頂端ドメインのノックインを含む非ヒトトランスジェニック動物は、様々な方法、例えば、ジンクフィンガーヌクレアーゼ(ZFN)、テールエフェクタードメインヌクレアーゼ(TALEN)、トランスポゾン媒介システム、及びCRIPSR/Cas9システムを使用して生成することができる。これらの方法は、典型的には、1つまたは複数のヌクレアーゼをコードする1つまたは複数のポリヌクレオチドを細胞に投与し、DNAが切断されてDNA鎖に5’及び3’切断末端が生じることにより内因性遺伝子の修飾を媒介することを含む。5’末端から5’方向に伸びる配列及び3’末端から3’方向に伸びる配列に実質的に相同な左右の相同アームと隣接しているドナー配列の存在下で、該ドナーは、相同性指向修復(homology−directed repair、HDR)を介してヌクレアーゼの標的となる内因性遺伝子に組み込まれる。いくつかの実施形態では、ノックインは、CRISPR/Cas9システムを使用して実施される。例えば、異種頂端ドメインをコードする核酸配列が、内因性TfR遺伝子に導入され、キメラTfRを生成し、その結果、頂端ドメインをコードする自然発生の配列が置換されるが、遺伝子の全体的な構造は維持される。

いくつかの実施形態では、キメラTfRは、ZFNを使用してhuTfR頂端ドメインをノックインすることによって生成される。ZFNは、FokIエンドヌクレアーゼの非特異的切断ドメイン(N)とジンクフィンガータンパク質(ZFP)とを含む融合タンパク質である。ZNFのペアは、標的遺伝子の特定の遺伝子座を認識するために関与しており、一方は修飾される部位の上流の配列を認識し、もう一方は修飾される部位の下流の配列を認識する。ZFNのヌクレアーゼ部分は、特定の遺伝子座で切断する。次に、上記のようなドナーDNAは、特定の遺伝子座に挿入できる。ZFNを使用して遺伝子発現を低減する方法は、例えば、米国特許第9,045,763号、及びさらにDurai et al.,“Zinc Finger Nucleases:Custom−Designed Molecular Scissors for Genome Engineering of Plant and Mammalian cells,”Nucleic Acid Research,33(18):5978−5990(2005)にも開示されているように周知であり、その開示はその全体が参照により本明細書に組み込まれる。

いくつかの実施形態では、本発明は、例えば、上記のキメラトランスフェリン受容体をコードする核酸配列を含むキメラTfRを発現する宿主細胞を提供する。いくつかの実施形態では、宿主細胞は非ヒト哺乳動物細胞である。上記のノックイン方法のいずれか、すなわち、CRISPR、TALEN、ジンクフィンガーヌクレアーゼを使用して、宿主細胞内の天然トランスフェリン受容体の頂端ドメインを、配列番号1と少なくとも80%同一のアミノ酸配列を有する異種頂端ドメインで置換することができる。いくつかの実施形態では、宿主細胞は真核生物、例えば、配列番号1と少なくとも80%同一のマウス細胞である。いくつかの場合では、宿主細胞は、sgRNA及びCas9、異種頂端ドメインをコードする核酸配列を含むドナーDNAと接触され、該核酸配列は、左右の相同アームと隣接している。sgRNA及び相同アームは、異種頂端ドメインコード配列がゲノム内の位置に挿入され、宿主細胞の天然トランスフェリン受容体の頂端ドメインのコード配列を置換するような配列を有する。いくつかの実施形態では、宿主細胞は、マウス、ラット、ウサギ、ウシ、ヒツジ、イヌ、ネコ、ウマ、ブタなどの非霊長類哺乳動物由来の細胞である。
本明細書で使用される「頂端ドメイン結合ポリペプチド」または「ADBP」は、配列番号1と少なくとも80%同一のアミノ酸配列を有する頂端ドメインに結合するポリペプチドを指す。ADBPは、キメラTfRのhuTfRの頂端ドメインに結合することができる抗体または任意のポリペプチドであり得る。いくつかの実施形態では、ADBPは、血液脳関門を通過して送達される薬剤である。いくつかの実施形態では、ADBPは、例えば共有結合によりそれに結合したエフェクター分子をさらに含む。エフェクター分子は、治療薬、標識薬、または診断薬であり得る。ある特定の実施形態では、エフェクター分子は、治療用または診断用抗体などのポリペプチド、または酵素もしくはシグナル伝達分子に対して酵素活性もしくは阻害活性を有するポリペプチドである。ある特定の実施形態では、エフェクター分子は、小分子、RNA、DNA、またはタンパク質を含む。
本明細書に開示されるキメラTfRを使用して、TfRに結合することができるADBPをスクリーニングすることができる。スクリーニングする方法は、候補ADBPを上記に開示されたキメラTfRと接触させるステップと、キメラTfRに結合する候補ADBPの量を決定するステップとを含む。いくつかの実施形態では、候補ADBPをキメラTfRと接触させるステップは、ADBPを、キメラTfRを発現する宿主細胞と接触させることを含む。いくつかの場合では、候補ADBPをキメラTfRと接触させるステップは、ADBPを、キメラTfRを発現する内皮と接触させることを含む。いくつかの実施形態では、内皮はBBB内皮である。
上記のキメラTfRを発現する非ヒトトランスジェニック動物を使用して、キメラTfRの頂端ドメインに結合するADBPの能力、及び最終的にBBBを通過する能力を特徴付けることができる。
いくつかの実施形態では、本明細書に記載のキメラトランスフェリン受容体ポリヌクレオチドまたはポリペプチド、またはそのようなポリペプチドを発現する細胞を含むキットが提供される。いくつかの実施形態では、キットは、上記のADBPのスクリーニングに使用するためのものである。
ノックイン/ノックアウトマウスを生成する方法は文献に発表されており、当業者には周知である。簡単に説明すると、C57Bl6マウスを使用して、単細胞胚への前核マイクロインジェクションを介してヒトの頂端TfRマウス系統のノックインを生成し、続いて偽妊娠雌へ胚移植した。具体的には、Cas9、sgRNA、配列番号10〜11、及びドナーDNA、配列番号4が胚に導入された。ドナーDNAは、マウスでの発現のためにコドン最適化されたヒト頂端ドメインコード配列、配列番号2を含んでいた。頂端ドメインコード配列は、左の相同アーム(配列番号4のヌクレオチド1〜817)及び右の相同アーム(配列番号4のヌクレオチド1523〜2329)と隣接していた。ドナー配列は、この方法で設計され、頂端ドメインが第4のマウスエクソンの後に挿入され、その3’末端に第9のマウスエクソンが直接隣接した。胚を移植された雌の子孫からの始祖の雄は、野生型の雌と交配させて、F1ヘテロ接合マウスを生成した。続いて、ホモ接合マウスは、F1世代のヘテロ接合マウスの繁殖から生成された。
ヒトTfRまたはヒト/マウスBACE1を標的とするツール抗体は、重鎖と軽鎖とをコードするDNAを含む発現プラスミドでExpi293またはExpiCHO細胞を形質転換し、当業者に周知のプロトコルを使用して生成された。二重特異的抗体は、「knobs−into−holes」技術を使用して生成され、knobとholeとのハーフ抗体は別々に発現され、公開された方法を使用して結合された。抗体は、最初にプロテインAで精製し、次にサイズ排除クロマトグラフィで精製された。これらの研究のために生成された抗体は以下である:
抗TfR:ヒトTfR頂端ドメインに結合するヒトIgG1抗体。
抗BACE1:ヒトBACE1に結合し、マウスBACE1と交差反応するヒトIgG1抗体。この抗体は、BACE1の酵素活性を阻害する。
抗TfR/BACE1:ヒトTfR頂端ドメインならびにヒト及びマウスBACE1に結合するヒトIgG1 knobs−into−holes二重特異的抗体。knobハーフ抗体は、抗BACE1抗体からの可変ドメインを有し、holeハーフ抗体は、抗TfR抗体からの可変ドメインを有する。
血液は、野生型C57Bl6、huTfR頂端+/−、huTfR頂端+/+マウス(n=3/グループ)から採取され、標準的な全血球算定(CBC)分析が実施された。遺伝子型特異的な違いは、総赤血球、ヘモグロビン、及びヘマトクリットレベルを含む全ての赤血球パラメータにおいて観察されなかった(図1)。
この例では、抗TfR抗体は、huTfR頂端+/−マウスにおけるTfR標的治療薬の脳への取り込みを評価するために生成された。huTfR頂端+/−マウスまたは野生型C57Bl6は、5mg/kgの抗TfR抗体を静脈注射された。1時間後、マウスは屠殺され、PBSで灌流された。半脳は、4%PFA内に一晩入れて固定した後、30%スクロースで保存した。矢状脳切片(35μm)はミクロトームを使用して切断され、5%BSA+0.3%Triton X−100でブロックされた後、Alexa488抗huIgG1(1:500)で蛍光二次染色した。脳画像は、20倍の対物レンズを備えたZeissの広視野顕微鏡を使用して撮影された。有意な血管染色がhuTfR頂端+/−マウスで観察され、TfRが高度に発現しているBBBの脳内皮細胞上のヒト頂端特異的抗TfRの強固な結合が示された(図2)。対照的に、ごく少量の染色が野生型マウスで観察された。
この実施例では、huTfR頂端+/+マウスは、50mg/kgの抗BACE1抗体または抗TfR/BACE1二重特異的抗体のどちらか一方を静脈注射された。24時間後、血液は心臓穿刺を介して採取され、マウスはPBSで灌流された。脳組織は、10倍の組織重量のPBS中に1%NP−40を含有する溶解緩衝液で均質化された。血液は、EDTAチューブに採取され凝固するのを防止し、血漿を分離するために14000rpmで7分間回転させた。マウス血漿及び脳溶解物中の抗体濃度は、製造元の指示に従って、一般的なヒトIgGアッセイ(MSD human IgG kit #K150JLD)を使用して定量された。簡単に説明すると、事前にコーティングしたプレートは、MSD Blocker Aで30分間ブロックされた。血漿試料は、Hamilton Nimbusリキッドハンドラを使用して1:10,000に希釈され、ブロックされたプレートに二重に添加された。脳試料は、1%NP40溶解緩衝液で均質化され、溶解物をPK分析用に1:10に希釈した。投与溶液もまた同じプレートで分析され、正確な投与量を確認した。0.78〜200ng/mLのIgGの標準曲線は、4パラメータロジスティック回帰を使用して適合された。
TfR発現レベルがhuTfR頂端+/+マウスで変化するかを決定するために、野生型及びhuTfR頂端+/+マウスから脳及び様々な末梢組織が分離された。脳、肝臓、肺、及び腎臓は、PBSの灌流後のマウスから採取された。組織は、10倍の組織重量のPBS中に1%NP−40を含有する溶解緩衝液で均質化された。試料はウエスタンブロットで実施され、TfR発現レベルはTfRの細胞内部分を認識し、その結果野生型とhuTfR頂端+/+との両方に対して交差反応するTfR抗体(1:2000;Thermofisher#13−6800)を使用して決定された。TfR発現の定量化は、アクチンとの比として表された(1:5000;Abcam 8227)。図5A〜5Dは、huTfR頂端+/+マウスにおけるTfR発現が、野生型マウスにおける脳(図5A)、肝臓(図5B)、腎臓(図5C)、及び肺(図5D)と非常に類似していることを示す。

Claims (67)
- 非ヒト哺乳動物トランスフェリン結合部位と、配列番号1と少なくとも80%同一のアミノ酸配列を有する異種頂端ドメインとを含むキメラトランスフェリン受容体(TfR)ポリペプチドをコードする核酸配列を含むポリヌクレオチド。
- 前記異種頂端ドメインが、配列番号1のアミノ酸配列を含む、請求項1に記載のポリヌクレオチド。
- 前記異種頂端ドメインが、配列番号7、配列番号8、または配列番号9のアミノ酸配列を含む、請求項1に記載のポリヌクレオチド。
- 前記非ヒト哺乳動物トランスフェリン結合部位が、天然マウストランスフェリン結合部位である、請求項1〜3のいずれか1項に記載のポリヌクレオチド。
- 前記キメラTfRポリペプチドが、配列番号3との少なくとも80%のアミノ酸配列同一性を有する、請求項1〜4のいずれか1項に記載のポリヌクレオチド。
- 前記キメラTfRポリペプチドが、配列番号3のアミノ酸配列を含む、請求項5に記載のポリヌクレオチド。

- 前記キメラTfRをコードする前記ポリヌクレオチドが、マウストランスフェリン受容体遺伝子のエクソンとイントロンとを含み、かつ前記異種頂端ドメインをコードする前記核酸配列が、マウストランスフェリン受容体遺伝子の頂端結合ドメインに取って代わるように前記マウストランスフェリン受容体遺伝子の第4のエクソンの後に配置されている、請求項1〜8のいずれか1項に記載のポリヌクレオチド。
- 請求項1〜9のいずれか1項に記載のポリヌクレオチドによってコードされるキメラトランスフェリン受容体(TfR)ポリペプチド。
- 請求項1〜9のいずれか1項に記載のポリヌクレオチドを含む、宿主細胞。
- マウス細胞である、請求項11に記載の宿主細胞。
- キメラトランスフェリン受容体(TfR)ポリペプチドを発現する宿主細胞であって、前記キメラTfRポリペプチドが、(a)TfRポリペプチドの内因性頂端ドメインに代わる異種頂端ドメインと、(b)内因性トランスフェリン結合部位とを含む、宿主細胞。
- 前記異種頂端ドメインが、配列番号1と少なくとも80%同一のアミノ酸配列を有する、請求項13に記載の宿主細胞。
- 前記異種頂端ドメインが、配列番号1のアミノ酸配列を含む、請求項13に記載の宿主細胞。
- 前記細胞内の前記異種頂端ドメインをコードする核酸配列が、配列番号2のヌクレオチド配列を含む、請求項13、14、または15のいずれか1項に記載の宿主細胞。

(b)前記非ヒトトランスジェニック動物の脳内の前記ADBPの存在または活性を測定するステップと

(b)前記雌より生まれた子孫から非ヒトトランスジェニック動物を選択するステップであって、前記非ヒトトランスジェニック動物は、内因性TfRの頂端ドメインが配列番号1との少なくとも80%の同一性のアミノ酸配列を有する異種頂端ドメインで置換されたキメラトランスフェリン受容体(TfR)ポリペプチドを含む、ステップと
を含む、方法。 - キメラトランスフェリン受容体(TfR)ポリペプチドを発現する非ヒトトランスジェニック動物を生成する方法であって、
(a)配列番号1との少なくとも80%の同一性を有する頂端ドメインをコードするポリヌクレオチドを、動物の胚細胞内に導入するステップであって、前記ポリヌクレオチドが、内因性TfR頂端ドメインをコードする内因性TfR遺伝子の領域を標的とし、かつ配列番号1との少なくとも80%の同一性を有する前記頂端ドメインをコードする前記ポリヌクレオチドが、前記内因性頂端ドメインをコードする前記内因性TfR遺伝子の前記領域を置換する、ステップと、
(b)前記細胞またはその子孫を非ヒトトランスジェニック動物へと発達させるステップと
