Abstract
Description
この実施例は実施例2〜9のデータを得るために使用された材料及び方法を説明するものである。
この例は、ナイーブCD8+T細胞のタイプIエフェクター分化を調整する指令が、mTOR活性を増強することを示すものである。タイプIエフェクター機能のためのナイーブCD8+T細胞の調製の基礎となる指令(各々、シグナル1、2、3−抗原[Ag]、B7.1[共刺激]、IL−12[サイトカイン])の機構を特徴づけるために、我々は、接着細胞株(H−2Kb、OVAp及びB7.1を発現しているBOK)で刺激されたOT−I細胞におけるタイプIエフェクターの成熟にIL−12が果たす決定的な役割を確定することから検討を開始した。IL−12の付加は、強度のIFN−γの産生、72時間のOT−I細胞における細胞傷害性T細胞(CTL)活性をもたらした(図1A,1B、一次)。
さらに、一次エフェクターOT−I細胞(72時間)をさらに72時間IL−7と静置し(144時間目に12%IFN−γが検出された)、Ag及びB7.1に刺激された場合、IL−12処理されたOT−I細胞のみがIFN−γ及びCTL活性が再度誘発されていた(図1A、1B、二次)。このように、IL−12はCD8+T細胞エフェクター成熟に決定的な役割を有している。
本実施例は、CD8+T細胞におけるIL−12増強性のmTOR活性はPI3K及びSTAT4を要求することを示すものである。CD8+T細胞におけるmTOR活性を支配する分子的な経路を決定するために、我々はAg−、B7.1−、及びIL−12−誘導性のホスホイノシチド3−キナーゼ(PI3K)−Aktキナーゼ経路がCD8+T細胞におけるmTORシグナルに要求されるかどうか分析した。Ag+B7.1±IL12で刺激されたOT−I細胞は、PI3K活性の機能的な測定としてAktリン酸化(Thr308)が測定された。IL−12の有無に関わらずAg+B7.1は30分までに同量のAktリン酸化を誘導したが、IL−12の存在下では48時間までAktリン酸化が増加した。このAktリン酸化はPI3K阻害剤(LY294002)によってブロックされた(図2A)。こうしてIL−12はOT−I細胞においてAg+B7.1誘導性PI3K活性を増加させることが確認された。さらに、IL−12で増強されたmTOR活性(2、12、24時間において観察されたS6Kリン酸化)は、PI3K阻害によってブロックされた(図2B)、このことは、抗原に刺激されたOT−I細胞における、Ag+B7.1及びIL−12に活性化されるPI3K活性は、mTORキナーゼ活性の誘導に要求されることを示している。
本実施例は、維持されたmTOR活性は遺伝的なタイプIエフェクター機能に対して本質的であることを示すものである。抗原刺激中のIL−12の存在はmTOR活性を増加させ、また、タイプIエフェクターの成熟に決定的であるので、我々は、維持されたmTORキナーゼ活性はOT−I細胞におけるIL−12調節性のタイプIエフェクター機能に要求されるかどうかについて分析した。そうするために、我々はナイーブOT−I細胞をBOK±IL−12、及びラパマイシンで刺激し、エフェクター機能が一次及び二次活性化されたOT−Iプールにおいて分析された。IL−12で調整されたOT−I細胞へのラパマイシンの追加は、一次活性化されたOT−I細胞からのIFN−γ産生に影響しなかったが、グランザイムB(GranzymeB)発現の減少に伴ってCTL活性を減少させた(図3A、3B、3C;一次)。対照的に、我々は、二次活性化されたプールからIL−12で調整されたエフェクター機能の完全な回復を認めた。(IFN−γ産生、CTL活性)(図3A、3B;二次)。このIL−12で調整されたタイプIエフェクター機能のブロックは、ラパマイシン誘導性の細胞増殖及び/又はタンパク質合成の阻害によるものでない。なぜなら、IL−12存在下でのこれらの細胞の再活性化は、IFN−γの顕著な産生を招いたからである。これらの結果は、IL−12誘導性のナイーブCD8+T細胞のタイプIエフェクター機能への関与はmTOR活性を要求することを示唆している。加えて、我々は、一次刺激後144時間において、ラパマイシン処理のIL−12誘導性のIFN−γ産生及びCTL活性のブロックを観測した。これらの結果はさらにラパマイシン処理はタイプIエフェクター機能をブロックすることを確認しており、また、二次活性化プール(168時間)において観察されたエフェクター機能の欠失は単独のものであると考えられた。なぜなら、これらの細胞はIFN−γ産生を再誘導する能力はないからであり、ラパマイシン処理された細胞の不応性によるのではないからである。
本実施例は、IL−12増強性のmTOR活性は持続的なT−bet発現に重要であることを示すものである。持続的なT−betの発現はタイプIエフェクター細胞の運命づけに必要かつ十分であり(松田ら、2007)、また、mTOR阻害はOT−I細胞においてIL−12刷り込みのタイプIエフェクター成熟を逆転させるため(図3)、我々は次に、ラパマイシン処理がOT−I細胞においてT−bet発現に影響するかどうか、T−betRNA発現の速度論的解析を行うことで、決定するために検討をおこなった(図4A)。試験された全ての時点(24−96時間)において、IL−12は、Ag+B7.1誘導性のT−bet発現を増強し、維持した。しかしながら、mTOR阻害は、Ag+B7.1及びIL−12誘導性の早期のT−bet発現(24−48時間)には影響しなかったが、IL−12誘導性の持続的なT−betmRNA発現をブロックし(96時間にわずかに観察される)、そしてそれに応じて、OT−I細胞はT−betタンパク質発現を喪失した(図4B)。さらに、12時間におけるmTOR活性の阻害はまた、観察されたタイプIエフェクター成熟の欠失と同様に、T−bet発現の欠失を生じた(図3E)。このように、IL−12に増大された(増強され、維持された)mTOR活性は、CD8+T細胞における継続的なT−bet発現に要求される。
本実施例は、CD4+及びCD8+細胞におけるmTORキナーゼの異なる要求を示すものである。CD4+T細胞のラパマイシン処理はFoxp3発現制御性T細胞に免疫不応答(anergy)及び/又は偏位(deviation)をもたらすので、我々は、Ag+B7.1及びIL−12誘導性のmTOR活性の阻害が、活性化や増殖をブロックし、及び/又は異なるサブタイプのエフェクターへの偏在を生じて、CD8+T細胞のタイプIエフェクター分化に干渉するかどうかを分析した。CD4+T細胞の公知の観察に従い、我々の結果も、ラパマイシン処理は活性化(CD44発現)、増殖(CFSE希釈)、及び、CD4+T細胞(OT−II)の細胞回復を顕著に低減することを示す。しかしながら、ラパマイシン処理はCD8+T細胞(OT−I)の早期(CD69、12時間)及び遅れた活性化(CD44)に影響せず、ただ増殖(CFSE)と細胞の回復にわずかに影響したのみであった。さらに、報告されているCD4+T細胞のFoxP3発現とは対照的に、ラパマイシン処理されたOT−I細胞は継続的にFoxP3を発現しなかったが、これはT細胞の調節的機能の一部である。さらに、mTOR阻害でのT−betの欠失は、タイプ2もしくはタイプ17サブセットへの偏位を導かなかった。これらの観察は、ラパマイシンの多様な投与量(20ng/mL〜2μg/mL)でも確認された。高投与量ではラパマイシンはOT−I細胞(S6Kp、S6p)のmTOR活性を効果的に阻害したが、CD4+T細胞とは異なって、活性化阻害、増殖及び調節性T細胞サブセットの偏在はなかった。これらの結果は、ラパマイシンはCD4+及びCD8+T細胞で違う効果を有することを示している。そして、IL−12誘導性のタイプI CDS+エフェクター分化のブロック能力は、他のエフェクターサブタイプに対する免疫不応答や偏位が理由ではないことをこれらの結果は示すものである。
本実施例は、mTOR阻害が持続的なエオメソデルミンの発現を誘導し、メモリー−前駆CD8+T細胞を産生することを示すものである。ラパマイシン処理は、タイプIエフェクター分化をブロックせず、また、免疫不応答や他の転写調節因子の発現を誘導しなかったので、我々は次に、ラパマイシン処理されたIL−12で調整されたOT−I細胞の運命を特徴づけることを探求した。近い転写調節因子である、T−betとエオメソデルミンとは、エフェクターやメモリーCD8+T細胞において逆に調節を受ける。T−betの発現を抑えるmTOR阻害が、エオメソデルミンの誘導を導くかどうか決定するために、我々は、OT−I細胞におけるエオメソデルミンmRNAの発現を体系的に分析した。我々は、ナイーブOT−I細胞においてエオメソデルミンのわずかな発現を観察したが、これはAg+B7.1で刺激されたときには増強され、IL−12付加時には低減された(図5A)。しかしながら、Ag+B7.1及びIL−12調整されたOT−I細胞へのラパマイシンの付加は、エオメソデルミンmRNAの発現を顕著に増強し、これは、試験されたすべての時間(24−96時間)において維持された(図5A)。エオメソデルミンmRNAの増加は、タンパク質レベルでも確認された。ラパマイシン処理されたIL−12調整OT−I細胞は、エオメソデルミンタンパク質の有意な増加を生じたからある(図5B)。我々が一貫して、Ag+B7.1及びIL−12調整されたOT−I細胞におけるmRNA誘導なしに、エオメソデルミンタンパク質の発現のわずかな増加(有意でない)を観測したことは特筆すべきである(図5A、5B)。OT−I細胞におけるラパマイシン仲介性のエオメソデルミンの上方調節は、mTOR阻害の直接的な帰結であるのか、或いは、持続的なT−bet発現を阻害する能力による帰結であるのか検討するために、我々は、ラパマイシンで調整されたOT−I細胞において、異所的にT−bet発現を誘導し、タモキシフェンの存在/非存在下でのエオメソデルミンの発現を分析した。実際に、ラパマイシン処理されたOT−I細胞におけるT−betの誘導は、エオメソデルミンの発現を減少させた(図5C)。さらに我々は、Ag+B7.1及びIL−12で処理されたTbx21−/−OT−I細胞におけるエオメソデルミン発現の増加を、継続的に観察した。これらを合わせると、これらの結果は、IL−12で調整されたOT―I細胞においてmTOR阻害は、転写プログラムを、持続的なT−betからエオメソデルミン発現へと選択的に切り替えることを示している。我々はまたOT−I細胞において、IFN−αもmTOR活性やT−bet発現を調整するかどうかを決定した。我々は、Ag+B7.1刺激OT−I細胞において、IFN−αは、mTOR活性やT−bet発現を増強することはできないことを確認した;しかしながら我々は、エオメソデルミンの発現とIFN−γの産生において、増加を観察した。これらの結果は、IL−12は、持続的なmTORとT−betの発現の促進によってタイプIエフェクター成熟を刷り込むことについて、独特の能力を有すること、また、IFN−αは、持続的なmTOR活性とmTOR依存的なT−bet発現を促進する能力を欠くので、この活性を欠いているかもしれないことを確認した。
本実施例は、mTOR阻害はメモリーCD8+T細胞の産生を増強することを示すものである。IL−12仲介性タイプIエフェクター機能をブロックする、エオメソデルミン発現のための持続的なT−betを切り替える、そして、OT−I細胞においてメモリー様表現型を誘導するというラパマイシンの能力に基づいて、我々は、ラパマイシン処理されたIL−12で調整されたOT−I細胞が養子移入の後にメモリー応答を生じるかどうか分析した。これを試験するため、我々は初めに、ラパマイシン処理されたOT−I細胞が、CD62L及びCD69発現の増加によって示唆されるように、二次性リンパ器官に局在する能力が変化をしめすかどうかを検討した。養子移入されたAg+B7.1±IL−12及びラパマイシンで調整されたOT−I細胞(Thy1.1+)は、24時間後にC57BL/6(Thy1.2+)レシピエントにおいて検出された。ラパマイシン処理されたOT−I細胞は、二次性リンパ器官(リンパ管及び脾臓)における局在が増加していることを示した。また、これに対応して、例えば肝臓(図6A)や血液のような、三次的な場所ではより少ない数が観察された。ラパマイシン処理されないOT−I細胞ではこの局在パターンを示さなかった(図6A)。しかしながら、我々は、肺での細胞数についていかなる有意な違いも観察しなかった。すなわち、mTOR活性阻害は、抗原及びIL−12調整CD8+T細胞の局在を、二次性リンパ区画にシフトさせるのである。
本実施例は、ラパマイシン処理されたIL−12調整OT−I細胞は、増強された腫瘍効力を有することを示す。養子細胞移入(ACT)において体外で創出された腫瘍抗原特異的なエフェクターCD8+細胞の使用は、臨床的な設定において腫瘍の退縮をもたらす(モーガンら、2006)。ラパマイシン処理されたIL−12調整OT−I細胞の腫瘍効力を試験するために、我々は、ラパマイシン処理された、又はされていないIL−12調整OT−I細胞(72時間)を、E.G7腫瘍細胞、腫瘍サイズ(s.c.)を有する完全なC57BL/6レシピエントに養子移入し、その後の生存が追跡された。ナイーブOT−I細胞のレシピエントと比較して、Ag+B7.1刺激OT−I細胞を移植されたマウスは明らかな効果を示し(30日までに、100%に対し80%死亡率)、IL−12調整OT−I細胞ではさらに効果があった(30日までに50%死亡率)。ラパマイシン処理IL−12調整OT−I細胞は、120日まで78%のレシピエントが腫瘍なく生存し、著しく増強された腫瘍効力を示した(図7B)。さらに、ラパマイシン処理IL−12調整OT−I細胞は、ラパマイシン処理されていない対照と比較した場合、腫瘍サイズの制御が著しく増強されている(図7A)。これらの結果は、抗原、及び、IL−12調整エフェクターCD8+T細胞よりも大きな腫瘍効力を示すメモリー応答のためのIL−12調整CD8+T細胞を、mTOR阻害が調節することを示している。
本実施例は、テムシロリムス及びラパマイシンは、RCC及びメラノーマのマウスモデルにおいて、がんワクチンの抗腫瘍効力を増強することを示す。RCCモデルにおいて、ヒートショックタンパク質(HSP)が免疫アジュバントして提供され、標的抗原であり、明細胞RCCでは90%の発現率である炭酸脱水素酵素(CA9)と複合化された。Balb/cマウスは、CA9を発現するように改変された同系のRENCA腫瘍が移植された。腫瘍移植された目的の処理モデルでは、マウスは、移植から10日後に、テムシロリムス有り/無しのがんワクチンで処置された(図8)。図8にみられるように、ワクチン単体では腫瘍の生長に対して緩い効果しかない。テムシロリムス単独では、腫瘍生長の減少はあるが、ワクチンとテムシロリムスの組み合わせは腫瘍の生長に対してより大きな効果がある(図9)。同様にメラノーマモデルでも、ワクチンとテムシロリムスとの組み合わせは腫瘍の生長に対してより大きな効果を有した。このモデルでは、CA9はメラノーマ抗原gp100と複合化された。C57/BL6マウスはgp100を発現するように改変された同系のB16腫瘍が移植された;マウスは10日後に腫瘍ワクチンで処置された。ワクチンがラパマイシンで増強されたマウス卵巣がんモデルでも、同様の結果が観察された。
本実施例は、テムシロリムスの増強効果は免疫仲介性であることを示す。特に、テムシロリムスはインビトロにおいてRENCA(腎臓がん細胞)の生長に直接的な影響を有したが、インビトロにおいてB16メラノーマの生長には影響を有さなかった(図10)。このことは、メラノーマモデルでは、テムシロリムスの一次的な効果は免疫仲介性であることを示した。この可能性に合致して、CA9+gp100による免疫は、ELISPOTアッセイを用いて、脾臓細胞からgp100特異的なIFN−γ−応答を誘発した。この応答は、テムシロリムスと同時に処理することで有意に増大した(p<0.05)。さらに、特異的な致死はインビボのCTLアッセイにおいて、テムシロリムス処理によって増加した。Pmel−1細胞がC57/BL6マウスに養子移入され、テムシロリムス有り/無しのgp100+CA9で免疫された。Pmel−1細胞は、gp100.13の25−33番目のアミノ酸に対応するH−2Db制限エピトープを認識する遺伝子改変細胞である。H−2Db制限エピトープを負荷された標的細胞は、注入され、14時間後にフローサイトメトリーで検出された。テムシロリムス非受容群での特異的な致死は66%であった。ワクチンとともにテムシロリムスが投与された場合、特異的致死は78%に増加した。
本実施例は、本発明の多様な態様を示すものであり、各例は、mTOR阻害剤の使用が、抗がん免疫応答を増強することを示している。各例において、ブラック6(Black6)マウスが使用された。
Claims (15)
- CD8+T細胞の個体群及び哺乳類ラパマイシン標的タンパク質(mTOR)阻害剤を含む組成物。
- 前記CD8+T細胞が前記組成物の細胞中に少なくとも10%含まれる、請求項1に記載の組成物。
- 前記CD8+T細胞が特異的で、かつ、個体において免疫応答の増強が所望される抗原をさらに含む、請求項1に記載の組成物。
- さらにインターロイキン12(IL−12)を含む、請求項3に記載の組成物。
- 抗原が腫瘍抗原である、請求項3に記載の組成物。
- 増強された免疫応答を必要とする個体において、抗原に対する増強された免疫応答を得るための方法であって、前記個体において前記抗原及び哺乳類ラパマイシン標的タンパク質(mTOR)阻害剤を投与することを含む方法。
- 前記個体が、がんを有すると診断されもしくは疑われており、前記がんが前記抗原を発現しているがん細胞を含む、請求項6に記載の方法。
- 個体において、前記抗原の投与に続いて前記mTOR阻害剤が投与される、請求項6に記載の方法。
- 少なくとも7日間、少なくとも1日1回、mTOR阻害剤が個体に投与される、請求項8に記載の方法。
- 継続して20日間を超えない期間、mTOR阻害剤が個体に投与される、請求項8に記載の方法。
- mTOR阻害剤が、ラパマイシン又はテムシロリムスである、請求項6に記載の方法。
- さらに、個体において、前記抗原に特異的なCD8+T細胞を含む組成物を投与することを含む、請求項6に記載の方法。
- 個体にCD8+T細胞を投与する以前に、前記抗原がCD8+T細胞に提示されている、請求項12に記載の方法。
- 個体にCD8+T細胞を投与する以前に、CD8+T細胞がmTOR阻害剤と接触させられている、請求項13に記載の方法。
- 個体にCD8+T細胞を含む前記組成物を投与する以前に、個体からCD8+T細胞が単離されている、請求項12に記載の方法。
