また、ナノESIでイオン化を行う場合、図2(b)に示すようなイオン化プローブ22bを用いる。このイオン化プローブ22bは、基本的には上記ESIの構成からネブライズガス及び乾燥窒素ガスを除いた構成であり、液体試料を噴霧するノズル221は、先端がガラスキャピラリに金属薄膜がコーティングされたもの又は金属製キャピラリ等によるキャピラリ管であって先端が細く絞られている。ナノESIではネブライズガスによる噴霧の補助が無く、ノズル221の先端から流出した同極性のイオンを多量に含んだ液体試料はクーロン力によって細く引き伸ばされ、テーラーコーンと呼ばれる円錐形状体を形成する。その進行に伴って電荷密度が高くなると臨界点でクーロン爆発が生じ、イオンの生成を伴いつつ円錐状に広がる。
