UV18−25のBlueSTAR遺伝子ライブラリーの構築
UV18−25の染色体DNAをSau3Aで部分的に消化し、12〜15kbのフラグメントを単離し、そしてクローニングベクターBlueSTARのBamHI部位に連

CBH1のアミノ酸配列から、われわれは、該タンパク質が大きさが約63kDであること、および該タンパク質のC末端部分にセルロース結合ドメイン(CBD)が存在することを結論した。興味深いことに、単離された55kDの主要タンパク質中にCBDの存在の証拠は見出されなかった。しかしながら、コードされるCBH1タンパク質(配列番号1、2)中での、この55kDの主要タンパク質からの単離されたペプチドの存在は、該55kDのタンパク質がクローン化された遺伝子によりコードされることを確認する。これらの結果の可能な説明は、該55kDのタンパク質がCBDを欠くCBH1タンパク質の短縮されたバージョンであることである。
Xyl1のアミノ酸配列から、われわれは、またここにCBDが、このタンパク質中でN末端側に(すなわち、シグナル配列にもしくはそれから5アミノ酸未満の距離に直接結合されて)存在することを結論した。文献では、CBDをもつもう少しのキシラナーゼのみが既知である(フサリウム オキシスポルム(Fusarium oxysporum)、フミコラ グリセア(Humicola grisea)およびネオカリマスティクス パトリシアルム(Neocallimastix patriciarum))。Xyl1タンパク質の推定される大きさは43kDであり、そして30kDのキシラナーゼから単離されるいくつかのペプチドはこのタンパク質から発する(配列番号3、4)。いくつかの単離されたペプチドはコードされる配列中で見出すことができなかった。これは、代替のキシラナーゼタンパク質がUV18−25中に存在することを示す可能性がある。以前の分析で、この30kDのタンパク質中でのCBDの存在の証拠は見出されなかった。また、これらの結果から、われわれは、該タンパク質のCBDがタンパク質分解により切断されることを仮定した。この仮説は今後分析されるであろう(活性、異なるC1株、すなわちC1野性型、NG7C、UV13−6、UV18−25、およびUV18−25のプロテアーゼ突然変異体中の異なるタンパク質のN末端配列および大きさの決定により)。また、酵素活性に対するCBDの存在もしくは非存在の影響を、より詳細に分析する。多様なC1宿主中での完全長の遺伝子の過剰発現を考慮してもよい。

Q/D/A−W/F/Y−Y−Y/S/H/A−Q C−L/I/Q/V/Tであり、ここでW/YはWもしくはYなどのいずれかを意味し、Xはいかなるアミノ酸も意味し、そして−は非存在を意味する。最も縮重のコンセンサス配列を伴う4つの差異がXyl1中に存在し、それは上の配列7中で下線をつけられる。本発明は、従って、このコンセンサスCBDと異なるN末端CBDを有しかつフサリウム オキシスポルム(Fusarium oxysporum)キシラナーゼ以外のキシラナーゼに関する。より具体的には、本発明のキシラナーゼは、上の配列7と最低55%、とりわけ最低65%、好ましくは最低75%の配列の同一性を有するCBDを有する。好ましくは、CBDは、位置23のアミノ酸Phe、TyrおよびTrpの1種、もしくは4種のアミノ酸、位置20のVal、位置22のGln、位置23のPheおよび位置24のValの最低1種を含有する。好ましい配列は、Cys−Xaa−Phe、Xaa−Phe−Val、Cys−Xaa−Phe−Val、Cys−Gln−Phe、Val−Cys−Xaa−Phe、Gln−Phe−Val、Gln−Trp−Val、Gln−Tyr−Val、Val−Cys−Gln、Val−Cys−Gln−Phe、およびVal−Cys−Xaa−Phe−Valを含んで成り、ここでXaaはいずれかのアミノ酸、または好ましくはVal、Thr、Arg、Glu、GlnもしくはLys、または最も好ましくはGlnもしくはGluである。
gpd1遺伝子のC末端部分のDNA配列は決定されていない。この遺伝子のプロモーター配列は本発明の好ましい一態様であり、そして配列番号5に描かれる。

Claims (15)
- 配列番号2のアミノ酸配列と[BLASTアルゴリズムで決定した時に]少なくとも75%のアミノ酸同一性を表すクリソスポリウム(Chrysosporium)のグリコシルヒドロラーゼ ファミリー7に対応するタンパク質、または配列番号2のアミノ酸配列1−246または396−526の対応する部分に同一である少なくとも20個の連続するアミノ酸を有するそれらの部分。
- 配列番号4のアミノ酸配列と[BLASTアルゴリズムで決定した時に]少なくとも70%のアミノ酸同一性を表すクリソスポリウム(Chrysosporium)のグリコシルヒドロラーゼ ファミリー10に対応するタンパク質、または配列番号4のアミノ酸配列1−133または252−383の対応する部分に同一である少なくとも20個の連続するアミノ酸を有するそれらの部分。
- 配列番号4のアミノ酸配列と[BLASTアルゴリズムで決定した時に]少なくとも65%のアミノ酸同一性を表すクリソスポリウム(Chrysosporium)のグリコシルヒドロラーゼ ファミリー10に対応するタンパク質、または配列番号4のアミノ酸配列1−383の対応する部分に同一である少なくとも20個の連続するアミノ酸を有するそれらの部分。
- グリコシルヒドロラーゼ ファミリー10に対応し、そして配列番号4のアミノ酸配列22−53と少なくとも55%のアミノ酸同一性を有し、かつ/またはアミノ酸配列22−53から少なくとも20個の連続するアミノ酸を有し、かつ/または相対位置44にアミノ酸F、WおよびYの1つを有するセルロース−結合ドメインを含んで成るタンパク質。
- 配列番号6の部分的アミノ酸配列と[BLASTアルゴリズムで決定した時に]少なくとも86%のアミノ酸同一性を表すクリソスポリウム(Chrysosporium)のグリセルアルデヒド リン酸 デヒドロゲナーゼに対応するタンパク質、または配列番号6のアミノ酸配列1−277の対応する部分に同一である少なくとも20個の連続するアミノ酸を有するそれらの部分。
- 請求項1に記載の酵素の使用を含んで成る、β-グルコシド結合の加水分解法。
- 請求項2、3または4に記載の酵素の使用を含んで成る、β-キシロシド結合の加水分解法。
- 請求項1ないし5のいずれか1項に記載のタンパク質をコードする核酸配列。
- 配列番号1、3および5の任意の1つの核酸配列の5'-非コード領域に含まれる少なくとも70%のヌクレオチドを含んで成る核酸配列。
- 目的のポリペプチドをコードする核酸配列に機能的に連結された配列番号1、3および5の任意の1つの核酸配列の5'-非コード領域に含まれる、クリソスポリウム(Chrysosporium)に由来する核酸発現−調節領域を含んで成る核酸構築物。
- 請求項8ないし10のいずれか1項に記載の核酸配列を含み、そしてコード核酸配列によりコードされるポリペプチドを発現することができる、組換え微生物株、好ましくは真菌株。
- 請求項10に記載の構築物または請求項11に記載の微生物株を使用するポリペプチドの生産法。
- 配列番号1、3および5のいずれか1つの核酸配列の少なくとも15個の連続するヌクレオチド、またはその相補体を含んで成るオリゴヌクレオチドプローブ。
- プローブが20〜50ヌクレオチド長である、請求項13に記載のオリゴヌクレオチドプローブ。
- プローブが検出可能な標識により標識されている、請求項13または14に記載のオリゴヌクレオチドプローブ。
