Description
本書面において用いられる「ポリヌクレオチド」という用語は、任意の長さのヌクレオチドの重合形態をいい、デオキシリボヌクレオチドまたはリボヌクレオチドでありうる。この用語はもっぱら、分子の一次構造をいう。したがって、この用語は二本鎖状および一本鎖状のDNA、ならびに二本鎖状および一本鎖状のRNAを含む。
以下に、本発明者らが行った、薬物、およびより好ましくはALSの処置用の薬物として用いるのに、HcTeTxおよびそのコード配列が効果的であることを実証する試験を通じて本発明を例証する。
種々の突然変異を有するヒトスーパーオキシドジスムターゼ-1 (SOD-1)の遺伝子を過剰発現するトランスジェニック動物の作出によって、疾患ALSの研究用の動物モデルを得た。これらの動物はALSに罹患するものなどの、臨床的および病理学的特徴を呈する。最も研究され特徴付けられているモデルの一つはSOD1G93Aトランスジェニックマウスであり、これは、SOD-1酵素の遺伝子上の93位にあるグリシンアミノ酸をアラニンと置き換えた突然変異を提示する。このモデル動物は各種の治療用化合物を用いて成功裏に試験されている。しかしこれは、不適切な投与経路によりおよび/または標的細胞に達する治療用分子の生物学的利用能の制限により、ヒト臨床試験では有効な治療法に結び付いていない。遺伝子治療戦略のなかにはアデノ随伴ウイルス(AAV)の使用を含むものがあり、このウイルスは筋肉内注射により運動ニューロンへ逆行的に輸送される。ウイルスベクターを使用する可能性は依然として存在するが、これは、処置される罹患者にさらなる損傷を与える可能性がある。ネイキッドDNAの使用は、特異的な遺伝子治療を患者へ届けるのに、より安全かつ適切な代替戦略である。
1.1 HcTeTxをコードするネイキッドDNA
HcTeTx (破傷風毒素の重鎖のC末端ドメイン−462アミノ酸のSEQ ID NO:2)をコードする遺伝子を、サイトメガロウイルス(CMV)プロモーターの制御下で、真核生物発現プラスミドpcDNA3.1 (Invitrogen)にクローニングした。このベクターを化学的にコンピテントな細菌である大腸菌(Escherichia coli) (DH5a)中で産生させ、Sigma-Aldrich maxiprep GenEluteキットを用いて精製した。
G93A突然変異を有するヒトSOD1を過剰発現するトランスジェニックマウス(B6SJL-TgN[SOD1-G93A]1Gur)は、Jackson Laboratory (Bar Harbor, ME)から入手した。実験の全てにおいてヘミ接合体変異体(非トランスジェニック雌性マウスに交配した変異体雄性マウス)を用いた。Gurneyら(Gurney et al., 1994. Motor neuron degeneration in mice that express a human Cu, Zn superoxide dismutase mutation. Science, 264 (5166): 1772-5)に記述されているように、尾部から抽出したDNAのPCR増幅によってトランスジェニックマウスを特定した。これらの動物をサラゴサ大学の混合研究部(Mixed Research Unit)に保管した。それらに適宜、水と餌を与えた。サラゴサ大学の基準および実験動物の使用に関する国際的指針にしたがって、動物に対して行う全ての実験および動物に施すケアを作り上げた。
8週齢の時点でSOD1G93AトランスジェニックマウスにpCMV-HcTeTx 300 μgを大腿四頭筋(筋肉につき50 μgの注射2回)におよび上腕三頭筋(筋肉につき50 μgの注射1回)に筋肉内注射した。対照群のマウスには同じ量の空のプラスミドを注射した。筋肉内プラスミド注射から10日後に、接種を受けた筋肉を摘出し、これを液体窒素中で予め凍結し、その後、-70℃で保存した。
これらの組織を液体窒素中で凍結し、その後、氷冷乳鉢中で挽いて粉末にした。全筋肉RNAをTRIzol試薬(Invitrogen)のプロトコルにしたがって抽出した。cDNA合成のため、SuperScript (商標) First-Strand Synthesis System (Invitrogen)キットを使用し、終量20 μL中RNA 1 μgから始めた。PCR反応を150 nMの各イニシエータ、150 μMのdNTPs、2 mMのMgCl2、1×緩衝液、0.2 UのTaqポリメラーゼおよびHcTeTx遺伝子断片の増幅のために10倍希釈したcDNA反応液各2 μLにより、終量20 μL中で行った。PCR反応は全てGeneAmp (登録商標) Thermal Cycler 2720 (Applied Biosystems, Foster City, CA, USA)中で行った。熱サイクルパラメータは次の通りであった: 94℃で3分間インキュベーション、ならびに94℃で30秒間、61℃で30秒間および72℃で30秒間を35サイクル。2%臭化エチジウムで染色した寒天ゲル中でHcTeTx遺伝子の増幅の存在を観察した。使用したフォワードおよびリバースプライマー配列は、それぞれ、SEQ ID NO:3およびSEQ ID NO:4であった。単位複製配列のサイズは355 pbであった。
グリッド試験を用いて、筋力およびALS症状の発症を判定した。動物に8週齢から週1回この試験を行った。通常檻の蓋として用いたグリッド上に各マウスを置いた。次にグリッドを180°回転し、軟表面からおよそ60 cmの距離に維持して、いかなる損傷も回避した。各マウスが落下するまでの待ち時間を計った。各マウスに最大180秒間、反転したグリッドにつかまるよう3回試験し、その最長の時間を記録した。
2.1 筋肉におけるプラスミド発現の検出
最初に、構築したpCMV-HcTeTxがSOD1G93Aトランスジェニックマウスの筋細胞においてコード化遺伝子を発現する能力について確認した。これらのマウスではHcTeTx遺伝子の内因性発現の存在は認められないので、該分子のmRNA発現を検出するために、この遺伝子の断片のPCR増幅を注射した筋肉に適用した。図1に示されるように、遺伝子HcTeTxの発現は、空のプラスミドを注射した対照群では認められなかった。しかしながらPCRによって、HcTeTx遺伝子のコード化ベクターを接種した筋肉ではHcTeTx遺伝子の増幅の存在が明らかとなり、ベクターが筋細胞に成功裏に到達したこと、および該遺伝子の転写の過程が行われたことが示された。
HcTeTxをコードするネイキッドDNAによる筋肉内処置は、症状の発症の遅延をもたらし、運動活動を改善し、G93A突然変異およびヒトSOD1遺伝子を含んだ、ALSモデルマウスにおける疾患の終点を引き延ばした。症状の顕在化を記録し、同様に、マウスが3分間、反転したグリッド上にとどまれなかった最初の日を記録した。症状の発症は、対照群と比較した場合、HcTeTxを注射した動物の群においておよそ8日で有意に低下した(図2および表1)。図3および表1において見られるように、最大の生存期間はHcTeTx処置群のマウスにおいて検出され、平均136日に達し、対照群よりも16日長かった。12週目と13週目の間に対照群ではローターロッド上での活動の発現で有意な減少が見られたが、これらの不足は処置動物の群では16週目まで観察されなかった(図4)。
材料および方法
1.1 HcTeTxをコードするネイキッドDNA
HcTeTxをコードする遺伝子(破傷風毒素の重鎖のC末端ドメイン−SEQ ID NO:1)を、サイトメガロウイルス(CMV)プロモーターの制御下で、真核生物発現プラスミドpcDNA3.1 (Invitrogen)にクローニングした。このベクターを化学的にコンピテントな細菌である大腸菌(DH5a)中で産生させ、Sigma-Aldrich Genelute maxiprepキットを用いて精製した。
G93A突然変異を有するヒトSOD1を過剰発現するトランスジェニックマウス(B6SJL-TgN[SOD1-G93A]1Gur)は、Jackson Laboratory (Bar Harbor, ME)から入手した。実験の全てにおいてヘミ接合体変異体(非トランスジェニック雌性マウスに交配した変異体雄性マウス)を用いた。Gurneyら(Gurney et al., 1994. Motor neuron degeneration in mice that express a human Cu, Zn superoxide dismutase mutation. Science, 264 (5166): 1772-5)に記述されているように、尾部から抽出したDNAのPCR増幅によってトランスジェニックマウスを特定した。これらの動物をサラゴサ大学の混合研究部に保管した。それらに適宜、水と餌を与えた。サラゴサ大学の基準および実験動物の使用に関する国際的指針にしたがって、動物に対して行う全ての実験および動物に施すケアを作り上げた。動物計12匹を用いた: 野生型(n=5)、pcDNA3.1を注射したSOD1G93Aマウス(対照、n=5)およびHcTeTxで処置したSOD1G93Aマウス(n=5)。
8週齢の時点でSOD1G93AトランスジェニックマウスにpCMV-HcTeTx 300 μgを大腿四頭筋(筋肉につき50 μgの注射2回)におよび上腕三頭筋(筋肉につき50 μgの注射1回)に筋肉内注射した。対照群のマウスには同じ量の空のプラスミドを注射した。
脊髄由来の全RNAをRNeasy (登録商標) Lipid Tissue Miniキット(Qiagen)のプロトコルにしたがって抽出した。cDNA合成のため、SuperScript (商標) First-Strand Synthesis System (Invitrogen)キットを使用し、終量20 μL中RNA2 μgから始めた。
リアルタイムのPCR反応を終量10 μLにて、1×TaqMan (登録商標) Universal PCR Master Mix, No AmpErase (登録商標) UNG (Applied Biosystems)、1×非標識プライマーミックスおよび分析を行う各遺伝子用のTaqMan (登録商標) MGB (Applied Biosystems)プローブならびに10倍希釈したcDNA反応液各1 μLにより行った。標準化のため、3種の内因性遺伝子(18s rRNA、GAPDHおよびβ-アクチン)を用いた。調べる各遺伝子を増幅するために用いたプローブおよびプライマーの混合物を以下に言及した: カスパーゼ(caspasa)-3 (Mm00438023_m1)、カスパーゼ-1 (Mm00438023_m1)、NCS-1 (Mm00490552_m1)、Rrad (Mm00451053_m1)、18s rRNA (Hs99999901)、GAPDH (4352932E)およびβ-アクチン(4352933E)。ABI Prism 7000 Sequence Detection Systemサーマルサイクラ(Applied Biosystems)中で全てのPCR反応を行った。熱サイクルパラメータは次の通りであった: 95℃で10分間インキュベーション、ならびに95℃で15秒間および60℃で1秒間を40サイクル。カスパーゼ-3、カスパーゼ-1、NCS-1およびRradに対する発現を、3種の内因性遺伝子の幾何平均を適用して標準化した。
野生型マウスの脊髄およびHcTeTxで処置したSOD1G93Aマウスのサンプルを、以下の組成を有する抽出用緩衝液により液体窒素中でホモジナイズした: 150 mM NaCl、50 mM Tris-HCl pH=7.5、1%デオキシコール酸(desoxicolate)、0.1% SDS、1% Triton X-100、1 mM NaOVa、1 mM PMSF、10 μg/mLロイペプチンおよびアプロチニンならびに1 μg/mLペプスタチン。これを4℃にて10分間3000 gで遠心分離した。BCA (9643 Sigma)法を用いて各サンプル由来の上清のタンパク質濃度を定量化した後に、タンパク質25 μgを10%アクリルアミドゲルにロードした。PVDF膜を転写過程に使用し、これを5%スキムミルクのTTBS溶液(20 mM Tris塩基、0.15 M NaCl、pH=7.5、0.1% Tween)で1時間ブロッキングした。その後、それらを終夜4℃で一次抗体(抗p-Akt (sc-7985R, Santa Cruz))とともにインキュベートした。GAPDH (グリセロアルデヒド3リン酸デヒドロゲナーゼ)を用いて、Aktで得た媒質を標準化した(抗GAPDH (sc-25778, Santa Cruz))。一次抗体とともにインキュベートした後に、膜をTTBSで洗浄し、それらを室温で1時間、二次抗体とともにインキュベートした。最後に、それらを化学発光(Western Blotting Luminol Reagent, sc-2048 Santa Cruz)によって発色させた。AlphaEase FC (Bonsai Technologies)ソフトウェアを用いてフィルムをスキャンし分析した。ANOVA検定およびStudent-Neuman-Keuls検定を用いて統計解析を行った。
本研究では、運動ニューロンの変性が存在している、SOD1G93A ALS疾患モデルマウスにおけるHcTeTxの適用から得た結果を示してある。発症期におけるこれらのマウスの脊髄の転写研究を図7に示す。野生型マウスおよびSOD1G93Aマウス脊髄での遅発性発症期(110日齢)におけるアポトーシスに関わるカスパーゼ-1、カスパーゼ-3、BaxおよびBcl2遺伝子の転写制御を比較した。これらの結果から、野生型と比較した場合、SOD1G93A対照マウスではカスパーゼ-1 (P<0.05)、カスパーゼ-3 (P<0.05)およびBcl2 (P<0.01)遺伝子の有意な誘導が示されたが、Bax (P>0.05)遺伝子のプロファイルに有意差は認められなかった(図7)。HcTeTx処置を受けたマウスの群において、カスパーゼ-1およびカスパーゼ-3発現のレベルは野生型で維持され、有意差は未処置マウスと比較した場合でしか認められなかった(それぞれP<0.05およびP<0.01)。しかしながら、これらのトランスジェニックマウスの脊髄において、BaxおよびBcl2遺伝子の発現は、HcTeTx処置(P>0.05)により影響されなかった(図7)。
材料および方法
1.1 破傷風毒素の重鎖のC末端ドメイン(HcTeTx)を含むポリペプチドの抽出
使用したポリペプチド(HcTeTxと呼ぶ)は、破傷風毒素の重鎖のC末端ドメインに相当し、SEQ ID NO:2の451アミノ酸の配列(SEQ ID NO:1)を含み、Gilら(Gil et al., 2003. Biochem J. 373,613-620)によって記述されているプロトコルにしたがって得られた。
G93A突然変異を有するヒトSOD1を過剰発現するトランスジェニックマウス(B6SJL-TgN[SOD1-G93A]1Gur)は、Jackson Laboratory (Bar Harbor, ME)から入手した。実験の全てにおいてヘミ接合体変異体(非トランスジェニック雌性マウスに交配した変異体雄性マウス)を用いた。Gurneyら(Gurney et al., 1994. Science, 264 (5166): 1772-5)に記述されているように、尾部から抽出したDNAのPCR増幅によってトランスジェニックマウスを特定した。これらの動物をサラゴサ大学の混合研究部(Mixed Research Unit)に保管した。それらに適宜、水と餌を与えた。サラゴサ大学の基準および実験動物の使用に関する国際的指針にしたがって、動物に対して行う全ての実験および動物に施すケアを作り上げた。
12週齢の時点で、SOD1G93Aトランスジェニックマウスに、破傷風毒素のC末端ドメイン(HcTeTx)を含むポリペプチドの濃度0.5 μMで250 μlを腹腔内注射した。この注射をその全生涯にわたって毎週繰り返した。
マウスの生涯の終点は、動物を仰臥位に置き、自らを回転させられなくなった時点とみなした。
1.1.- HcTeTxはSOD1G93Aトランスジェニックマウスの生存期間を延長した
図10および表2において見られるように、最大の生存期間はHcTeTx処置群のマウスにおいて検出され、平均135日に達し、対照群よりも9日長かった。
筋萎縮性側索硬化症(ALS)に関連した異常な細胞内カルシウム恒常性の証拠がある。ニューロンタンパク質NCS1は、神経分泌をカルシウム依存的に制御することが示されており(McFerran et al., 1998. J. Biol. Chem. 273: 22768-22772)、ニューロンのシグナル伝達に関与するカルシウム/カルモジュリン依存的な酵素の調節とも関連付けられている(Schaad et al., 1996. PNAS. 93: 9253-9258)。SOD1G93Aマウスの脊髄におけるNCS1の発現をHcTeTxによる処置後に50回試験した。RT-PCR実験において、NCS1遺伝子の発現は同齢の野生型マウスに関して遅発性総体症状を有するトランスジェニックマウスで抑制されることが分かった(P<0.05)。さらに、筋肉内HcTeTx処置を受けたマウスは、いっそう高いレベルのNCS1を有し(P<0.05)、野生型マウスのものに近づいた。同じサンプルを用い、伝令RNAのレベルをRas関連遺伝子において分析し、糖尿病遺伝子(Rrad)と関連付けた。本実施例において、Rradレベルは、同程度の年齢の野生型マウスと比較した場合、対照トランスジェニックマウスの脊髄においてほぼ2倍に増えた。しかしながら、対照と比べて、SOD1G93AマウスでのHcTeTxによる処置は、Rradの発現を有意に低減し(P<0.05)、野生型マウスで得られたものと同様の値を達成した(図11)。
Claims (10)
- 破傷風毒素の重鎖サブユニットのカルボキシル末端ドメイン(HcTeTx)のコード配列、その対立遺伝子変種またはそれらの機能的断片を含む単離ポリヌクレオチドの、薬物の作出のための使用。
- HcTeTxがSEQ ID NO:1によってコードされる、前記の請求項記載の使用。
- 破傷風毒素の重鎖サブユニットのカルボキシル末端ドメインのコード配列断片がSEQ ID NO:6を含む、請求項1記載の使用。
- 前記請求項のいずれか一項記載のポリヌクレオチドのいずれかを含むベクターの、薬物の作出のための使用。
- 請求項1〜3のいずれか一項記載のポリヌクレオチドのいずれかを含むトランスジェニック細胞の、薬物の作出のための使用。
- 薬物が筋萎縮性側索硬化症(ALS)の処置のためである、前記請求項のいずれか一項記載の使用。
- 破傷風毒素の重鎖サブユニットのカルボキシル末端ドメイン(HcTeTx)、その対立遺伝子変種またはそれらの機能的断片を含む単離ポリヌクレオチドの、ALS処置用の薬物の作出のための使用。
- HcTeTxがSEQ ID NO:2を含む、前記請求項記載の使用。
- HcTeTxがSEQ ID NO:5を含む、前記請求項記載の使用。
- 薬物が経口経路、非経口経路、筋肉内経路または鼻腔経路を通じて投与されるようにデザインされる、前記請求項のいずれか一項記載の使用。
