試験例1:ヒトFXR受容体に対する転写活性化試験
10%脱脂牛血清を含むダルベッコ変法イ-グル培地(FCS/DMEM)にて培養したヒト胎児腎細胞(HEK293)に、酵母の転写因子(GAL4)のDNA結合領域とヒト型FXRのリガンド結合領域との融合蛋白質を発現する受容体プラスミド及びそのレポータープラスミド、さらに内部標準用のβガラクトシダーゼプラスミドをリン酸カルシウム法にて無血清状態にてコトランスフェクションした。その後、被検化合物を添加して16時間後にルシフェラ-ゼ活性及びβガラクトシダーゼ活性を測定し、内部標準により補正してFXR転写活性化活性を求めた。
また、合成FXRリガンドであるGW−4064(30nM)共存下での転写活性の抑制度を求めることでFXR転写抑制活性を求めた。
結果を表1に示す。本発明の化合物はヒトFXRに対して天然のFXRアゴニストであるケノデオキシコール酸以上のEC50値を示し、ヒトFXR受容体に対して強力な転写活性化作用を有することが示された。また、別の本発明化合物(実施例15の化合物)はヒトFXRに対して強い転写抑制作用を有することが示された。
Test Example 1: Transcriptional activation test for human FXR receptor
Human fetal kidney cells (HEK293) cultured in Dulbecco's modified Eagle's medium (FCS / DMEM) containing 10% defatted bovine serum, DNA binding region of yeast transcription factor (GAL4) and ligand binding of human FXR A receptor plasmid expressing the fusion protein with the region, its reporter plasmid, and β-galactosidase plasmid for internal standard were co-transfected in the serum-free state by the calcium phosphate method. Thereafter, the luciferase activity and β-galactosidase activity were measured 16 hours after the addition of the test compound, and the FXR transcription activation activity was determined by correcting with an internal standard.
Moreover, the FXR transcription inhibitory activity was calculated | required by calculating | requiring the suppression degree of the transcriptional activity in GW-4064 (30nM) coexistence which is a synthetic FXR ligand.
The results are shown in Table 1. The compound of the present invention showed an EC 50 value higher than that of chenodeoxycholic acid, which is a natural FXR agonist, for human FXR, and was shown to have a strong transcriptional activation effect on human FXR receptor. Moreover, it was shown that another this invention compound (compound of Example 15) has a strong transcriptional inhibitory effect with respect to human FXR.
