実施例2
本実施例は、実施例1で得られた2種類のセンサーチップに対し、カルボン酸のpKaよりも高く、タンパク質の等電点(pI)よりも低い溶液pHにおける、タンパク質のプレコンセントレーションおよび固定化に関するものである。
タンパク質としては、ニュートラルアビジン(PIERCE製)を用いた。ニュートラルアビジンの等電点(pI)は、ATTO社のAE-8150を用いた電気泳動実験で、同時測定したマーカー(Broad pI Kit, pH 3.5-9.3:Amersham Biosciences社製)との比較から、pI=6.0程度であることを確認した。

カルボキル基のみが結合している試料1、およびその一部がスルホン酸基に置換されている試料2は、カルボン酸のpKa以上かつニュートラルアビジンの等電点以下であるpH5.0における固定量はそれぞれ13000RUと10500RU程度であり、このpHでは、ほぼ同等のプレコンセントレーションおよび固定効果があることが証明された。
実施例3
本実施例は、実施例1で得られた2種類のセンサーチップに対し、カルボン酸のpKaおよびニュートラルアビジンのpIよりも低い溶液pHにおける、ニュートラルアビジンのプレコンセントレーションおよび固定化に関するものである。
酢酸バッファー(ビアコア社製、pH5.0)をグリシンバッファー(ビアコア社製、pH1.5)に変更した以外は実施例2と同一の操作を行なった。得られたセンサーグラムを図2に示す。
