It relates to eye drops that can be used.
Pranoprofen is useful for the symptomatic treatment of inflammatory diseases of the external eye and anterior eye such as blepharitis, conjunctivitis, keratitis, scleritis, suprasclitis, anterior uveitis, postoperative inflammation, etc. A non-steroidal anti-inflammatory agent. However, pranoprofen is unstable in an aqueous solution, and there is a problem that it is difficult to stably maintain an aqueous preparation such as an eye drop containing pranoprofen over a long period of time (Patent Document 1, Patent Document 2, (See Patent Document 3 and Patent Document 4). In particular, when a nonionic surfactant coexists, the stability of pranoprofen is significantly reduced. As a method for improving the stability of an aqueous liquid preparation containing pranoprofen, in the presence of a nonionic surfactant, alkylphenols such as dibutylhydroxytoluene and butylhydroxyanisole, L-ascorbic acid 2- [3, 4-dihydro-2,5,7,8-tetramethyl-2- (4,8,12-trimethyltridecyl) -2H-1-benzopyran-6-yl-hydrogen phosphate] and the like, methionine, Techniques containing an amino acid such as tryptophan and histidine, an antioxidant such as sodium thiosulfate (see Patent Document 1), an organic amine such as tromethamine or 4- (2-hydroxyethyl) -1- (2-sulfoethyl) piperazine Technology for combined stable and less irritating pranoprofen ophthalmic solution ( Patent Document 2), technology related to stable eye drops with low irritation (see Patent Document 3), a heterocyclic base containing a propylene glycol or a nonionic surfactant having an HLB value of 14 to 18 in pranoprofen. The technique (refer patent document 4) etc. regarding aqueous liquid preparations, such as eye drops which stabilized arylcarboxylic acid, such as a planoprofen, by mix | blending are disclosed. However, these disclosed technologies have a problem that additives that are undesirable from the viewpoint of safety must be used.
On the other hand, flavin adenine dinucleotide sodium, cyanocobalamin, pyridoxine hydrochloride, panthenol, calcium pantothenate, sodium pantothenate and the like are known as water-soluble vitamins generally incorporated in eye drops, and these components are ophthalmic drugs. It is listed in the manufacturing (import) approval standard (see Non-Patent Document 1). Of these, cyanocobalamin is known as a component with anti-anemic action, and it promotes the regeneration of the corneal epithelium and activates the optic nerve function for the eyes, thus improving the fine movement control in regulatory eye strain Tetrahydrozoline hydrochloride, a decongestant component, neostigmine methylsulfate, an antihistamine component, chlorpheniramine maleate, an antihistamine component, pyridoxine hydrochloride, a normal factor component that promotes mucosal tissue metabolism, No. 5 is commercially available (see Non-Patent Document 2). Pyridoxine hydrochloride is a component known as a prophylactic and therapeutic agent for glossitis, cheilitis, eczema, peripheral neuritis, etc., and is used for eye drops for the purpose of promoting the metabolism of the cornea. Commercially available eye drops with neostigmine methylsulfate, a regulatory component, allantoin with tissue repair activity, tetrahydrozoline hydrochloride, a decongestant component, chlorpheniramine maleate, an antihistamine component, and sodium chodoitine sulfate with corneal protection (See Non-Patent Document 3). These water-soluble vitamins significantly decrease stability when a compound having a polyoxyethylene chain, such as polyethylene glycol, coexists, but can be stabilized by adding alcohols thereto (non-patent) Reference 4 and Non-Patent Document 5). In addition, there is a technique relating to eye drops that stabilize flavin adenine dinucleotide sodium or cyanocobalamin by blending polyhydric alcohol with flavin adenine dinucleotide sodium or cyanocobalamin in the presence of a nonionic surfactant having a polyoxyethylene chain. It is disclosed (see Patent Document 5).

[Test example]
The eye drop preparations of Examples 1 to 15 and Comparative Examples 1 to 15 were subjected to a storage stability test for 6 months under conditions of 40 ° C. and a relative humidity of 75%. The stability test items were 1) quantification of pranoprofen, 2) quantification of cyanocobalamin, and 3) quantification of pyridoxine hydrochloride, and the results are shown in Table 1, Table 2 and Table 3, respectively. Each component was quantified by measuring the content of each component by reverse phase partition high performance liquid chromatography, and then calculating the residual rate by the following formula.
Residual rate (%) = Ws / Wo × 100
Ws; component content after storage Wo; component content immediately after production
Further, the stability was evaluated with ◎ when the residual ratio of each component was 95% or more, ◯ when 85% or more and less than 95%, △ when 40% or more and less than 85%, and × when less than 40%.
The stability of the eye drop is preferably ◯, particularly preferably ◎. Δ and × cannot stabilize each component, and are not suitable as the stability of eye drops. In particular, “x” is not suitable because of extremely poor stability.
