Abstract
【解決手段】 配列表に示される遺伝子は、間葉系幹細胞で特異的に発現する。該遺伝子を間葉系幹細胞検出用のマーカーとして用いることよりなる。更に、本発明は、該間葉系幹細胞マーカー遺伝子検出用プローブ、及び該間葉系幹細胞遺伝子マーカーの検出に際して、被検細胞の該遺伝子を増幅するためのPCR用プライマーを含み、また、本発明の間葉系幹細胞マーカー遺伝子が発現するポリペプチドからなる間葉系幹細胞検出用ポリペプチドマーカー、該ポリペプチドマーカーを検出するための、該ポリペプチドマーカーに特異的に結合する抗体、更には、間葉系幹細胞マーカー遺伝子検出用プローブ、ポリペプチドマーカーに特異的に結合する抗体を用いた間葉系幹細胞の識別、分離方法を含む。

(細胞)
ヒト由来間葉系幹細胞(human mesenchymal stem cell)、およびヒト繊維芽細胞(human fibroblast)はDalbecco's modified Eagle's medium(low glucose)(Sigma社製)に10% fetal calf serum、1ng/mlのbFGFを加えた培地を用いて37℃にて培養した。
10cmディッシュ1枚の各細胞からTRIZOL reagent(Invitrogen社製)を用いて全RNAを抽出後、micro poly (A) purist(Ambion社製)を用いてmRNAを精製し、DNA microarray(Incyte Genomics社:Kurabo Life Array analysis servis;Lot. No. KL01081)を行った。なお、DNAマイクロアレイ解析は、Kurabo社に委託した。
各細胞からTRIZOL reagent(Invitrogen社製)を用いて全RNAを抽出後、各全RNA1μgを鋳型として、SuperScript first-strand synthesis system for RT-PCR(Invitrogen社製)を用いて、42℃、50分間の逆転写反応を行ってcDNAを合成した。合成されたcDNAを鋳型としてAdvantage 2 PCR enzyme system(Clontech社製)を用いて各遺伝子を増幅した。94℃30秒間で変性、68℃1分間でアニールを30サイクル行った。増幅に用いた各遺伝子のプライマー配列を表1にまとめた。増幅した遺伝子を1%アガロースゲルを用いた電気泳動にて分離し、エチジウムブロマイド染色によって検出した。
(DNAマイクロアレイ)
DNAマイクロアレイの結果、9400遺伝子中、63遺伝子が繊維芽細胞と比較して間葉系幹細胞において有意に発現が高く、141遺伝子が低いことが明らかになった。また、それ以外の遺伝子では有意な差は認められなかった。このうち、3倍以上の差が認められた87遺伝子中、間葉系幹細胞の方が発現が高かった遺伝子が29遺伝子、低かった遺伝子が58遺伝子であった。
3個体由来の間葉系幹細胞と3個体由来の繊維芽細胞から全RNAを抽出し、これらを鋳型に用いて、上記87遺伝子について半定量的RT−PCR法によりその発現量を調べた。その結果、発現量に差が見られない遺伝子がほとんどであった。また、異なる個体の間葉系幹細胞、あるいは繊維芽細胞の間でも発現量に差があり、個体差によって発現が異なると考えられるものも多くあった(図1)。
実施例1と同様にして、本発明の遺伝子マーカーのヒト由来間葉系幹細胞及びヒト繊維芽細胞における発現を測定した。用いたRT−PCR及びリアルタイムPCRのプライマーとリアルタイムPCR用のプローブを表2に示す。
実施例1と同様にして、本発明の各遺伝子マーカーのヒト由来間葉系幹細胞及びヒト繊維芽細胞における発現を測定した。用いたRT−PCRのセンス及びアンチセンスプライマーは、配列表の配列番号71〜116に示す。間葉系幹細胞とヒト繊維芽細胞とにおける遺伝子の発現の状況を図9及び図10に示す。また、間葉系幹細胞/繊維芽細胞の発現の割合を、表4に示す。
実施例1と同様にして、本発明の各遺伝子マーカーのヒト由来間葉系幹細胞及びヒト繊維芽細胞における発現を測定した。間葉系幹細胞/繊維芽細胞の発現の割合を、表5に示す。

実施例1と同様にRT−PCRを用い、間葉系幹細胞の分化誘導における各遺伝子の発現の変化を測定した。間葉系幹細胞の骨芽細胞への分化誘導における各遺伝子の発現の状況を図12に示す。各マーカー遺伝子について、骨分化誘導4日〜28日での遺伝子発現の変化を図13〜16に示す。

- 請求項1〜9のいずれか記載の塩基配列のアンチセンス鎖の全部又は一部からなる請求項10記載の間葉系幹細胞マーカー遺伝子検出用プローブ。
- 請求項10又は11記載のDNAの少なくとも1つ以上を固定化させたことを特徴とする間葉系幹細胞マーカー遺伝子検出用マイクロアレイ又はDNAチップ。
- 配列表の配列番号1、2、4、6、7、9、10、11、12、14、15、17、19、又は46に示される塩基配列を有する遺伝子のグループ、及び請求項2〜9のいずれか記載の遺伝子のグループの2以上の遺伝子を検出するプローブを固定化させたことを特徴とする請求項12記載の間葉系幹細胞マーカー遺伝子検出用マイクロアレイ又はDNAチップ。
- 配列表の配列番号3、5、8、13、16又は18に示されるアミノ酸配列を有するポリペプチドであることを特徴とする間葉系幹細胞検出用ポリペプチドマーカー。
- 請求項14記載のポリペプチドを用いて誘導され、該ポリペプチドに特異的に結合することを特徴とする抗体。
- 抗体が、モノクローナル抗体であることを特徴とする請求項15記載の抗体。
- 抗体が、ポリクローナル抗体であることを特徴とする請求項15記載の抗体。
- 被検細胞における請求項1〜9のいずれか記載の間葉系幹細胞マーカー遺伝子の発現を検出することを特徴とする間葉系幹細胞の識別方法。
- 間葉系幹細胞マーカー遺伝子の発現の検出を、ノーザンブロッティング法を用いて行うことを特徴とする請求項18記載の間葉系幹細胞の識別方法。
- 間葉系幹細胞マーカー遺伝子の発現の検出を、請求項10又は11記載の間葉系幹細胞マーカー遺伝子検出用プローブを用いて行うことを特徴とする請求項18記載の間葉系幹細胞の識別方法。
- 間葉系幹細胞マーカー遺伝子の発現の検出を、請求項12又は13記載の間葉系幹細胞マーカー遺伝子検出用マイクロアレイ又はDNAチップを用いて行うことを特徴とする請求項18記載の間葉系幹細胞の識別方法。
- 請求項18〜21のいずれか記載の間葉系幹細胞マーカー遺伝子の発現の検出が、定量的又は半定量的PCRの使用を含んでいることを特徴とする間葉系幹細胞の識別方法。
- 定量的又は半定量的PCRの使用が、RT−PCR法であることを特徴とする請求項22記載の間葉系幹細胞の識別方法。

- 被検細胞における遺伝子を、請求項24〜26のいずれか記載のセンスプライマー及びアンチセンスプライマーの少なくとも1対以上のプライマーを用いて増幅し、増幅した遺伝子を請求項10又は11記載の間葉系幹細胞マーカー遺伝子検出用プローブを用いて検出することを特徴とする請求項18記載の間葉系幹細胞の識別方法。
- 請求項15〜17のいずれか記載の抗体を用いて、被検細胞における間葉系幹細胞マーカーポリペプチドの発現を検出することを特徴とする間葉系幹細胞の識別方法。
- 被検細胞における、配列表の配列番号1、11及び19からなる間葉系幹細胞マーカー遺伝子のグループの1遺伝子以上の発現と、配列表の配列番号2、4、6、7、9、10、12、14、15、及び17からなる間葉系幹細胞マーカー遺伝子のグループの1遺伝子以上の発現とを組み合わせて、評価することを特徴とする間葉系幹細胞の識別方法。
- 請求項10又は11記載の間葉系幹細胞マーカー遺伝子検出用プローブ及び/又は請求項15〜17のいずれか記載の抗体を用いて、被検細胞における間葉系幹細胞検出用マーカー遺伝子及び/又は間葉系幹細胞検出用マーカーポリペプチドの発現を検出し、識別した間葉系幹細胞を分離することを特徴とする間葉系幹細胞の識別分離方法。
- 請求項15〜17のいずれか記載の抗体を用いた蛍光抗体法により被検細胞中における間葉系幹細胞を標識化し、該標識化した間葉系幹細胞を分離することを特徴とする請求項30記載の間葉系幹細胞の識別分離方法。
- 請求項10又は11記載の間葉系幹細胞マーカー遺伝子検出用プローブ、該プローブを固定した請求項12又は13記載の間葉系幹細胞マーカー遺伝子検出用マイクロアレイ又はDNAチップ、及び請求項15〜17のいずれか記載の抗体の少なくても一つ以上を装備してなる間葉系幹細胞の識別用キット。
