本発明は、アレルギー性疾患の検査方法に関する。

【従来の技術】
アレルギー性疾患は、多因子性の病気(multifactorial diseases)と考えられている。つまりアレルギー性疾患は多くの異なる遺伝子の発現の相互作用によって起こり、これらの個々の遺伝子の発現は、複数の環境要因によって影響を受ける。このため、アレルギー性疾患を起こす特定の遺伝子を解明することは、非常に困難であった。
たとえば、気管支喘息は、気道における慢性の炎症性疾患として位置付けられている。そして気管支喘息の病態形成には、気道粘膜や気管支平滑筋におけるアレルギー反応の密接な関与が指摘されている。したがって気管支喘息の診断においては、これらの組織や全身性のアレルギー反応の状態を把握することが重要な課題である。また気管支喘息の治療においては、アレルギー反応の制御が課題となる。

一方、アレルギー性疾患には、変異や欠損を有する遺伝子の発現や、特定の遺伝子の過剰発現や発現量の減少が関わっていると考えられている。病気に関して遺伝子発現が果たしている役割を解明するためには、遺伝子が発症にどのように関わり、薬剤などの外的な刺激が遺伝子発現をどのように変化させるのかを理解する必要がある。

さて、気管支喘息の患者の多くに、IgE抗体の産生亢進を伴うアトピー素因が見られる。気管支喘息には、多様な原因が考えられているが、アトピー素因が多くの患者において過敏症の原因となっていることは疑いを入れない。喘息発作の気道閉塞の機序には、気管支平滑筋の収縮、あるいは気道粘膜の浮腫や気道内分泌亢進が予想されている。このような気道の変化には、病因アレルゲンへの曝露によるI型アレルギー反応が重要な役割を果たしている。

さて、現在アレルギー性疾患の診断においては、一般に、問診、家族歴、そして本人の既往症の確認が重要な要素となっている。またアレルギーをより客観的な情報に基づいて診断するために、血液を試料とする試験方法や、アレルゲンに対する患者の免疫学的な応答を観察する方法も実施されている。前者の例として、アレルゲン特異的IgE測定、白血球ヒスタミン遊離試験、あるいはリンパ球幼若化試験等が挙げられる。アレルゲン特異的IgEの存在は、そのアレルゲンに対するアレルギー反応の証明である。しかし患者によっては、必ずしもアレルゲン特異的なIgEを検出できるとは限らない場合もある。また、その測定原理上、診断に必要なアレルゲンの全てに対して、試験を実施しなければならない。白血球ヒスタミン遊離試験やリンパ球幼若化試験は、免疫システムのアレルゲンに対する反応をin vitroで観察する方法である。これらの方法は、操作が煩雑である。
一方、患者を実際にアレルゲンに接触させたときに観察される免疫応答をアレルギーの診断に役立てる方法(後者)も公知である。プリック・テスト、スクラッチ・テスト、パッチ・テスト、皮内反応、あるいは誘発試験等が、この種の試験に含まれる。これらの試験では、患者のアレルギー反応を直接診断することができる反面、実際に被検者をアレルゲンに曝露する侵襲性の高い検査であると言うことができる。

この他、アレルゲンに関わらず、アレルギー反応の関与を証明するための試験方法も試みられている。たとえば、血清IgE値が高値である場合、その患者にはアレルギー反応が起きていると推定することができる。血清IgE値は、アレルゲン特異IgEの総量に相当する情報である。アレルゲンの種類に関わらずIgEの総量を決定することは容易であるが、非アトピー型気管支炎等の疾患を持つ患者では、IgEが低値となる場合がある。

したがって、患者に対する危険が少なく、しかも診断に必要な情報を容易に得ることができる、アレルギー性疾患のマーカーが提供されれば有用である。このようなマーカーは、アレルギー性疾患の発症に深く関与していると考えられるので、診断のみならず、アレルギー症状のコントロールにおいても、重要な標的となる可能性がある。

【発明が解決しようとする課題】
本発明は、アレルギー性疾患の検査を可能とする指標の提供を課題とする。さらに、本発明は該指標に基づく、アレルギー性疾患の検査方法および治療薬候補化合物のスクリーニング方法を提供することを課題とする。

【課題を解決するための手段】
IL−4とIL−13がアレルギー反応に深く関与していることは、いくつかの報告により示唆されている。たとえばIL−4(Yssel, H and Groux,H: Int. Arch. Allergy Immunol., 121; 10−18, 2000)や、STAT6(Akimoto, T. et al.: J.Exp. Med., 187, 1537−1542, 1998)をノックアウトしたマウスでは、気道過敏性が消失する。モデルマウスにおいては、IL−13がIgE産生やTh2型に関係なく喘息様病態の形成に関与している(Wills−Karp,M. et al.: Science, 282, 2258−2261, 1998; Grunig, G. et al.: Science, 282,2261−2263, 1998; Zhu, Z. et al.: J. Clin. Invest., 103, 779−788, 1999)。

またヒト気道上皮細胞、気管支平滑筋に、IL−4受容体及びIL−13受容体が高発現している(Heinzmann, A. et al.: Hum. Mol. Genet.,9 : 549−559, 2000)。このことから、これらの組織はIL−4及びIL−13の標的細胞と思われる。一方、IL−4受容体α及びIL−13に存在するSNPが、アレルギー疾患の遺伝的要因の1つであることが示された(Mitsuyasu,H., et al.: Nature Genet., 19, 119−120, 1998; Mitsuyasu, H., et al.: J.Immunol., 162: 1227−1231, 1999; Kruse, S., et al.: Immnol., 96, 365−371, 1999;Heinzmann, A. et al.: Hum. Mol. Genet., 9 : 549−559, 2000)。更に、可溶型IL−4受容体αによりIL−4あるいはIL−13の作用を阻害することが気管支喘息の治療として有効であることも示された(Borish,L. C. et al.: Am. J. Respir. Crit. Care Med., 160: 912−922, 1999)。

以上のように、IL−4とIL−13には、特に呼吸器症状を中心とするアレルギー反応との深い関係が示唆されている。つまりIL−4及びIL−13によるシグナル伝達経路を構成する遺伝子は、アレルギー反応に深い関連性を有する遺伝子と言える。したがって、これらの遺伝子を単離し、アレルギー反応との関連性を明らかにすることにより、アレルギー性を含む気管支喘息のような疾患の治療の新たな標的を見出すことができる。

本発明者らは、このような考えかたに基づいて、ヒト気管支上皮細胞をIL−4及びIL−13で処理したときに、発現レベルに変化を示す遺伝子を探索すれば、アレルギー反応に関連する遺伝子を単離することができるのではないかと考えた。同様のアプローチにより、IL−4やIL−13の処理によって発現レベルが変化する遺伝子の単離を試みた報告もある(Wang et al., Immunology 2000, Seattle, May 12−16, 2000)。しかし公知の探索方法においては、解析に用いた細胞のロット数が少ない上、発現レベルの変化の幅が明らかでないので、IL−4やIL−13の刺激に対する特異性が期待できない。

そこで本発明者らは、IL−4やIL−13の刺激に対してより特異的に応答する遺伝子を単離するために、発現レベルの変動が2倍以上に及ぶ遺伝子としてアリル炭化水素受容体(以下、AHRと記載する)遺伝子を選択した。更に、AHR遺伝子のアレルギー症状との関連性を明らかにするために、IL−4、CD40リガンド、あるいは抗IgM抗体で刺激した末梢血B細胞におけるAHR遺伝子の変動を観察した。CD40リガンドとしては抗CD40抗体を用いた。その結果、AHR遺伝子の発現レベルは、これらの処理を施した末梢血B細胞においても有意に上昇することが確認された。

B細胞から抗体が産生されるようになる前に、全てのB細胞は再編成を終えた同一のV遺伝子によりコードされる膜型免疫グロブリン分子IgMをB細胞レセプターとして細胞表面に発現する。イソタイプスイッチを終了したメモリーB細胞は、IgM以外の新たな免疫グロブリンを産生できるようになる。
B細胞表面免疫グロブリン分子の架橋によって細胞内シグナル伝達経路が活性化されることは、良く知られており、in vitroでは、例えば抗IgM抗体を作用させることによりIgMを架橋すると、blk, lck, fyn, lyn等のチロシンキナーゼの活性化が起こる。

また、B細胞の免疫反応で重要なのは、ヘルパーT細胞との相互作用による活性化である。抗原特異的ナイーブB細胞が抗原のみによって活性化されることはない。B細胞の場合、ヘルパーT細胞由来の補助シグナルが必要となる。B細胞表面のCD40は、ヘルパーT細胞表面のCD40リガンドと結合し、また、ヘルパーT細胞の産生するサイトカインによりB細胞は活性化される。In vitroでは、B細胞にCD40リガンドを加えることにより活性化を促すことができる。また、CD40リガンドの効果は、抗CD40抗体を作用させることでも同様に観察可能である。したがって、IL−4、CD40リガンド、あるいは抗IgM抗体などで刺激されたB細胞において発現レベルが変動する遺伝子は、B細胞の活性化と関連する遺伝子であると考えることができる。

以上の知見に基づいて本発明者らは、AHR遺伝子、並びにAHR遺伝子によってコードされる蛋白質を指標とすることによって、アレルギー性疾患の検査が可能となることを見出し本発明を完成した。また本発明者らは、AHR遺伝子の発現レベル、あるいはAHR遺伝子によってコードされる蛋白質の活性を指標とすることによって、アレルギー性疾患の治療薬をスクリーニングできることを見出し、本発明を完成した。
すなわち、本発明は、以下の検査方法、並びにスクリーニング方法に関する。
〔1〕次の工程を含む、アレルギー性疾患の検査方法であって、指標遺伝子がアリル炭化水素受容体遺伝子である方法。
a)被検者の生体試料における、指標遺伝子の発現レベルを測定する工程
b)アレルギー性疾患ではない生体の生体試料における指標遺伝子の発現レベルと比較する工程
〔2〕生体試料が血液である〔1〕に記載の検査方法。
〔3〕遺伝子の発現レベルを、cDNAのPCRによって測定する〔1〕に記載の検査方法。
〔4〕遺伝子の発現レベルを、前記遺伝子によってコードされる蛋白質の検出によって測定する〔1〕に記載の検査方法。
〔5〕アリル炭化水素受容体遺伝子の塩基配列を含むポリヌクレオチド、またはその相補鎖に相補的な塩基配列を有する少なくとも15塩基の長さを有するオリゴヌクレオチドからなる、アレルギー性疾患の検査用試薬。
〔6〕アリル炭化水素受容体蛋白質のアミノ酸配列を含むペプチドを認識する抗体からなる、アレルギー性疾患の検査用試薬。
〔7〕次の工程を含む、アレルギー性疾患の治療薬のスクリーニング方法であって、指標遺伝子が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子である方法。
(1)指標遺伝子を発現する細胞に候補化合物を接触させる工程
(2)指標遺伝子の発現レベルを測定する工程、
(3)対照と比較して指標遺伝子の発現レベルを低下させる化合物を選択する工程
〔8〕細胞が株化B細胞である〔7〕に記載の方法。
〔9〕次の工程を含む、アレルギー性疾患の治療薬のスクリーニング方法であって、指標遺伝子が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子である方法。
(1)被験動物に候補化合物を投与する工程、
(2)被験動物の生体試料における指標遺伝子の発現レベルを測定する工程、および
(3)対照と比較して指標遺伝子の発現レベルを低下させる化合物を選択する工程
〔10〕次の工程を含む、アレルギー性疾患の治療薬のスクリーニング方法であって、指標遺伝子が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子である方法。
(1)指標遺伝子の転写調節領域と、この転写調節領域の制御下に発現するレポーター遺伝子を含むベクターを導入した細胞と候補物質を接触させる工程、
(2)前記レポーター遺伝子の活性を測定する工程、および
(3)対照と比較してレポーター遺伝子の発現レベルを低下させる化合物を選択する工程
〔11〕次の工程を含む、アレルギー性疾患の治療薬のスクリーニング方法であって、指標蛋白質が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子によってコードされる蛋白質である方法。
(1)指標蛋白質と候補物質を接触させる工程、
(2)指標蛋白質の活性を測定する工程、および
(3)対照と比較して指標蛋白質の活性を低下させる化合物を選択する工程
〔12〕〔7〕、〔9〕、〔10〕、および〔11〕のいずれかに記載のスクリーニング方法によって得ることができる化合物を有効成分として含有する、アレルギー性疾患の治療薬。
〔13〕指標遺伝子のセンス鎖の塩基配列の連続する少なくとも15塩基の配列に対して相補的な配列を含むアンチセンスDNAを主成分として含む、アレルギー性疾患の治療薬であって、指標遺伝子がアリル炭化水素受容体遺伝子または該遺伝子と機能的に同等な遺伝子である治療薬。
〔14〕アリル炭化水素受容体に結合する抗体を主成分として含む、アレルギー性疾患の治療薬。
〔15〕指標遺伝子のB細胞における発現強度を上昇させたトランスジェニック非ヒト脊椎動物のアレルギー性疾患のモデル動物としての使用であって、指標遺伝子が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子である使用。
〔16〕指標遺伝子の塩基配列を含むポリヌクレオチド、またはその相補鎖に相補的な塩基配列を有する少なくとも15塩基の長さを有するオリゴヌクレオチドと、指標遺伝子を発現する細胞からなる、アレルギー性疾患の治療薬候補化合物をスクリーニングするためのキットであって、指標遺伝子が、アリル炭化水素受容体遺伝子、または該遺伝子と機能的に同等な遺伝子であるキット。

【発明の実施の形態】
本発明において、アレルギー性疾患(allergic disease)とはアレルギー反応の関与する疾患の総称である。より具体的には、アレルゲンが同定され、アレルゲンへの曝露と病変の発症に深い結びつきが証明され、その病変に免疫学的な機序が証明されることと定義することができる。ここで、免疫学的な機序とは、アレルゲンの刺激によって白血球細胞が免疫応答を示すことを意味する。アレルゲンとしては、ダニ抗原や花粉抗原等を例示することができる。
代表的なアレルギー性疾患には、気管支喘息、アレルギー性鼻炎、花粉症、あるいは昆虫アレルギー等を示すことができる。アレルギー素因(allergic diathesis)とは、アレルギー性疾患を持つ親から子に伝えられる遺伝的な因子である。家族性に発症するアレルギー性疾患はアトピー性疾患とも呼ばれ、その原因となる遺伝的に伝えられる因子がアトピー素因である。喘息は、アトピー性疾患のうち、特に呼吸器症状を伴う疾患に対して与えられた総称である。

本発明のアレルギー性疾患の検査方法は、被検者の生体試料におけるAHR遺伝子の発現レベルを測定し、アレルギー性疾患ではない生体の測定値と比較する工程を含む。両者の比較の結果、アレルギー性疾患ではない生体よりも発現が亢進している場合には、被検者はアレルギー性疾患患者であると判定される。本発明において、アレルギー性疾患ではない生体とは健常者のほか、健常とは言えないが明らかにアレルギー性疾患を有さないと診断された生体が含まれる。

本発明においてAHR遺伝子とは、本実施例において同定されたヒトAHR遺伝子およびそれと同一の遺伝子座または他生物において相同の遺伝子座にある遺伝子を言う。例えば本発明においてAHR遺伝子は、本実施例において同定されたヒトAHR遺伝子と同じまたは相同の遺伝子座にある任意の対立遺伝子が含まれる。本発明においてAHR遺伝子には、具体的には、本実施例において同定されたヒトAHR遺伝子、その多型(SNPを含む)、変異体、スプライシングバリアント、および他生物のホモログを含む。これらの遺伝子は、ヒトAHR遺伝子と実質的に同一または類似の発現制御を受けていると考えられることから、それらの発現を検出することにより、本発明のアレルギー性疾患の検査を行うことが可能である。

具体的には、本発明においてAHR遺伝子は、以下の核酸からなる内在性遺伝子と実質的に同一の遺伝子である。内在性遺伝子とは、自然界の生物が元来保持している、人の手により改変されていない遺伝子を言い、それと同一の塩基配列からなる遺伝子は、内在性遺伝子と実質的に同一の遺伝子である。また「遺伝子」とは、ここでは転写産物またはそれをコードする核酸(DNAまたはRNAなど)を言う。
(a)AHR遺伝子の塩基配列(GenBank Acc.# NM_001621)から選択された塩基配列を含む核酸。
(b)AHR遺伝子のコード配列の塩基配列において、1または複数の塩基が置換、欠失、および/または挿入した塩基配列を含む核酸。
(c)AHR蛋白質のアミノ酸配列の連続した少なくとも15アミノ酸をコードする核酸。
(d)AHR蛋白質のアミノ酸配列において、1または複数のアミノ酸が置換、欠失、および/または挿入したアミノ酸配列を含む蛋白質をコードする核酸。
(e)AHR遺伝子の連続した少なくとも50塩基を含み、AHR遺伝子の塩基配列以外の配列を含まない核酸と、ストリンジェントな条件下でハイブリダイズする核酸。

(a)に記載の核酸には、好ましくはAHR遺伝子(より好ましくはそのコード配列)に記載の塩基配列の連続した少なくとも40塩基、より好ましくは60塩基以上、より好ましくは100塩基以上、より好ましくは200塩基以上、より好ましくは500塩基以上、より好ましくは1000塩基以上、最も好ましくは全長を含む核酸が含まれる。このような核酸には、主に同一種内の遺伝子の多型、変異体、およびスプライシングバリアントが含まれ得る。また(a)に記載の核酸としては、AHR遺伝子の塩基配列の連続した少なくとも30塩基、より好ましくは35塩基以上、より好ましくは40塩基以上、より好ましくは45塩基以上、より好ましくは50塩基以上の塩基配列を含む核酸が挙げられる。塩基配列は、AHR遺伝子のコード配列から選択することが好ましい。塩基配列は、任意の数を選択することができる。複数の塩基配列を選択する場合には、オーバーラップしないように2箇所以上、好ましくは3箇所以上、より好ましくは4箇所以上、より好ましくは5箇所以上の連続した塩基配列を含む核酸がより好ましい。

Biol. 215: 403−410)を用いて決定することができる。具体的には、塩基配列の同一性を決定するにはblastnプログラム、アミノ酸配列の同一性を決定するにはblastpプログラムを用い、例えばNCBI(National Center for Biothchnology Information)のBLASTのウェブサイトにおいてLow complexityを含むフィルターは全てOFFにして、デフォルトのパラメータを用いて検索を行う(Altschul, S.F. et al. (1993) Nature Genet. 3:266−272; Madden, T.L. et al. (1996) Meth. Enzymol. 266:131−141; Altschul, S.F. et al. (1997) Nucleic Acids Res. 25:3389−3402; Zhang, J. & Madden, T.L. (1997) Genome Res. 7:649−656)。例えば2つの配列の比較を行うblast2sequencesプログラム(Tatiana A et al. (1999) FEMS Microbiol Lett. 174:247−250)により、2配列のアライメントを作成し、配列の同一性を決定することができる。ギャップはミスマッチと同様に扱い、AHR遺伝子のコード配列全体に対する同一性の値を計算する。

また、モノクローナル抗体を得るには、上記抗原を感作した哺乳動物から免疫細胞を取り出して骨髄腫細胞などと細胞融合させる。こうして得られたハイブリドーマをクローニングして、その培養物から抗体を回収しモノクローナル抗体とすることができる。

また本発明によるアレルギー性疾患モデル動物は、後に述べるアレルギー性疾患の発作の治療または予防のための医薬品のスクリーニングに加えて、アレルギー性疾患のメカニズムの解明、さらにはスクリーニングされた化合物の安全性の試験に有用である。
たとえば本発明によるアレルギー性疾患モデル動物が気管支喘息を発症したり、何らかのアレルギー性疾患に関連した測定値の変化を示せば、それを回復させる作用を持った化合物を探索するスクリーニングシステムが構築できる。

更に本発明は、アレルギー性疾患の治療薬候補化合物のスクリーニング方法に関する。本発明における指標遺伝子は、IL−4、CD40リガンド、および抗IgM抗体で刺激したB細胞において有意に発現レベルが上昇する。したがって、これらの遺伝子の発現レベルを低下させることができる化合物を選択することによって、アレルギー性疾患の治療薬を得ることができる。本発明において遺伝子の発現レベルを低下させる化合物とは、遺伝子の転写、翻訳、タンパク質の活性発現のいずれかのステップに対して阻害的に作用する作用を持つ化合物である。
本発明のアレルギー性疾患の治療候補化合物のスクリーニング方法は、in vivoで行なうこともin vitroで行うこともできる。このスクリーニングは、たとえば以下のような工程にしたがって実施することができる。なお本発明のスクリーニング方法における指標遺伝子とは、先に指標遺伝子として挙げたAHR遺伝子に加え、それらの遺伝子と機能的に同等ないずれかの遺伝子を含む。
(1)被験動物に、候補化合物を投与する工程、
(2)被験動物の生体試料における前記指標遺伝子の発現レベルを測定する工程、
(3)対照と比較して、指標遺伝子の発現レベルを低下させる化合物を選択する工程

本発明のスクリーニング方法における被験動物としては、たとえばヒトの指標遺伝子を強制発現させたトランスジェニック動物からなるアレルギー性疾患モデル動物を利用することができる。発現ベクターに使用するプロモーターとして、適当な薬剤等の物質により転写が調節されるプロモーターを用いれば、該物質の投与によってトランスジェニック動物における外来性の指標遺伝子の発現レベルを調整することができる。

このようにして指標遺伝子を強制発現させたモデル動物に薬剤候補化合物を投与し、モデル動物の生体試料における指標遺伝子の発現に対する化合物の作用をモニターすることにより、指標遺伝子の発現レベルに与える薬剤候補化合物の影響を検出することができる。被験動物の生体試料における指標遺伝子の発現レベルの変動は、前記本発明の検査方法と同様の手法によってモニターすることができる。更にこの検出の結果に基づいて、指標遺伝子の発現レベルを低下させる薬剤候補化合物を選択すれば、薬剤候補化合物をスクリーニングすることができる。
より具体的には、被験動物から、生体試料を採取し、前記指標遺伝子の発現レベルを対照と比較することにより、本発明によるスクリーニングを実施することができる。血液細胞や血液細胞から単離されたB細胞を生体試料として用いることができる。実際には、全血、末梢血単核球、あるいは末梢血リンパ球など試料としてを利用することができる。これらの生体試料の採取方法、および調製方法は公知である。

このようなスクリーニングにより、指標遺伝子の発現に様々な形で関与する薬剤を選択することができる。具体的には、たとえば次のような作用点を持つ薬剤候補化合物を見出すことができる。

また前記スクリーニング方法において、候補化合物の投与前および/または投与後に、被験動物をアレルゲンで刺激する工程を含む方法も本発明に含まれる。候補化合物の投与前にアレルゲンで刺激した場合には、候補化合物の、アレルゲン刺激後の免疫応答を抑制する作用を検出することができる。このようなスクリーニングによって得ることができる化合物には、アレルギー性疾患の治療効果を期待できる。

あるいは候補化合物の投与後にアレルゲンで刺激した場合には、候補化合物の、アレルゲン刺激による免疫応答の開始を抑制する作用を検出することができる。このようなスクリーニングによって得ることができる化合物には、アレルギー性疾患の予防効果を期待できる。本発明のスクリーニング方法に用いることができるアレルゲンとしては、公知のアレルゲン性の物質を用いることができる。具体的には、ダニ、ハウスダスト、植物の花粉、あるいは各種の食品由来のタンパク質等がアレルゲン性物質として公知である。これらのアレルゲンは、天然に由来するものであっても良いし、遺伝子組換えなどの手法によって合成されたものであっても良い。またアレルゲンは、タンパク質の断片であることもできる。精製アレルゲンの調製方法も公知である。

本発明において、指標遺伝子を発現する細胞は、指標遺伝子を適当な発現ベクターに挿入し、該ベクターを適当な宿主細胞に導入することにより得ることができる。利用できるベクター、および宿主細胞は、本発明の遺伝子を発現し得るものであればよい。宿主−ベクター系における宿主細胞としては、大腸菌、酵母、昆虫細胞、動物細胞等が例示でき、それぞれ利用できるベクターを適宜選択することができる。

さらに本発明の開示に基づいて本発明の指標遺伝子の転写調節領域を取得し、レポーターアッセイ系を構築することができる。レポーターアッセイ系とは、転写調節領域の下流に配置したレポーター遺伝子の発現量を指標として、該転写調節領域に作用する転写調節因子をスクリーニングするアッセイ系をいう。
すなわち本発明は、次の工程を含む、アレルギー性疾患の治療薬のスクリーニング方法であって、指標遺伝子が、AHR遺伝子またはAHR遺伝子と機能的に同等な遺伝子である方法に関する。
(1)指標遺伝子の転写調節領域と、この転写調節領域の制御下に発現するレポーター遺伝子を含むベクターを導入した細胞と候補物質を接触させる工程、
(2)前記レポーター遺伝子の活性を測定する工程、および
(3)対照と比較してレポーター遺伝子の発現レベルを低下させる化合物を選択する工程

転写調節領域としては、プロモーター、エンハンサー、さらには、通常プロモーター領域に見られるCAATボックス、TATAボックス等を例示することができる。またレポーター遺伝子としては、CAT(chloramphenicol acetyltransferase)遺伝子、ルシフェラーゼ(luciferase)遺伝子、成長ホルモン遺伝子等を利用することができる。
転写調節領域は、ゲノムの塩基配列から推測することができる。AHR遺伝子は、ヒト7番染色体の7p21.1の約47kbにマッピングされている。この領域の塩基配列はQV40以上(10000bに1ヶ所以下のエラー頻度)の精度で既に解析され、塩基配列が明らかにされている(GeneBank accession# NT_007819)。AHR遺伝子は、NT_007819の塩基配列上で塩基番号16631771から16681241に位置している。転写調節領域は一般に転写開始点付近に位置しているので、当該領域を中心として上記転写調節領域に特徴的な塩基配列を検索することによって、目的とする転写調節領域を予測することができる。予測された領域をクローニングし、実際に転写因子によって認識され、転写が開始されることを確認して、転写調節領域を単離することができる。

得られた転写調節領域を、レポーター遺伝子の上流に位置するようにクローニングしてレポーターコンストラクトを構築する。得られたレポーターコンストラクトを培養細胞株に導入してスクリーニング用の形質転換体とする。この形質転換体に候補化合物を接触させ、レポーター遺伝子の発現を制御する化合物のスクリーニングを行うことができる。

また、指標遺伝子の転写調節領域の制御下に連結されたレポーター遺伝子を含む細胞は、いわゆるノックイン動物の細胞であってもよい。ノックイン動物とは、内在性標的遺伝子の蛋白質コード領域に他の外来遺伝子が挿入された遺伝子改変動物を言う。ノックインされたこの外来遺伝子は、標的遺伝子の転写調節領域の下流に挿入されるため、内在性標的遺伝子と同様の発現制御を受けて発現する。ノックイン遺伝子としてレポーター遺伝子を用い、得られたノックイン動物またはその動物から得た細胞を使ってスクリーニングを行うことができる。ノックイン動物を用いてスクリーニングを行う方法は、(a)指標遺伝子部位にレポーター遺伝子がノックインされた動物に候補化合物を投与する工程、(b)該レポーター遺伝子の発現レベルを測定する工程、および(c)該候補化合物を投与しない対照と比較して該レポーター遺伝子の発現レベルを低下させる化合物を選択する工程、を含む。

(1)指標遺伝子によってコードされる蛋白質と候補物質を接触させる工程、

ここで指標遺伝子と機能的に同等な遺伝子としては、上記のように内在性AHR遺伝子の人為的な改変体などが含まれる。

本発明による各種のスクリーニング方法に必要な、ポリヌクレオチド、抗体、細胞株、あるいはモデル動物は、予め組み合わせてキットとすることができる。より具体的には、たとえば指標遺伝子を発現する細胞と、これらの指標遺伝子の発現レベルを測定するための試薬とで構成される。指標遺伝子の発現レベルを測定するための試薬としては、たとえば少なくとも1つの指標遺伝子の塩基配列を含むポリヌクレオチド、若しくはその相補鎖に相補的な塩基配列を有する少なくとも15塩基の長さを有するオリゴヌクレオチドが用いられる。あるいは、少なくとも1つの指標蛋白質のアミノ酸配列を含むペプチドを認識する抗体を試薬として用いることができる。これらのキットには、標識の検出に用いられる基質化合物、細胞の培養のための培地や容器、陽性や陰性の標準試料、更にはキットの使用方法を記載した指示書等をパッケージしておくこともできる。

本発明のスクリーニング方法によって選択される化合物は、アレルギー性疾患の治療薬として有用である。あるいは、本発明における指標遺伝子の発現を抑制することができるアンチセンスDNAも、アレルギー性疾患の治療薬として有用である。好ましくはこのようなアンチセンスDNAは、本発明における指標遺伝子のセンス鎖の塩基配列の連続する少なくとも20塩基、より好ましくは25塩基以上、より好ましくは30塩基以上、より好ましくは40塩基以上、より好ましくは50塩基以上、より好ましくは100塩基以上の配列に対して相補的な配列を含むアンチセンスDNAである。アンチセンス領域としては、指標遺伝子の転写産物(プロセッシング前、中間産物、またはプロセッシング後のいずれの転写産物でもよい)の任意の部位のアンチセンスであってもよい。このような部位としては、例えば翻訳開始コドンを含む領域などが挙げられる。

更に、本発明における指標遺伝子によってコードされる蛋白質に結合する抗体は、アレルギー性疾患の治療薬として有用である。蛋白質の活性を抑制する抗体の調製方法は公知である。前記のドミナントネガティブ効果を確認した報告(Mohan BK. et.al. J. Biol. Chem., 276, 42302−42310, 2001)においては、Q−richを欠失した変異体(AhR1−637,AhR1−663)で、XRE−ルシフェラーゼレポーター遺伝子の発現誘導が抑制されている。したがって、Q−richサブドメインの適当な配列をエピトープとする抗体がAhRの転写調節活性を抑制する可能性が考えられる。抗体をヒトに投与する場合には、キメラ抗体やヒト化抗体、あるいはヒト型抗体とすることにより、安全性の高い薬剤とすることができる。

本発明のアレルギー性疾患の治療薬は、前記スクリーニング方法によって選択された化合物、該アンチセンスDNA、または該抗体を少なくとも1つの主成分として含み、生理学的に許容される担体、賦形剤、あるいは希釈剤等と混合することによって製造することができる。本発明のアレルギー性疾患の治療剤は、アレルギー症状の改善を目的として、経口、あるいは非経口的に投与することができる。
経口剤としては、顆粒剤、散剤、錠剤、カプセル剤、溶剤、乳剤、あるいは懸濁剤等の剤型を選択することができる。注射剤には、皮下注射剤、筋肉注射剤、あるいは腹腔内注射剤等を示すことができる。

投与量は、患者の年齢、性別、体重および症状、治療効果、投与方法、処理時間、あるいは該医薬組成物に含有される活性成分の種類などにより異なるが、通常成人一人あたり、一回につき0.1 mgから500 mgの範囲で、好ましくは0.5 mgから20 mgの範囲で投与することができる。しかし、投与量は種々の条件により変動するため、上記投与量よりも少ない量で充分な場合もあり、また上記の範囲を超える投与量が必要な場合もある。

また、投与すべき化合物がタンパク質からなる場合には、それをコードする遺伝子を遺伝子治療の手法を用いて生体に導入することにより、治療効果を達成することができる。治療効果をもたらすタンパク質をコードする遺伝子を生体に導入し、発現させることによって、疾患を治療する手法は公知である。
あるいはアンチセンスDNAは、適当なプロモーター配列の下流に組み込み、アンチセンスRNA発現ベクターとして投与することができる。この発現ベクターをアレルギー性疾患患者のB細胞へ導入すれば、これらの遺伝子のアンチセンスを発現し、当該遺伝子の発現レベルの低下によってアレルギー性疾患の治療効果を達成することができる。B細胞への発現ベクターの導入としては、in vivo、あるいはex vivoで行う方法が公知である。

本発明においては、アレルギー性疾患の指標遺伝子としてAHR遺伝子が選択された。AHRは、ダイオキシン類に非常に感受性の高いβHLH−PAS型転写因子ファミリーに属する受容体型因子として知られている。AHRに関してはダイオキシン類に対する応答についての様々な報告があるが、アレルギー性疾患との関連についての報告は無い。以下に、AHRについて得られている代表的な知見についてまとめた。以下にまとめたように、AHRとアレルギー性疾患の関連を示唆する報告は無い。

細胞質中で、AHRはHsp90等と複合体を形成し存在している(文献1/Carver, LA & Bradfield CA. J. Biol.Chem., 272, 11452−11456, 1997; 文献2/Mimura, J. & Fujii−Kuriyama, Environ. Sci., 9, 71−81, 2002)。ダイオキシン類の強力な生体毒性から、AHRの生理機能について興味が持たれている。AHRは、そのリガンドであるダイオキシン類と結合することによって核内に移行する。リガンドとして作用するダイオキシン類としては、例えば2,3,7,8−tetrachlorodibenzo−p−dioxin (TCDD)が同定されている。

核内に移行したAHRは、予め核内に存在するARNTと出会うとHsp90を解離し、ARNTとヘテロ二量体を形成する。AHR−ARNTヘテロ二量体はDNA結合型転写因子として働き、プロモーター領域にXRE(xenobiotic responsive element)と呼ばれるエンハンサーエレメント(TNGCGTG, Nは任意)を認識して結合する。AHR−ARNTヘテロ二量体がXREに結合すると、プロモーター領域のクロマチン構造がほどかれ、基本転写因子の結合が促進される結果、転写が活性化される(文献3/KO,HP., Okino, ST., Ma, Q., Whitlock, JP. Mol. Cell. Biol., 16, 430−436, 1996)。

AHRの転写制御を受けている代表的な遺伝子として、シトクロムP4501A1,1A2,1B1等の薬物代謝酵素遺伝子が挙げられる。薬物代謝酵素遺伝子以外にもTCDDによりJun−B等の細胞周期調節因子、インターロイキン1β、トランスフォーミング増殖因子(TGF−α、β2)などの成長因子やアルデヒドデヒドロゲナーゼ3(ADH−3)等の発現が誘導される。これらの遺伝子の中にプロモーター領域にXREが存在し、AHR依存的であることが示されているものもあるが、Jun−Bのように不明なものもある(文献4/Wilk,r.,Weizman,I.,&Shilo,BZ., Genes Dev., 10, 93−102, 1996; 文献5/Kolluri, SK Weiss, C., Koff, A. & Gottlicher, M., Genes Dev., 13, 1742−1753, 1999)。

AHR関連遺伝子群の一つであるAHRRは、一次構造がAHRとN末側で似ているがC末側の構造は全く異なる。AHRRはAHRのパートナー分子であるARNTと核内でヘテロ二量体を形成し、XREに結合することができる。このときリガンドは必要ない。AHRR遺伝子はプロモーター領域にXREを有し、AHR/ARNTによる転写制御を受けることから、AHRRはAHRのネガティブフィードバック制御に働くことが示唆されている(文献6/Miura J et.al. Genes Dev. 13, 20−25, 1999)。

マウスにおけるAHR発現の組織分布に関する報告がある(文献7/Abbott, BD., Birnbaum, LS. & Perdew, GH., Dev. Dyn., 204, 133−143, 1995)。AHR mRNAは成体マウスでは、肺、肝臓での発現が高いが、その他の組織でも量的には低いものの普遍的に発現している。ARNTは、殆どの組織で普遍的に発現している(文献8/Abbott, BD.& Probst, MR., Dev. Dyn., 204, 144−155, 1995)。

AHRは、長らくオーファンレセプターとされており、その機能や生理的リガンドが不明であった。最近のAHRノックアウトマウスの研究では、生殖能力の低下や肝臓での血管形成異常が報告されている(文献9/Abbott, BD. et.al., Toxicol. Appl. Pharmacol., 155, 62−70, 1999)。AHRホモノックアウトマウスは、週齡を重ねると心筋症などの病変や、免疫機能の低下により感染症を起こす(文献10/Fernandez−Salguero,PM. et.al. Vet. Pathol., 34, 605−614, 1997)。これらの知見は、少なくともマウスではAHRが何等かの生理機能維持に関与していることを示している。
機能解析の糸口として内因性リガンドの探索が試みられている。その結果、紫外線やオゾン処理されたトリプトファン産物がAHRのアゴニストして働くことが明らかにされた(文献10/Sindhu,RK.Mitsuhashi,& Kikkawa,Y.,J.Pharmacol.Exp.Ther.,292,1008−1014, 2000)。また尿中に***されるインディルビンがAHRと高親和性のリガンドとして機能することが報告されている(文献11/Adachi, J. et.al., J. Biol. Chem., 276, 31475−31478, 2001)。

ダイオキシンの受容体であるAHRの機能研究は、ダイオキシンの毒性研究の一環として研究されてきた経緯がある。AHRホモノックアウトマウスにおけるTCDD毒性が検討されている。ノックアウトマウスでは、TCDDによる薬物代謝酵素の発現誘導はみられない。妊娠12.5日の野生型マウス母体にTCDDを経口投与すると、ほぼ100%の胎児に口蓋裂、水腎症という奇形が生じる。しかしAHR−/−あるいは+/−の交配で生まれたホモ欠損マウス胎仔は、口蓋裂も水腎症も発症しない(文献12/Mimura, J. et.al., Genes Cells, 2, 645−654, 1997)。野生型のマウスにTCDDを投与すると胸腺の萎縮やリンパ球の減少がみられるが、ホモ欠損マウスでは変化がない(文献13/Fernandez−Salguero,PM. et.al., Toxicol. Appl. Pharmacol., 140, 173−179, 1996)。これらのことはTCDDの毒性にはAHRが必須であることを示している。

リガンドの結合したAHRとNF−κBが相互作用し、NF−κBシグナル伝達経路を阻害することが報告されている(文献14/Tian,Y.et.al. J.Biol.Chem.,274,510−515, 1999)。この阻害作用によりアポトーシスが誘導されることからTCDDによる免疫の抑制はAHRによるNF−κBの阻害が関係している可能性がある。ダイオキシンを曝露したマウスES細胞とコントロールの遺伝子発現パターンを比較したところ、ヒスタミン放出因子がダイオキシンにより誘導、分泌される報告がある(文献15/Oikawa, K. et.al., Biochem Biophys Res Commun., 290, 984−987, 2002)。これは、炎症メディエーターとダイオキシンの関連を示す最初の報告であるが、この現象にAHRが関与しているかどうかは明らかでない。
以下、本発明を実施例により具体的に説明するが、本発明はこれら実施例に制限されるものではない。

【実施例】
[実施例1]DNAマイクロアレイを使った候補遺伝子の選択
1. 正常ヒト気管支上皮細胞の培養とIL−4あるいはIL−13刺激
Clonetics社より販売している正常ヒト気管支上皮細胞を3ロット購入した(8F1756, 8F1548, 8F1805)。1バイヤル中に入っている5x105細胞を未刺激、IL−4刺激、IL−13刺激用に3等分し(1.67x105/75 cm2 flask)、SABM培地(Clonetics社)にて培地交換しながら約8−10日間培養した。その際に培地にBPE(ウシ脳下垂体抽出液)、Hydrocortisone, hEGF, Epinephrine, Transferrin, Insulin, Retinoic Acid, BSA−FAF, Triiodothyronine, GA−1000(Gentamicin/Amphotericin−B)を添付のプロトコールに従い添加した。
細胞はサイトカイン刺激前に、PBSで洗浄後、添加因子を除いたSABMに交換した。そこにIL−4 (10 ng/ml), IL−13 (50 ng/ml)を添加し(両者ともPeprotech社製)、24時間培養した。

2. GeneChip用RNAの調製
上記のように処理した気道上皮細胞を Isogen(日本ジーン;和光純薬)に溶解し、この溶液から、Isogenに添付されているプロトコルに従ってRNAを分離した。クロロホルムを加え、攪拌遠心して水層を回収した。次にイソプロパノールを加え、攪拌遠心して沈殿の全RNAを回収した。

3.GeneChip用のcDNA合成
lot 8F1548、および8F1805の細胞より調製した全RNA 5μgから、T7−(dT)24 (Amersham Pharmacia社)をプライマーとして、Affymetrix社のExpression Analysis Technical Manualの方法に従いSuperscript II Reverse Transcriptase(Life Technologies社)を用いて逆転写し1本鎖 cDNAを作製した。T7−(dT)24プライマーは、以下のようにT7プロモーターの塩基配列にd(T)24を付加した塩基配列からなる。

次に、Expression Analysis Technical Manualに従い、DNA Ligase, DNA polymerase I及びRNase Hを加え、2本鎖cDNAを合成した。cDNAをフェノール・クロロホルム抽出後、Phase Lock Gelsに通し、エタノール沈澱し精製した。
さらに、BioArray High Yield RNA Transcription Labeling Kitを用い、ビオチンラベルしたcRNAを合成した。RNeasy Spin column (QIAGEN)を用いてcRNAを精製し、熱処理により断片化した。
そのうち12.5 μgのcRNAをExpression Analysis Technical Manualに従いHybridization Cocktailに加えた。これをアレイに入れ、45℃ 16時間ハイブリダイゼーションした。
アレイを洗浄した後、Streptavidin Phycoerythrinを加え染色した。洗浄後、normal ヤギIgGとビオチン化ヤギIgGの抗体混合液をアレイに加えた。さらに、蛍光強度を増強する目的で、再度Streptavidin Phycoerythrinを加え染色した。洗浄後、スキャナーにセットし、GeneChip Softwareにて解析した。

4.GeneChip解析
GeneChip解析ソフトであるSuiteを用いてデータ解析を行った。Absolute analysisで、Average Intensity (1)とBackground Average (2)を調べ、(1)から(2)を引いた、未刺激、IL−4刺激、またはIL−13刺激の3つの平均を補正値(Scale Factor)としてComparison Analysisを行った。
まず、Absolute Analysisを行い1個のチップデータの解析をした。プローブセットのパーフェクトマッチとミスマッチの蛍光強度を比較して、positiveとnegativeを決定した。Pos Fraction, Log Avg, Pos/Negの値から判定されるAbsolute CallであるP(present)、A (absent)、およびM(marginal)の3区分の判定をした。
Pos Fraction; Positiveなペアの割合。
Log Avg; パーフェクトマッチとミスマッチのプローブセルの蛍光強度比の対数の平均

また、それぞれの遺伝子において、パーフェクトマッチとミスマッチのプローブセルの蛍光強度の差の平均値であるAverage Difference (Avg Diff) も計算した。

次に2つのデータを比較解析(Comparison Analysis)した。未刺激とIL−4刺激あるいは未刺激とIL−13刺激で比較し、発現レベルの差を以下のようにランキングした。Inc/Dec,Inc Ratio, Dpos−Dneg Ratio, Log Avg Ratio Changeの値から判定されるDifference CallであるI, D, MI, MD, NCの5区分の判定をした。

[実施例2]候補遺伝子の発現レベルの確認
実施例1で選択されたAHR遺伝子の発現量を定量的に確認するために、ヒト末梢血B細胞、およびヒト正常気管支上皮細胞を用いて更にABI 7700による定量的PCRを行った。ABI 7700による測定に用いたプライマーおよびTaqManプローブは、AHR遺伝子の配列情報に基づいてPrimer Express(PEバイオシステムズ)により設計した。TaqManプローブの5’末端はFAM(6−carboxy−fluorescein)で、また3’末端はTAMRA(6−carboxy−N,N,N’,N’−tetramethylrhodamine)で標識されている。フォワードプライマー(F)、リバースプライマー(R)、およびTaqManプローブ(TP)に用いたオリゴヌクレオチドの塩基配列は、以下に示すとおりである。

末梢血B細胞は、全血よりCD19ポジティブセレクションにより単離した。B細胞の単離方法はDynal Biotech社のDynabeads M−450 CD19のプロトコールに従った。すなわち、健常ヒトボランティアから採血した全血100mlに対し、100μlの抗CD19抗体結合磁気ビーズ(Dynabeads M−450 CD19, Dynal Biotech, Oslo, Norway)を加え、氷冷下で30分、穏やかに混合しながらインキュベートした。インキュベート後、試料を磁気濃縮装置(Dynal MPC, Dynal Biotech, Oslo, Norway)に静置し、ビーズ−細胞結合物を管壁に集め上清を除去した。磁気濃縮装置から試験管を外し、冷PBSを加えビーズ−細胞結合物を再懸濁し、再度磁気濃縮装置に静置し上清を除去した。

ビーズに結合したCD19陽性B細胞を、ビーズから剥離させるため、DETACHaBEA(Dynal Biotech, Oslo, Norway)を使用した。ビーズ−細胞結合物を100μlのwashing buffer(終濃度2%のFBSを含むPBS)に再浮遊させ、10μlのDETACHaBEAD加え、2秒間ボルテキシングで完全に混合した。室温で穏やかに混合しながら1時間放置してビーズを離脱させた。試験管を磁気濃縮装置に静置しビーズを回収し、剥離した細胞を含む上清を回収した。回収した1×106細胞のB細胞を1.5mlの10%FCS含有RPMI1640培地中でIL−4単独、あるいはIL−4、CD40リガンド及び抗IgM抗体で刺激し、24時間培養した。
一方ヒト気管支上皮細胞については、実施例1と同様の処理を与えた細胞を試料とした。

各細胞サンプルからの前述の方法で抽出した全RNAをDNase(ニッポンジーン)処理した。その後、randam hexamer (GIBCO BRL)をプライマーとして逆転写したcDNAを鋳型とした。コピー数を算出する標準曲線のために両プライマーで増幅される塩基配列領域を含むプラスミドクローンを各々の遺伝子について準備し、その段階希釈を鋳型として反応を行った。PCR増幅のモニタリングのための反応液の組成は表2に示した。

定量的PCRの結果、実施例1で選択したAHR遺伝子のB細胞における発現レベルは、抗IgM抗体及び抗CD40抗体で活性化した細胞に対するIL−4刺激で10倍近く上昇した。ヒト正常気管支上皮細胞でも若干の上昇が認められたが、B細胞で観察されたような変化はなかった。これらの結果に基づいて、活性化したB細胞ではIL−4の刺激に応答して、AHR遺伝子の発現レベルが上昇することが予測された。
アレルギー反応との密接な関連が知られているIL−4やIL−13の刺激によって、発現レベルが上昇するAHR遺伝子は、アレルギー反応の進行を制御する重要な遺伝子であると考えられる。

[実施例3]AhR遺伝子発現誘導のWestern blottingによる確認
上述のようにして回収したB細胞をIL−4、抗CD40抗体あるいは抗IgM抗体で刺激し、AhR蛋白質の発現をウェスタンブロッティングにより確認した。
1×106細胞のB細胞を1.5mlの10%FCS含有RPMI1640培地中で24時間培養した。このとき、次の5つの条件で培養する系をそれぞれ設定した。
1)無処理、
2)IL−4(10ng/ml)、
3)IL−4 + 抗IgM抗体(10μg/ml, Cappel #55055)、
4)IL−4 + 抗CD40抗体(0.5μg/ml, Immunotech #1374)、および
5)IL−4 + 抗IgM抗体 + 抗CD40抗体を添加

培養後の細胞をPBSで洗浄後、30μlのLysis buffer、10μlの4×SDS−PAGE sample bufferに懸濁し、4℃で30分間インキュベートした。得られた細胞溶解液のうち25μlを常法に従いSDS−PAGEに供した。泳動後のゲルをメンブレンにトランスファー後、一次抗体として抗AhR抗体(TOYOBO SC−5579)あるいは抗ARNT抗体(TOYOBO SC−5580)と反応させた後、二次抗体としてanti rabbit Ig HRP linked Fab(AmershamPharmacia NA9340)で処理した。メンブレンをECLplus(AmershamPharmacia)に1分間イクスポーズした後、ルミノイメージアナライザーLAS100(富士写真フィルム)で画像を取得し、AhRあるいはARNTタンパク質の発現を確認した。

その結果、AhRの発現はB細胞をIL−4単独で処理しても誘導されないが、IL−4で刺激した際に、抗IgM抗体あるいは抗CD40抗体が共存させると明らかに発現の誘導が認められた。さらに、AhRの発現は抗IgM抗体、抗CD40抗体及びIL−4で刺激した場合、最も強く誘導された。

【発明の効果】
本発明により、炎症性のサイトカインの刺激によってB細胞において発現レベルが増強される遺伝子が見出された。本発明においてアレルギー性疾患との関連性が見出された遺伝子AHRは、アレルギー関連サイトカインである、IL4+CD40リガンド+抗IgM抗体の刺激によっても、末梢血B細胞において、発現レベルが10倍近く増大する、アレルギーに密接に関連した遺伝子である。このような特徴を有する遺伝子は、アレルギー性疾患の本質的な原因となっている可能性が高い。したがって本発明によって提供された指標遺伝子は、炎症性の症状がアレルギー症状によってもたらされたものかどうかを確実に知ることができる有用な指標となる。アレルギー性の免疫応答によってもたらされた疾患を確実に診断できることにより、的確な治療方法を早期に選択することができる。

IL−4あるいはIL−13は、アレルギー反応を増強する重要な因子である。したがって、これらの因子の刺激に伴って発現が増加する遺伝子は、アレルギー症状の病態形成において重要な役割を果たしていると考えられる。既に述べたように遺伝子発現解析に用いた気管支上皮細胞は、アレルギー性の喘息患者においては、IL−4やIL−13の刺激によってアレルギー性の応答を示す細胞である。またB細胞は、IL4のほか、CD40リガンドや抗IgM抗体による刺激により活性化される。この過程で発現レベルが増強されたAHR遺伝子は、アレルギー性反応の特徴であるIgE産生の過程で発現レベルが上昇する遺伝子と言うことができる。本発明者らか明らかにしたこれらの知見は、本発明の指標遺伝子が、アレルギー性疾患に欠かせない存在であることを裏付けている。
このように、本発明の指標遺伝子は、いずれもその発現亢進が炎症性サイトカインによる刺激と結びついていることから、それを抑えることがアレルギー性疾患の治療戦略のターゲットとなるとともに、そのような新しい治療法におけるモニタリングのための新しい臨床診断指標としての有用性が期待できる。

本発明によって提供された指標遺伝子は、アレルゲンの種類に関わらず、簡便にその発現レベルを知ることができる。したがって、アレルギー反応の病態を総合的に把握することができる。
また本発明によるアレルギーの検査方法は、生体試料を試料としてその発現レベルを解析することができるので、患者に対する侵襲性が低い。しかも遺伝子発現解析に関しては、微量サンプルによる高感度な測定が可能である。遺伝子解析技術は、年々ハイスループット化、低価格化が進行している。したがって本発明によるアレルギーの検査方法は、近い将来、ベッドサイドにおける重要な診断方法となることが期待される。この意味でこれらの病態関連遺伝子の診断的価値は高い。

Allergic diseases are considered to be multifactorial diseases. Thus, allergic diseases are caused by the interaction of the expression of many different genes, and the expression of these individual genes is affected by multiple environmental factors. For this reason, it has been very difficult to elucidate the specific genes that cause allergic diseases.
For example, bronchial asthma is positioned as a chronic inflammatory disease in the airways. It has been pointed out that allergic reactions in airway mucosa and bronchial smooth muscle are closely involved in the pathogenesis of bronchial asthma. Therefore, in diagnosing bronchial asthma, it is important to understand these tissues and the state of systemic allergic reactions. In the treatment of bronchial asthma, control of an allergic reaction becomes an issue.

On the other hand, allergic diseases are considered to involve the expression of genes having mutations or deficiencies, or overexpression or reduction of the expression level of specific genes. To understand the role of gene expression in disease, it is necessary to understand how genes are involved in the pathogenesis and how external stimuli such as drugs alter gene expression.

By the way, many patients with bronchial asthma have an atopic predisposition accompanied by enhanced production of IgE antibodies. Although there are a variety of causes for bronchial asthma, there is no doubt that atopic predisposition causes hypersensitivity in many patients. As a mechanism of airway obstruction due to asthma attack, contraction of bronchial smooth muscle, edema of airway mucosa, and enhancement of airway endocrine secretion are expected. Type I allergic reactions due to exposure to etiological allergens play an important role in such airway changes.

Now, in the diagnosis of allergic diseases, interviews, family histories, and confirmation of the patient's history are important factors in general. In order to diagnose allergy based on more objective information, a test method using blood as a sample and a method of observing a patient's immunological response to an allergen have been implemented. Examples of the former include an allergen-specific IgE measurement, a leukocyte histamine release test, and a lymphocyte blastogenesis test. The presence of allergen-specific IgE is evidence of an allergic reaction to the allergen. However, in some patients, it is not always possible to detect allergen-specific IgE. In addition, due to its measurement principle, tests must be performed for all allergens required for diagnosis. The leukocyte histamine release test and lymphocyte blastogenesis test are methods for observing the response of the immune system to allergens in vitro. These methods are complicated in operation.
On the other hand, a method of using an immune response observed when a patient is actually contacted with an allergen to diagnose allergy (the latter) is also known. Such tests include prick tests, scratch tests, patch tests, intradermal reactions, or provocation tests. While these tests can directly diagnose a patient's allergic reaction, they can be said to be highly invasive tests that actually expose the subject to an allergen.

In addition, test methods for proving the involvement of allergic reactions regardless of allergens have been attempted. For example, when the serum IgE level is high, it can be estimated that the patient has an allergic reaction. The serum IgE value is information corresponding to the total amount of allergen-specific IgE. Although it is easy to determine the total amount of IgE regardless of the type of allergen, IgE may be low in patients with diseases such as non-atopic bronchitis.

Therefore, it would be useful to provide a marker for an allergic disease, which has a low risk to the patient and which can easily obtain information necessary for diagnosis. Since such a marker is considered to be deeply involved in the development of allergic diseases, it may be an important target not only in diagnosis but also in control of allergic symptoms.

Several reports have suggested that IL-4 and IL-13 are deeply involved in allergic reactions. For example, IL-4 (Yssel, Hand and Groux, H: Int. Arch. Allergy Immunol., 121; 10-18, 2000) and STAT6 (Akimoto, T. et al .: J. Exp. Med., 187, 1537-1542, 1998), the airway hyperresponsiveness disappears. In model mice, IL-13 is involved in the formation of asthma-like pathologies irrespective of IgE production or Th2 type (Wills-Karp, M. et al .: Science, 282, 2258-2261, 1998; Grunig, G. et al .: Science, 282,2261-2263, 1998; Zhu, Z. et al .: J.
