口腔内で迅速に崩壊・溶解する固形製剤の開発が進めら
れている。例えば、国際公開番号WO93/12769
号には、医薬物質と乳糖およびマンニトールを寒天水溶
液に縣濁させ、成形ポケット等に充填後、減圧乾燥させ
て得られる口腔内崩壊錠が記載されている。この成型物
は急速な崩壊性が得られる反面、通常の錠剤に比べると
強度が弱く、割れ、カケ等が発生し易いことおよび製造
に長時間を要することから生産性に劣る製造法である。
また、特開平6−218028号、特開平8−1958
9号には、練合した湿潤粉体を錠剤成形用の穴に充填し
湿式成形後、乾燥させて錠剤を製造する方法が記載され
ている。得られた錠剤は多孔性で適度な空隙率を有して
いることから、速やかな崩壊性が得られている。しかし
この製造法は、流動性の悪い湿体を充填・圧縮するた
め、充填バラツキが大きいことに加え、特殊な乾燥機が
必要なこともあり、工業的な生産性に劣る。このため工
業的な生産性に優れた乾式打錠法による口腔内崩壊錠の
製造についても報告されている。例えば、国際公開番号
WO95/20380には、成形性の高い糖類に成形性
の低い糖類を組み合わせた乾式打錠法による口腔内崩壊
錠の製造法が記載されている。また、国際公開番号WO
98/02185には、賦形剤と糖アルコールであるエ
リスリトールを組み合わせ、乾式あるいは湿式造粒した
顆粒を用いた乾式打錠法による口腔内崩壊錠の製法が記
載されている。さらに、国際公開番号WO97/472
87には、平均粒子径30μm以下の糖または糖アルコ
ールと活性成分および崩壊剤を組み合わせ、口腔内で速
やかに崩壊する錠剤の製造法が記載されている。これに
記載されている製造法によれば、D−マンニトールや乳
糖等の糖または糖アルコールを微粉砕したのち、崩壊剤
等を加えて圧縮成形した成型物には、速やかな崩壊性が
得られるが、粉砕前の粗い糖(乳糖,平均粒子径80μ
m)あるいは糖アルコール(D−マンニトール,平均粒
子径60μm)を用いた成形物の場合、低い打錠圧では
成形が困難であり、高い打錠圧で成形しても十分な機械
的硬度が得られていない。また、D−マンニトールは圧
縮成形を受けた場合、臼壁面との間で極めて高い摩擦
(バインディング)を生ずることが知られている。さら

かりでなく、錠剤製造時の流動性低下といったハンドリ
