ピリジノントロンビンインヒビター 発明の背景
トロンビンは、血漿中、前駆体、プロトロンビンの形態で存在するセリンプロ
テアーゼである。トロンビンは、溶液血漿蛋白質、フィブリノーゲンを不溶性フ
ィブリンに転化することによる血液凝固のメカニズムにおいて中心的な役割を果
たしている。
Edwardsら、J.Amer.Chem.Soc.,(1992)vol.114,1854-63頁では、セリンプ
ロテアーゼヒト白血球エラスターゼ及びブタすい臓エラスターゼの可逆性インヒ
ビターであるペプチジルα-ケトベンズオキサゾールについて記載している。
欧州特許公開第363284号は、基質ペプチドの切断性アミド基の窒素原子が水素
または置換カルボニル部分により置き換わったペプチターゼ基質の類似物につい
て記載している。
オーストラリア特許公開第86245677号も、活性化求電子性ケトン部分、例えば
、フルオロメチレンケトンまたはα-ケトカルボキシル誘導体などを有するペプ
チターゼインヒビターについ
て記載している。
R.J.Brownら、J.Med.Chem.,Vol.37,1259-1261頁(1994)は、トリフルオロメ
チルケトン及びピリジノン部分を含有するヒト白血球エラスターゼの経***性な
、非-ペプチド性インヒビターについて記載している。
H.Mackら、J.Enzyme Inhibition,Vol.9,73-86頁(1995)は、中心核構造
としてピリジノン部分を含有する硬質アミジノ-フェニルアラニントロンビンイ
ンヒビターについて記載している。発明の概要
本発明は、医薬的に許容可能なキャリヤ中に本発明の化合物を含む、ほ乳類に
おいて血小板の損失を阻止し、血小板凝集物の形成を阻止し、フィブリン形成を
阻止し、血栓形成を阻止し、且つ塞栓形成を阻止するための組成物を包含する。
これらの組成物は、場合により、抗凝血薬、抗血小板薬及び血栓溶解剤も包含し
得る。本組成物は、所望の阻害作用を得るために、血液、血液製品またはほ乳類
臓器にも添加し得る。
本発明は、医薬的に許容可能なキャリヤ中に本発明の化合物を含む、ほ乳類に
おける、不安定アンギナ、難治療性アンギナ、
心筋骨屈折(myocardinal infraction)、一過性虚血性発作、心房細動、血栓性
脳卒中、塞栓性脳卒中、深静脈血栓症、播種性血管内凝血、フィブリンの眼球内
集積(ocular build up)及び疎通化血管の再閉塞または再狭窄症を予防または治
療するための組成物も包含する。これらの組成物は場合により、抗凝血薬、抗血
小板薬及び血栓溶解剤も包含し得る。
本発明は、本発明の化合物を共有的または非共有的のいずれかで、表面に付与
することによる、ほ乳類の表面の凝塊形成を減少させる方法も包含する。
本発明は、ほ乳類における血栓形成の阻害、血栓形成の予防、トロンビンの阻
害、フィブリン形成の阻害及び血小板凝集物の形成を阻害するために薬剤の製造
時に、本発明の化合物または医薬的に許容可能なその塩の使用も包含する。

本明細書中、他に特記しないかぎり、「アルキル」とは、特定数の炭素原子を

はメチル、Etはエチル、Prはプロピル、Buはブチルである);「アルコキシ」と
は、酸素橋を介して結合した表示数の炭素原子の直鎖または分岐アルキル基を表
すものとする。本明細書中で使用する「ハロ」とは、フルオロ、クロロ、ブロモ

本明細書中で使用する「アリール」なる用語は、他に記載しない限り、安定な
6-〜10-員の-または二環性環系、例えば、フェニルまたはナフチルなどを表す。
アリール環は、1個以上のC1-4低級アルキル;ヒドロキシ;アルコキシ;ハロゲ
ン;アミノで置換または非置換であってもよい。
本明細書中で使用する「複素環(heterocycle)」または「複素環式環(heterocy
clic ring)」なる用語は、他に記載しない限り、任意の環が飽和または不飽和で
あり得且つN、O及びSからなる群から選択される1〜4個のヘテロ原子及び炭素
原子
からなる安定な5-〜7-員の一若しくは二環性または、安定な7-〜10-員の二環性
複素環式環系を表し、窒素及び硫黄ヘテロ原子は、場合により酸化され得、窒素
ヘテロ原子は場合により四級化され得、上記定義の任意の複素環式環は、ベンゼ
ン環に融合されている任意の二環性基を包含する。
1個の酸素若しくは硫黄、1〜4個の窒素原子または、1若しくは2個の窒素原
子と結合している1個の酸素若しくは硫黄を含有する環が特に有用である。複素
環式環は、任意のヘテロ原子または炭素原子と結合し得、これにより安定な構造

式Iの化合物の医薬的に許容可能な塩(水-若しくは油-溶性または分散性生成
物の形態)としては、例えば、無機若しくは有機酸または塩基から形成した慣用
の非毒性塩または第4級アンモニウム塩が挙げられる。このような酸付加塩の例
としては、酢酸塩、アジピン酸塩、アルギン酸塩、アスパラギン酸塩、安息香酸
塩、ベンゼンスルホン酸塩、重硫酸塩、酪酸塩、クエン酸塩、樟脳酸塩、樟脳ス

及びヨーダイド;ジメチル、ジエチル、ジブチルなどのジアルキルサルフェート
;及びジアミルサルフェート、長鎖ハライド、例えば、デシル、ラウリル、ミチ
スチル及びステアリルクロリド、ブロミド及びヨーダイド;アラルキルハライド
、例えば、ベンジル及びフェネチルブロミドなどの薬剤で四級化されていてもよ
い。トロンビンインヒビター治療的使用-使用法
抗凝血剤治療は、種々の血栓性疾患、特に冠状動脈及び脳血管疾患の治療及び
予防用に示されたものである。この分野の当
業者らは、抗凝血治療を必要とする状況に容易に気づく。本明細書中で使用する
「患者」なる用語は、ヒトを含む霊長類、羊、馬、牛、豚、犬、猫、ラット及び
ネズミ等のほ乳類を指すものとする。
トロンビン阻害は、血栓性疾患の個人の抗凝血剤治療に有効なだけでなく、貯
蔵全血の凝集防止及び試験または貯蔵用の他の生物学的サンプル中の凝集を防止
するなどの、血液凝固の阻害に何時でも有用である。従って、トロンビンインヒ
ビターは、トロンビンを含有しているかまたはその疑いのある任意の媒体であっ
て、血液凝固を阻害することが望ましいもの、例えば、ほ乳類の血液を管移植、
ステント、整形外科プロテーゼ、心臓プロテーゼ及び体外循環系(extracorporea
l circulation system)からなる群から選択される材料と接触させる際などに、
これらの媒体に添加またはこれと接触させることができる。
本発明のトロンビンインヒビターは、錠剤、カプセル(その各々は、継続的な
放出または時間指定で放出する配合物を包含する)、ピル、パウダー、粒子、エ
リキシル、チンキ、懸濁液、シロップ及びエマルションなどの経口形で投与し得
る。同様に、これらは、全て医薬業界の当業者には公知の形態を使用する、
静脈内(丸塊または注入)、腹腔内、皮下または筋肉内形で投与し得る。所望の化
合物の有効であるが非毒性量を、抗凝集薬として使用し得る。フィブリンの眼球
内集積(ocular build up)の治療には、本化合物は、眼球内または局所的並びに
経口または非経口で投与し得る。
トロンビンインヒビターは、活性成分の継続的放出ができるような状態で配合
し得るデポー注射またはインプラント調製物の形態で投与し得る。活性成分は、
ペレットまたは小さなシリンダー内に圧縮し、次いでデポー注射またはインプラ
ントとして皮下または筋肉内にインプラントし得る。インプラントは、生分解性
ポリマーまたは合成シリコーン、例えば、Silastic)シリコーンラバーまたはDo
w Chemical Corporationにより製造された他のポリマーなどの不活性材料を使用
し得る。
トロンビンインヒビターは、リポソームデリバリーシステム、例えば、小さな
ユニラメラ小胞、大きなユニラメラ小胞及びマルチラメラ小胞等の形状でも投与
し得る。リポソームは、種々のリン脂質、例えば、コレステロール、ステアリル
アミンまたはホスファチジルコリンから形成し得る。
トロンビンインヒビターは、化合物分子が結合する個々のキ
ャリヤとしてモノクローナル抗体を使用することによっても搬送し得る。トロン
ビンインヒビターは、標的可能なドラッグキャリヤとして溶解性ポリマーとも結
合させ得る。そのようなポリマーの例としては、ポリビニルピロリドン、ピラン
コポリマー、ポリヒドロキシ-プロピル-メタクリルアミド-フェノール、ポリヒ
ドロキシエチル-アスパルタミド-フェノールまたはパルミトイル残基で置換した
ポリエチレンオキシド-ポリリシンが挙げられる。さらに、トロンビンインヒビ
ターは、薬(drug)の放出を制御するために有用な一種の生分解性ポリマー、例え
ば、ポリ乳酸、ポリグリコール酸、ポリ乳酸とポリグリコール酸とのコポリマー
、ポリイプシロンカプロラクトン、ポリヒドロキシ酪酸、ポリオルソエステル、
ポリアセタール、ポリジヒドロビラン、ポリシアノアクリレート及びヒドロゲル
の架橋または両親媒性ブロックコポリマーと結合させ得る。
トロンビンインヒビターを用いる用量治療方式(dosageregimen)は、患者のタ
イプ、種、年齢、体重、性別及び健康状態;治療すべき疾患の重篤度;投与方法
;患者の腎臓及び肝機能;及び用いる特定の化合物またはその塩に依存して選択
する。当業者である医師または獣医師は、疾患の進行を阻止、対抗ま
たは抑制すのに必要な薬の有効量を直ちに決定し、処方することができる。
表示作用のために使用する際、トロンビンインヒビターの経口用量は、1日当
たり体重1キログラム当たり約0.1g(mg/kg/日)〜約100mg/kg/日であり、1.0〜10
0mg/kg/日が好ましく、1〜20mg/kg/日が最も好ましい。1日に1回投与するため
の好適に調製した薬剤は、9mg〜9gを含み、90mg〜9gが好ましく、90mg〜1.8gが
最も好ましく、例えば100mg、500mg及び1gである。好都合には、トロンビンイン
ヒビターは、1日当たり2、3または4回に分けて投与し得る。そのような複数回
の投与のためには、個々の薬剤強度は、例えば、25mg、33mgまたは50mgであり
得る。静脈内の場合には、最も好ましい用量は、一定速度の注入時に約0.01〜約
10mg/kg/分を変動する。さらに、これらは、当業者に公知の経皮スキンパッチの
形態を使用する経皮経路または好適な鼻腔内ビヒクルの局所使用を介する鼻腔内
形で投与し得る。経皮デリバリーシステムの形態で投与するためには、投与用量
または治療単位(course)は、用量治療方式を通して間欠的であるよりもむしろ連
続的である。
トロンビンインヒビターは、通常、投与の目的形、例えば、
経口錠剤、カプセル、エリキシル、シロップなど及び慣用の医薬的な実務と合致
するように好適に選択された、好適な医薬的希釈剤、賦形剤またはキャリヤ(以
後、集合名詞的に「キャリヤ」材料と称する)と混合した活性成分として投与する
。
例えば、錠剤またはカプセルの形態での経口投与の場合には、活性薬剤成分は
、経口の、非毒性の医薬的に許容可能な不活性キャリヤ(例えば、ラクトース、
スターチ、蔗糖、グルコース、メチルセルロース、ステアリン酸マグネシウム、
リン酸二カルシウム、硫酸カルシウム、マンニトール、ソルビトールなど)と混
合され得;液状で経口投与するためには、経口薬剤成分は、任意の経口の、非毒
性の、医薬的に許容可能な不活性キャリヤ(例えば、エタノール、グリセロール
、水など)と混合され得る。さらに、所望によりまたは必要により、好適なバイ
ンダー、潤滑剤、崩壊剤及び着色剤も混合物中に含ませ得る。好適なバインダー
としては、スターチ、ゼラチン、天然の糖(例えば、グルコース、β-ラクトース
、コーン-甘味料)、天然及び合成ガム(例えば、アカシア、トラガカントまたは
アルギン酸ナトリウム)、カルボキシメチルセルロース、ポリエチレングリコー
ル、蝋などが挙げられる。これらの用量形で使用する潤滑剤としては、
オレイン酸ナトリウム、ステアリン酸ナトリウム、ステアリン酸マグネシウム、
安息香酸ナトリウム、酢酸ナトリウム、塩化ナトリウム等が挙げられる。崩壊剤
としては、これらに限定されないが、スターチメチルセルロース、寒天、ベント
ナイト、キサンタンガム等が挙げられる。
トロンビンインヒビターは、種々の病理学(ascular pathologies)の治療で
相乗効果を得るために、これらに限定されないが、フィブリノーゲンレセプター
アンタゴニスト、ヘパリン、アスピリン若しくはワルファリンを含む好適な抗凝
集剤または、プラスミノーゲン活性剤若しくはストレプトキナーゼなどの血栓溶
解剤と共に投与し得る。例えば、トロンビンインヒビターは、組織プラスミノー
ゲン活性剤-介在血栓溶解再潅流の効率を促進する。トロンビンインヒビターを
、まず最初に血栓形成後に投与し得、次いでその後組織プラスミノーゲン活性剤
または他のプラスミノーゲン活性剤を投与する。製造法
以下の合成例は、本発明の化合物を製造するために使用し得る。
実施例1による例示として(スキーム1)
出発物質の6-メチル-2-ヒドロキシ-ピリジン-3-カルボン酸を、段階Aでジフ
ェニルホスホリルアジド(DPPA)及びベンジルアルコールと反応させて、保護化ア
ミノピリジンを得る。これを段階Bにてt-ブチルブロモアセテートでアルキル化
させ、CBZ基を段階Cで触媒による水素化で除去する。得られたアミンを次いで
、段階Dで酸掃去剤としてピリジンと、好適な試薬、この場合にはベンジルスル
ホニルクロリドと反応させ、次いでt-ブチルエステルを段階Eで酸性条件下で除
去する。次いで、酸を段階Fでアミン、例えば、2,5-ジクロロベンジルアミンと

は、試薬、例えば、ジシクロヘキシルカルボジイミドまたは1-エチル-3-(3-ジメ
チル-アミノプロピル)カルボジイミドとのカルボジイミド法により実施し得る。
アミドまたはペプチド結合を形成する他の方法としては、本方法に限定されない
が、酸クロリド、アジド、混合酸無水物または活性化エステルを介する合成法が
挙げられる。典型的には、溶液相アミド結合化を実施するが、伝統的なMerrifie
ld法による固相合成法も代わりに実施し得る。1種以上の保護基の添加または除
去も典型的な実施である。
本方法の変形では、表示合成段階で好適な試薬または好適に置換された出発物
質の使用により、以下の広範な請求の範囲により考慮されたW、R3、R4及びR5基
が存在することができる。例えば、段階Aの出発物質は、R3の異なる操作可能な
値とするために、6-置換基として、エチル、イソプロピル、シクロプロピル、ト
リフルオロメチルなどを有し得る。同様に、段階Dで好適なアルキル化剤、カル
ボニル化剤、スルホニル化剤、尿素化剤などを使用することにより異なるW基が
存在し得る。例えば、アルキルハライド、アルコキシカルボニルハライド、アシ
ルハライド、アルキルスルホニルハライドまたはアルキルイソシアネートを使用
すると、W(ここで、WはR1、R1OCO、R1CO、R1SO2または(R1)m(CH2)nNCOである)の
対応する値を与える。同様に、段階Fのアミンを好適に選択すると、R4及びR5の
異なる操作可能な値とすることができる。同様の及び自明の変形をするための本
方法の自明の変形及び変異は、当業者には明らかであろう。
末端基として2-ヒドロキシベンジルアミンを含むか、またはその誘導体である
化合物の製造法は、スキーム2に示されており、実施例2により例示される。
4-クロロサリアルデヒド(chlorosalicaldehyde)をエタノール性炭酸ナトリウ
ム水溶液中、ヒドロキシルアミン塩酸塩と濃縮した。オキシムをロジウムなどの
触媒による水素化により還元し、アミンを標準条件下でそのBOC誘導体として保
護する。フェノールを段階Dでエチルブロモアセテートなどの酢酸塩等価物でア
ルキル化し、得られたエステルを水酸化リチウムで加水分解する。生成物カルボ
ン酸を段階Fでエチルアミン等のアミンと結合させ、BOC基を強酸により除去す
る。アミンをスキーム1、段階Eの生成物と結合させ、最終生成物を得る。スキーム2 本方法の変形により、以下の広範な請求の範囲により考慮された異なるR4及び
R5基が、表示合成段階の好適な試薬または好適に置換された出発物質を使用する
ことにより可能になる。例えば、段階Fのアミンを好適に選択することにより、
R7及びR8の異なる値とすることができる。本方法の種々の変異及び変形により、
同様及び異なる変異が得られることは当業者には公知である。
3-アミノ-4-トリフルオロメチル-2-ピリジノンの誘導体である化合物の製造法
をスキーム3に示す。段階Aにおいては、1,1,1-トリフルオロメチル-2,4-ペン
タンジオンをエタノール性ジエチルアミン中ニトロアセトアミドと濃縮させると
、2-ヒドロキシ-6-メチル-3-ニトロ-4-トリフルオロメチルピリジンが得られる
。これを段階Bで塩基として水素化ナトリウムを使用してTHF中、t-ブチルブロ
モアセテートなどの酢酸塩等価物でアルキル化し、ニトロ基を段階Cで、炭素上
のパラジウムなどの触媒を使用する水素化により還元する。t-ブチル基を段階D
でHClなどの強酸により除去し、得られたカルボン酸を段階Eでエチル-2-アミノ
メチル-4-クロロフェノキシアセトアミドと結合させ、最終生成物を得る。
本方法の変形では、表示合成段階の好適な試薬または好適に置換された出発物
質の使用により、以下の広範な請求の範囲により考慮された異なるR3、R4及びR5
基が存在することができる。例えば、段階Aの好適なジオン(dione)を使用する
ことにより、R3の異なる値とすることができる。同様に、段階Eのアミンを好適
に選択することにより、R4及びR5の異なる操作可能な値とすることができる。本
方法の自明の変異及び変形により、同様及び異なる変異が得られることは当業者
には公知である。スキーム3 以下の実施例は、本発明者らにより考慮されたものとして本発明を例示するた
めのものであり、本発明の趣旨または範囲を限定するものではない。
実施例1 3- ベンジルスルホニルアミノ-6-メチル-1-(メチレン-カルボキシアミドメチル-2 ,5-ジクロロフェニル)-2-ピリジノンの合成 段階A:3-ベンジルオキシカルボニルアミノ-6-メチル-2-ピリジノン
DPPA(35.6ml,165mmol)を2-ヒドロキシ-6-メチルピリジン-3-カルボン酸(22.9
7g,165mmol)及びトリエチルアミン(23.0ml,165mmol)の乾燥ジオキサン(300ml)
中の撹拌溶液に添加し、得られた溶液を加熱して還流した。16時間後、追加のト
リエチルアミン(23.0ml,165mmol)及びベンジルアルコール(17.1ml,165mmol)を
添加し、溶液をさらに24時間還流した。反応を真空濃縮させて、揮発分の殆どを
除去した。残渣を塩化メチレン
(500ml)と塩水(500ml)の間で分配し、1M HCl(165mmol)でpH1の酸性にした。水性
層を塩化メチレンで(2回)抽出し、混合した有機層を炭酸水素ナトリウム溶液と
塩水で洗浄し、乾燥(Na2SO4)し、真空蒸発させると茶色の固体が得られた。これ
をメタノールから再結晶させると、褐色固体の標記化合物が得られた。
段階B:3-ベンジルオキシカルボニルアミノ-6-メチル-1-(t-ブチル-メチレンカ ルボキシ)-2-ピリジノン
水素化ナトリウム(5.3g,0.22mol)を3-ベンジルオキシカルボニルアミノ-6-
メチル-2-ピリジノン(53.2g,0.20mol)の撹拌スラリーに0℃で添加した。t-ブチ
ルブロモアセテート(45ml,0.27mol)を得られた溶液に添加すると、すぐに沈澱
が形成した。反応を1時間かけて温め、2時間後、溶媒を真空除去し、残渣を1:
1の水/塩水(200ml)及び6:1のTHF/塩化メチレン(700ml)の間で分配した。有機層
を乾燥(Na2SO4)させ、真空蒸発させると固体が得られ、これをヘキサンですり砕
くと結
晶性固体の標記化合物が得られた。
段階C:3-アミノ-6-メチル-1-(t-ブチル-メチレンカルボキシ)-2-ピリジノン
3-ベンジルオキシカルボニルアミノ-6-メチル-1-(t-ブチル-メチレンカルボキ
シ)-2−ピリジノン(5.59g,15.0mmol)及びパールマン(Pearlman)触媒(0.56g)の4
:1エタノール/水(200ml)中の混合物をH2(50psi)下、パール装置中2時間振盪さ
せた。反応混合物をセライトを通して濾過し、真空蒸発させ、エタノールで共沸
させると固体の標記化合物が得られた。
段階D:3-ベンジルスルホニルアミノ-6-メチル-1-(t-ブチル-メチレンカルボキ シ)-2-ピリジノン
塩化ベンジルスルホニル(3.15g,16.5mmol)を3-アミノ-6-
メチル-1-(t-ブチル-メチレンカルボキシ)-2-ピリジノン(3.55g,14.9mmol)のピ
リジン(30ml)中撹拌溶液に0℃で添加し、得られた溶液を撹拌するうちに、濃厚
な沈澱が形成した。1時間後、反応混合物を真空蒸発させてペースト状とし、こ
れを塩化メチレンと10%硫酸水素カリウム溶液の間で分配した。有機層を乾燥(N
a2SO4)し、真空蒸発させると、淡桃色固体の標記化合物が得られた。
段階E:3-ベンジルスルホニルアミノ-6-メチル-1-メチレンカルボキシ-2-ピリ ジノン
3-ベンジルスルホニルアミノ-6-メチル-1-(t-ブチル-メチレンカルボキシ)-2
−ピリジノン(5.70g,14.52mmol)の酢酸エチル(60ml)中の撹拌溶液内に0℃で、H
Clで飽和した溶液が形成されるまで、HClを泡立たせた。室温で1.5時間後、濃厚
懸濁液が形成した。混合物を窒素で脱気し、濾過すると、固体の標記化合物が得
られた。
段階F:3-ベンジルスルホニルアミノ-6-メチル-1-(N-2,5-ジクロロベンジル-ア セトアミド)-2−ピリジノン
3-ベンジルスルホニルアミノ-6-メチル-1-メチレンカルボキシ-2-ピリジノン(
70mg,0.21mmol)、HOBT(34mg,0.25mmol)、2,5-ジクロロベンジルアミン(44mg,
0.25mmol)及びトリエチルアミン(0.070ml,0.50mmol)のDMF(2.5ml)中の撹拌混合
物にEDC塩酸塩(48mg,0.25mmol)を添加し、混合物を16時間撹拌した。反応を酢
酸エチルで希釈し、塩水で洗浄し、乾燥(Na2SO4)し、真空蒸発させると固体とな
った。粗な生成物をシリカ上のフラッシュカラムクロマトグラフィー(50%酢酸エ
チル/ヘキサン)で精製すると、結晶固体の標記化合物が得られた。
実施例2 3- ベンジルスルホニルアミノ-6-メチル-1-[エチル-(2-メチレンカルボキシアミ ドメチル-4-クロロフェノキシ)-アセトアミド]-2-ピリジノンの合成 段階A:4-クロロサリックアルデヒドオキシム
ヒドロキシルアミン塩酸塩(16.7g,0.24mol)及び炭酸ナトリウム(12.7g,0.12
mol)の水(120ml)中の溶液を、4-クロロサリックアルデヒド(25.0g,0.16mol)の
エタノール(160ml)中撹拌溶液に添加し、得られた溶液を加熱して還流させた。
1時間後、反応を冷却し、水(320ml)を添加し、得られた結晶性沈殿物を濾過に
より単離した。第2収穫分を同様に集め、混合した固体を乾燥させると、標記化
合物が得られた。
段階B:2-ヒドロキシ-5-クロロベンジルアミン
4-クロロサリアルデヒドオキシム(10g,58.3mol)及び5%Rh/C(2.0g)の濃硫酸(1
0ml)含有エタノール(100ml)中の混合物を、24時間、H2(60psi)下パール装置中
で振盪した。水(100ml)を添加し、混合物をセライトを通して濾過した。濾液を
生成物が溶液から結晶化するまで濃縮した。固体を濾過により集め、濾液をさら
に濃縮し、水を添加して第2収穫分を得、これを第1収穫分と混合して乾燥させ
ると、標記化合物が得られた。
段階C:N-t-ブトキシカルボニル-2-ヒドロキシ-5-クロロベンジルアミン
2-ヒドロキシ-5-クロロベンジルアミン(1.22g,4.77mmol重硫酸塩として)、(B
OC)2O(1.56g,7.16mmol)及びN-メチルモルフォリン(1.05ml,9.54mmol)のDMF(10
ml)中混合物を室温で5時間撹拌した。反応を水と酢酸エチルとの間で分配し、
有機層を5%KHSO4溶液で2回、炭酸水素ナトリウム溶液及び塩水で洗浄し、乾燥(
Na2SO4)させ、真空蒸発させると固体となった。粗な生成物を酢酸エチル/ヘキサ
ン(1:5,12ml)で再結晶さ
せると標記化合物が得られた。 段階D:エチル(2-t-ブトキシカルボニルアミノメチル-4-クロロフェノキシ)-ア セテート
N-t-ブトキシカルボニル-2-ヒドロキシ-5-クロロベンジルアミン(730mg,2.83
mmol)、Cs2CO3(923mg,2.83mmol)及びエチルブロモアセテート(0.314ml,2.83mm
ol)のDHF(5ml)中混合物を2時間撹拌した。粗な反応混合物を水と酢酸エチルと
の間で分配し、有機層を塩水で洗浄し、乾燥(Na2SO4)させ、真空蒸発させると油
状となった。これを次の段階で使用した。段階E:2-t-ブトキシカルボニルアミノメチル-4-クロロフェノキシ酢酸
段階Dからの生成物を1:1:1メタノール/THF/水(9ml)中に溶解させ、水酸化リ
チウム水和物(126mg,3.0mmol)を添加した。16時間後、揮発成分を真空除去し、
溶液を水で希釈し、酢酸エチルで洗浄し、十分量の塩水を添加してエマルション
を分散さ
せた。水性層を5%KHSO4溶液で酸性とし、塩化メチレンで抽出し、乾燥(Na2SO4)
させ、真空蒸発させると固体の標記化合物が得られた。
段階F:エチル-(2-t-ブトキシカルボニルアミノメチル-4-クロロフェノキシア セトアミド
EDC塩酸塩(249mg,1.3mmol)を、2-t-ブトキシカルボニルアミノメチル-4-クロ
ロ-フェノキシ酢酸(316mg,1.0mmol)、HOBT(176mg,1.3mmol)、エチルアミン塩
酸塩(106mg,1.3mmol)及びN-メチルモルフォリン(0.396ml,3.6mmol)のDHF(4ml)
中撹拌混合物に添加し、混合物を16時間撹拌した。反応を酢酸エチルと5%KHSO4
溶液との間で分配し、有機層を5%KHSO4溶液、水、NaHCO3溶液及び塩水で洗浄し
、乾燥(Na2SO4)させ、真空蒸発させると固体の標記化合物(333mg)が得られた。
これを次の段階で使用した。段階G:エチル-(2-アミノメチル-4-クロロフェノキシ)-アセトアミド
段階Fからのエチル-(2-t-ブトキシカルボニルアミノメチル-4-クロロフェノ
キシ)-アセトアミドを2:1の塩化メチレン/TFA(3ml)中に溶解させ、15分後、溶液
を真空蒸発させた。残渣を水に溶解させ、溶液を塩化メチレンで(2回)洗浄した
。水性層を飽和炭酸ナトリウム溶液で塩基性化し、NaClで飽和させた。混合物を
酢酸エチルで抽出し、有機層を乾燥(Na2SO4)させ、真空蒸発させると結晶性固体
の標記化合物が得られた。
段階H:3-ベンジルスルホニルアミノ-6-メチル-1-[エチル-(2-メチレンカルボ キシアミドメチル-4-クロロフェノキシ)-アセトアミド
DCC(103mg,0.50mmol)をエチル-(2-アミノメチル-4-クロロフェノキシ)-アセ
トアミド(137mg,0.57mmol)及び3-ベンジルスルホニルアミノ-6-メチル-1-メチ
レンカルボキシ-2-ピリ
ジノン(168mg,0.0.50mmol)の塩化メチレン(3ml)中の撹拌溶液に添加した。3時
間後、得られた濃厚な混合物をセライトを通して濾過し、生成物があまり溶解性
でないのでパッドを大容量の塩化メチレンで洗浄し、濾液を真空蒸発させると固
体となった。粗な生成物をシリカ上のフラッシュクロマトグラフィー(3:1ヘキサ
ン/アセトン、次いでメタノール/クロロホルム勾配液、2-4%メタノール)で精製
すると、結晶性固体の標記化合物が得られた。融点210〜213℃。
実施例2の方法を使用して以下の化合物を製造した。
実施例3 3- ベンジルスルホニルアミノ-6-メチル-1-[シクロプロピル-(2-メチレン-カルボ キシアミドメチル-4-クロロフェノキシ)-アセトアミド]-2-ピリジノン DCC(103mg,0.50mmol)をシクロプロピル-(2-アミノメチル-4-クロロフェノキ
シ)-アセトアミド(127mg,0.50mmol)及び3-ベンジルスルホニルアミノ-6-メチル
-1-メチレンカルボキシ-2-ピリジノン(168mg,0.50mmol)の塩化メチレン(2ml)中
の撹拌溶液に添加した。16時間後、得られた濃厚な混合物をセライトを通して濾
過し、1:1メタノール/クロロホルムで洗浄し、濾液を真空蒸発させると固体とな
った。粗な生成物をシリカ上のフラッシュクロマトグラフィー(3:1ヘキサン/ア
セトン、次いでメタノール/クロロホルム勾配液、2-4%メタノール)で精製すると
、結晶性固体の標記化合物が得られた。融点>200℃。 実施例4 3- ベンジルスルホニルアミノ-6-メチル-1-(2-メチレンカルボキシアミドメチル- 4-クロロフェノキシアセトアミド)-2-ピリジノン DCC(63mg,0.307mmol)を2-アミノメチル-4-クロロフェノキシアセトアミド(10
1mg,0.307mmol)及び3-ベンジルスルホニルアミノ-6-メチル-1-メチレンカルボ
キシ-2-ピリジノン(103mg,0.307mmol)の塩化メチレン(2ml)中の撹拌溶液に添加
した。66時間後、得られた濃厚な混合物をセライトを通して濾過し、1:1メタノ
ール/塩化メチレンで洗浄し、濾液を真空蒸発させると固体となった。粗な生成
物をシリカ上のフラッシュクロマトグラフィー(3:1ヘキサン/アセトン、次いで
メタノール/クロロホルム勾配液、2-5%メタノール)で精製すると、結晶性固体の
標記化合物が得られた。融点>200℃。
さらに、以下の実施例を実施した。実施例5 3- ベンジルスルホニルアミノ-6-メチル-1-(2-メチレンカルボキシアミドメチル- 4-クロロフェノキシ)-2-ピリジノン EDC塩酸塩(236mg,1.23mmol)を3-ベンジルスルホニルアミノ-6-メチル-1-メチ
レンカルボキシ-2-ピリジノン(415mg,1.23mmol)、HOBT(167mg,1.23mmol)、2-
ヒドロキシ-5-クロロベンジルアミン(195mg,実施例B段階Bの生成物)及びNMM(
0.136ml,1.23mmol)のDHF(1.2ml)中の撹拌混合物に添加し、混合物を3時間撹拌
した。反応を酢酸エチルで希釈し、水で洗浄し、乾燥(NaSO4)させ、真空蒸発さ
せると固体となった。
粗な生成物をシリカ上のフラッシュクロマトグラフィー(酢酸エチル/ヘキサン/2
%酢酸勾配液、50-80%酢酸エチル)で精製すると、結晶性固体の標記化合物が得ら
れた。融点>200℃。
実施例6 3- ベンジルスルホニルアミノ-6-メチル-1-[2,2,2-トリフルオロエチル-(2-メチ レンカルボキシアミドメチル-4-クロロ)フェノキシ]-2-ピリジノン DCC(25mg,0.12mmol)を2-(2,2,2-トリフルオロエチル)-5-クロロベンジルアミ
ン(29mg,0.12mmol)及び3-ベンジルスルホニルアミノ-6-メチル-1-メチレンカル
ボキシ-2-ピリジノン(61mg,0.18mmol)の塩化メチレン(0.7ml)中の撹拌溶液に添
加した。16時間後、得られた濃厚な混合物をセライトを通して濾過し、塩化メチ
レンで洗浄し、濾液を真空蒸発させると固体となった。粗な生成物をシリカ上の
フラッシュクロマトグラフィー(3:1ヘキサン/アセトン、次いで50%酢酸エチル/
ヘキサン)で精製すると、結晶性固体が得られ、これを塩化メチレンですり砕く
と、結晶性固体の標記化合物が得られた。融点>200℃。
実施例7 3- ベンジルスルホニルアミノ-6-メチル-1-(メチレンカルボキシアミドメチル-3- クロロフェニル)-2-ピリジノン DCC(43mg,0.21mmol)を3-クロロベンジルアミン(0.025ml,0.21mmol)及び3-ベ
ンジルスルホニルアミノ-6-メチル-1-メチ
レンカルボキシ-2-ピリジノン(70mg,0.21mmol)の塩化メチレン(1.0ml)中の撹拌
溶液に添加した。16時間後、得られた濃厚な混合物をセライトを通して濾過し、
塩化メチレンで洗浄し、濾液を真空蒸発させると固体となった。粗な生成物をシ
リカ上のフラッシュクロマトグラフィー(3:1ヘキサン/アセトン、次いで酢酸エ
チル/ヘキサン勾配液、60-100%酢酸エチル)で精製すると、結晶性固体が得られ
、これをメタノールですり砕くと、固体の標記化合物が得られた。融点176-17℃
実施例8 プロテイナーゼ阻害を測定するin vitroアッセイ
ヒトα-トロンビン及びヒトトリプシンのアッセイを、S.D.LewisらのThrombos is Research
,Issue No.70,173頁(1993)の実質的に記載通りに実施した。
アッセイは、25℃、0.05M TRIS緩衝液中、pH7.4、0.15M NaCl、0.1%PEGで実施
した。トリプシンアッセイも1mM CaCl2を含有していた。p-ニトロアニリド(pn
a)基質の加水分解速度を測
定するアッセイでは、Thermomax 96-ウェルプレートリーダーを使用してp-ニト
ロアニリンの時間に依存した発生を(405nmで)測定した。sar-PR-pnaを使用して
ヒトα-トロンビン(Km=125μM)及びウシトリプシン(Km=125μH)をアッセイ
した。p-ニトロアニリド基質濃度を8270cm-1M-1の吸光係数を使用して342nmの吸
収を測定することにより決定した。
トロンビンの阻害度が非常に高い、強い阻害剤(Ki<10nM)の特定の研究におい
ては、より感受性の高い活性アッセイを使用した。本アッセイにおいては、蛍光
基質Z-GPR-afc(Km=27μM)のトロンビン触媒加水分解速度を、7-アミノ-4−ト
リフルオロメチルクマリンの産生を伴う500nm(400nm励起)における蛍光の増加か
ら決定した。Z-GPR-afcの貯蔵溶液の濃度は、トロンビンによる貯蔵溶液のアリ
コートの加水分解完了時に産生した7-アミノ-4-トリフルオロメチルクマリンの3
80nmにおける吸収を測定することにより決定した。
活性アッセイを、終濃度≦0.1Kmに少なくとも10倍で基質の貯蔵溶液を酵素ま
たはインヒビターで平衡化した酵素含有溶液に希釈することにより実施した。酵
素とインヒビターとの間の平衡を達成するのに必要な倍率を対照実験より決定し
た。イン
ヒビターの非存在(V0)または存在(Vi)での産生物形成の最初の速度を測定した。
競合阻害及び単一体(unity)がKm/[S]、[I]/e、及び[I]/e(式中、[S]、[I]及びe
は、各々、基質濃度、インヒビター濃度及び酵素濃度を示す)と比較して無視し
得ると仮定し、酵素由来のインヒビターの解離の平衡定数(Ki)を、方程式1に示
される[I]のV0/Viの依存性から得ることができる。

本アッセイにより示される活性は、本発明の化合物が、不安定アンギナ、難治
療性アンギナ、心筋骨屈折、一過性虚血性発作、心房細動、血栓性脳卒中、塞栓
性脳卒中、深静脈血栓症、播種性血管内凝血及び再疎通化管の再閉塞または再狭
窄症に罹患している患者の種々の状態を治療するのに治療的に有効であることを
示している。Kiにより表される、以下の化合物の各々のヒトトロンビンに対する
阻害活性は、15nM未満である。これらは、少なくとも1800nMであるヒトトリプシ
ン(Kiにより表される)に対するそれらの阻害活性により明示されるように、選択
的化合物である。 実施例9 血栓閉塞を測定するためのIn Vivo研究
本出願人は、KurtzらのThrombosis Research,No.60,269頁(1990)に記載の
以下のラット塩化鉄アッセイを実質的に使
用して、本明細書中に請求する本化合物のin vivo研究を実施した。
トロンビンインヒビターまたは本発明のin vivo活性を決定するために使用す
るアッセイにおいて、オスのSprague-Dawleyラット(体重200-350g)をダイアル-
ウレタン溶液(0.1ml/100g体重、最初の時点)で麻酔をかけ、側面尾静脈(lateral
tailvein)に12インチ長のPE50チューブと結合させた23ゲージ針でカニューレ挿
入した。チューブをチューブアダプターにより三方弁に結合させた。塩水(対照)
または試験化合物を好適には、尾静脈カテーテルを介して投与した。気管開口術
を0.75インチ長のPE205チューブで実施した。右頸動脈を露出させ、1.33mm径の
ドップラーフロープローブを血管内に設置した。ヒートランプを使用して、体温
を37℃に保持した。
ラット(8-10匹/グループ)を無作為に0.028ml/分の速度で尾静脈を介して投与
した塩水または試験化合物の連続静脈内注入にかけた。処理注入は、フロープロ
ーブ末端の露出頸動脈上に35%FeCl3で飽和させたワットマンNo.1濾紙の3mm2片
をおく前に60分イニシエーションにかけた。血栓閉塞が起きなかった場合または
血管の血栓閉塞後30分後に停止した場合、FeCl3適
