AGENTE FITOSANITARIO ED UN SUO PARTICOLARE USO NEL TRATTAMENTO DELLE PIANTE INFETTE DA Xylella Fastidiosa. PLANT HEALTH AGENT AND ITS PARTICULAR USE IN THE TREATMENT OF PLANTS INFECTED BY Xylella Fastidiosa.
DESCRIZIONE DESCRIPTION
The present invention relates to a product and its particular use as a phytosanitary agent in plants, in particular in the treatment of Xylella fastidiosa bacterial infections in olive trees.
In recent years in Italy, numerous harmful organisms have been unintentionally introduced into the agricultural sector through the importation of infected plant material, such as the black mange of the potato (Synchytrium endobioticum), the bacterial cancer of the actinidia (Pseudomonas syringae) and the bacterium gram negative Xylella fastidiosa. It is estimated that since 2013, in the Puglia region in southern Italy, the bacterium Xylella fastidiosa has infected about 6.5% of the olive trees monitored, causing significant environmental and economic repercussions for farmers, as well as a dramatic decline in the production of oil from the region.
X. fastidiosa has been recognized as a quarantine pathogen capable of causing serious phytosanitary pandemics in the territories in which it establishes and was therefore included in the A1 list of the European and Mediterranean plant protection organization EPPO, directive 2000/29 / EC.
This bacterium, particularly dangerous in the agricultural sector, belongs to the Xanthomonadaceae family and is characterized by high genetic and phenotypic variability. In fact, four subspecies of Xylella are known which are capable of infecting about 150 different types of plants.
However, the attack mechanism is similar for all varieties of the bacterium, in fact Xylella, not being a sporogenous bacterium, is transmitted through vector insects, in particular those of the Cicadellidae family, which feed by sucking from the lymphatic vessels of the plants. These insects, feeding on an infected plant, then transmit the bacterium to a healthy plant.
Once transmitted to the healthy host plant, the bacterium multiplies in the xylem vessels, whose function is to carry the nutrient-rich raw sap from the roots to the stem and leaves.
Bacterial multiplication continues until these vessels are obstructed by creating a sort of gel that prevents the regular flow of raw sap.
As a consequence of this colonization, a series of biological alterations develop in the plant which can also determine the death of the plant itself.
Among the diseases caused by X. fastidiosa stand out the “Pierce's disease”, which on the vine can be destructive, the variegated chlorosis of citrus fruits and the complex of rapid drying of the olive tree. Equally serious diseases also affect almond, apple, pear, oleander and various species of forest species and ornamental plants.
The typical and most frequent symptoms of these diseases include leaf flaking, reduced growth and drying of branches and shoots.
Although this bacterium has been known for over a century, there is still no effective therapy to cure diseased plants.
To date, a sort of containment of the infection in the environment is used through essentially two practices.
The first practice is based on the fact that, not being able to act on the bacterium, an attempt is made to target the vector, that is the insects that spread the bacterium, through insecticidal treatments and often cutting the grass, to eliminate larvae and insects that are still young. .
However, the drawbacks related to the use of insecticides are well known which, at high doses, can cause significant damage also in humans, such as dizziness, headache, nausea, muscle spasms, convulsions, breathing difficulties, skin rashes, alteration of the endocrine system, reproductive problems, learning disabilities etc.
Furthermore, although the use of insecticidal products leads to a reduction in the spread of the bacterium, it is still necessary to use the second practice in combination to contain the propagation of the microorganism.
This second practice consists in reducing the bacterium reservoir by killing infected plants.
However, the felling involves huge losses both from the environmental point of view due to the eradication of plants, the olive trees, which for the most part are secular trees, and from the point of view of the supply of oil, because the number of olives decreases. they can be squeezed. Last but not least, especially from the point of view of farmers, the felling causes huge economic losses both for the lack of production and sale of oil and for the necessary waiting time between the cultivation of a new olive tree and the moment in which it reaches production regime.
The present invention intends to overcome the limitations and drawbacks of the treatments for the eradication of X. Fastidiosa, indicated above.

The coordination complex (I) of the invention was found to be particularly suitable for use as a phytosanitary agent for the treatment of bacterial and fungal pathogens in agriculture.
The term "phytosanitary agent" means an active substance or compositions of active substances, intended to exert a beneficial action on plants and vegetables.
Further, the active substance represents that compound or mixture of compounds which acts against a target pathogen. Advantageously, the coordination complex (I) of the invention applied to the infected plant is absorbable by the plant itself and can reach the xylem vessels where it exerts its antibacterial and antifungal function.
This ability to be absorbed by the plant is particularly advantageous especially for use in the treatment of plants infected with Xylella fastidiosa, in fact this bacterium nestles in xylem vessels and it is therefore necessary for the plant protection agent to be absorbed in order to perform its function.
According to the preferred embodiment of the invention, in the coordination complex (I) R <1> is oxygen, R <2> and R <3> are identical or different and independently of each other they are selected from hydrogen or a linear or branched C1-C6 alkyl group, i.e. having from 1 to 6 carbon atoms such as for example a methyl, ethyl, butyl, propyl, isopropyl, butyl, isobutyl, sec-butyl, terbutyl group etc .; R <6>, R <7> and R <8> are identical or different and independently of each other they are chosen from hydrogen, a hydroxyl group, a linear or branched C1-C6 alkyl group.
Some examples not to be considered limiting of the coordination complex (I), are reported below and indicated with general formula (II), (III), (IV), (V) and (VI), in which M is a metal of transition.

In the coordination complex (I) according to the preferred embodiment of the invention, the transition metal M is selected from the metals of the fourth period of the periodic table.
Advantageously, the transition metals of the fourth period have been shown to have particular relevance in the biological field, as they are often present in the catalytic sites of enzymes, favoring the action of these enzymes.
In particular, according to the preferred embodiment of the invention, the coordination complexes (I) of the invention in which the transition metal is selected from manganese, iron, cobalt, nickel, copper or zinc.
Preferably, the aforementioned transition metal is selected from copper and zinc.
Advantageously, the choice of copper as the transition metal allows the coordination complex (I) of the invention to be used as an anti-fungal phytosanitary agent, in fact copper is a known metal with antifungal properties.
As previously indicated, the coordination complex (I) of the invention is absorbable by the plant where it is applied and can therefore act as a vehicle for the copper complexed to it.
Still advantageously, the use of copper in the agricultural sector is also authorized in organic farming, and for some particular applications also in biodynamic agriculture.
With regard to zinc, experimental analyzes of the use of the coordination complex (I) complexed with zinc have shown that this complex has interesting antibacterial activity when applied to plants infected with Xylella fastidiosa.
Particularly preferred in the present invention is the use of the coordination complex of formula (II) 5-hydroxy-naphthalene-1,4-dione complexed with the aforementioned transition metal.
Advantageously, the organic part of the coordination complex of formula (II) is a compound already present in nature, its use as a phytosanitary agent therefore has a limited impact from the environmental point of view.
Still advantageously, the complexation with the aforementioned transition metal to form the coordination complex enhances the antibacterial activity of the 5-hydroxy-naphthalene-1,4-dione itself.
Furthermore, according to the preferred embodiment of the invention, the coordination complex is 5-hydroxy-naphthalene-1,4-dione complexed with copper.
According to a variant of the preferred embodiment of the invention, the coordination complex is 5-hydroxy-naphthalene-1,4-dione complexed with zinc.
The coordination complex (I) of the invention can be prepared starting from the C5-hydroxyl derivative of the coordination complex (I) in the non-complexed form, indicated in the following reaction with the number 1 and for which R <1>, R <2>, R <3>, R <6>, R <7>, R <8> have been defined previously.
The hydroxyl derivative 1 is reacted with a hydroxide of general formula Me (OH) n, in which Me is a metal and n is the oxidation number of this metal Me, so as to allow the hydrogen to be released from the hydroxyl group in C5 and the formation of the corresponding metal salt 2. To thus obtain the coordination complex (I) of the invention, the metal salt 2 is reacted in an equimolar quantity with a salt MXm, in which M is the transition metal which will form the coordination complex (I), X is a halogen and m is the oxidation number of the transition metal M.
Come sottoprodotto si ottiene il sale MeXn. MeXn salt is obtained as a by-product.
Particolarmente preferito è l’utilizzo dell’etanolo. The above reactions are carried out in a protic solvent indicated in the reaction scheme as Sol, such as for example water, ethanol or methanol. The use of ethanol is particularly preferred.
Preferably, the reaction between the metal salt 2 and the MXm salt is carried out under reflux conditions.
According to the embodiment in which the coordination complex has general formula (II) complexed with copper, the aforementioned complex is prepared starting from an ethanolic solution of 5-hydroxynaphthalene-1,4-dione, to which an ethanolic solution of sodium hydroxide. To this mixture CuCl2 dissolved in ethanol is slowly added. At the end of this addition it is refluxed for about 3-7 hours, preferably about 5 hours, then the solvent is cooled and evaporated at reduced pressure, thus obtaining 5-hydroxy-naphthalene-1,4-dione complexed with copper. The reaction by-product is instead NaCl.
The product obtained by the aforementioned reaction was structurally characterized by infrared IR spectrometry and MS mass spectrometric analysis, as detailed in example 2.
As previously indicated, the compound (VII) and the coordination complex (I) of the invention advantageously have antibacterial activity.
According to the preferred embodiment of the invention, the compound (VII) and the aforementioned coordination complex (I) are used in the treatment of bacterial infections from gram negative bacteria in plants.
Particolarmente preferito è l’utilizzo come agente fitosanitario nelle piante di olivo. Particularly preferred is the use as a phytosanitary agent in olive trees.
In particular, the compound (VII) and the coordination complex (I) of the invention were advantageously suitable for use as phytosanitary agents in the treatment of Xylella fastidiosa infections, preferably in the treatment of Xylella fastidiosa infections of olive trees.
In this treatment, the compound (VII) and / or the aforementioned coordination complex (I) can be applied on the trunk and / or on the leaves of the infected plants. Preferably, they are also applied on the soil surrounding the infected plant, up to a radius of about 1-2 meters from the trunk of the plant.
Still preferably, the compound (VII) and / or the coordination complex (I) are dissolved in an aqueous solvent prior to application on plants, to favor its dispersion on the plant itself and, if necessary, in the surrounding soil.
They are also applied each preferably in an amount comprised between 100-250 mg, preferably about 200 mg for each treated plant. Examples not to be considered limiting, of the application of the aforementioned coordination complex (I) on olive trees infected with Xylella fastidiosa, are shown in examples 3.1 to 3.3.
Furthermore, examples not to be considered limiting, of the application of compound (VII) on olive trees infected with Xylella fastidiosa, are reported in examples 3.4 and 3.5.
As already indicated above, it is hypothesized that part of the effectiveness of the compound (VII) and of the coordination complex (I) of the invention is due to the ability of their organic part to be absorbed through the trunk of the plant up to the xylem vessels, in which the bacterium lurks and subsequently destroying or promoting, through internal mechanisms of the bacterium itself, the destruction of the biofilm surrounding the microorganism itself, causing its death.
Regardless of the mechanism of action, the coordination complex (I) of the invention also has the advantage of being able to convey the transition metal to which it is coordinated, in particular within the trunk of the plant.
Further advantageously, when the transition metal is copper or other transition metal with antifungal activity, it can also exert this antifungal action on the treated plant.
Indeed, the coordination complex (I) acts as a vehicle for the transition metal. This vehicle, being absorbable by the trunk of the plant, allows the metal to exert its fungicidal action in depth. In particular, this ability of the coordination complex (I) to convey the metal within the trunk of the plant allows to obtain a fungicidal efficacy equal to or greater than that of the metal in the form of, for example, sulphate, oxychloride, hydroxide, etc. but, advantageously, using smaller quantities of free metal and therefore with a lower risk of phytotoxicity.
Tale caratteristica è particolarmente vantaggiosa soprattutto in agricoltura biologica. This feature is particularly advantageous especially in organic farming.
Another advantage of using the compound (VII) and the coordination complex (I) as plant protection agents is that they are harmless to insects, such as bees, as they are not toxic by contact or by ingestion.
Returning to the phytosanitary composition of the invention, the preferred vehicle is water, in particular double-distilled water, so as to be easily absorbed by the plant and for its non-existent environmental impact.
Furthermore, the phytosanitary composition of the invention may also include one or more excipients among the conventional ones that will be chosen according to the usual practice.
Such excipients may include, among others, co-formulant compounds which serve to reduce the concentration of the coordination complex, such as inert substances and diluents.
Furthermore, it is not excluded that the phytosanitary composition is in the form of an emulsion in water, preferably in the presence of surfactants, dispersants or other stabilizers to ensure its stability.
The recorded spectrum shows a good correspondence with the simulated one obtained from DFT calculations at the B3LYP / 6-311G (d) theory level, both as regards the number of bands and the respective absorption frequencies.
The processing of the spectra was performed using the OPUS 4.2 software, supplied with the instrument.
The value of the absorption frequencies must be considered with an error of ± 1cm-1.
The structural characterization of the coordination complexes used in the IR measurements was performed by mass spectrometry (MS) measurements with electrospray ionization (ESI).
The MS analysis was performed at atmospheric pressure by nebulizing a solution of the coordination complex in polar protic solvent, methanol and water through a high electric potential needle, capable of donating or acquiring a proton from the analyte molecule. Pseudo-molecular ions are formed, detectable under negative or positive ion conditions.
An ion trap analyzer (IT) was then used to differentiate the ions generated based on their mass / charge ratio (m / z).
The MS spectra were recorded with a BrukerEsquire-LCTM instrument, equipped with an electrospray source and an APCI analyzer, by carrying out the measurements in both positive and negative ions by direct infusion of a solution in water / acetonitrile.

From the assignment it results that the oxygen of C (4) = O coordinates through one of its free electron doublets with the empty d-type orbitals of the metal ion forming a bond that reduces its double bond character.
