Claims (7)
1. Postupak višestruke (multiplex) lančane reakcije polimeraze (PCR) koji obuhvaća:
umnožavanje više ciljnih nukleinskih kiselina pomoću ciljno-specifičnih ugniježdenih (engl. nested) parova početnica koji obuhvaćaju nizvodnu (engl. forward) vanjsku, nizvodnu unutarnju, reverznu unutarnju i reverznu vanjsku početnicu za svaki cilj u prvoj reakciji umnožavanja, gdje je koncentracija ugniježdene početnice 100-1000nM, i gdje barem jedna od navedenih unutarnjih početnica ima dodatne nukleotide koji omogućavaju dodatnu sekvencu koja nije specifična za ciljne nukleinske kiseline tako da će umnožavanje ciljnih nukleinskih kiselina takvom početnicom također u dobiveni amplikon uključiti vezno mjesto za zajedničku početnicu koja se, za razliku od ciljno-specifičnih početnica, može koristiti za daljnje umnožavanje nesrodnih amplikona ciljnih nukleinskih kiselina, čime se dobiva barem jedan amplikon nukleinske kiseline koji sadrži barem jedno vezno mjesto zajedničke početnice;
odvajanje barem jednog amplikona nukleinske kiseline od barem dijela početnica iz prvog umnožavanja; i
umnožavanje barem jednog amplikona nukleinske kiseline u drugoj reakciji umnožavanja pomoću barem jedne zajedničke početnice koja se veže za barem jedno vezno mjesto zajedničke početnice.
2. Postupak prema zahtjevu 1 gdje se jedna ili više ciljnih nukleinskih kiselina odabire iz skupine koju čine virusne, bakterijske i gljivične nukleinske kiseline.
3. Postupak prema zahtjevu 1 ili zahtjevu 2 gdje se jedna ili više ciljnih nukleinskih kiselina dobiva iz ljudskog kliničkog uzorka.
4. Postupak prema zahtjevu 1 ili zahtjevu 2 gdje se jedna ili više ciljnih nukleinskih kiselina dobiva iz životinjskog kliničkog uzorka.
5. Postupak prema bilo kojem od zahtjeva 1 do 4 gdje se barem jedna zajednička početnica odabire iz skupine koju čine SEQ ID NO: 1 (identifikacijski broj sekvence), SEQ ID NO: 2 i njihove kombinacije.
6. Postupak prema bilo kojem od zahtjeva 1 do 5 gdje korak odvajanja barem jednog amplikona nukleinske kiseline od barem dijela početnica iz prvog umnožavana dalje obuhvaća uzimanje malog uzorka iz dovršenog umnožavanja u prvom reakcijskom sustavu da se dobiju amplikoni za drugo umnožavanje u drugom reakcijskom sustavu.
7. Postupak prema bilo kojem od zahtjeva 1 do 6 gdje se ciljno-specifične početnice odabiru iz skupine koju čine SEQ ID NO: 3, SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 9, SEQ ID NO: 10, SEQ ID NO: 11, SEQ ID NO: 13, SEQ ID NO: 14, SEQ ID NO: 15, SEQ ID NO: 16, SEQ ID NO:18, SEQ ID NO: 19, SEQ ID NO: 20, SEQ ID NO: 21, SEQ ID NO: 23, SEQ ID NO: 24, SEQ ID NO: 25, SEQ ID NO: 26, SEQ ID NO: 28, SEQ ID NO: 29, SEQ ID NO: 30, SEQ ID NO: 31, SEQ ID NO: 33, SEQ ID NO: 34, SEQ ID NO: 35, SEQ ID NO: 36, SEQ ID NO: 38, SEQ ID NO: 39, SEQ ID NO: 40, SEQ ID NO: 41.
