Abstract
Cellulose acetate hydrogen phthalate containing 5-25 per cent weight of free carboxyl is prepared by dissolving phthalic anhydride in pyridine, adding cellulose acetate containing 33 per cent acetyl and heating on a steam bath for 14-18 hours. The reaction mixture is diluted with acetone, poured into 5 per cent hydrochloric acid and the resultant precipitate washed and dried. In the form of its water-soluble salts it may be incorporated in gelatin capsules which, after subsequent tanning in 1 per cent aqueous formaldehyde, to render them resistant to dissolution in the stomach, are thereby capable of rapid disintegration in the intestinal secretions. The sodium, potassium, ammonium and lithium salts and salts of organic amines such as diethanolamine, ethylene diamine, diethylene triamine and triethylene tetramine are specified.ALSO:A medicinal capsule shell consists of a tanned gelatin envelope which contains in dispersed admixture with the gelatin, particles of a material resistant to dissolution in the stomach but capable of dissolving in the intestinal secretions. Such materials are the water-soluble salts of cellulose acetate hydrogen phthalate containing 5-25 per cent free carboxyl and they may be used in the proportion of 5-10 per cent wt. of the shell. Examples are the sodium, potassium, ammonium and lithium salts, and the salts of organic amides such as diethanolamine, ethylene diamine, diethylene triamine and triethylene tetramine. In an example finely ground cellulose acetate hydrogen phthalate is mixed with sufficient sodium bicarbonate to neutralize the phthalate and the mixture dissolved in water Gelatin is soaked in aqueous glycerin, the phthalate mixture added and the product maintained at 600 DEG C. The phthalate should dissolve and the pH of the solution should not exceed 7-7.5. This solution is suitable for preparing capsules which are subsequently tanned as, for example, by immersion in 1 per cent aqueous formaldehyde solution followed by washing and drying.
