1281499 Benzodiazepine derivatives F HOFFMANN-LA ROCHE & CO AG 19 April 1971 [6 April 1970] 26331/71 Heading C2C Compounds of the general formula (A=CH 2 , CO ; Z=O, NH, N-(C 1-7 alkyl), N acyl; R<SP>1</SP>=H, halogen, NO 2 , CF 3 , C 1-7 alkyl, alkylthio or alkoxy ; R<SP>2</SP> = H, C 1-7 alkyl, hydroxyalkyl or aminoalkyl, C 4-6 cycloalkylalkyl, C 1-7 alkoxy-(C 1-7 alkyl), C 1-7 alkylamino-(C 1-7 alkyl), di(C 1-7 alkyl)amino-(C 1-7 alkyl); R<SP>3</SP>=C 2- 8 alkoxycarbonyl; Y = dimethylene or trimethylene optionally substituted by C 1-7 alkyl, chloromethyl, bromomethyl, C 1-7 alkoxy-(C 1-8 alkyl), di(C 1-7 alkyl)amino; R<SP>5</SP>=H, C 1-7 alkyl, pyridyl, phenyl optionally substituted by halogen, NO 2 , CF 3 , C 1-7 alkyl) are prepared by (a) cyclizing a compound of the formula or (b) reacting a compound of the formula with a compound of the formula (R<SP>41</SP>=H or optional substituent in Y group) in the presence of an acidic agent, optionally followed by alkylation or acylation (Z = NH), introduction of R<SP>2</SP> and/or epimer separation. The preparation of ethyl 2<SP>1</SP>-benzoyl-4<SP>1</SP>-chloromesoxalanilate 2-oxime and 7-chloro-1,3- dihydro - 2 - oxo - 5 - phenyl - 2H - 1,4 - benzodiazepine-3-carboxylic acid ethyl ester is described. The above compounds are sedatives, muscle relaxants and anticonvulsants and may be administered in the form of pharmaceutical preparations containing them, with or without other (unspecified) active ingredients, in association with a carrier.
