Abstract
The invention relates to novel polypeptides which partially or entirely correspond to the primary structure of, and possess one or more of the biological properties of, mammalian erythropoietin ("EPO"), which is characterised, in preferred forms, in that it constitutes the product of prokaryotic or eukaryotic host expression of an exogenous DNA sequence. The invention is illustrated by the fact that genomic DNA, cDNA or prepared DNA sequences, which partially or entirely encode the sequence of the amino acid residues of EPO, or analogues thereof, is/are incorporated into autonomously replicating plasmid or virus vectors, which are used for transforming or transfecting suitable prokaryotic or eukaryotic host cells such as bacteria, yeast or vertebrate cells in culture. After isolation from culture media or cellular lysates or fragments, the expression products of the DNA sequences exhibit, for example, the immunological properties and the biological activities of human or monkey EPO both in vitro and in vivo. The invention also relates to chemically synthesised polypeptides which share the biochemical and immunological properties of EPO. The invention also relates to improved methods for detecting specific single stranded polynucleotides in a heterologous cell or virus sample which has been prepared, for example, from DNA which is present in a plasmid-generated or virusgenerated cDNA or genomic DNA "library"
