La presente invención se relaciona con un
compuesto para el suministró de agentes activos, y en particular

organismo. Los ejemplos de barreras físicas son la piel y
diferentes membranas de órganos que deben ser atravesadas antes de
alcanzar un objetivo. Las barreras químicas incluyen, pero no se
limitan a, variaciones de pH, bicapas lipídicas, y enzimas de
degradación.
Estas barreras son particularmente
significativas en el diseño de sistemas orales de suministro. El
suministro oral de muchos agentes biológica o químicamente activos
sería la ruta de escogencia para administración a los animales si
no fuera por barreras biológicas, químicas y físicas tales coma la
variación del pH en el tracto gastcoiutestinai (GI), enzimas
digestivas poderosas y membranas gastrointestinales impermeables a
un agente activo. Entre los numerosos agentes que no están
típicamente sujetos a administración oral se encuentran los péptidos
biológica o químicamente activos, tal como la calcitonina y la
insulina; los polisacáridos, y en particular los mucopolisacáridos
incluyen, pero no se limitan a, heparina; heparinoides;
antibióticos; y otras sustancias orgánicas. Estos agentes se toman
rápidamente inefectivos o son destruidos en el tracto
gastrointestinal por causa de hidrólisis ácida, enzimas, o
similares.
Los primeros métodos para administración oral de
agentes farmacológicos vulnerables contaban con la administración
conjunta de adyuvantes (por ejemplo, resorcinoles y tensoactivos no
iónicos tales como el polioxietilén oleil éter y el
n-hexadecilpolietilén éter) para incrementar
artificialmente la permeabilidad de las paredes intestinales, así
como la administración conjunta de inhibidores enzimáticos (por
ejemplo, inhibidores pancreáticos de tripsina,
diisoprópilfluorofosfato (DFF) y trasilol) para inhibir la
degradación enzimática.
También han sido descritos liposomas como
sistemas para el suministro de droga para insulina y heparina. Ver,
por ejemplo, la patente estadounidense No. 4.239.754; Patel y
colaboradores (1976), FEBS Letters, Vol. 62, pg. 60; y Hashimoto y
colaboradores (1979), Endocrinology, Japón, Vol.. 26, pg. 337.
Sin embargo, se evita el amplio espectro de uso
de tales sistemas de suministro de droga debido a que: (1) los
sistemas requieren de cantidades tóxicas de adyuvantes o
inhibidores; (2) las cargas adecuadas de bajo peso molecular, esto
es de agentes activos, no están disponibles; (3) los sistemas
exhiben pobre estabilidad y una vida útil inadecuada; (4) los
sistemas son de difícil fabricación; (5) los sistemas fallan en
proteger al agente activo (carga); (6) los sistemas afectan en
forma adversa al agente activo; o (7) los sistemas fallan en
permitir o en promover la absorción del agente activo.
Más recientemente, se han utilizado microesferas
de polímeros artificiales de aminoácidos mezclados (proteinoides)
para suministrar productos farmacéuticos. Por ejemplo, la patente
estadounidense No. 4.925.673 describe microesferas portadoras
proteinoides que contienen droga así como métodos para su
preparación y uso. Estas microesferas proteinoides son útiles para
el suministro de una cantidad de agentes activos.
Aún existe la necesidad en el arte por sistemas
simples y económicos de suministro que se preparan fácilmente y que
pueden suministrar un amplio rango de agentes activos.
Se proveen compuestos y las composiciones que
son útiles en el suministro de agentes activos. Estas composiciones
incluyen al menos un agente activo, preferiblemente un agente
biológica o químicamente activo, y al menos uno de los siguientes
compuestos o sales de los mismos, especialmente un compuesto que
tiene la siguiente estructura:
La presente invención provee modalidades como
las identificadas más abajo y como las reivindicadas en las

aminoácidos por medio de métodos conocidos por aquellos capacitados
en el arte, con base en la presente divulgación y los métodos
descritos en las solicitudes estadounidenses de patente con seriales
Nos. 60/017.902, presentada el 29 de marzo de 1996; 08/414.654,

Por ejemplo, se puede preparar el compuesto por
medio de la reacción del ácido solo con el agente apropiado que

La cantidad de agente activo es una cantidad
efectiva para lograr el propósito del agente activo particular. La

La cantidad total del agente activo, y

a ser utilizado pude ser determinada por aquellos capacitados en el
arte. Sin embargo, sorprendentemente se ha encontrado que con
algunos agentes biológica o químicamente activos, el uso de los
portadores actualmente divulgados provee un suministro
extremadamente suficiente, particularmente en sistemas como el
oral, intranasal, sublingual, intraduodenal, rectal, vaginal, bucal,
oftálmico, o subcutáneo así como sistemas para atravesar la barrera
hematoencefálica. Por lo tanto, se pueden administrar cantidades
menores del agente biológica o químicamente activo que aquellas
utilizadas en las formas unitarias previas de dosificación o

La cantidad de portador en la presente
composición es una cantidad de suministro efectiva y se puede

Las formas unitarias de dosificación pueden
incluir también a cualquiera de los excipientes; diluyentes;

El compuesto y las composiciones objeto de la
invención son útiles para la administración de agente biológica o
químicamente activos a cualquiera de los animales tales como aves;
mamíferos, tales como primates y particularmente humanos; e
insectos. El sistema es particularmente conveniente para el
suministro de agentes química o biológicamente activos que de otra
manera serían destruidos o se volverían menos efectivos por las
condiciones encontradas por el agente activo en su zona objetivo
(esto es, el área en la cual se va a liberar el agente activo de la
composición para suministro) y dentro del organismo del animal para
el cual se administran ellos. Particularmente el compuesto y las

aquellos que no pueden ser suministrados ordinariamente por vía
oral.
Los siguientes ejemplos ilustran la invención

Preparación General de los Portadores. Se

Preparación del Compuesto 79. Método A.
Se cargó un balón fondo redondo de 1 L acondicionado con un
agitador magnético con el ácido
3-(4-aminofenil)propiónico (46,3 g, 0,28
moles, 1,17 equivalentes) e hidróxido de sodio acuoso 2 M (300 mL).
Se añadió en porciones cloruro de
2,3-dimetoxibenzoilo (48,0 g, 0,24 moles, 1,00
equivalentes) durante 1 hora a la solución agitada. Después de la
adición, se agitó la reacción durante 2,5 h a temperatura ambiente,
y se mantuvo el pH de la solución aproximadamente en 10 por medio
de la adición de hidróxido de sodio 10 M. Se aciduló entonces la
solución con ácido clorhídrico 1 M (3 x 100 mL), agua (100 mL), y se
secó al aire. Se redisolvió en acetona en ebullición
(aproximadamente 500 mL), se decoloró con carbón activado (3 g), y
se filtró. Se añadió agua (1,5 L) al filtrado para inducir la
formación de un aceite de color marrón. El aceite de color marrón
solidificó con agitación a temperatura ambiente durante 10 min. Se

Preparación del Compuesto 79. Método B.
Se acondicionó un balón de fondo redondo de tres bocas de 2 L con
un agitador magnético y dos embudos adicionales bajo una atmósfera
de argón. Se añadió al balón una suspensión del ácido
3-(4-aminofenil)propiónico (46,3 g, 0,28
moles, 1,17 equivalentes) en acetato de etilo (700 mL). Se cargó
una solución de cloruro de 2,3-dimetoxibenzoilo
(48,0 g, 0,24 moles, 1,00 equivalentes) en acetato de etilo (250 mL)
en uno de los embudos para adición, en adiciones gota a gota
durante 1 h. Se cargó posteriormente trietilamina (28,29 g, 0,28
moles, 1,00 equivalentes) a través del segundo embudo añadiendo gota
a gota durante 15 min. Se agitó la reacción a temperatura ambiente
durante 3 h, y se evaporó el solvente al vacío para producir un
aceite residual de color marrón. Se añadió agua (600 mL) al residuo
seguido por hidróxido de sodio (2 M, 500 mL), y se agitó la mezcla
a temperatura ambiente durante 3 horas. Se aciduló la solución de
color marrón resultante con ácido clorhídrico 2 M (aproximadamente 1
L). Después de enfriar la mezcla en un baño de hielo durante 1 h,
se formó un sólido de color amarillo y se lo recolectó por medio de
filtración. Se lavó el sólido con agua (3 x 1,5 L) y se recristalizó
a partir de una mezcla al 50% de etanol en agua (v/v) para producir
un compuesto 79 como un sólido de color canela (59.2 g, 68%).
Se prepararon también los compuestos 18, 32, 37.

Preparación de Compuesto 79. Método C. Se
cargó un balón de fondo redondo de 2 L equipado con un agitador
magnético y un condensador de reflujo con una suspensión del ácido
3-(4-aminofenil)propiónico (46,3 g, 0,28
moles. 1,17 equivalentes) en diclorometano (560 mL). Se añadió
clorotrimetilsilano (62,36 g, 0,57 moles, 2,05 equivalentes) en una
porción. y se calentó la mezcla a reflujo durante 1 h bajo
atmósfera de argón. Se le permitió a la reacción enfriarse hasta
temperatura ambiente y se la colocó en un baño de hielo
(temperatura interna <10ºC). Se colocó nuevamente el condensador
de reflujo con un embudo adicional que contenía trietilamina (42,50
g. 0,42 moles, 1,50 equivalentes). Se añadió gota a gota la
trietilamina durante 15 min, y el sólido de color amarillo que se
había formado durante la adición. Se reemplazó el embudo por otro
embudo para adición que contenía una solución de cloruro de
2,3-dimetoxibenzoilo (48,0 g, 0,24 moles, 1,00
equivalentes) en diclorometano (100 mL). Se añadió gota a gota la
solución durante 30 min. Se agitó la reacción en el baño de hielo
durante otros 30 min y a temperatura ambiente durante 1 h. Se
evaporó el diclorometano al vacío para producir un aceite de color
marrón. Se enfrió el aceite de color marrón en un baño de hielo y
se añadió una solución de hidróxido de sodio 2 M enfriada en hielo
(700 mL). Se removió el baño de hielo, y se agitó la reacción
durante 2 h hasta producir una solución clara de color marrón. Se
aciduló la solución con ácido sulfúrico 2 M (400 mL) y se almacenó
aproximadamente a 5ºC durante 1 hora. Se formó un sólido de color
amarillo y se lo recogió por medio de filtración. Se lavó el sólido
con agua (3 x 100 mL) y se lo recristalizó a partir de etanol en
agua al 50% (v/v) para producir el compuesto 79 como agujas de
color canela (64,7 g, 82%).

Preparación del Compuesto 35. Se enfrió
una solución de cloruro de O-acetilsaliciloilo
(24,68 g, 124 mmol, 1 equivalente) en tetrahidrofurano (300 mL) en
un baño de hielo. Se añadió gota a gota trietilamina (25 g, 2,2
equivalentes) a través de un embudo adicional. Se disolvió
clorhidrato de
metil-9-aminononaoato en DMF (190
mL, calentando ligeramente para disolver), cargado a través de un
embudo de adición y añadido gota a gota a la mezcla anterior. Se
agitó la reacción en el baño de hielo durante 20 min y a
temperatura ambiente durante 2 h. La evaporación del THF a presión
reducida produjo una solución de color rosa de DMF. Se enfrió la
solución de color rosa en un baño de hielo, y se añadió hidróxido

recristalizó a partir de etanol en agua al 50% para producir el
compuesto 35 (32 g, 87%) como un sólido completamente blanco.
Preparación del Compuesto 49. Se coloca
1-(2-hidroxifenil)-3-(4-metilbenzoato)-1,3-propano
diona (3,00 g,
0,0101 mmol) en un balón de fondo redondo de 100 mL que tiene acoplado un sistema de purga con argón, una barra de agitación magnética y un condensador enfriado con agua. Se añadieron ácido acético glacial (20 mL) y ácido sulfúrico concentrado (5 mL), y se inició el calentamiento de la mezcla. Se le permitió a la mezcla de reacción calentarse a reflujo durante 6 h antes de suspender el calentamiento. Se le permitió a la mezcla de reacción enfriarse hasta temperatura ambiente, y luego se la vertió en 100 mL de agua/hielo. Se agitó aproximadamente durante ½ h antes de filtrar la mezcla, y se aisló un sólido de color marrón. Se recristalizó el sólido de color marrón dos veces a partir de ácido acético, produciendo un compuesto 49 como un sólido de color canela (1,44 g. 53,8%).

Preparación del Compuesto 167. Se
disolvió 2-cumaranona (4,21 g. 0,0314 mol), con
agitación, en acetonitrilo (75 ml) en un balón fondo redondo de 250
m1 equipado con una barra de agitación magnética, purga de argón y
condensador de agua fría. Se añadieron trietilamina (3,18 g, 0.0314
mol) y ácido 8-aminocaprílico (5,00 g. 0,0314 mol),
y se formó una suspensión de color canela. Se inició el
calentamiento y se dejó la mezcla de reacción durante la noche a
reflujo. Después de calentar durante la noche, la cromatografla en
capa delgada de la mezcla de reacción (50% acetato de etilo/50%
hexano) indicó que la reacción se había completado. Se suspendió el
calentamiento, se le permitió a la mezcla de reacción enfriarse
hasta temperatura ambiente y se concentró al vacío. Se recogió el
residuo resultante en cloruro de metileno, y se lo lavó con dos
porciones de 100 ml de una solución de ácido clorhídrico 1 N. Se
secó la capa de cloruro de metileno con sulfato de sodio y se la
concentró al vacío. Se le permitió al sólido color canela resultante
secarse al vacío durante la noche, produciendo el compuesto 167
como un sólido de color canela (8,35 g, 70,4%).
Preparación del Compuesto 171. Se
disolvió
1,4-benzodioxan-2-ona
(3,93 g, 0,0262 mol), con agitación, en acetonitrilo (70 ml) en un
balón fondo redondo de 250 ml equipado con una barra de agitación
magnética, purga de argón y condensador de agua fría. Se añadieron
trietilamina (2,64 g, 0,0262 mol) y ácido
8-aminocaprílico (5,00 g, 0,0262 mol), y se formó
una suspensión de color canela. Se inició el calentamiento y se
dejó la mezcla de reacción aproximadamente durante 3 horas a
reflujo. En ese momento, la cromatografia en capa delgada de la
mezcla de reacción (50% acetato de etilo/50% hexano) indicó que la
reacción se había completado. Se suspendió el calentamiento, y se le
permitió a la mezcla de reacción enfriarse hasta temperatura
ambiente y se concentró al vacío. Se recogió el residuo resultante
en cloruro de metileno, y se lo lavó con una porción de 100 m1 de
una solución de ácido clorhídrico 1 N. En ese momento se notó que
se precipitó un sólido de color canela y se lo aisló por medio de
filtración. Se lavó el sólido de color canela con una porción
adicional de 100 ml de agua. Se le permitió al sólido color canela
resultante secarse al vacío durante la noche, produciendo el
compuesto 171 como un sólido de color canela (7,73 g, 95,6%).
Preparación del Compuesto 120. Se vertió
gota a gota una solución de 3,00 g (18,3 mmol) de
2-nitrofenilisocianato y 5 mL de tetrahidrofurano
durante 10 min en una solución enfriada en baño de hielo de 2,08
(13,1 mmol) de ácido 8-aminocaprílico, 1,40 mL de
NaOH 10 N y 40 mL de agua. Se agitó la mezcla de reacción 30 min
adicionales, se calentó a 25ºC y se la trató con una solución de
HCl al 3% hasta un pH de 5. Se filtró el precipitado de color
amarillo y se lo enjuagó con 100 ml de agua. Se recristalizó el
sólido de color amarillo en 2-propanol y agua para
producir 3,7 g del compuesto 120 como cristales de color amarillo
pálido.
Los compuestos 104-106 también
fueron preparados por medio de este procedimiento.
Preparación del Compuesto 133. Se calentó
una suspensión de 2,40 g (16,3 mmol) y 2,80 g (15,6 mmol) del ácido
4-(4-aminofenil)butírico en 20 mL de propilén
glicol, 2,40 mL (1,74 g, 17,3 mmol) de trietilamina y 10 mg (0,08
mmol) de dimetilaminopiridina a 140ºC. La mezcla se tomó en una
solución clara después de 5 min a 140ºC. Después de agitar durante
330 min, se enfrió la mezcla de reacción a 25ºC y se diluyó con 20
mL de agua. Se filtró la ftalimida sólida que se había formado. Se
aciduló el filtrado con una solución de HCl al 3%. Se filtró el
sólido resultante y se lo recristalizó a partir de
2-propanol y agua para producir 0,62 g del compuesto
133 como un sólido de color canela.
Preparación del Compuesto 138. Se calentó
a reflujo una solución de 1,73 g (12,9 mmol) de dialdehído ftálico,
2,04 g del ácido 8-aminocaprílico y 20 mL de ácido
acético, durante 10 min. Se enfrió la mezcla de reacción a 40ºC, se
diluyó con agua y se extrajo con CH_{2}Cl_{2} (2 x 20 mL). Se
lavó la fase orgánica con agua y salmuera, se secó sobre Na2SO4 y
se evaporó. Se disolvió el residuo en éter y se extrajo con NaOH 2
N. Se separaron las capas. Se aciduló la capa acuosa con HCl al 3% y
se extrajo con CH_{2}Cl_{2}. Se secó la fase orgánica sobre
Na_{2}SO_{4} y se evaporó. Sé recristalizó el residuo de color
amarillo a partir de acetonitrilo y agua para producir 1,25 g del
compuesto 138 como un sólido de color amarillo.
Preparación del Compuesto 140. Se calentó
una mezcla de 1,40 g (9,48 mmol) de anhídrido ftálico y 1,51 g
(9,48 mmol) del ácido 8-aminocaprílico a 150ºC
durante 5 min. Después de enfriar se obtuvieron 2,61 g del
compuesto 140 sólido.
También se preparó el compuesto 150 por medio de
este procedimiento.
Preparación del Compuesto 145. Se trató
una suspensión de 2,11 g (10,1 mmol) del ácido etil
carbamoilantranílico y 5 mL de CH_{2}Cl_{2} con 2,20 mL de
cloruro de oxalilo. Después de agitar durante 1 hora se removieron
los volátiles. En ese mismo momento, se trató una suspensión de 1,60
g (10,1 mmol) del ácido 8-aminocaprílico y 15 mL de
CH_{2}Cl_{2} con 2,60 mL (2,23 g, 20,5 mmol) de TMSCI. Se
calentó esta mezcla a reflujo durante 90 min, se la enfrió en baño
de hielo y se la trató con 4,30 mL (3,12 g, 30,9 mmol) de
trietilamina. Cinco minutos después, se añadió una suspensión del
residuo de la reacción del cloruro de oxalilo en 20 mL de
CH_{2}Cl_{2}. Se calentó la mezcla de reacción a 25ºC y se
agitó durante la noche. Después de acidulación de la mezcla con HCl
al 3%, se formó u sólido de color blanco. Se filtró el sólido y se
recristalizó a partir de EtOH y agua para producir 1,88 g del
compuesto 145. El compuesto 153 fue preparado también por medio de
este procedimiento.
Preparación del Compuesto 154. Se calentó
a reflujo durante 12 h una suspensión de 4,02 g (25,6 mmol) del
ácido
trans-4-aminometilciclohexanocarboxílico,
4,18 g (25,6 mmol) de anhídrido isatóico, 20 mL de
CH_{2}Cl_{2}, 20 mL de dioxano, y 4 mL de agua. Se enfrió la
solución a 25ºC y se extrajo con éter (4 x 20 mL). Se secó la capa
orgánica sobre Na_{2}SO_{4} y se concentró. Se recristalizó el
sólido resultante a partir de EtOH y agua para producir 4,95 g del
compuesto 154.
El compuesto 103 está disponible con Aldrich
Chemical Company, Inc., Milwaukee, WI.
Se prepararon composiciones para dosificación
intracolónica ("IC") que contenían 100 mg/kg del portador y 25
\mug/kg de la hormona paratiroidea en propilén glicol acuoso al
25% o una solución para dosificación por medio de alimentación oral
forzada ("PO") que contenía 400 mg/kg del portador y 100
\mug/kg de hormona paratiroidea en agua, con los portadores 9, 33,
35, 77, 79, 109, 110, 123, 136, 141 y 169. Las soluciones para
dosificación se designan como P-número del
portador-DS.

Claims (15)
1. Una composición farmacéutica que
comprende:
(A) al menos un agente activo; y
(B) un compuesto que tiene la fórmula
y/o una sal del
mismo.
2. La composición farmacéutica como se define en
la reivindicación 1, en donde dicho agente activo se selecciona del
grupo que consiste de un agente biológicamente activo, un agente
químicamente activo, o una combinación de los mismos.
3. La composición farmacéutica como se define en
la reivindicación 2, en donde dicho agente biológicamente activo
comprende al menos un péptido, un mucopolisacárido, un
carbohidrato, o un lípido.
4. La composición farmacéutica como se define en
la reivindicación 2, en donde dicho agente biológicamente activo se
selecciona del grupo que consiste de hormona de crecimiento humano,
hormona de crecimiento bovino, hormona de liberación del
crecimiento, un interferón, interleuquina-II,
insulina, heparina, heparina de bajo peso molecular, calcitonina,
eritropoyetina, factor naturético atrial, un antígeno, un anticuerpo
monoclonal, somatostatina, adrenocorticotropina, hormona que libera
gonadotropina, oxitocina, vasopresina, cromolín sódico,
vancomicina, hormona paratiroidea, desferrioxamina (DFO), o
cualquier combinación de los mismos.
5. La composición farmacéutica como se define en
la reivindicación 4, en donde dicho agente biológicamente activo
comprende un interferón, interleuquina-II, insulina,
heparina, heparina de bajo peso molecular, calcitonina, oxitocina,
vasopresina, vancomicina, DFO, hormona paratiroidea, o
combinaciones de los mismos.
6. La composición farmacéutica de acuerdo con la
reivindicación 1, en donde al menos un agente activo es una hormona
paratiroidea.
7. La composición farmacéutica de acuerdo con la
reivindicación 1, en donde al menos un agente activo es una hormona
de crecimiento humano recombinante.
8. La composición farmacéutica de acuerdo con la
reivindicación 1, en donde al menos un agente activo es
heparina.
9. Una forma de dosificación unitaria que
comprende
- (A)
- la composición farmacéutica como se define en cualquiera de las reivindicaciones 1 a 8; y
(B)
- (a)
- un excipiente,
- (b)
- un diluyente,
- (c)
- un desintegrante,
- (d)
- un lubricante,
- (e)
- un plastificante,
- (f)
- un colorante,
- (g)
- un vehículo de dosificación, o
- (h)
- cualquier combinación de los mismos.
10. La forma de dosificación unitaria como se
define en la reivindicación 9, que comprende una tableta, una
cápsula, o un líquido.
11. La forma de dosificación unitaria como se
define en la reivindicación 9 ó 10. en donde dicho vehículo de
dosificación se selecciona del grupo que consiste de agua,
1,2-propano diol, etanol, aceite de oliva, o
cualquier combinación de los mismos.
12. La composición farmacéutica como se define
en cualquiera de las reivindicaciones 1 a 8 para ser administrada
en forma oral, intranasal, sublingual, intraduodenal, subcutánea,
rectal, vaginal, bucal y/o oftálmica.
13. El compuesto
o una sal del
mismo.
14. Un método para preparar una composición
farmacéutica, dicho método comprendiendo mezclar:
(A) al menos un agente activo;
(B) el compuesto como se define en la
reivindicación 13; y
(C) opcionalmente, un vehículo de
dosificación.
15. La composición farmacéutica como se define
en cualquiera de las reivindicaciones 1 a 8 capaz de pasar un
agente biológicamente activo a través de la barrera hematoencefálica
de un animal.
