Opfindelsen angår en fremgangsmåde til fremstilling af nico-tinsyre ved hydrolyse af nicotinonitril i nærværelse af ammoniak og eventuelt et ammoniumsalt.
Nicotinsyre er en af bestanddelene af vitamin B-komplekset og fremstilles almindeligvis ved oxidation af en tilsvarende monoeller disubstitueret pyridin substitueret med lavere alkylgrupper, f.eks. beta-picolin eller 2,5-dimethylpyridin, med salpetersyre, svovlsyre, dichromat eller ved katalytisk oxidation med luft.
Nicotinamid fremstilles sædvanligvis enten ved dehydrering af nicotinsyre under tilstedeværelse af ammoniak eller ved partiel hydrolyse af nicotinonitril i svagt alkalisk eller vandig opløsning.
I beskrivelsen til USA-patent nr. 2.904.552 beskrives f.eks. anvendelse af svagt alkaliske katalysatorer, navnlig magnesiumoxid og 150480 2 j ordalkalimetalkarbonater, til katalyse af denne partielle hydrolyse. Det nævnes, at store mængder alkali vil reagere med den ækvivalente mængde nicotinamid under dannelse af salt af nicotinsyre sammen med ammoniak, og at alkalimaterialet også katalyserer yderligere hydrolyse af amidet til carboxylsyrens ammoniumsalt. Øjensynligt fremgår det dog af dette patentskrift, at der kun er tale om en partiel omdannelse til ammoniumsaltet, og der er således ikke anvist midler til opnåelse af en kvantitativ omdannelse af nicotino-nitril til ammoniumnicotinat og videre til fri nicotinsyre, hvilket da heller ikke er målet med de i dette patentskrift beskrevne metoder.
Fra Kirk-Othmer: Encyclopedia of Chemical Technology, 1952, bd.9, side 305, er det kendt, at ammoniumnicotinat ved opvarmning giver nicotinsyre, men heller ikke dette litteratursted anfører noget om, hvorledes man kvantitativt omdanner nicotinonitril til ammoniumnicotinat - specielt anføres det ikke, hvorledes dette kan gøres ved en kontinuerlig fremgangsmåde.
Som følge af at nicotinsyren skal anvendes som en del af et vitamin B-kompleks, er det nødvendigt, at den er af ret høj renhed. Fremstilling af nicotinsyre ved direkte oxidation af alkylpyridiner resulterer i, at produktet indeholder oxidationsprodukter eller nitrogenholdige forbindelser (når oxidationen gennemføres med salpetersyre), som alle skal adskilles fra syren, inden den anvendes som emæringstilskud. I kommercielle anlæg, der fremstiller nicotinsyre og/eller derivater heraf, er rensningsopgaven stor og udgør en stor del af produktionsprisen.
Formålet med den foreliggende opfindelse er at tilvejebringe en fremgangsmåde til fremstilling af nicotinsyre med høj renhed i tilnærmelsesvist kvantitativt udbytte ud fra nicotinonitril.
Dette opnås ved fremgangsmåden ifølge opfindelsen, hvilken fremgangsmåde er ejendommelig ved, at nicotinonitrilet og en vandig opløsning af ammoniak og eventuelt et ammoniumsalt opvarmes i en hydrolysereaktor til en temperatur på 121-205°C og i et tidsrum, der er tilstrækkeligt til at frembringe en i det væsentlige kvantitativ hydrolyse af nicotinonitrilet til ammoniumnicotinat, fortrins-. vis 2-8 timer, hvorefter reaktionsproduktet strippes med damp eventuelt tilsat en anden inert stripningsgas ved en temperatur på 93-232°C til en i det væsentlige kvantitativ omdannelse af ammonium-nicotinatet til ammoniak, der fjernes med stripningsgassen, og en 150480 3 nicotinsyreholdig opløsning, fra hvilken nicotinsyren fraskilles ved afkøling.
Fremgangsmåden kan udføres batchvis eller kontinuerligt. Det foretrækkes at den udføres kontinuerligt, og at filtratet fra nicotinsyrekrystallisationen recirkuleres, hvorved der kan opnås et udbytte af nicotinsyre på 100% baseret på den tilførte nico-tinonitril.
Fortrinsvis bringes nicotinonitril sammen med ammoniak og et vandigt recirkulationsprodukt indeholdende nicotinamid og ammo-niumnicotinat under dannelse af en reaktionsblanding. Reaktionsblandingen opvarmes til en temperatur på 121-205°C til gennemførelse af hydrolysen af nicotinonitrilet og nicotinamidet til ammoniumnicotinat. Ammoniumnicotinatet opvarmes for at dekomponere dette til nicotinsyre, idet dekomponeringen gennemføres under kontinuerlig fjernelse af udviklet ammoniak. Et produkt indeholdende nicotinsyre og en vandig opløsning af ammoniumnicotinat og nicotinamid udtages, og nicotinsyre skilles fra den vandige opløsning. Den vandige opløsning af ammoniumnicotinat og nicotinamid recirkuleres.
For at danne nicotinsyre indføres nicotinonitrilet i en hydrolysereaktor og hydrolyseres i nærværelse af en katalysator bestående af ammoniak og eventuelt ammoniumsalte til ammoniumsaltet af nicotinsyre. Hydrolysen gennemføres ved temperaturer på 121-205°c og under systemets autogene tryk. Ammoniumnicotinatet omdannes derefter til nicotinsyre ved stripning af ammoniak med damp eventuelt tilsat en anden inert gas.
Omsætningen kan gennemføres kontinuerligt i et trin, idet reaktionsblandingen kontinuerligt indføres i en øvre del af en hydrolysereaktor, medens damp eller anden inert stripningsgas kontinuerligt indføres i en nedre del af hydrolysereaktoren, og ammoniak kontinuerligt udtages (sammen med stripningsgassen) som topprodukt fra reaktoren, og en produktstrøm indeholdende nicotinsyre kontinuerligt fjernes fra den lavere del af reaktoren.
Reaktoren er fortrinsvis anbragt vertikalt.
Omsætningen kan også gennemføres som en totrinsproces, idet nicotinonitrilet hydrolyseres til ammoniumnicotinat, og ammoniumnicotinatet omdannes derefter til fri syre i et termisk dekompo-neringstrin under kontinuerlig fjernelse af ammoniak ved hjælp af damp som stripningsgas tilsat eventuelt andre inerte gasser. Den 150480 4 termiske dekomponering gennemføres ved temperaturer på mellem 93 og 232°C.
Hydrolysen gennemføres ved temperaturer på mellem 121 og 205°C med en opholdstid, der er tilstrækkelig til at frembringe en i det væsentlige kvantitativ hydrolyse af nicotinonitrilet til ammoniumnicotinat, fortrinsvis 2-8 timer, og især 2-6 timer.
Systemets autogene tryk vil variere alt efter damptrykket og temperaturen og koncentrationen af materialer i fødningen, men 2 ligger sædvanligvis mellem 7,0 og 21,1 kg/cm (100 og 300 psia).
I en foretrukken udførelsesform indføres ammoniakken i hydrolysereaktoren enten ved indsprøjtning i hydrolysefødningen eller ved særskilt indføring i reaktoren. Hydrolysereaktionen kan også fremmes ved recirkulering af ammoniumnicotinat eller ved indføring af andre ammoniumsalte i reaktoren eller begge dele. Ammoniakkens virkning kan øges ved at holde en høj ammoniakkoncentration i reaktorens øverste del, f.eks. ved indsprøjtning af yderligere ammoniak i reaktoren eller ved udtagning af en del af stripningsgassen, inden den når søjlens top (når stripningen og hydrolysen foregår samtidig). Tilsætning af ammoniak i en mængde på fra omkring 2 til 10 mol% baseret på det vand, der er tilstede i den totale blanding, resulterer i en betydelig forøgelse af hydrolysereaktionens hastighed.
Stripningsgassen er damp, men andre gasser, der er inerte i forhold til reaktanterne og materialerne i hydrolyseapparaturet, og som ikke kondenserer under de anvendte driftstemperaturer og -tryk, kan tilsættes. Egnede gasser omfatter nitrogen, carbonhy-drider, såsom methan og propan, og carbonfluorider, såsom Freonserierne.
Nicotinsyreproduktet udvindes ved afkøling af hydrolyseprodukterne, således at den fri syre udfælder. Syren renses derefter ved filtrering, vask, centrifugering og/eller lignende teknik. Genvundet modervæske indeholdende ammoniumnicotinat og intermediære hydrolyseprodukter, såsom nicotinamid, kan recirkuleres til hydrolysetrinnet.

Nicotinonitrilet indføres gennem røret 55 og indføres i blandingstanken 60, hvori det blandes med ammoniak (indført 5 150430 gennem røret 57) og en recirkuleret vandig opløsning af ammonium-nicotinat og andre intermediære hydrolyseprodukter indført gennem røret 59. Den samlede fødning passerer gennem røret 61 ind i den øverste del af en hydrolysereaktor 70. Damp indsprøjtes i fød-ningsrøret gennem røret 63, og også i et nedre område af reaktoren 70 gennem røret 65 for at tjene som stripningsgas. I reaktoren udsættes fødningen fra røret 61 for samtidig hydrolyse og * dampstripning. Topproduktet indeholder primært ammoniak og stripningsgas, som fjernes gennem røret 67 og 69. Ammoniakken kan føres til et ammoniakdestillationsapparat og renset recirkuleres til røret 57. Nicotinsyre er flygtig, og en del kan føres med ud i topproduktet; det kan genvindes i en refluxkondensator 68 og returneres til reaktoren gennem røret 71. En produktstrøm fjernes fra det nederste område af reaktoren gennem røret 73. Produkt-

strømmen indføres i rensningssektionen 72, hvori nicotinsyre udvindes på sædvanlig måde, såsom ved afkøling af produktet for at udfælde syren efterfulgt af centrifugering, filtrering, tørring, o.s.v. Nicotinsyreproduktet fjernes fra rensningssektionen gennem røret 75. Filtrat eller modervæske kan fjernes gennem røret 59 og recirkuleres til blandingstanken 60. Filtratet eller modervæsken kan være egnet til hydrolyse, da det indeholder intermediære hydrolyseprodukter, såsom nicotinamid og ammonium-nicotinat, der enten kan hydrolyseres til yderligere nicotinsyre eller i forbindelse med ammoniumnicotinat medvirke til katalyse af hydrolysereaktionen.
I ovennævnte anlæg gennemføres hydrolysen i en vertikalt anbragt reaktor med samtidig hydrolyse og dampstripning. Som tidligere nævnt, kan hydrolysen imidlertid også gennemføres i andre reaktortyper. I så tilfælde kan hydrolysen og dampstripningen blive gennemført i adskilte områder i reaktoren. Den totale proces forandres imidlertid ikke.
Til yderligere belysning af opfindelsen er følgende eksempler tilvejebragt.
Eksempel 1
En blanding indeholdende 30 vægtprocent nicotinonitril i 4M vandig ammoniak blev hydrolyseret ved 171°C og et overtryk på 11,9 atm. i 4 timer. Reaktionsproduktet, en vandig opløsning af ammoniumnicotinat (svarende til 84,1% af den påfyldte nicotino- 150480 6 nitril) og nicotinamid (svarende til 15,9% af den påfyldte nicotinonitril ), blev strippet med damp ved 171°C. Nicotinsyren blev indvundet ved afkøling af den strippede opløsning, filtrering og rekrystallisation fra vand og blev ved væskechromatografi analyseret til 100% ren nicotinsyre (tør). Smeltepunkt: 234-235°C (ifølge litteraturen: 235,2°C). Tungmetalindhold: 0. Den udvundne nicotinsyre udgjorde 57,9 mol% af den påfyldte nicotinonitril, medens 26,2 mol% blev tilbage i opløsningen som ammoniumnicotinat.
Det opløste ammoniumnicotinat og nicotinamid blev recirkuleret til hydrolysøren og tilført frisk nicotinonitril og vandig ammoniak» , hvorved et udbytte på 100% baseret på forbrugt nicotinonitril kunne opnås.
Eksempel 2
En blanding indeholdende 17 vægtprocent nicotinonitril i 4M vandig ammoniak blev hydrolyseret ved 171°C i 4 timer. Reaktionsproduktet, en vandig opløsning af ammoniumnicotinat (svarende til ca. 90 mol% af det påfyldte nicotinonitril) og nicotinamid, blev strippet med en blanding af damp/nitrogen ved 1 atm. og 100°C.
Ved afkøling af den strippede opløsning udskiltes et rent, hvidt nicotinsyreprodukt svarende til et udbytte på ca. 20 mol% af den påfyldte nicotinonitril. Den resterende del forekom i filtratet som opløst ammoniumnicotinat.
Eksempel 3
Fremstillingen af nicotinsyre ud fra nicotinonitril blev udført i et kontinuerligt arbejdende pilot-hydrolyseanlæg og illustreres af følgende eksempel.
Frisk nicotinonitril, vand, ammoniak og recirkuleret filtrat fra krystallisationsanlægget blev med de i tabel I angivne hastigheder pumpet til hydrolysøren, der havde en udvendig diameter på 7,6 cm og var 86,4 cm lang. Varme blev tilført eksternt. Hydrolysøren blev holdt ved en indledningstemperatur på 179°C og ved 192°C i toppen. Opholdstiden var 5,5 timer, og nicotino-nitrilomdannelsen var fuldstændig.
Hydrolysørens afgivelse blev pumpet til en pakket søjle, der var 2,5 cm i diameter og 267 cm i længden, og som virkede som stripper. Heri blev det i den nedre del af apparatet kontaktet med damp ved en temperatur på 171°C. Stripperens øvre del inde- 150480 7 holdt ammoniak og vand, som kontinuerligt blev fjernet fra appa-ratet. Stripperens nedre del indeholdt vand, nicotinsyre, nico-tinamid og noget ammoniumnicotinat. Der blev ikke opnået andre produkter.
Nicotinsyren blev udvundet fra sin opløsning ved hurtig afkøling til 0°C, centrifugering, vaskning af centrifugekagen med vand med en temperatur på under 4,4°C, og derpå tørret. Nicotinsyren blev, som vist i tabel I, udvundet i et udbytte på 100% baseret på tilførslen af frisk nicotinonitril. Filtratet og vand-vaskningerne, der indeholdt nogen nicotinsyre og ammoniumnicotinat, blev recirkuleret til hydrolysøren til undgåelse af produkttab .
