Claims (14)
1. Isolierte DNA, die ein Protein kodiert mit einem N-terminalen
Domäne und einer ligandenbindenden Domäne, worin
die Aminosäuresequenz der genannten DNA-bindenden Domäne des
genannten Proteins mindestens eine 80%ige Homologie mit der in SEQ ID Nr. : 3 gezeigten Aminosäuresequenz aufweist und,
die Aminosäuresequenz der genannten ligandenbindenden Domäne
des genannten Proteins mindestens eine 70%ige Homologie mit
der in SEQ ID Nr.: 4 gezeigten Aminosäuresequenz aufweist.
2. Isolierte DNA nach Anspruch. 1, dadurch gekennzeichnet,
dass die Aminosäuresequenz der genannten DNA-bindenden Domäne
des genannten Proteins mindestens eine 90%ige, vorzugsweise 95%ige, bevorzugter 98%ige und am bevorzugtesten eine 100%ige
Homologie mit der in SEQ ID Nr.: 3 gezeigten Aminosäuresequenz
aufweist.
3. Isolierte DNA nach Anspruch. 1 oder 2, dadurch gekennzeichnet,
dass die Aminosäuresequenz der genannten ligandenbindenen Domäne des genannten Proteins mindestens eine
75%ige, vorzugsweise 80%ige, bevorzugte 90%ige und am bevorzugtesten
eine 100%ige Homologie mit der in SEQ ID Nr.: 4 gezeigten Aminosäuresequenz aufweist.
4. Isolierte DNA nach einem der Ansprüche 1 bis 3, wobei
die genannte DNA ein Protein kodiert, das die Aminosäuresequenz der SEQ ID Nr. : 5, SEQ ID Nr.: 6, SEQ ID Nr.: 21 oder
SEQ ID Nr.: 25 umfasst. :v
5. Isolierte DNA nach einem der Ansprüche 1 bis 4, dadurch
gekennzeichnet; dass die genannte DNA die Nukleinsäuresequenz
der SEQ ID Nr.l:, SEQ IDNr. : 2, SEQ ID Nr.: 20 oder SEQ ID
Nr.: 24 umfasst. ..;..-'■

6. ReJcombinanter Expressionsvektor umfassend die DNA nach
einem der Ansprüche 1 bis 5.
7. Zelle, die mit der DNA nach einem der Ansprüche 1 bis 5 oder dem Expressionsvektor nach Anspruch 6 transfiziert wurde.
8. Zelle nach Anspruch 7, die eine stabile transfizierte
Zelllinie ist, die das Steroid-Rezeptorprotein nach einem der Ansprüche 9 bis 11 ausdrückt.
9. Protein, das durch die DNA nach einem der Ansprüche l
bis 5 oder einen Expressionsvektor nach Anspruch 6 kodiert wird.
10. Protein nach Anspruch 9, wobei das genannte Protein die Aminosäuresequenz der SEQ ID Nr -: 5, SEQ ID Nr.: 6, SEQ ID
Nr.: 21 oder SEQ ID Nr.: 25 umfasst.
11. Chimäres Protein mit einer N-terminalen Domäne, einem
DNA-bindenden Domäne und einer ligandenbindenden Domäne, dadurch
gekennzeichnet, dass mindestens eine der Domänen des Chimären Proteins aus einem Protein nach Anspruch 9 oder 10
stammt und wenigstens eine der anderen Domänen des Chimären Proteins aus einem anderen Rezeptorprotein der nuklearen Rezeptor-
Superfamilie stammt, unter der Voraussetzung, dass die DNA-bindende Domäne und die ligandenbindende Domäne des genannten
Chimären Proteins aus verschiedenen Proteinen stammt.
12. DNA, die ein Protein nach Anspruch Ii kodiert.
13. Verwendung einer DNA nach einem der Ansprüche 1 bis 5
oder 12, eines Expressionsvektors nach Anspruch 6, einer Zelle
nach Anspruch 7 oder 8 oder eines Proteins nach einem der Ansprüche 9 bis ii in einem Screening-Assay zur Identifikation
neuer Arzneimittel.
14. Verfahren zur Identifizierung funktioneller Liganden für
das Protein nach einem der Ansprüche 9 bis 11, wobei das genannte Verfahren die folgenden Schritte umfasst:
a) Einfügen in eine geeignete Wirtszelle 1) von DNA nach einem der Ansprüche 1 bis 5 oder 12, und 2) eines geeigneten
Reportergens, das funktionell an ein operatives Hormonantwort-Element gebunden ist, wobei das genannte
HRE durch das DNA-bindende Domäne des von der genannten DNA codierten Proteins aktiviert werden
kann,-
b) Die Wirtszelle aus Schritt a in Kontakt mit potentiellen
Liganden bringen, die sich wahrscheinlich mit dem ligandenbindenden Domäne des von der genannten DNA aus
Schritt a) kodierten Protein verbindet;
c) Überwachen der Expression des vom genannten Reportergen aus Schritt a) kodierten Proteins.
