Description
- Die Erfindung betrifft Polypeptide mit hervorragenden medizinischen Wirkungen, ein Verfahren zu ihrer Herstellung, sie enthaltende pharmazeutische Zusammensetzungen, als auch ihre Anwendung.
- Insbesondere betrifft die Erfindung Polypeptide der folgenden allgemeinen Formel (I):
- worin R¹ und R² gleich oder verschieden sein können, und jeweils ausgewählt sind aus Wasserstoff, Niederalkylgruppen mit 1 bis 6 Kohlenstoffatomen oder Niederalkenylgruppen mit 2 bis 6 Kohlenstoffatomen,
- A ist ausgewählt aus D-Met, D-Ser oder D-Thr,
- C ist ausgewählt aus L-Leu, L-Ile, L-Nle, L-tertleu, L-Met, L-Met(O), L-Ser oder L-Val,
- D und E können gleich oder verschieden sein und sind jeweils ausgewählt aus L-Arg, L-Lys, L-homoArg oder L-Orn und
- F ist ausgewählt aus -OH oder -NH&sub2;.
- Oder pharmazeutisch annehmbare Salze dieser Polypeptide und sie enthaltende Arzneimittel.
- Die Aminosäuren, die die vorliegenden Peptide bilden, umfassen D- und L-Aminosäuren. Sofern nicht anders angegeben sind die Aminosäuren L-Aminosäuren. Die hier verwendeten Symbole haben die gleichen Bedeutungen wie im Bereich der Peptidchemie allgemein üblich. Sie sind nämlich wie folgt:

- Die bei Untersuchungen des Mechanismus des von analgetischen Wirkungen von Morphin erhaltenen Ergebnisse legen nahe, daß der lebende Körper sogenannte endogene Morphin-ähnliche Substanzen enthält, die verschiedene vitale Empfindungen kontrollierten, wie solche von Schmerz und Geistestätigkeit. Nach einer Reihe von Untersuchungen wurden Enkephalin und Endorphin als opioide Peptide isoliert, und deren Strukturen wurden aufgeklärt. Hierauf wurden in diesem Bereich weitere intensive Untersuchungen gemacht und verschiedene opioide Peptide, wie β-Neoendorphin, β-Casomorphin, Kyotorphin, Dermorphin und Dynorphin wurden gefunden.
- Unter diesen ist Dynorphin ein Opioidpeptid mit der folgenden Strukturformel, die von einigen der vorliegenden Erfinder aufgeklärt wurde:
- H-Tyr-Gly-Gly-Phe-Leu-Arg-Arg-Ile-Arg-Pro-Lys-Leu-Lys-Trp-Asp-Asn-Gln- OH
- Dynorphin ist ein natürliches Opioidpeptid mit spezifischen Wirkungen auf einen K-Rezeptor und daher erwartet man seine Verwendung als Analgetikum ohne Nebenwirkungen, wie Toleranz oder Abhängigkeit.
- Dynorphin zeigt jedoch den Mangel, daß es als solches keine analgetische Wirkung ausübt, wenn es durch intravenöse Injektion verabreicht wird, da es im Blut instabil ist.
- Ferner besteht das Bedürfnis nach einem hoch aktivem Peptid mit einer kürzeren Kette, da Dynorphin ein Heptadecapeptid mit einer relativ langen Kette ist.
- Die Erfinder haben intensive Untersuchungen an Peptiden mit kürzeren Ketten als bei Dynorphin, die in der Lage sind, einen analgetischen Effekt bei Verabreichung durch sowohl intravenöse als auch subkutane Injektion auszuüben, und haben schließlich entdeckt, daß diese Aufgabe durch Verwendung von Peptiden der folgenden Formel (I), umfassend 7 Aminosäuren, gelöst werden kann.
- Die Erfindung stellt Polypeptide der folgenden allgemeinen Formel zur Verfügung:
- worin R¹ und R² gleich oder verschieden sein können, und jeweils ausgewählt sind aus Wasserstoff, Niederalkylgruppen mit 1 bis 6 Kohlenstoffatomen oder Niederalkenylgruppen mit 2 bis 6 Kohlenstoffatomen,
- A ist ausgewählt aus D-Met, D-Ser oder D-Thr,
- C ist ausgewählt aus L-Leu, L-Ile, L-Nle, L-tertLeu, L-Met, L-Met(O), L-Ser oder L-Val,
- D und E können gleich oder verschieden sein und sind jeweils ausgewählt aus L-Arg, L-Lys, L-homoArg oder L-Orn und
- F ist ausgewählt aus -OH oder -NH&sub2;,
- oder pharmazeutisch verträgliche Salze dieser Polypeptide.
- Beispiele von pharmakologisch verträglichen Salzen umfassen Salze anorganischer Säuren, wie Hydrochlorid, Sulfat, Hydrobromid und Hydrojodid wie auch Salze organischer Säuren, wie Maleat, Fumarat, Succinat, Acetat, Malonat, Citronat und Benzoat.
- Demnach ist eine Aufgabe der Erfindung die Bereitstellung neuer als Arzneimittel, insbesondere als Analgetika, geeigneter Peptide.
- Eine weitere erfindungsgemäße Aufgabe ist die Bereitstellung eines Verfahrens zur Herstellung der als Arzneimittel geeigneten neuen Peptide.
- Eine weitere erfindungsgemäße Aufgabe ist die Bereitstellung neuer Analgetika, die die neuen Peptide enthalten.
- Der Begriff "Niederalkylgruppe" in der obigen Definition von R¹ bis R¹³ betrifft geradkettige, verzweigte, zyklische oder Ring-haltige Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, wie Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, iso-Butyl, 2-Methylpropyl, tert-Butyl, Cyclopropylmethyl, n- Pentyl, 1-Ethylpropyl, Isoamyl und n-Hexylgruppen. Der Begriff "Niederalkenylgruppe" in der obigen Definition von R¹ und R² betrifft solche entsprechend den obigen Alkylgruppen jedoch mit einer Doppelbindung an einer Position. Der Begriff "Alkyl" in den 'α-N-Alkylderivaten' bei der Definition der Aminosäuren hat die gleiche Bedeutung wie bei der obigen Niederalkylgruppe.
- Die erfindungsgemäßen Verbindungen, bei denen mindestens einer der konstituierenden Aminosäuren eine α-N-Alkylaminosäure oder eine D-Aminosäure ist, sind frei von den ernsthaften Mängeln des Dynorphins und seinen Derivaten, nämlich, daß sie keine analgetischen Wirkungen als solche ausüben können, wen sie durch intravenöse Injektion verabreicht werden, da sie im Blut instabil sind. Die erfindungsgemäßen Verbindungen sind sehr wertvoll, da sie eine hohe Stabilität in vivo haben und praktisch als Analgetika geeignet sind.
- Die erfindungsgemäßen Peptide können nach jedem geeigneten Verfahren synthetisiert werden. Die geschützten Peptide können durch ein konventionelles Flüssigphasenverfahren oder Festphasenverfahren synthetisiert werden. Es ist im allgemeinen bevorzugt, daß die in den Seitenketten der Aminosäure vorkommenden funktionalen Gruppen geschützt sind. Alle Schutzgruppen werden auf der Endstufe entfernt. Die Schutzgruppen für die funktionalen Gruppen in den Seitenketten der Aminosäuren umfassen alle im Stand der Technik berichteten Schutzgruppen. Typische Beispiele davon umfassen Tosyl (TOS), Nitro (NO&sub2;), Benzyl (Bzl), tert-Butyl (But), Benzyloxycarbonyl (Z) und tert-Butoxycarbonyl (BOC) Gruppen.
- Als Schutzgruppen für die α-Aminogruppen in den Aminosäuren können alle Schutzgruppen, wie im Stand der Technik berichtet, verwendet werden. Es ist jedoch bevorzugt, die Kombination von Schutzgruppen so auszuwählen, daß die Schutzgruppen nur für die α-Aminosäuren selektiv ohne Einfluß auf der Schutzgruppen der funktionalen Gruppen in den Seitenketten entfernt werden können. Wenn z. B. eine tert-Butoxycarbonylgruppe als Schutzgruppe für die α-Aminogruppe verwendet wird, ist eine Benzyl- oder eine Benzyloxycarbonylgruppe zum Schutz der funktionalen Gruppe(n) in der (den) Seitenkette(n) bevorzugt. Wenn eine Benzyloxycarbonylgruppe als Schutzgruppe für die α-Aminogruppe verwendet wird, ist eine tert-Butyl- oder tert-Butoxycarbonylgruppe zum Schutz der funktionalen Gruppe(n) in der der (den) Seitenkette(n) bevorzugt. Wenn die Aminogruppe von Tyr am N-terminalen Ende dialkyliert ist, kann die Aminogruppe so wie sie ist ohne weiteren Schutz verbleiben. Zur Verhinderung einer Razemisierung wird das geschützte Peptid vorzugsweise durch ein Schrittverfahren synthetisiert, worin alle Aminosäuren in der Sequenz ausgehend vom C-terminalen Ende gebunden sind, oder ein Verfahren, worin die Fragmentkondensierung an der Gly-Position durchgeführt wird. Es ist auch möglich, eine Fragmentkondensation an jeder erwünschten Stelle durchzuführen.
- Entweder im Festphasenverfahren oder im Flüssigphasenverfahren gemäß der Erfindung werden die Schutzgruppen vom Peptid entfernt, und das letztere wird durch Wiederholung der Reaktionen in den folgenden Schemata unter Synthetisierung des Peptids gereinigt. Die Schritte dieses Verfahrens werden unter Bezugnahme auf das Flüssigphasenverfahren erläutert: Kondensation Entfernen der Schutzgruppe von der α-Aminogruppe
- worin X¹ und X² jeweils eine H oder eine Alkylgruppe bedeuten, Y¹ und Y² bedeuten jeweils eine Aminosäureseitenkette und R' und R" bedeuten eine Schutzgruppe oder einen Peptidrest.
- Die Peptidbindung kann nach jedem bislang berichteten Verfahren gebildet werden. Ein üblicher Verfahren umfaßt die Aktivierung der Carboxylgruppe einer Säurekomponente der allgemeinen Formel:
- R" - - HCOOH
- wie dem Azidverfahren, dem Dicyclohexylcarbodiimid (DCC) Verfahren, dem gemischten Anhydridverfahren oder dem aktivierten Esterverfahren, und Umsetzen der aktivierten Verbindung einer Aminkomponente der allgemeinen Formel:
- H - HCOR'.
- Die Reaktionsbedingungen, wie als Reaktionslösungsmittel und Temperatur, können je nach dem gewählten Verfahren zur Aktivierung der Carboxylgruppe variieren. Das gemischte Anhydridverfahren, welches ein typisches Kondensationsverfahren ist, wird wie folgt durchgeführt:
- eine Säurekomponente der allgemeinen Formel:
- R"- - HCOOH
- wird in einem aprotischen Lösungsmittel, wie Dimethylformamid, Tetrahydrofuran oder Ethylacetat gelöst, die erhaltene Lösung wird auf ca. -20ºC abgekühlt und dann werden äquimolare Mengen an N-Methylmorpholin und Ethylchlorcarbonat der Reihe nach zur Lösung zugegeben. 5 Minuten wird eine Aminkomponente der allgemeinen Formel:
- H - HCOR'
- in einer äquimolaren Menge zugegeben, und die Mischung wird bei -15 bis 0ºC 2 bis 5 Stunden gerührt und dann auf übliche Weise unter Erhalt eines geschützten Peptids der allgemeinen Formel
- behandelt.
- Das Entfernen einer Schutzgruppe erfolgt nach einem üblichen Verfahren, wie einem katalytischen Reduktionsprozeß, einem Verfahren, worin eine Säure verwendet wird, ein Verfahren, worin eine Base verwendet wird und ein Verfahren, worin Hydrazin verwendet wird. Ein bevorzugtes Verfahren ist ausgewählt aus diesen Verfahren je nach Typ der für die α-Aminogruppe verwendeten Schutzgruppe. Typische Verfahren umfassen solche, worin die Schutzgruppe durch katalytische Reduktion der Benzyloxycarbonylgruppe entfernt wird, und eines worin die tert-Butoxycarbonylgruppe mit Trifluoressigsäure entfernt wird. Nun wird eine Ausführungsform des Verfahrens zur Entfernung der tert-Butoxycarbonylgruppe mit Trifluoressigsäure in Einzelheiten beschrieben: 0,25 ml Anisol und 5 ml Trifluoressigsäure werden zu 1 g eines α-N-Butoxycarbonylpeptids der allgemeinen Formel zugegeben:
- unter Kühlung mit Eis. Die Mischung wird 60 Minuten gerührt und dann mit Ether unter Erhalt eines Trifluoracetats des Peptids der allgemeinen Formel
- behandelt.
- Dieses Produkt wird in einem Lösungsmittel gelöst und mit einem Amin, wie Triethylamin, unter Erhalt einer Verbindung, die der folgenden Reaktion unterworfen wird, neutralisiert.
- Nach Wiederholen der obigen Kondensation und Entfernen der Schutzgruppen für die α-Aminogruppen zur Verlängerung der Peptidkette, werden alle Schutzgruppen entfernt unter Erhalt des gewünschten Rohpeptids. Die Schutzgruppen werden nach einem üblichen Verfahren entfernt, wie einem katalytischen Reduktionsverfahren, einem Verfahren, worin flüssiges Ammoniak/Alkalimetall verwendet wird, ein Verfahren worin eine Säure verwendet wird, ein Verfahren worin eine Base verwendet wird, oder ein Verfahren, worin Hydrazin verwendet wird. In der Praxis wird des Verfahren je nach Typ der Schutzgruppe, die entfernt werden soll, ausgewählt. Eines der häufig verwendeten Verfahren umfaßt das Entfernen der Schutzgruppe mit Wasserstoffluorid (HF) wie folgt:
- 1 g des geschützten Peptids wird in ca. 30 ml HF in Gegenwart von 0,5 ml Anisol bei einer Temperatur von -15 bis 0ºC in einem geschlossenen Reaktionsgefäß gelöst. Die Lösung wird 60 Minuten gerührt und HF wird aus dem Reaktionssystem ausdestilliert. Der Rückstand wird mit Ether gewaschen und in Wasser gelöst. Die Lösung wird mit Amberlite IRA-93 (Essigsäuretyp) behandelt und unter Erhalt eines Rohpeptids gefriergetrocknet, aus dem die Schutzgruppen entfernt wurden.
- Das Rohpeptid kann nach einem konventionellen Verfahren, wie Ionenaustauschcromatographie, Gelfiltration, Partitionschromatographie, Gegenstromverteilungs- und Hochdruck-Flüssigchromatographie gereinigt werden. Die Reinigung wird z. B. nach dem folgenden Hochdruck-Flüssigchromatographie verfahren durchgeführt: 100 mg des Rohpeptids werden auf eine Säule mit einem Durchmesser von 20 mm und einer Länge von 250 mm, enthaltend Nudeosil 5 c 18 als Träger, beladen und dann mit 0,05 % HCl (H&sub2;O-CH&sub3;CN) eluiert. Fraktionen mit Peaks, entsprechend dem gewünschten Peptid, wurden durch Detektion bei einer UV von 210 nm gesammelt und unter Erhalt des gewünschten Peptids gefriergetrocknet.
- Die erfindungsgemäßen Verbindungen zeigen eine sehr starke Aktivität im Vergleich zu Dynophin A.
- Ferner haben sie eine starke Aktivität zur Inhibierung der Kontraktion der ilealen Longitalmuskeln bei Meerschweinchen und der Differentialleiter bei der Maus, verursacht durch elektrische Stimulierung.
- Die erfindungsgemäß erhaltenen Peptidverbindungen haben eine Opioidaktivität ähnlich zu solchen von Dynorphin, ihre Wirkungen sind ziemlich stark und es zeigen sich deutliche analgetische Wirkungen nach intravenöser oder subkutaner Injektion.
- Es ist sehr wertvoll, daß die erfindungsgemäßen Verbindungen starke analgetische Wirkungen nach systemischer Verabreichung durch intravenöse oder subkutane Injektion zeigen, wogegen Dynorphin und seine Derivate, wie soweit berichtet, kaum analgetische Wirkungen nach intravenöser Injektion zeigen, daß sie in Blut instabil sind.
- Die nach dem erfindungsgemäßen Verfahren erhaltenen Peptidverbindungen haben deutliche analgetische Wirkungen und sie sind als Arzneimittel in der therapeutischen Anwendung geeignet.
- Bei Verwendung der erfindungsgemäßen Verbindungen als Analgetika werden sie peroral oder parenteral verabreicht. Üblicherweise werden sie parenteral in Form von intravenöser, subkutaner oder intramuskulärer Injektion oder als Suppositorien oder Sublingualtabletten gegeben. Die Dosis variiert je nach Grad der Symptome, Alter des Patienten, Geschlecht, Körpergewicht, Empfindlichkeit, Verabreichungsart, Stadium der Krankheit, Abständen, Eigenschaften der medizinischen Präparation, Formulierung, Typ der Präparation und Typ des Wirkstoffes. Obwohl daher die Dosis nicht besonders eingeschränkt ist, ist sie im allgemeinen ca. 0,1 bis 1000 mg/Tag, vorzugsweise ca. 1 bis 300 mg/Tag für Erwachsene.
- Die erfindungsgemäßen Verbindungen können Injektionen, Suppositorien, Sublingualtabletten, Tabletten und Kapseln nach üblichen Verfahren, wie im Bereich der Pharmakologie eingesetzt, formuliert werden.
- Bei der Herstellung einer Injektion, werden Additive wie pH-Kontrollmittel, Puffer, Suspendierungsmittel, Solubilisierungsmittel, Stabilisatoren, Isotonisierungsmittel und Konservierungsmittel zum Wirkstoff zugegeben, und die erhaltene Mischung wird zu einer intravenösen, subkutanen oder intramuskulären Injektionsflüssigkeit nach einem üblichen Verfahren formuliert. Falls notwendig, kann die Mischung nach üblichen Verfahren lyophilisiert werden.
- Beispiele von Suspendierungsmitteln umfassen Methylzellulose, Polysorbat 60, Hydroxyethylzellulose, Acacia, Tragacanthpulver, Natriumcarboxymethylzellulose und Polyoxyethylensorbitanmonolaureat.
- Beispiele von Solubilisatoren umfassen Polyoxyethylen gehärtetes Castor-Öl, Polysorbat 80, Nicotinamid, Polyoxyethylensorbitanmonolaureat, Macrogol und Ethylester von Castor-Öl-Fettsäuren.
- Beispiele von Stabilisatoren umfassen Natriumsulfit, Natriumetasulfit und Ether. Beispiele der Konservierungsmittel umfassen Methyl-p-hydroxybenzoat, Ethyl-p-hydroxybenzoat, Sorbinsäure, Phenol, Cresol und Chlorcresol.
Claims (3)
1. Polypeptide mit der Formel (I):
worin R¹ und R² gleich oder verschieden sein können, und jeweils ausgewählt
sind aus Wasserstoff, Niederalkylgruppen mit 1 bis 6 Kohlenstoffatomen oder
Niederalkenylgruppen mit 2 bis 6 Kohlenstoffatomen,
A ist ausgewählt aus D-Met, D-Ser oder D-Thr,
C ist ausgewählt aus L-Leu, L-Ile, L-Nle, L-tertLeu, L-Met, L-Met(O),
L-Ser oder L-Val,
D und E können gleich oder verschieden sein und sind jeweils
ausgewählt aus L-Arg, L-Lys, L-homoArg oder L-Orn und
F ist ausgewählt aus -OH oder -NH&sub2;
oder ein pharmazeutisch verträgliches Salz des Polypeptids.
2. Analgetische Zusammensetzung, umfassend als Wirkstoff eine
therapeutisch wirksame Menge des Polypeptids der allgemeinen Formel (I) gemäß
Anspruch 1 oder ein pharmazeutisch verträgliches Salz des Polypeptids in
Kombination mit einem pharmazeutisch verträglichen Vehikel.
3. Verwendung der Polypeptide gemäß Anspruch 1 zur Herstellung einer
pharmazeutischen Zusammensetzung, die als Analgetikum geeignet ist.
