Die Erfindung bezieht sich auf Platinkomplexverbindungen, auf pharmazeutische Stoffzusammensetzungen, die sie enthalten,
und auf ihre Verwendung bei der Behandlung von bösartigen Tumoren oder bösartigen Neoplasmen.
Gemäß einem ersten Aspekt der vorliegenden Erfindung enthält eine entsprechende Stoffzusammensetzung eine Platinkomplexverbindung
mit der Formel:

worin χ 1,2 oder 3 ist und die R-gruppen gleich oder unterschiedlich
sind und ausgewählt sind aus folgenden Verbindungen: Wasserstoff, substituierten oder nicht substituierten gerade-
oder verzweigtkettigen Alkyl-, Aryl-, Alkaryl-, Aralkyl-, Alkenyl-, Cycloalkyl- und Cycloalkenyl-, Halogen-, Pseudohalogen-
(wie nachfolgend definiert), Hydroxyl-, Formyl-, Amido-, Amino-, Alkoxy-, Aryloxy-gruppen, Sulfonsäuren oder
-Salzen, Carbonsäuren, -Estern oder -Salzen oder wobei zwei R-gruppen zusammen durch Sauerstoff oder Schwefel ersetzt sind,
und die vorhandenen X-gruppen in der obigen Formel II) Halogen-, Pseudohalogen- oder Hydroxylgruppen sind.
Wo eine oder mehrere der R-gruppen Carboxylate oder ein Derivat davon sind wie Ester, deren allgemeine Formel CR2 ..CO- ist,
kann es ein substituiertes Carboxylat sein, so daß η eine ganze
Zahl zwischen 1 und einschließlich 9 ist und die R -gruppen gleich oder unterschiedlich sind und ausgewählt werden aus folgenden
Verbindungen: Wasserstoff, substituierten oder nicht substituierten gerad- oder verzweigtkettigen Alkyl-, Aryl-,
Alkaryl-, Aralkyl-, Alkenyl-, Cycloalkyl- und Cycloalkenyl-, Halogen-, Pseudohalogen- (wie nachfolgend definiert), Hydroxyl-,
Alkoxy-, Aryloxy-, Formyl-, Nitro-, Amido-, Amino-gruppen und Sulfonsäuresalzen. Die obenstehende Definition soll auch Sauerstoff
und Schwefel einschließen, so daß ein doppeltgebundenes Sauerstoff- oder Schwefelatom zwei R -gruppen ersetzt.

Beispiele für einzelne Aminosäuren, die verwendet werden können, um einen Komplex mit Platin gemäß der Erfindung zu
bilden, sind Glycin (NH2CH2CO2H), Alanin (CH3CH(NH2)(CO2H),
Valin (CH3J3 CHCH(NH2) CO3H), Phenylalanin
Asparaginsäure (HO2CCH2CH(NH2)CO2H), Asparagin
(CO2H) und Cystein (HSCH2CH(NH2)CO2H), Lencin (CH3J3 CHCH2CH
(NH0)CO0H und Glutaminsäure HO0C(CH0)OCH(NH0)CO0H.

3-Aminobutyrylsäure und 4-Aminobutyrylsäure sind Beispiele
für β - und oC- Aminosäuren beziehungsweise für solche, die
verwendet werden können, um einen Platinkomplex gemäß der Erfindung bilden zu können.
Der Ausdruck Pseudohalogen in dieser Spezifikation hat die
Bedeutung, die auf S. 560 von "Advanced Publishers, 1966" angegeben ist als "ein Molekül, das aus mehr als zwei elektro
negativen Atomen besteht, die in freiem Zustand den Halogenen ähneln; diese Pseudohalogene bilden Anionen, die im Verhalten
den Halogenidionen ähneln". Beispiele für geeignete Pseudohalogene sind Cyanid, Cyanat, Thiocyanat und Azid.
Man fand heraus, daß Verbindungen der vorliegenden Erfindung aktiv wirksam gegen Krebs, hösartige Tumoren oder bösartige
Neoplasmen sind. Normalerweise wird die Verbindung deshalb in Zusammenhang mit einem pharmazeutisch annehmbaren Träger
verwendet.

Demgemäß sieht die Erfindung in einem zweiten Aspekt eine
pharmazeutische Stoffzusammensetzung vor, die eine Verbindung
gemäß dem ersten Aspekt der Erfindung und einen pharmazeutisch
für besagte Verbindung annehmbaren Träger enthält/ vor; diese Stoffzusammensetzung kann so formuliert werden, daß sie sich
z.B. für parenterale oder orale Anwendung bei Tieren eignet, die von einem bösartigen Tumor oder bösartigem Neoplasmä
befallen sind.
Präparative Einzelheiten von Platinkomplexen gemäß der Erfindung werden nun an Hand von Beispielen beschrieben werden.
cis-bis-(Glycinato)-platin (II)

K2 ιPtCl. J (50g) in 200 ml heißem Wasser wurde mit
Aktivkohle vernetzt und durch ein vorgeheiztes Sinterfilter
in Glycin (53,52 g) und KOH (40 g) (6 VaI) in 100 ml warmem
Wasser filtiert. Die Lösung wurde in ein Becherglas überführt und auf einer Heizplatte 20 Minuten lang erhitzt, um die Lösung
zu entfärben. Die Lösung wurde dann auf Raumtemperatur abgekühlt und heftig gerührt, während langsam konzentrierte
HCl (30 ml) zugegeben wurde. Der ausgefällte weiße Komplex wurde durch ein Sinterfilter mit der Porenweite 3 abfiltriert,
reichlich mit Wasser gewaschen und bei 50° C im Vakuum getrocknet.

Der weiße Feststoff wurde in 250 ml Wasser bei 80° C drei Stunden lang suspendiert, die abgekühlte Lösung durch ein
Sinterfilter mit der Porenweite 3 filtriert und das Produkt mit Wasser gewaschen und im Vakuum bei 60° C getrocknet.
Ausbeute = 29.90 g (73 %)

Eine Lösung von K2 jPtCl.J (50 g) in heißem Wasser (250 ml)
wurde zu Alanin (42,34 g) und Kaiiumhydroxid (26,68 g) in
heißem Wasser (75 ml) gegeben und die sich ergebende Lösung auf einer Heizplatte erhitzt, bis sie blaßgelb geworden war
(annähernd eine Stunde). Dann wurde tropfenweise konzentrierte HCl (9,9 ml) zugegeben, um den pH-Wert der Lösung auf annähernd

drei zu bringen und die Lösung auf einer Heizplatte vier Stunden lang auf ein Volumen von 325 ml erhitzt. Der weiße
Niederschlag aus der gekühlten Lösung wurde auf einem Sinterfilter mit der Porenweite drei abfiltriert, mit Wasser,
Äthanol und Äther gewaschen und im Vakuum bei 60° C getrocknet.
Ausbeute = 8,8 g (20 %)
Weitere Kristalle von eis- iPt (an)2j wurden durch Eindampfen
der Mutterflüssigkeit auf 225 ml und nachfolgendes Abkühlen auf Raumtemperatur erhalten. Die Gesamtausbeute waren 13 g (30 %)

Stoffzusammensetzung, dadurch gekennzeichnet, daß eine Platinkomplexverbindung die Formel hat:I.R1-FUN NR1R05 6 \ /12.,PtII.C Il 0-O 0R5R6N(R7R8C)x Pt'C 1! 0- 019.4.79- C 0(CR3R4).0 - C Jl 0909844/0904worin χ 1,2 oder 3 ist und die R-gruppen gleich oder unterschiedlich sind und ausgewählt sind aus folgenden Verbindungen: Wasserstoff, substituierten oder nicht substituierten gerad- oder verzweigtkettigen Alkyl-, Aryl-, Älkaryl-, Aralkyl-, Alkenyl-, Cycloalkyl- und Cycloalkenyl-, Halogen-, Pseudohalogen-, Hydroxyl-, Formyl-, Amido-, Amino-, Alkoxy-, Aryloxy-gruppen, Sulfonsäure oder -Salz und Carbonsäure, -Ester oder -Salz, oder worin zwei R-gruppen zusammen durch Sauerstoff oder Schwefel ersetzt sind, und die in Formel II) vorhandenen X-gruppen Halogen-, Pseudohalogen- oder Hydroxylgruppen sind.2. Stoffzusammensetzung nach Anspruch 1, dadurch gekennzeichnet, daß eine oder mehrere R-gruppen Carboxylat oder ein Derivat davon sind.3. Stoffzusammensetzung nach Anspruch 2, dadurch gekennzeichnet, daß die R-gruppe ein Ester mit der allgemeinen Formel ist:CR1 ... C0~
η 2η+Ί l*worin η eine ganze Zahl zwischen 1 und einschließlich 9 ist und die R-gruppen gleich oder unterschiedlich sind und ausgewählt sind aus folgenden Verbindungen:J 21 P 20019.4.79 - 3 -909344/0904- 3 - 2916U3Wasserstoff, substituierten oder nicht substituierten gerad- oder verzweigtkettigen Alkyl-, Aryl-, Alkaryl-, Aralkyl-, Alkenyl-Cycloalkyl- und Cycloalkaryl-, Halogen-, Pseudohalogen-, Hydroxyl-, Alkoxy-, Aryloxy-, Formyl-, Nitro-, Amido-, Amino-gruppen und SuIfonsäuresalzen.4. Stoffzusammensetzung nach Anspruch 1 oder Anspruch 3, dadurch gekennzeichnet, daß zwei R -gruppen durch ein doppelt gebundenes Sauerstoff- oder Schwefelatom ersetzt sind.5. Stoffzusammensetzung nach Anspruch 1, dadurch gekennzeichnet, daß die Aminosäure, die dazu verwendet wird, um einen Komplex mit dem Platin zu bilden, aus den Gruppen ausgewählt ist, die bestehen aus: Glycin (NH2CH2CO2H), Alanin (CH3CH(NH2)CO2H), Valin ((CH3J3 CHCH (NH2)CO2H), Phenylalanin (PhCH2CH(NH2)CO2H), Asparaginsäure (HO2CCH2CH(NH2)CO2H), Asparagin (NH2COCH2Ch(NH2)CO2H und Cystein6.
