Description
Die Erfindung betrifft Cyclopenten-Derivate der ίο allgemeinen Formel I
worin R1 eine Alkylgruppe mit 1 —3 Kohlenstoffatomen und R2 eine Hydroxymethyl-, Acetoxymethyl-
oder Formylgruppe darstellen und die geschlängelte Linie am C-Atom 1 epimere Formen bedeutet
2. Verbindung nach Anspruch 1, worin R1 eine
Methyl-, Ethyl- oder n-Propylgruppe bedeutet und R2 und die weiteren Symbole die in Anspruch 1
angegebenen Bedeutungen besitzen.
3. Verwendung der Verbindungen nach Anspruch 1 oder 2 als Riechstoffe und in Parfürnkornpositionen
in üblichen Mengen, insbesondere 1 bis 10%.
4. Verfahren zur Herstellung von Verbindungen
nach Anspruch 1 und 2, dadurch gekennzeichnet, daß man Campholenaldehyd in an sich bekannter Weise
durch Wittig-Reaktion mit tx-Brom-alkansäureestern mit 3 bis 5 Kohlenstoffatomen umsetzt und
die erhaltene Verbindung nach an sich bekannter katalytischer Hydrierung der Doppelbindung in der
Seitenkette, in an sich bekannter Weise zum entsprechenden Carbinol reduziert oder Campholenaldehyd in an sich bekannter Weise mittels
basenkatalysierter Aldolkondensation mit aliphatischen Aldehyden mit 3 bis 5 Kohlenstoffatomen
oder mit den entsprechenden substituierten Vinylal- 3s
lcyläthern unter BF3-Katalyse umsetzt und die auf diese Weise erhaltenen ungesättigten Aldehyde
nach an sich bekannter katalytischer Hydrierung der Doppelbindung der Seitenkette zu den gesättigten
worin R1 eine niedere Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, R2 eine Hydroxymethyl-, Acetoxymethyl- oder Formylgruppe und die geschlängelte Linie
am C-Atom 1 epimere Formen bedeuten.
Die Erfindung betrifft zudem die Verwendung von Verbindungen der allgemeinen Formel I als Riechstoffe
und somit auch Riechstoffkompositionen, gekennzeichnet durch einen Gehalt an einer Verbindung der
allgemeinen Formel I sowie die Herstellung solcher Verbindungen.
Es wurde gefunden, daß die neuen mehrfach substituierten Cyclopenten-Derivate der allgemeinen
Formel I wertvolle und stabile Riechstoffe sind. Insbesondere weisen die in der Seitenkette gesättigten
Derivate mit R1 = Methyl, Ethyl oder n-Propyl sowie R2
= Hydroxymethyl ausgeprägte Sandelholznoten auf.
Das in der Parfümerie vielfach eingesetzte ostindische Sandelholzöl enthält hauptsächlich die Sesquiterpene x- und /?-Santalol (A bzw. B), die auch den
charakteristischen mildholzigen Geruch des Öles bewirken.

Die begrenzte Verfügbarkeit und der hohe Preis des Sandelholzöles veranlaßten die Entwicklung von chemi- so
sehen Synthesen für A, B, die jedoch industriell nicht
ausgewertet werden konnten. Als synthetische Ersatzprodtikte werden zur Zeit hauptsächlich Terpenylcyciohexanole verwendet (Übersicht in: I. S. Aulchenko und
L A. Kheifits, American Perfumer and Cosmetic 1970, 37), die durch säurekatalysierte Umsetzung von
Camphen mit Phenol-Derivaten und nachfolgende katalytische Hydrierung erhalten werden. Hierbei
entstehen komplexe Gemische, die nur zum geringen Teil aus wirksajnen Riechstoffen vom Sandelholz-Typ $q
bestehen und hkufig durch Terphenylphenole verunreinigt sind
Es wäre grundsätzlich von der Überlegung auszugehen, daß die Erzielung einer möglichst natürlichen
Sandelholznote von Verbindungen zu erwarten ist, die den oben gezeigten Santalolen A und B strukturell
ähnlich sind und eine vergleichbare Molekülgröße bzw. Kohlenstoff zahl aufweisen. Dies bedingt allerdings den
Aufbau eines verhältnismäßig komplizierten Ringsystems, was, wie oben gezeigt, komplizierte Verfahren
und Reinigungsmaßnahmen oder die Einführung verhältnismäßig langer Seitenketten mit entsprechender
Partialstruktur an ein einfacheres Grundgerüst erfordert Bei den zuletzt genannten Verfahren waren die
Ergebnisse hinsichtlich der Geruchsnuancen wenig erfolgreich (Übersicht in G. Ohloff, Fortschr. Chem.
Forsch, Bd. 12/2, Seite 212,1969). So ist in der US-PS
40 52 341 neben höhermolekularen bzw. längerkettigen Verbindungen auch die Verbindung 3-Methyl-5-{2£3-trimethy!cyclopent-3-en-I-yl)-pentan-2-g| gezeigt, also
eine Verbindung, bei welcher an das einfache Trimethylcyclopenten-Ringgerüst eine längere Seitenkette, eine
sekundäre Hydroxylgruppe enthaltend, gebunden ist Das für die technische Darstellung dieser Substanz
angegebene Verfahren ergibt ein Isomerengemisch, das in größerem Anteil 6-(Trimethyl-cyclopent-3-en-l-yl)-hexan-3-ol enthält, so daß Geruchs-Intensität und
-Qualität des Produkts weniger befriedigend sind. Die
genannte Patentschrift sagt aus, daß man derartige
längere Ketten mit sekundären Hydroxylgruppen einfahren muß, um die gewünschten Geruchsnoten zu
erzielen.
In der DE-OS 19 22 391 werden Campholenyliden-Alkanole, nämlich Campholenyüdenpropanol, 2-Campho-Ienylidenbutanol und Campholenylidenisopropanol beschrieben, die an Moschus, Sandelholz bzw. Feigenduft
erinnernde Duftnoten aufweisen. Der Nachteil auch dieser Verbindungen ist, daß sie wegen ihrer Unsättigung in der Seitenkette, wie dies bei derartigen
ungesättigten Verbindungen häufig ist, chemisch weniger beständig sind, zu Alterungsreaktionen neigen und
daher als Riechstoffe weniger gut geeignet sind.
Es wurde nun aber überraschenderweise gefunden, daß die gesättigte Partialstruktur mit primärer Hydroxylgruppe (l-Hydroxy-2-methyl-alkanyl) der Santalole
auch in Kombination mit dem Trimethylcyclopentengerüst ausgezeichnete Geruchsnoten ergibt, wenn man
kürzere Seitenketten einführt, also insgesamt die Kohlenstcffzah] der Verbindung verringert So weist die
Verbindung 4a (Schema 1) eine ausgeprägte und intensive Sandelholz-Note auf. Dies ist um so überraschender, da nach dem allgemeinen Stand der Kenntnis
nur solche Verbindungen Sandeiholz-Duftnoten aufweisen sollten, die 14 oder mehr Kohlenstoffatome
enthalten (G. Ohloff, Fortschr. Chem. Forsch, Bd. 12/2,
S. 213,1969).
Damit kann man als Ausgangsmaterial Campholenaldehyd (1) verwenden, der einen Aufbau geeigneter
Seitenketten ermöglicht und nach bekannter Vorschrift (z. B. L C. King und H. Farber, J Org. Chem. 26, 326
[1961]) durch Umlagerung von «-Pinen-epoxid auch in großen Mengen erhalten werden kann. Auch den
natürlich vorkommenden optischeir Antipoden des (X-Pinens resultieren die enantiomeren Formen des
Campholenaldehyds, die einzeln oder als Gemisch eingesetzt werden können. Der Aufbau der Seitenketten wird nach an sich bekannten Methoden durchgeführt (Schemata 1 und 2) und ergibt im Gegensatz zur
US-PS 40 52 341 einheitliche Produkte.
Durch Wittig-Reaktion von 1 mit «-Brom-alkansäure-estern mit 3 bis 5 Kohlenstoffatomen im Alkansäureteil (einschließlich der Carboxylgruppe), vorzugsweise
den Ethylestern, nach an sich bekannten Verfahren (HeIv. Chim. Acta 40, 1242 [1957]) können die
Cyclopenten-Derivate der allgemeinen Formel I leicht hergestellt werden.
Die für die Wittig-Reaktion benötigten Phosphorylide können in an sich bekannter Weise durch Einwirkung
starker Basen auf die durch Umsetzen von «-Hal-Alkansäureestern (Hai — Chlor, Brom, Jod) mit Triphenylphosphin erhaltenen Phosphoniumsalzen dargestellt werden

Als Lösungsmittel für die Herstellung der Phosphorylide und die Durchführung der Wittig-Reaktion
kommen, wie üblich, aliphatische oder aromatische

Benzol, Toluol und Xylol), deren Halogenierungsprodukte, sowie Alkohole (z.B. Methanol, Ethanol,
Isopropanol, Butanole, Hexanole, Cyclohexanol, Cyclooctanol), Glykole und Ether (z.B. Düsopropyl-,
Ethylenglykoldimethylether, Tetrahydrofuran, Dimethyltetrahydrofuran und Dioxan bzw. Gemische hieraus
in Frage. Besonders geeignet sind polare aprotische Lösungsmittel wie Methanol, Ethanol, Formamid,
ίο Dimethylformamid, N-Methylpyrrolidon, Hexamethyl
phosphorsäuretriamid, Acetonitril und Dimethylsulfoxid
oder deren Gemische. Das Verfahren ist auch in
is von λ-Haifettsäuren (Hai = Chlor, Brom, Jod) mit
stöchiometrischen Mengen einer starken Base zu den jeweiligen Phosphoryliden umgesetzt Geeignete Basen
sind Alkalihydroxide, Alkalihydride, Alkaliamide, Alkali- und Erdalkalialkoholate, Phenyllithium oder Butyl-li
thium. Die so erhaltenen Phosphorylide werden mit
Campholenaidehyd in den genannten Lösungsmitteln bei Temperaturen von 10 - 40° C umgesetzt
Die Wittig-Reaktion kann auch einstufig durchgeführt werden, indem die Phosphoniumsalze und-etwa
stöchiometrische Mengen Campholenaidehyd in einem Lösungsmittel vorgelegt, bei Temperaturen von — 20 bis
+ 700C portionsweise mit etwa stöchiometrischen Mengen einer starken Base versetzt und 1 bis 2 Stunden
bei Temperaturen von 10—40° C gehalten werden.
So wurden die Verbindungen 2a, b und c des
nachfolgenden Schemas auf diese Weise hergestellt und aus diesen Verbindungen durch katalytische Hydrierung
3a, b und c erhalten. Reduktion der Ester, beispielsweise mit Alkali- oder Erdalkalimetallen in Alkoholen oder
mit gemischten Hydriden von Metallen der 1. und 3. Hauptgruppe, ergab die Carbinole 4a, b und c, die einen
intensiven Sandelholzgeruch aufweisen.
Die Hydrierung erfolgt in an sich bekannter Weise unter Verwendung von Übergangsmctall-Katalysato
ren, vorzugsweise Platin (Adams-Katalysator, Pt auf
Aktivkohle), Palladium (auf Aktivkohle) oder Raney-Nickel in neutralem Medium, vorzugsweise Methanol
oder Ethylacetat bei Temperaturen von 10—800C und
Drucken von 1-30 at, vorzugsweise 20-400C und
1-5 at
Die Esterreduktion wird nach Bouveault-Blanc unter
Verwendung von Alkali- oder Erdalkalimetallen in Alkoholen, vorzugsweise Natrium in Amylalkohol, oder
unter Verwendung von gemischten Hydriden der
Elemente der 1. und 3. Hauptgruppe in einem polaren
aprotischen Lösungsmittel, vorzugsweise mit Li[AlHJ in Diethylether ausgeführt
Die erwähnten und soeben besprochenen Reduktionsmethoden sowie die katalytische Hydrierung sind,
wie schon erwähnt, an sich bekannt und bedürfen keiner näheren Erläuterung. Sie werden aber auch in den
später folgenden Hersteilungsbeispielen noch gezeigt

Ein weiterer Syntheseweg geht aus von der basenkataiysierten Aldolkondensation von t mit aliphatischen C3- bis C5-Aldefiyden (Schema 2). Die auf diese
Weise erhaltenen ungesättigten Aldehyde 5a—c wurden durch katalytische Hydrierung zu 6a-c und
nachfolgende Reduktion in die gesättigten Carbinole 4a—c überführt Auch diese Reaktionen sind an sich
bekannt
Als Basen für Aldolkondensationen von Camphoenaldehyd mit aliphtischen Aldehyden werden Alkalihydro-

geeignet ist Borsäureanhydrid. Ais Lösungsmittel dienen Alkohole, vorzugsweise Methanol oder Ethanol.
Die Aldehyde 6a-c sind auch durch Umsetzung von 1 mit substituierten Vinylalkyläthern unter BF3-Katalyse
zugänglich (analog S. Satsumabayshi, K. Nakajo, R. Soneda und S. Motoki, BuIL Chem. Soc. Japan 43, 1586
[1970].

Die Alkohole 4a—c wurden in an sich bekannter
Weise mit Essigsäure zu den Acetoxymethylverbindungen 7a-c verestert
Für die neuen Verbindungen (2a—c-» 7a— c) liegen
zutreffende IR-, NMR- und MS-Spektren vor.
Die Verbindungen der allgemeinen Formel I zeichnen
sich durch besondere Riechstoffeigenschaften aus, insbesondere durch mild-holzige (4a-ςλ grün-holzige
(6a-ςλ auch etwas leicht harzige fruchtige, esterartige
Geruchsnoten {Ta—cjL Vor allem die Alkohole 4a-c
zeichnen sich durch zum Teil intensive Sandelholznoten aus; so übertrifft 4a das strukturanaloge 3-MethyI-5-(2,2,3-triinethylcyclopent-3-en-l-yl)-penten-2-ol (US-PS
40 52 34?) in der Geruchsintensi-tSt und somit auch in
der Ergiebigkeit in Kompositionen. Verbindungen der allgemeinen Formel Γ können demgemäß als Riechstoffe und als Bestandteil von Riechstoffkompositionen wie
Parfüms und Parfumbasen bzw. zur Parfümierung kosmetischer und technischer Produkte aller Art
verwendet werden.
Die folgenden Beispiele zeigen die Herstellung der erfindungsgemäßen Verbindungen.
l-(3-Carbethoxi-but-2-enyI)-2,2,3-trimethylcyclopent-3-en (2a)
150 g (IMoI) Campholenaldehyd (1) und «70g
(2-Carbethoxi-ethyliden)-triphenylphosphoran in 431
Benzol wurden unter N2 6 Stunden bei Siedetemperatur gerührt. Nach Abkühlung, Waschen mit Wasser und
Abdestillieren des Benzols wurde in Petrolether aufgenommen und filtriert. Aus dem nach Abdestillieren
des Lösungsmitteis verbleibenden Rohprodukt (210 p gelbes Öl) wurde durrh fraktionierte Destillation 176 g
(75%) 2a vom Kp«,8105-108° C gewonnen.

l-(3-Carbethoxi-butyl)-2iß-trimethylcyclcpent-3-en (3a)
Eine Lösung von 100 g (0,42MoI) 2a in 300 ml Methanol wurde mit 5 g Raney-Nickel unter Normalbedingungen hydriert (Ca. 10 Stunden). Filtration, Abdestillieren des Lösungsmittels und Fraktionierung ergaben 94 g (94%) 3a vom Kp^7 92 - 95° C
l-(3-Hydroxymethyl-butyl-2,2,3-trimethylcyclopent-3-en (4a)
50 g 3a in 200 ml absol. Ether wurden bei 15° C zu
einer Aufschlämmung von 9 g Li[AlH4] in 250 ml absol.
Ether getropft Nach 4 Stunden Rühren bei Raumtemperatur wurden 20 ml Eisessig zugegeben. Das Reaktionsgemisch wurde auf ca. 300 g Eis gegossen,
aufgearbeitet und fraktioniert Erhalten wurden 22 g (83%) 4a vom Κρ-ο,β 96 - 99" C.
l-(3-Formyl-2-aIkanyl)-2iP3-tiimethylcyclopent-3-ene (Sa- c)
Bei ca. 0°C wurden Suspensionen von 17 g Natriummethylat in 450 ml Methanol jeweils 368 g (2,4 Mol)
Campholenaldehyd (1) zugegeben. Innerhalb von 1
Stunde wurden jeweils ca SMoI dts Aldehyds (Propion-, Butyr- und n-Valeraldehyd) zugetropft
(Innentemperatur 40°C). Nach 1 Stunde Rühren bei Raumtemperatur, Ansäuren mit jeweils 50 ml Eisessig,
Abdestillieren des Methanols, Aufarbeitung und fraktionierter Destillation verblieben 60 bis 80% des
entsprechenden Aldehyds (5a — c).
Herstellungsbeispiel 5
l-(3-Formyl-2-alkyl)-2.2.3-trimethylcyciopent-ene(6iic)
Jeweils 100 g der Aldehyde 5a — f (aus Herstellungsbeispiel
4) in 200 ml Methanol wurden mil 5 g Raney-Nickel 3 Stunden unter Normalbedingungen
hydriert. Filtration und Abdestillieren des Methanols ergaben 80 bis 90% der Aldehyde 6a-r Bei
Durchführung der Hydrierung unter erhöhtem Druck, vorzugsweise 2O-6O;Hü erfolgte weitergehende Reduktion
/u 4a- c.

Zu Lösungen von jeweils 150e der Aldehyde ba - c
(aus Herstellungsbeispiel 5) in !ÖOml Ethanol wurden
bei 35- i0 C jeweils Lösungen von 38 g Natriumborhy drid und 0.6 g NaOH in 75 ml Wasser innerhalb von
30 min zugetropft. Nach 3 Stunden Rühren bei Raumtemperatur und Destillation lagen die Alkohole
4,i -cin Ausbeuten von 75-85% vor.
Nachstehende Beispiele zeigen die Verwendung der erfindungsgemäßen Verbindungen.
