Description
Neue 2-(4-Phenyl-4-cyanobutyl)-l,2,3,4-tetrahydro-5(H)-pyrido-Γ4,3-b]-indole und Verfahren zu ihrer Herstellung
In den US-Patentschriften Nr. 3 419 568, Nr. 3 448 114 und Wr.
3 466 293 sind Butyrophenonderivate von Tetrahydropyridoindolen der allgemeinen Formel
NjI-(CH2) ,CO-A~\-F
beschrieben, die analgetische und neuroleptische Eigenschaften besitzen.
Es wurde nun gefunden, dai3 durch Abwandlung der oben gezeigten
MoleküTstruktur neue Verbindungen geschaffen werden können, die in
ihren phamiakologischen Eigenschaften den vorbekannten Derivaten überlegen sind. Gegenstand der Erfindung sind daher neue 2-(4-Phenyl-4-cyanobutyl)-l,2,3,4-tetrahydro-5(H)-pyrido-[4,3-b]-indole
der allgemeinen Formel

und ihre physiologisch verträglichen Säureadditionssalze.
In der Formel I bedeuten R^ ein Wasserstoff- oder Halogenatom oder
eine Methylgruppe und R2 ein Wasserstoff- oder Halogenatom oder
eine Methyl- oder Methoxygruppe.
Die Erfindung betrifft ferner ein Verfahren zur Herstellung der
neuen Verbindungen der Formel I und ihrer Säureadditionssalze durch Umsetzung eines Pyridoindols der allgemeinen Formel

in der R-, wie oben angegeben definiert ist, mit einem Halogenid
oder aktivem Ester eines substituierten 2-Phenylvaleronitrils der
allgemeinen Formel

worin R2 die oben genannte Bedeutung besitzt und Y eine bei der
Umsetzung austretende Gruppe, z. ß. ein Halogenatom oder eine Arylsulfonyloxy- oder Alkylsulfonyloxygruppe bezeichnet.

Man verwendet die berechnete Menge des Alkylierungsmittels.der
Formel III oder einen Überschuß davon und arbeitet zweckmäßig in Gegenwart eines säurebindenden Stoffes, wie z. B. Triäthylamin,
Dicyclohexyläthylamin, Natriumcarbonat, Caliumcarbonat, Calciumoxyd
oder vorzugsweise Natriumhydrogencarbonat. Obwohl man auf Lösungsmittel verzichten kann, ist die Durchführung in inerten Lösungsmitteln,
wie Chloroform, Toluol, Äthanol, Nitrometan, Tetrahydrofuran oder vorzugsweise Dimethylformamid zweckmäßiger. Die Reaktionstemperatur
ist in weiten Grenzen variabel. Zweckmäßig sind Temperaturen zwischen 50 ° und 150 0G,vorzugsweise 100 0C. Die Zugabe von katalytischen
bis molaren Mengen Kaliumiodid oder Natriumiodid erweist
sich als günstig.
Die erhaltenen Reaktionsprodukte werden aus den Reaktionsansätzen mit Hilfe bekannter Methoden isoliert und gegebenenfalls können
die so hergestellten Rohprodukte noch unter Anwendung besonderer Verfahren, z. B. durch Säulenchromatographie gereinigt werden, ehe
man sie in Form der Basen oder geeigneter Säureadditionsverbindungen kristallisiert.
Für die Herstellung der Säureadditionssalze kommen solche Säuren in Frage, die physiologisch unbedenkliche Salze liefern, z. B.
Halogenwasserstoffsäure, Salpetersäure, Schwefelsäure, o-Phosphorsäure,
Oxalsäure, Zitronensäure, Weinsäure, Fumarsäure, Malein-*
säure, Propionsäure, Buttersäure, Essigsäure, Methansulfonsäure,
Toluolsulfonsäure, Sulfanilsäure, Bernsteinsäure, Äthanphosphonsäure
und dergleichen.
Die Ausgangsverbindungen der allgemeinen Formel II können nach den
Angaben in der DT-OS 1 670 010 hergestellt werden. Ausgangsverbindungen der allgemeinen Formel III werden durch Umsetzung eines
substituierten Benzylcyanids der allgemeinen Formel

worin R2 die oben genannte Bedeutung besitzt, mit l-Chlor-3-bromn-propan
gewonnen. Die erhaltenen Chloride können durch Finkelsteinreaktion in andere Halogenide überführt werden. Aus den Halogeniden
sind durch Umsetzung mit Sulfonsäureester entsprechende Sulfonsäureester der Formel III zugänglich.
Nach dem oben angeführten Verfahren lassen sich beispielsweise die
folgenden Endprodukte, gegebenenfalls in Form ihrer Säureadditionssalze, herstellen:

Die neuen Verbindungen der allgemeinen Formel I und deren Säureadditionssalze
zeigen das typische Wirkungsbild der Neuroleptika und sind daher als ZNS-dämpfende Mittel, als Sedativa und Tranquilizer
verwendbar. Bekannte Neuroleptika zeichnen sich u. a. durch antagonistische Wirkungen gegen Adrenalin (P. A. J. Janssen

et al, Arzneimittelforschung (Drug Research) 1J5 (1963) 205), Amphetamin
(J. H. Burn, R. Hobbs, Arch, inter. Pharmacodyn. 113 (1958) 290) und Apomorphin im Tierversuch aus. Außerdem haben sie
in Verhaltenstesten, wie dem Lochbrettversuch hemmende Wirkung auf Exploration (J. R. Borssin, P. Simin, J. M. Lwoff, Therapie 19
(1964) 571) und Lokomotion (P. A. J. Janssen, A. H. M. Jagenau, K.
H. L. Schellekens, Psychopharmacologia 1 (i960) 389). Schließlich hemmen sie auch das konditionierte Verhalten von Kleintieren, z.
B. im Dobrin Test (P. B. Dobrin et al. Arch, inter. Pharmacodyn
178 (1969) 351).
Die erfindungsgemäßen Verbindungen sind den vorhin genannten vorbekannten
Butyrophenonen bezüglich ihrer neuroleptischen Wirkung überlegen. Hervorzuheben ist insbesondere die wesentlich verringerte
Toxizität. So besitzt z. B. das 2-[4-(4-Fluorphenyl)-4-oxybutyl]
8-fluor-l,2,3,4-tetrahydro-5(H)-[4,3-b]-indol eine akute orale Toxizität von 270 mg/kg, wohingegen das erfindungsgemäße 2-(4-
-henyl-4-cyanobutyl)-1,2,3,4-tetrahydro-5(H)-pyrido-[4,3-b]-indol
eine LD von 1400 mg/kg p. o. hat.
Die erfindungsgemäß erhältlichen Verbindungen können allein oder
in Kombination mit anderen erfindungsgemäßen Wirkstoffen zur Anwendung
gelangen. Geeignete Anwendungsformen sind beispielsweise Tabletten, Kapseln, Zäpfchen, Lösungen, Säfte, Emulsionen oder dispersible
Pulver. Entsprechende Tabletten können beispielsweise durch Mischen des oder der Wirkstoffe mit bekannten Hilfsstoffen, beispielsweise
inerten Verdünnungsmitteln, wie Calciumcarbonat, CaI-ciumphosphat
oder Milchzucker, Sprengmittel, wie Maisstärke oder Alginsäure, Bindemitteln, wie Stärke oder Gelatine, Schmiermitteln,
wie Magnesiumstearat oder Talk und/oder Mitteln zur Erzielung eines Depoteffektes, wie Carboxymethylen, Carboxymethylcellulose,
Celluloseacetphthalat oder Polyvinylacetat erhalten werden.
Die Tabletten können auch aus mehreren Schichten bestehen.

Entsprechend können Dragees durch Überziehen von analog den Tabletten
hergestellten Kernen mit üblicherweise in Drageeüberzügen verwendeten
Mitteln, beispielsweise Kollidon oder Schellack, Gummi arabicum, Talk, Titandioxid oder Zucker, hergestellt werden. Zur
Erzielung eines Depoteffektes oder zur Vermeidung von Inkompatibilitäten
kann der Kern auch aus mehreren Schichten bestehen. Desgleichen kamauch die Drageehülle zur Erzielung eines Depoteffektes
aus mehreren Schichten bestehen, wobei die oben bei den Tabletten erwähnten Hilfsstoffe verwendet werden können.
Säfte der erfindungsgemäßen Wirkstoffe bzw. Wirkstoffkombinationen
können zusätzlich noch ein Süßungsmittel, wie Saccharin, Cyclamat, Glycerin oder Zucker sowie ein geschmacksverbesserndes Mittel,
zum Beispiel Aromastoffe, wie Vanillin oder Orangenextrakt, enthalten. Sie können außerdem Suspendierhilfsstoffe oder Dickungs-r
mittel, wie Natriumcarboxymethylcellulose, Netzmittel, beispielsweise Kondensationsprodukte von Fettalkoholen mit Äthylenoxid.oder
Schutzstoffe, wie p-Hydroxybenzoate, enthalten.
Injektionslösungen werden in üblicher Weise,.z. B. unter Zusatz
von Konservierungsmitteln, wie p-Hydroxybenzoaten oder Stabilisatoren, wie Alkalisalzen oder Athylendiamintetraessigsäure hergestellt
und in Injektionsflaschen oder Ampullen abgefüllt.
Den oder die Wirkstoffe bzw. Wirkstoffkombinationen enthaltende Kapseln können beispielsweise hergestellt werden, indem man die
Wirkstoffe mit inerten Trägern, wie Milchzucker oder Sorbit, mischt und in Gelatinekapseln einkapselt.
Geeignete Zäpfchen lassen sich beispielsweise durch Vermischen der
dafür vorgesehenen Wirkstoffe bzw. Wirkstoffkombinationen mit üblichen
Trägermitteln, wie Neutralfetten oder Polyäthylenglykol bzw. dessen Derivaten, herstellen.
Die neuen Verbindungen können bei oraler Verabreichung in einer Dosierung von 0,5 bis 10, vorzugsweise 1 bis 5 mg, zur Anwendung
gelangen.

A. Herstellungsbeispiele
Beispiel 1
2-1~ 4- (p-Fluorphenyl) -4-cyanobutyl['-8-f luor-1,2,3.4-tetrahyro-5 (H)-pyrido-Γ 4,3-b1-indol
3,8 g (20 mMol) 8-Fluor-l,2,3,4-tetrahydro-5(H)-pyrido(4,3-b)indol,
4,2 g (20 mMol) 5-Chlor-2-(p-fluorphenyl)valeronitril, 2,52 g
(30 mMol) Natriumbicarbonat und 200 mg Kaliumiodid werden in 50 ml
Dimethylformamid bei 100 0C so lange gerührt, bis im Dünnschichtchromatogramm
keine weitere Umsetzung festzustellen ist (2 bis 4 Stunden). Die Suspension wird bei 70 0C am Rotationsverdampfer
eingeengt. Der Rückstand wird zwischen 250 ml Methylenchlorid und 100 ml Wasser verteilt. Die organische Phase wird danach 5 mal mit
je 100 ml Wasser kräftig geschüttelt, über Natriumsulfat getrocknet und eingeengt. Es hinterbleibt ein gelbliches Öl, das in 15 ml
Äthanol gelöst wird. Nach Zugabe von 20 mMol äthanolischer Salzsäure wird vorsichtig mit Äther so lange versetzt, bis die Trübung
gerade wieder verschwindet. Nach einiger Zeit kristallisiert das Hydrochlorid obiger Verbindung, das aus Alkohol/Äther umkristallisiert
wird. Man erhält 6,8 g Hydrochlorid entsprechend 85,2 % der Theorie mit einem Schmelzpunkt von 235 bis 237 °C.
Analog Beispiel 1 wurden die in der folgenden Tabelle aufgeführten
Derivate in Form ihrer Hydrochloride hergestellt:

B. Anwendungsbeispiele
a) Dragees
1 Drageekern enthält:
Wirkstoff gemäß vorliegender Erfindung
Milchzucker Maisstärke Gelatine Magnesiumstearat
Die Mischung der Wirksubstanz mit Milchzucker und Maxsstärke
wird mit einer 10%igen wässrigen Gelatinelösung durch ein Sieb mit 1 mm Maschenweite granuliert, bei AO0C getrocknet und
nochmals durch ein Sieb getrieben. Das so erhaltene Granulat wird mit Magnesiumstearat gemischt und verpreßt. Die so erhaltenen
Kerne werden in üblicher Weise mit einer Hülle überzogen,
die mit Hilfe einer wässrigen Suspension von Zucker, Titandioxyd, Talkum und Gummi arabictim aufgebracht wird. Die fertigen
Dragees werden mit Bienenwachs poliert. Dragee-Endgewicht: 100 mg.

Wirkstoff und Magnesiumatearat werde» mit einer wässrigen
Lösung der löslichen Stärke granuliert, das Granulat getrocknet und innig mit Milchzucker und Maisstärke vermischt. Das Gemisch
wird sodann zu Tabletten von 100 mg Gewicht verpreßt, die 2 mg Wirkstoff enthalten.
c) Suppositorien
1 Zäpfchen enthält:

Die feingepulverte Substanz wird mit Hilfe eines Eintauch-Homogenisatore
in die geschmolzene und auf 400C abgekühlte

vorgekühlte Formen gegossen.
d) Ampullen

destilliertes Wasser ad 2,0 ml
Wirkstoff und Natriumchlorid werden in Wasser gelöst, die Lösung frei von suspendierten Partikeln filtriert und in
2 ecm-Ampullen unter aseptischen Bedingungen abgefüllt. Zuletzt
werden die Ampullen sterilisiert und verschlossen.
Jede Ampulle enthält 2 mg Wirkstoff.

Claims (1)
- PATENTANSPRÜCHEl) Neue 2-(4-Phehyl-4-cyanobutyl)-1,2,3,4-tetrahydro-5(H)pyrido-[4,3-t·]indole der allgemeinen Formel■« ^-CH0-CH0-CH0-CH- /I \ 2 2 2,worin R1 ein Wasserstoff- oder Halogenatom oder eine Methylgruppe und R2 ein Wasserstoff- oder Halogenatom oder eine Methyloder Methoxygruppe bedeutet sowie deren Säureadditionssalze.2. Verbindungen der Formel I nach Anspruch 1, worin R1 ein Fluoratom bedeutet.3. 2-(4-Phenyl-4-cyanobutyl)-1,2,3,4-tetrahydro-5(H)pyrido-[4,3-b] indole.4. Verfahren zur Herstellung neuer 2-(4-Phenyl-4-cyanobutyl)-l,2, 3,4-tetrahydro-5(H)pyrido[4,3-b]-indole der allgemeinen Formelworin R^ ein V/assers toff- oder Halogenatom und R2 ein Wasserstoff- oder Halogenatom und R eine Methyl- oder Methoxygruppe bedeutet und von deren Säureadditionssalzen, dadurch gekennzeichnet, daß man ein Pyridoiridol der allgemeinen Formel709R49/0032ORfGfNAt INSPECTEDIIworin R-, die oben genannte Bedeutung besitzt, mit einem Halogenid oder aktiven Ester eines substituierten 2-Phenylvalero nitrils der allgemeinen FormelY-CH9-CHp-CHp-CH - // \ IIIί
C=:Nworin R2 wie oben angegeben definiert ist und Y eine bei der Umsetzung austretende Gruppe, z. B. ein Halogenatom oder eine Arylsulfonyloxy- oder Alkylsulfonyloxygruppe bedeutet, umsetzt und daß man gewünschtenfalls das erhaltene Endprodukt der Formel I in ein physiologisch unbedenkliches Säureadditionssalz überführt.5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, daß man die Alkylierung in Gegenwart eines säurebindenden Mittels durchführt.6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß man als säurebindende Mittel Triäthylamin, Dicyclohexyläthylamin, Natriumcarbonat, Caliumcarbonat, Calciumoxyd oder Natriumhydrogencarbonat verwendet.7. Verfahren nach Anspruch 4 bis 6, dadurch gekennzeichnet, daß man die Ums
durchführt.man die Umsetzung bei einer Temperatur zwischen 50 und 150 0C8.
