Halogenacetanilide als mikrobizide Wirkstoffe
Die vorliegende Erfindung betrifft Verbindungen der Formel I

CO-CH2-X

worin Z1 Methyl oder Chlor,
Z„ Chlor oder Brom, Ζ« Wasserstoff oder C,-C~~Alkyl, und
X Fluor, Chlor, Brom oder Jod bedeuten,
Verfahren zur Herstellung dieser Verbindungen, sowie tnikrobizide Mittel, die diese neuen Verbindungen als
Wirkstoffe enthalten, ferner auch Verfahren zur Bekämpfung phytopathogener Pilze und Bakterien unter Verwendung der
Verbindungen der Formel I.
Als C1-C3-Alkyl sind Methyl, Aethyl, Propyl und Isopropyl
zu verstehen.
In der Deutschen Offenlegungsschrift Nr. 2, 212, 268 wird

in allgemeiner Form angegeben, dass N-haloacylierte Anilinoalkancarbonsäurecster
selektive herbizide Wirkung besitzen. Es werden jedoch nur einige N-haloacetylierte ^,ö-üialkylanilinoessigsäuren
und ihre Ester genannt und als Herbizide belegt. Hinweise auf mikrobizide, insbesondere pflanzenfungizide
Wirkung werden nicht gegeben.
In der Deutschen Offenlegungsschrift Nr. 2'311'897 wird eine
Vielzahl substituierter Phenylamine mit ausserordentlich stark variierenden chemischen Strukturen als Herbizide beschrieben.
Hinweise auf mikrobizide Wirkung werden in dieser Anmeldung ebenfalls nicht gegeben. Die im Text aufgeführte nächstvergleichbare
Verbindung N-Chloracetyl-N-(2,6-dichlorphenyl)-glycinäthylester ist
gegen phytopathogene Pilze unwirksam.
Die von der Formel I umfassten Verbindungen dieser Erfindung sir.5
bisher nicht in der Literatur beschrieben worden und somit neu. Ausserordentlich überraschend ist, dass sie im Gegensatz zu den
vorbeschriebenen"Anilinoessigsäuren und -essigsäureestern der
DOS 2,212,268 und der DOS 2'311'897 starke pflanzenfungizide
Eigenschaften aufweisen.
So besitzen sie sowohl präventive als auch kurative Wirkung gegen phytopathogene Pilze an Kulturpflanzen wie beispielsweise Getreide,
Mais, Reis, Gemüse, Zuckerrüben, Soja, Erdnüsse, an Obstbäumen, Zierpflanzen, vor allem aber an Reben, Hopfen, Gurkengewächsen
(Gurken, Kürbis, Melonen) und Soianaceen wie Kartoffeln, Tabak und Tomaten,sowie auch an Bananen-, Kakao- und Naturkautschuk-Gewächsen,
Mit diesen Wirkstoffen können an Pflanzen oder Pflanzenteilen (Früchte, Blüten, Laubwerk, Stengel, Knollen, Wurzeln) auftretende
Pilze eingedämmt oder vernichtet werden, wobei auch später zuwachsende Pflanzenteile von derartigen Pilzen verschont bleiben.
Die Wirkstoffe sind gegen die den folgenden Klassen angehörenden phytopathogenen Pilze wirksam: Ascomycetes; Basidiomycetes wie vor
allem Rostpilze; Fungi imperfecti; dann aber besonders gegen die

der Klasse der Phycomycetes angehörenden Oomycetes wie
Phytcphthora, Peronospora, Pseudoperonospora, Pythiura oder
Plasmopara. Ueberdies wirken die Verbindungen der Formel I
systemisch. Sie können ferner zur Behandlung von Saatgut (Früchte, Knollen, Körner) und Pflanzenstecklingen zum Schutz vor
Pilzinfektionen soxtfie gegen im Erdboden auftretende phytopathogene
Pilze eingesetzt werden.-
Zur Verbreiterung oder Veränderung des Wirkungsspektrums können
die Wirkstoffe der Formel I mit weiteren vorbekannten Fungiziden, Bakteriziden, Fungistatika, Bakteriostatika, aber auch mit
Insektiziden, Akariziden, Herbiziden, und auf Grund ihrer systemischen Wirkung, die eine Erdbodenapplikation erlaubt, auch
mit Nematiziden, Molluskiziden oder Rodentiziden gemischt werden, wobei teilweise Synergistisch gesteigerte Wirkungen erzielt werden.
Als Mischkomponenten seien beispielsweise folgende bekannte Mikrobizide genannt, die je nach Einsatzgebiet verwendet werden

Die Herstellung der Verbindungen erfolgt erfindungsgernäss durch
Reaktion einer Verbindung der Formel II

mit einem Halogenacetylierungsmittel, vorzugsweise mit dem
Halogenid oder Anhydrid der jeweils in Frage kommenden Monofluor-, Monochlor-jMonobrom- bzw. Monojodessigsäure.
Die Umsetzungen können in An- oder Abwesenheit von gegenüber den Reaktionsteilnehmerη inerten Lösungs- oder Verdünnungsmitteln
durchgeführt werden. Es kommen beispielsweise folgende in Frage:
Aliphatische oder aromatische Kohlenwasserstoffe wie Benzol, Toluol, Xylole, Petroläther; halogenierte Kohlenwasserstoffe
wie Chlorbenzol, Methylenchlorid, Aethylenchlorid, Chloroform; Aether und ätherartige Verbindungen wie Dialkyläther,
Dioxan, Tetrahydrofuran; Nitrile wie Acetonitril; Ν,Ν-dialkylierte
Amide wie Dimethylformamid; ferner wasserfreie Essigsäure, Ketone wie Methyläthy!keton, Dimethylsulfoxid und Gemische solcher
Lösungsmittel untereinander.
Als geeignete Halogenacetylierungsmittel werden vorzugsweise Halogenessigsäureanhydride wie z.B. Chloressigsäureanhydrid und
Halogenessigsäurehalogenide, vorteilhaft Halogenessigsäurechlorid ode -bromid,verwendet. Die Reaktion kann jedoch auch mit den obengenannten
freien Halogenessigsäuren, ihren Estern oder Amiden durchgeführt werden.
Die Reaktionstemperaturen liegen zwischen 0° und 180° C, vorzugsweise zwischen 20° und 100° C. In manchen Fällen, insbesondere
bei der Verwendung von Halogenacety!halogeniden,

wird die Haloacetylierung in Gegenwart eines säurebindenden
Mittels bezw. Kondensationsmittels durchgeführt. Als solche kommen tertiäre Amine wie Trialkylamine (z.B. Triethylamin),
Pyridin und Pyridinbasen, oder anorganische Basen, wie die Oxide und Hydroxide, Hydrogencarbonate und Carbonate von Alkali-
und Erdalkalimetallen sowie Natriumacetat in Betracht.
Als säurebindendes Mittel kann ausserdem ein Ueberschuss des Anilinderivats der Formel II dienen.
Das erfindungsgemässe Herstellungsverfahren kann auch ohne
säurebindende Mittel durchgeführt werden, wobei in einigen Fällen
das Durchleiten von Stickstoff zur Vertreibung des gebildeten Halogenwasserstoffs angezeigt ist. In anderen Fällen ist ein
Zusatz von Dimethylformamid als Reaktionskatalysator sehr vorteilhaft.
Einzelheiten zur Herstellung der Zwischenprodukte der Formel II kann man den Methoden entnehmen, wie sie allgemein für die
Herstellung von Anilino-alkansäureestern in folgenden Publikationsorganen angegeben werden:

Zur Herstellung reiner optischer Antipoden der Formel I stellt
man zweckmässig aus dem Anilin der Formel III und der

Bedeutung haben, und setzt sie in an sich bekannter Weise mit einer

N-haltigen optisch aktiven Ease zum entsprechenden Salz um. Durch fraktionierte Kristallisation des Salzes'und nachfolgende
Freisetzung der mit dem optischen D-Antipoden angereicherten Säure der Formel IV und gegebenenfalls Wiederholung (auch
mehrfache Wiederholung) der Salzbildung, Kristallisation und Freisetzung der a-Anilinopropionsäure der Formel IV gewinnt man
stufenweise die reine D-Form. Aus dieser lässt sich dann auf übliche Art, z.B. in Gegenwart von HCl oder H9SO,, mit Methanol
die optische D-Konfiguration des Esters der Formel II herstellen,
der, wie oben angegeben, weiter zu den D-Formen der Verbindungen I acyliert wird. Als optisch aktive organische Base kommt z.B.
a-Phenyläthylareitn in Frage.
Die gewünschte enantiomere D-Form der Formel II lässt sich aber auch aus dem natürlich vorkommenden L-Almin dadurch gewinnen
(J. Am. Chem. Soc. 76_, 6056), dass man die Amino-Gruppe des
Alanins in Gegenwart von HCl oder HBr diazotiert und damit unter N9-Abspaltung gegen das Halogen zur (L)-a-Halogenpropionsäure
austauscht, die danach mit Methanol verestert wird. Durch Reaktion mit dem Anilin der Formel III erhält man unter Inversion
überwiegend die gewünschte D-Form der Verbindung II, aus der man durch nachfolgende Haloacetylierung zur D-Form des Endprodukts
der Formel I gelangt.
Soweit im folgenden Teil jedoch nichts anderes vermerkt wird, ist
bei der Nennung eines der Wirkstoffe der Formel I stets das racemische D,L-Gemisch gemeint.
Die Herstellung der Wirkstoffe der Formel I wird durch die
folgenden Beispiele 1-3 veranschaulicht. Die Temperaturen sind in Celsiusgraden angegeben.

a) Herstellung von oc-(2-Chlor-6-methylanilino) -propionsäuremethylester
(D,L)
35,4 g 2-Chlor-6-methylanilin, 21,3 g Natriumhydrogencarbonat
und 84 ml DL-a-Brompropionsäuremethylester
werden 17 Std. bei 140° Badtemperatur gerührt. Nach dem Abkühlen wird das dunkel gefärbte Reaktionsgemisch
filtriert, und der überschüssige a-Brompropionsäuremethylester
am Wasserstrahlvakuum abdestilliert. Der verbleibende Rückstand wird am Hochvakuum fraktioniert
destilliert. Man erhält 25,9 g a-(2-Chlor-6-methylanilino)-propionsäuremethylester,
Siedepunkt 72-75°/ 0.001 Torr.
b) Herstellung von N-(I1-Methoxycarbonyl-äthyl)-N-chloracetyl-2-chlor-6-methylanilin
(D,L)
25,9 gdes gemäss a) erhaltenen Esters, 1 ml Dimethylformamid
und 200 ml Chlorbenzol werden auf 90°(Innentemperatur) erwärmt. Man lässt unter Rühren bei dieser
Temperatur 10 ml Chloracetylchlorid zutropfen. Nachdem die HCl-Entwicklung aufgehört hat, wird noch
1 Std. bei 120° Badtemperatur ausgerührt und das Reaktionsgemisch am Rotationsverdampfer eingeengt. Der
Rückstand wird aus Diisopropyläther umkristallisiert. Man erhält 24,2 g N-(I1-Methoxycarbonyläthyl)-N-chloracetyl-2-chlor-6-methylanilin,
Schmelpunkt 75-78°.

und 200 ml Chlorbenzol werden auf 90°(Innentemperatur) erwärmt. Man lässt unter Rühren bei dieser
Temperatur 9,6 ml Fluoracetylchloiidzutropfen.
Nachdem die HCl-Entwicklung aufgehört hat, wird noch
1 Std. bei 120° Badtemperatur ausgerührt und das Reaktionsgemisch am Rotationsverdampfer eingeengt. Der
Rückstand wird aus Diisopropyläther umkristallisiert. Man erhält 19,5 g N-(I' -Methoxycarbonyläthyl) -N-fluor·
acetyl-2-chlor-6-methylanilin, Schmelzpunkt 101-103°.

a) Herstellung von α-(2,6-Dichloranilino)-propionsäuremethylester
(D5L)
40,5 g 2,6-Dichloranilin, 21 g Natriumhydrogencarbonat und
84 ml DL - a-Brompropionsauremethylester v?erden in einem
SuIfierkolben während zwei Tagen bei 140 Badtemperatur
gerührt. Nach dem Abkühlen wird das Reaktionsgemisch auf Wasser gegossen und mit Essigester extrahiert. Die
vereinigten Essigesterfraktionen werden einmal mit: gesättigter Kochsalzlösung ausgeschüttelt, über Natriumsulfat
getrocknet und am Rotationsverdampfer eingeengt. Am Wasserstrahlvakuum wird der überschüssige a-Brompropionsäurenethylester
zurückgewonnen. Der verbleibende dunkelgefärbte Rückstand wird am Hochvakuum fraktioniert
destilliert. Man erhält 23,6 g <x-(2,6-Dichloranilino)-propionsäuremethylester;
Siedepunkt 85 - 87 /0,001 Torr.
b) Herstellung von N-(I1-Methoxycarbonyl-äthyl)-N-chloracetyl-2,6-dichloranilin
(D,L)
18,4 g a-(2,6-Dichloranilino)-propionsäuremethylester,
1 ml Dimethylformamid und 100 ml Chlorbenzol werden auf 100 (Innentemp.) erwärmt. Unter Rühren lässt man
bei dieser Temp. 8 ml Chloracetylchlorid zutropfen. Nachdem
die HC!-Entwicklung aufgehört hat, wird 1 Std. bei 130 - 140 Badtemperatur ausgerührt. Man lässt auf
Raumtemperatur abkühlen, entfernt am Rotationsverdampfer Lösungsmittel und kristallisiert das als Rückstand
verbleibende gelbe OeI aus Diisopropyläether. Man erhält
16 g Endprodukt vom Fp. 82 - 85°. (Verb.Nr. 10)

Die Verbindungen der Formel I können zur Verbreiterung ihres
Wirkungsspektrums mit anderen geeigne :en pestiziden oder den
Pflanzenwuchs fördernden Wirkstoffen eingesetzt werden.
Die Verbindungen der Formel I können für sich allein oder zusammen mit geeigneten Trägern und/oder anderen Zuschlagstoffen
verwendet werden. Geeignete Träger und Zuschlagstoffe können fest
oder flüssig sein und entsprechen den in der Formulierungstechnik üblichen Stoffen wie z.B. natürlichen oder regenerierten mineralischen
Stoffen, Lösungs-, Dispergier-, Netz-, Haft-, Verdickungs-, Binde- oder Düngemitteln.
Der Gehalt an Wirkstoff in handelsfähigen Mitteln liegt zwischen 0,1 bis 90 %.
Zur Applikation können die Verbindurgen der Formel I in den
folgenden Aufarbeitungsformen vorliegen (wobei die Gewichts-Prozentangaben in Klammern vorteilhafte Mengen an Wirkstoff
darstellen):
Feste Aufarbeitungsformen: Stäubemittel und Streumittel (bis zu 10',
Granulate, Umhüllungsgranulate, Imprägiiievungsgranulate
und Homogengranulate (1 bis 80 %);
Flüssige Aufarbeitungsformen:
a) in Wasser dispergierbare Wirkstoffkonzentrate:
Spritzpulver (wettable powders) und Pasten (25-90% in der HändeIspackung, 0,01 bis I^ %

Die Aktivsubstanz wird mit Epichlorhydrin vermischt und mit
6 Teilen Aceton gelöst, hierauf wird Polyäthylenglykol und Cety!polyglykolether zugesetzt. Die so erhaltene Lösung wird auf
Kaolin aufgesprüht, und anschliessend wird das Aceton im Vakuum verdampft. Ein derartiges Mikrogranulat wird vorteilhaft zur
Bekämpfung von Bodenpilzen verwendet.
Spritzpulver: Zur Herstellung eines a) 70 7oigen b) 40 7oigen
c) und d) 25 7eigen e) 10 7oigen Spritzpulvers werden
folgende Bestandteile verwendet:

Benetzbarkeit und Schwebefähigkeit, die sich mit Wasser zu Suspensionen jeder gewünschten Konzentration verdünnen und
insbesondere zur Blattapplikation verwenden lassen.

Ia) Residual-präventive Wirkung
Solanum lycopersicum- Pflanzen der Sorte "Roter Gnom"
werden nach 3-wöchiger Anzucht nach dem Besprühen
mit einer 0,05 7O Aktivsubstanz enthaltenden
Brühe (hergestellt aus der zu einem Spritzpulver aufgearbeiteten Wirksubstanz) und deren Antrocknen mit
einer Zoosporensuspension von Phytophthora infestans infiziert. Sie bleiben dann während 6 Tagen in einer
Klimakammer bei 18 bis 20° und hoher Luftfeuchtigkeit,
die mittels eines künstlichen Sprühnebels .erzeugt wix*d- Nach dieser Zeit zeigen sich typische Blattflecken.
Ihre Anzahl und Grosse sind der Bewertungsmassstab für die geprüfte Substanz.

Ib) Kurative Wirkung
Tomatenpflanzen der Sorte "Roter Gnom" werden nach dreiwöchiger
Anzucht mit einer Zoosporensuspension des Pilzes besprüht und in einer Kabine bei 18 bis 20 und
gesättigter Luftfeuchtigkeit inkubiert. Unterbruch
der Befeuchtung nach 24 Stunden. Nach dem Abtrocknen der Pflanzen werden diese mit einer Brühe besprüht,
die die als Spritzpulver formulierte Wirksubstanz in einer Konzentration von 0,05 % enthält. Nach dem
Antrocknen des Spritzbelages werden die Pflanzen wieder ■ in der Feuchtkabine während 4 Tagen aufgestellt.
Anzahl und Grosse der nach dieser Zeit auftretenden typischen Blattflecken sind der Bewertungsmassstab
für die Wirksamkeit der geprüften Substanzen.
II) Präventiv-Systemische Wirkung
Die als .Spritzpulver formulierte Wirksubstanz wird in
einer Konzentration von 0,05 "U (bezogen auf das Bodenvolumen) auf die Bodenoberfläche von drei Wochen
' alten eingetopften Tomatenpflanzen der Sorte "Roter
Gnom" gegeben. Nach dreitägiger Wartezeit wird die Blattunterseite der Pflanzen mit einer Zoosporensuspension
von Phytophthora infestans besprüht. Sie werden dann 5 Tage in einer Sprühkabine bei 18 bis
20 und gesättigter Luftfeuchtigkeit gehalten. Nach dieser Zeit bilden sich typische Blattflecken, deren
Anzahl und Grosse zur Bewertung der Wirksamkeit der geprüften Substanzen dienen.

Die Verbindungen Nr. 1,10,13,14 und 15 minderten in den
gleichen Versuchen bei Anwendungskonzentrationen von nur 0,02 "L
den Pilzbefall auf <20 %, die Verbindungen Nr. la und 10a (D-Konfigurationen) auf nur 0 - 5 %.
Wirkung gegen Plasmopara viticola (Bert, et Curt.) (Berl. et
DeToni) auf Reben
a) Residual-präventive Wirkung
Im Gewächshaus wurden Rebenstecklinge der Sorte "Chasselas"
herangezogen. Im 10-Blatt-Stadium wurden 3 Pflanzen mit einer
aus der als Spritzpulver formulierten Wirksubstanz hergestellten Brühe (0,05 °L Wirkstoff) besprüht. Nach dem Antrocknen des
Spritzbelages wurden die Pflanzen auf der Blattunterseite mit der Sporensuspension des Pilzes gleichmässig infiziert. Die
Pflanzen wurden anschliessend während 8 Tagen in einer
Feuchtkammer gehalten. Nach dieser Zeit zeigten sich deutliche KrankheitsSymptome an den Kontrollpflanzen. Anzahl und Grosse

der Infektionsstellen an den behandelten Pflanzen dienten als
Bewertungsmassstab für die Wirksamkeit der geprüften Substanzen.
b) Kurative Wirkung
Rebenstecklinge der Sorte "Chasselas" wurden im
Gewächshaus herangezogen und im 10-Blatt-Stadium mit
einer Sporensuspension von Plasmopara viticola an der Blattunterseite infiziert. Nach 24 Std. Aufenthalt in
der Feuchtkabine wurden die Pflanzen mit einer 0,05%igcn Wirkstoffbrühe
besprüht, die aus einem Spritzpulver des Wirkstoffs hergestellt worden war. Anschliessend
wurden die Pflanzen 7 Tage weiterhin in der Feuchtkabine gehalten. Nach dieser Zeit zeigten
sich die KrankheitsSymptome an den Kontrollpflanzen.
Anzahl und Grosse der Infektionsstellen an den be-
- handelten Pflanzen dienten als Bewertungsmassstab für die Wirksamkeit der geprüften Substanzen.
Die Verbindungen der Formel I zeigten überwiegend gute blattfungizide
Wirkungen in diesen beiden Versuchen. Mit den Verbindungen Nr. 1, la, 9, 10, 10a, 13, 14 und 15 wurde der Pilzbefall fast
vollständig reduziert (0-5 % Befall).
a) Wirkung nach "odenapplikation
Der Pilz wird auf sterilen Haferkörnerη kultiviert und einer
Erde-Sand-Mischung beigegeben. Die so infizierte Erde wird in Blumentöpfe abgefüllt und mit Zuckerrübensamen besät. Gleich
nach der Aussaat werden die als Spritzpulver formulierten Versuchspräparate als wässerige Suspensionen über die Erde
gegossen (20 ppm Wirkstoff bezogen auf das Erdvolumen).
Die Töpfe werden darauf während 2-3 Wochen im Gewächshaus bei
20-24° C aufgestellt. Die Erde wird dabei durch leichtes

Besprühen mit Wasser gleichmär-sig feucht; gehalten. Bei der
Auswertung der Tests wird der Auflauf der Zuckerrfibenpf lanzen sowie der Anteil gesunder und kranker Pflanzen bestimmt.
b) Wirkung nach Beizapplikation
Der Pilz wird auf sterilea HaCerkörnern kultiviert und einer
Erde-Sand-Mischung beigegeben. Die so infizierte Erde wird in Blumentöpfe abgefüllt und mit Zuckerrübensamen besät, die mit
den als Eeizpulver formulierten Versuchspräparaten gebeizt worden sind (1000 ppm Wirkstoff bezogen auf das Samengewicht) .
Die besäten Töpfe werden während 2-3 Wochen im Gewächshaus bei 20-24° C aufgestellt. Die Erde wird dabei durch leichtes
Besprühen mit Wasser gleichmässig feucht gehalten. Bei der Auswertung wird der Auflauf der, Zuckerrtibenpf lanzen sowie
der Anteil gesunder und kranker Pflanzen bestimmt.
Nach der Behandlung mit den Wirkstoffen der Formel I liefen, sowohl unter den Testbedingungen a) wie b), mehr als 80 % der
Zuckerrübenpflanzen auf und hatten ein gesundes Aussehen. Bei Applikation mit den Verbindungen Nr. 7,8,12, 20>
22 und 24 erhöhte sich der Anteil an gesund aufgelaufenen Pflanzen auf 90 °L und mehr,
Bei der unbehandelten Kontrolle liefen weniger als 20 % Pflanzen mit zum TeiL kränklichem Aussehen auf.

worin Z,, Z~ und Z~ die im Anspruch 1 gegebene Bedeutung haben,
mit einem Halogenacetylierungsmittel bei einer Temperatur zwischen 0° und 180° C.
15. Verfahren gemäss Anspruch 14, wobei die Acylierung mit dem Halogenid oder Anhydrid der zu verwendenden Monofluor-,
Monochlor-, Monobrom- bzw. Monojod-essigsäure durchgeführt wird.

16. Verfahren gemäss Anspruch 15, wobei als Halogenid das
Chlorid oder Bromid verwendet wird.
17. Mikrobizide Mittel enthaltend als Wirkstoff eine Verbindung
der Formel I

26. Mittel gemäss Anspruch 17 mit dem Wirkstoff N-(I'-Methoxycarbonyl-äthyl)-N-chloracetyl-2,S-dimethyl-ö-chloranilin.
27. Mittel gemäss Anspruch 17 mit dem Wirkstoff N-(I'-Methoxycarbonyl-äthyl)-N-chloracetyl-2,5-dimethyl-6-chloranilin.
28. Mittel gemäss Anspruch 17 mit dem Wirkstoff N-(I1-Methoxycarbonyl-äthyl)
-N-chloracetyl-2, G-dichlor-S-rnethylanilin.
29. Verwendung von Verbindungen gemäss einem der Ansprüche 1 bis
